RSVアタッチメントタンパク質(RSV-Gタンパク質)は、4つの保存されたシステイン残基を有し、これらのシステイン残基が、アミノ酸169〜191の中に位置する2つの内部ジスルフィド結合を形成する。この領域を認識する抗体はRSVを無力化することができるので、このセグメントに由来する立体構造限定合成ペプチドは、効果的なワクチン成分であり得る。以前の研究では、タンパク質消化、HPLC、および質量分析の組合せを使用して、Cys173がCys186に結合しており、Cys176がCys182に結合していることが示された。RSV-Gタンパク質の残基169〜191を含むペプチドを、固相ペプチド合成によって合成した。中性pHでこれらのペプチドは容易に酸化し、結果として生じる産物は、ESMSスペクトラムにおいて4つのプロトンが消失していること、および、エルマン(DTNB)アッセイにおける陰性シグナルによって測定されるとおり、2つの内部ジスルフィドを含んでいると見られる。上記酸化反応が起こる容易さ、および、変換の明快さは、自然本来のジスルフィドが形成されていることを強く示唆している。典型的な実験では、還元されたRSV-Gペプチド(例えば、配列番号8、11、13、14、15)を、2.5 mMのグルタチオンおよび2.5 mMのグルタチオールを含むTris pH 7.4中に、1〜5 mg/mLの濃度で溶解する。酸化産物は還元ペプチドと比べて滞留時間が短い方にわずかにシフトすることから、折りたたみ反応はC18逆相HPLCによってモニターされる。この基準によれば、折りたたみ反応は、室温では2時間後には完了し、4℃では18時間後には完了すると判断される。
CaCO3ナノ粒子(NPCC-111)は、NanoMaterials Technology社(シンガポール)から入手した。走査型電子顕微鏡(SEM)実験は、粒子は立方体形態を有し、直径約50 nmであることを示した。ポリペプチドであるポリ-l-リシン15 kDa(PLL、カタログ番号P6516)およびポリ-l-グルタミン酸14.5 kDa(PGA、カタログ番号P4636)、ならびに1 M HEPES緩衝液(カタログ番号H-3662)は、Sigma-Aldrich社(米国)から入手した。反対に荷電したポリペプチドを、連続的な吸着工程において、CaCO3ナノ粒子コア上の多層フィルムへと自己会合させた。手短に述べると、PLL、PGA、または設計されたポリペプチド(DPと表すこともある)を、10 mM HEPES、pH 7.4中に1 mg/ml(重量/体積)となるように溶解させ、0.22μmフィルターを通してフィルター処理した。CaCO3ナノ粒子コアを、内毒素費含有水で3回洗浄し、16,000 gで1分間、微量遠心機中で遠心分離した。ナノ粒子コアを、第1の層としての1 mg/ml PGA中に、6%(重量/体積)となるように再懸濁させた。中性pHでは、PGAは負の実効電荷を示し、CaCO3粒子は正の実効電荷であるから、静電的相互作用、および、第1の層の堆積が可能となる。上記混合物に対して室温で10分間、超音波処理をし、10 mM HEPESバッファーで2回洗浄し、48,700 x gで1分間、遠心分離した(TL-100超遠心機、Beckman社)。第2の層を堆積するために、ナノ粒子を、1 mg/mlのPLL(正に荷電)中に、6% (w/v)となるように再懸濁し、第1の層の場合と同じように、室温で10分間、超音波処理をした。表2または表3に示すように、PGA、PLL、またはDPを使用して、後に続く層のそれぞれを同じ方法で堆積させた。典型的には、設計されたペプチドは0.5〜1.0 mg/mLの濃度でコーティングされた。最後の層を堆積させた後、ナノ粒子を、10 mM HEPES、pH 7.4で2回洗浄し、分注したものを遠心で沈殿させて液を吸引し、使用時まで、湿ったペレットとして4℃で貯蔵した。本質的に同じ手順を使用して、そして設計されたペプチドの配列と位置を変えて、いくつかの設計を作製した。すべてのナノ粒子は、ポリペプチドバイオフィルム中に合計8つの層を含む。ナノ粒子の設計を表2に要約する。ナノ粒子上のポリペプチドまたはDPの濃度は、アミノ酸分析により決定した。ナノ粒子のサイズは、HEPES、pH 7.4中の0.06% (w/v)懸濁液の動的光散乱(Malvern Nano S-90)により決定し、測定前に超音波ウォーターバス(Branson 1510、米国)中で20分間超音波処理した。ナノ粒子中のエンドトキシンのレベルは、エンドポイント発色性アッセイであるLimulus Amebocyte Lysateアッセイ(#50-647U, Lonza, Walkersville, MD)によって決定した。調製したナノ粒子は、使用時まで、湿ったペレットとして4℃で貯蔵した。
実施例3に記述した標準的な手順に従った。設計されたペプチドACT-2031(RSV-M281-98(KVKA)4(配列番号12))の1.0 mg/mLの溶液を、2番目のELBLコーティング工程において使用し、設計されたペプチドACT-2044(RSV-G164-191K21Y(配列番号8))の1.0 mg/mLの溶液を、8番目のELBL工程に使用した。各々の加層工程の後に、ナノ粒子を洗浄し、6%となるように再懸濁した。表面(ゼータ)電位解析のために、10μLの分量を取り出し、1.0 mLの10 mM HEPESバッファー中に希釈した。図3は、ELBLプロセスの各工程におけるナノ粒子について測定された表面ゼータ電位を示す。コーティングする前には、CaCO3粒子は、約+28 mVの正の表面電位を示した。1層のPGAでコーティングした後には、-18 mVの負の表面電位が測定された。2番目の層におけるACT-2031(配列番号12)によるコーティングは、ゼータ電位を約0 mVまで上昇させたが、これはそのペプチドが吸着されたことを示している。その後、各々の加層工程が、ゼータ電位を負または正にシフトさせたが、これはELBL吸着が成功していることを示している。製造後、吸着された各々の設計されたペプチドの量を、アミノ酸クロマトグラムにおける固有アミノ酸のシグナルから計算した。グリシンはACT-2031に固有であって、ACT-2031の濃度を決定するために使用できる一方、アルギニン、ヒスチジン、およびフェニルアラニンはACT-2044に固有であって、ACT-2044の濃度を決定するために使用できる。4つの別々のバッチについて、1% CaCO3粒子懸濁液に吸着されたACT-2031およびACT-2044の平均量は、それぞれ21μg/mLおよび53μg/mLであった。
実施例3で記述したELBLコーティング手順を使用して、50 nm CaCO3ナノ粒子を7層のPGAおよびPLLでコーティングした。10 mM HEPESバッファー中の設計されたペプチドACT-2086(配列番号13;RSV-M281-98G164-191 K20Yアミド)の0.5 mg/mL溶液を、最終層のコーティングに使用した。吸着された設計されたペプチドの量は、アミノ酸分析で測定した。3つの別々のバッチについて、1% CaCO3粒子懸濁液に吸着されたACT-2086の平均量は、37μg/mLであった。
実施例3で記述したELBLコーティング手順を使用して、50 nm CaCO3ナノ粒子を7層のPGAおよびPLLでコーティングした。最終濃度がそれぞれ0.25 mg/mLである、設計されたペプチドACT-2033(配列番号16;RSV M2 81-98 K20Yアミド)およびACT-2044(配列番号8; RSV-G161-191 K21Yアミド)の10 mM HEPES溶液を、最終層のコーティングに使用した。ナノ粒子を洗浄し、遠心で沈殿させ、湿ったペレットとして4℃で貯蔵した。吸着された各々の設計されたペプチドの量は、アミノ酸クロマトグラムにおける固有アミノ酸のシグナルから計算された。グリシンはACT-2033に固有であってACT-2033の濃度を決定するために使用できる一方、アルギニン、ヒスチジン、およびフェニルアラニンはACT-2044に固有であってACT-2044の濃度を決定するために使用できる。この解析から、1%粒子懸濁液は25μg/mLのACT-2033および24μg/mLのACT-2044を含むことが明らかとなった。
メソ多孔性の3μm CaCO3マイクロ粒子は、PlasmaChem GmbH社(ベルリン、カタログ番号PL-CA3)から入手した。粒子を、10 mM HEPES中で6%(重量/体積)となるように懸濁した。CaCO3ナノ粒子について実施例3で記述したELBLコーティング手順を、以下の変更を加えて使用した。3μm CaCO3マイクロ粒子は、水性懸濁液中で通常の重力で容易に沈下するので、加層工程中に、超音波処理をするかわりに、懸濁液をローテーター上で10分間穏やかに混合した。また、加層工程および洗浄工程の後にマイクロ粒子をペレットとするために、より遅い遠心速度を使用することができる。従って、マイクロ粒子懸濁液は1500 gで1分間の遠心で沈殿させた。ACT-1145の製造のためには、PGAの1.0 mg/mL溶液を使用して1番目の層をコーティングし、フルオレセインで標識したPLL(PLL-FITC、Sigma社カタログ番号 P3543)の1.0 mg/mLの層を2番目の層に使用した。続く5回の加層工程でPGAおよびPLLを使用して、マイクロ粒子を合計7つのポリペプチド層でコーティングした(これらの粒子は、実施例8に記述するように、アミド架橋実験に使用することができる)。ACT-2086(配列番号13;RSV-M281-98G164-191 K20Yアミド)の0.5 mg/mL溶液を、最終層のコーティングに使用した。マイクロ粒子を洗浄し、遠心で沈殿させ、湿ったペレットとして4℃で貯蔵した。粒子に吸着されたACT-2086の量は、アミノ酸分析によって計算され、1%粒子懸濁液について43μg/mLであることが見出された。粒子を蛍光顕微鏡で検査したところ、3.0 (+/- 1.5) μmの直径を有する球状であることが見出された。粒子はよく分散された個々の粒子であり、2つまたは3つの粒子の集合体が少数あった。
実施例7の手順を使用して、7つのポリペプチド層(PGA/PLL-FITC/PGA/PLL/PGA/PLL/PGA)を有する3μm CaCO3マイクロ粒子を製造した。38 mg/mL (0.20 M)の1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド (EDC、Sigma社カタログ番号E6383)および11 mg/mL (0.05 M)のN-ヒドロキシスルホスクシンイミド・ナトリウム塩(スルホ-NHS、Sigma社カタログ番号56485)を含む0.2 Mリン酸ナトリウムpH 6.5バッファーの溶液を新たに調製した。7層マイクロ粒子をEDC/スルホ-NHS溶液に懸濁し、3%懸濁液とした。懸濁液を室温で30分間、穏やかに混合し、それから粒子を遠心で沈殿させて10 mM HEPESバッファーで3回洗浄した。ACT-2086(配列番号13;RSV-M281-98G164-191 K20Yアミド)の0.5 mg/mL溶液を使用して最終層をコーティングした。マイクロ粒子を洗浄し、遠心で沈殿させ、湿ったペレットとして4℃で貯蔵した。粒子に吸着されたACT-2086の量は、アミノ酸分析によって計算され、1%粒子懸濁液について49μg/mLであることが見出された。
実施例8のCaCO3マイクロ粒子ACT-1146を、0.5 M EDTAナトリウム、pH 8.0溶液中に6%(重量/体積)となるように懸濁した。粒子を室温で30分間、穏やかに混合し、1500 gで3分間遠心分離し、EDTA溶液を吸引した。結果として得られたマイクロカプセルを、2回、10 mM HEPESバッファーに再懸濁させて1500 gで3分間遠心分離し、過剰な塩を除去した。マイクロカプセルを遠心で沈殿させ、液を吸引し、湿ったペレットとして4℃で貯蔵した。カプセルに吸着されたACT-2086の量はアミノ酸分析によって計算され、1%粒子懸濁液について41μg/mLであることが見出された。カプセルを蛍光顕微鏡で検査したところ、3.0 (+/- 1.5) μmの直径を有する球状であることが見出された。カプセルはよく分散された個々のカプセルであり、2つまたは3つのカプセルの集合体が少数あった。
BALB/cマウスを、足蹠注射により、ナノ粒子ACT-1042(DP配列番号8;RSV-G164-191)で3回免疫し、血清を採取してELISAで試験した。血清は、立体構造性RSV-G CX3CエピトープペプチドACT-1042を認識したが(図4)、システイン残基をキャッピングすることにより直線化した型の同じペプチド(ACT-2054)は認識しなかった(図5)。この血清はまた、未変性RSV-Gタンパク質も認識し(図6)、RSV-G ELBLナノ粒子による免疫処置は立体構造依存的抗体応答を誘発させたことが示唆された。ACT-1042によって誘発された抗体応答の生物学的活性は、RSV-G CX3Cケモカイン結合の阻害(図7)、および、精製RSV-Gへと向かうヒトPBMCの遊走の阻害(図8)を測定するアッセイによって確認された。従って、立体構造が制限されたRSV-G CX3Cエピトープに基づく、設計されたペプチドを組み入れた、新規のナノ粒子ワクチンの設計は、生物学的に意味のある抗体応答を誘発させることができる。
BALB/cマウスは、s.c.(皮下)、i.p.(腹腔内)、i.n.(鼻腔内)、または足蹠を介して、ナノ粒子ACT-1023(RSV-M281-98)(DP配列番号12)で免疫した。陽性対照のマウスは、フロイントの完全アジュバント(CFA)中のペプチドACT-2019(RSV-M281-95;配列番号7)でs.c.で免疫し、フロイントの不完全アジュバント(IFA)中のACT-2019でブースト接種を行った。未免疫のマウスを陰性対照とした。脾臓におけるT細胞応答は、免疫後14日目に、IL-4およびIFNγELISPOTアッセイで測定した。図9のデータは、ACT-1023を用いた免疫が弱いIL-4 ELISPOTsを誘発したことを示しているが、これはそのDPに含まれるCD8エピトープから予測される通りである。対照的に、足蹠を介して免疫したマウスでは、一回の免疫後に強力なIFNγ応答が生じた。s.c.群およびi.p.群ではより弱い応答が生じたが、それでも陽性対照のCFA群に匹敵していた。この実験では、鼻腔内送達では免疫原性が見られなかった。ペプチド単独での免疫は、低いレベルのIFNγ応答が生じた。従って、RSV-M2ペプチドの効力は、ELBLナノ粒子中にそれを埋め込むことによって著しく上昇するということを、本発明者らは確証した。
どちらかの一価ナノ粒子ワクチンを用いたマウスの免疫化では、予測された免疫応答が誘発された(図4〜8および9を参照のこと)。図4〜8に図示されている抗体応答は、3回の免疫処置を必要とした(第一次免疫処置+2回のブースト接種)一方、図9に図示されているT細胞応答は単一の(第一次の)免疫処置を要するのみであったことは、興味深い。RSV-GおよびRSV-M2の両方を含む多価ナノ粒子ワクチンは、RSV-G成分の免疫効力を向上させることができるか否かを決定するために、RSV-G(ACT-1042、投与あたり1μg DP)ナノ粒子とRSV-M2(ACT-1023、投与あたり5μg DP)ナノ粒子との混合物を用いて、足蹠または鼻腔内を介した送達によってマウスを免疫化した。第一次免疫処置後およびブースト後の抗体力価をELISAによって測定し、ブースト後のT細胞応答をELISPOTによってモニターした。いずれの構築物の投与を受けたマウスも、測定できるほどの第一次抗体応答を有していなかった(データは示していない)。図10は、RSV-G特異的IgGを測定する、ブースト後ELISAの結果を示す。ACT-1042のみの投与を受けたマウスでは、足蹠を介した投与の場合にのみ、検出可能な力価がもたらされた。足蹠群においてRSV-M2ナノ粒子を追加すると、CFA中5μgのRSV-Gペプチドによって誘発される場合と同等の抗体力価が生じた。この混合物の鼻腔内投与は、RSV-Gナノ粒子単独をf.p.注射した場合とほぼ同じ力価を誘発したが、RSV-Gナノ粒子単独のi.n.投与は、ブースト後に検出可能な抗体力価を誘発できなかった。これらの結果は、RSV-M2ナノ粒子を含めることが、RSV-G中の抗体エピトープの効力を増加させると見られること、および、ナノ粒子のi.n.投与は免疫応答を誘発することができることを示している。
同じ粒子内に一緒に加層された別個のDPとして、または、RSV-GエピトープとRSV-M2エピトープとの両方を含む融合DPとして、複数のエピトープが同一粒子内に含まれるように、さらなるナノ粒子を設計した。表3は、複数エピトープRSVナノ粒子の構成を記載し、表4は、それぞれにおいて用いられたDP配列を記載する。ナノ粒子ACT-1077から-1079までは、1つまたは複数の層にRSV-M2のT細胞エピトープを含み、8番目の層にRSV-GのB細胞エピトープを含む。ナノ粒子ACT-1086から-1088までは、それぞれ二重エピトープペプチドACT-2086から-2088までを含み、それらのペプチドは8番目の層のみに堆積されている。一緒に加層されるかまたは単一の融合ペプチド中にある、上記のまたはその他のRSVエピトープを使用し、さらなる設計を想定することができる。
同じ投与量の設計されたペプチド(1μg)を、陽性対照(CFA)およびナノ粒子(1042)群の両方において使用した。以前の実験では、CFAには5〜10μgを用い、ナノ粒子には1μgを使用していた。これらの結果は、投与量が同等ならば、ナノ粒子はCFA対照よりも免疫原性が高いことを示している。
32 mgのミディアム分子量PGAナトリウム塩(Sigma社により市販されている)を、1.0 mLの0.1 Mリン酸ナトリウムpH 7.2バッファーに溶解した。22 mgの1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(EDC)および15 mgのスルホ-NHSを加え、溶液を室温で15分間維持した。4.6 mgのシスタミンHCl塩を加え、溶液を90分間反応させた。生成物を、希釈酢酸バッファーでpH 4.5に事前平衡化したDesalt(登録商標)カラム(Pierce社によって市販されている)に通すことにより、精製した。溶出物を凍結し、-80℃で保存した。チオール化の程度はエルマン(DTNB)アッセイにより推定し、全グルタミン酸残基の約24%であると見出された。
RSV Gタンパク質からの既知のT細胞エピトープ(RSV G残基187〜198、配列番号25 KRIPNKKPGKKT)を含む合成ペプチドは、固相ペプチド合成樹脂上で合成することができる。樹脂を切り離す前に、樹脂を無水ブロモ酢酸で処理することにより、N末端にブロモアセチル基を取り付けることができる。樹脂の切り離し、および精製を行った後、1.5 mg(約1 umol)のペプチド(ブロモアセチル-KRIPNKKPGKKT)を、15 mgのPGA-SHのリン酸バッファーpH 7中の溶液に加えることができる。生成物は、5000 MWカットオフ透析チューブを使用した透析により部分的に精製でき、ペプチドが装着されたことはアミノ酸分析により確認できる。
使いやすい濃度(典型的には0.5〜50 mg/mL)およびほぼ中性のpH(典型的にはpH 6〜8)を有する、ミディアムMWのPLLナトリウム塩(Sigma社により市販されている)のストック溶液を、スルホスクシンイミジル4-[N-マレイミドメチル]シクロヘキサン-1-カルボキシレート(スルホ-SMCC)のような架橋剤と混合する。スルホ-SMCCの量は変え得るが、PLL中のリシン残基の1〜25%を修飾するために十分な量が使用される。典型的な反応時間は0.5〜5時間である。その後、反応溶液をゲルろ過カラムに通すか、または透析を行うことによって、過剰なスルホ-SMCC剤を除去する。修飾されたPLLをその後、ほぼ中性のpH(典型的にはpH 6〜8)において、わずかに過剰量であるシステイン含有エピトープペプチドと反応させる。最終的なPLL-エピトープコンジュゲートを、溶液をゲルろ過カラムに通すか、または透析を行うことによって、精製する。この試薬は、PLLについて同様に使用される方法によって、高分子電解質ELBLフィルムへ組み入れることに適している。
第28日目において、鼻腔内を通じてマウスをRSVの攻撃にさらし、5日後に犠牲にした。肺を採取してホモジェナイズし、ベロ細胞アッセイ、および、RSV-M遺伝子のqPCR増幅により、ウイルスのタイターを測定した。この結果(図17)は、RSV-Gを含むいずれの組成物を用いても(第3〜6群)、実質的に完全な保護が得られたことを示している。この試験では、RSV-M2単独では保護が得られず、多価ワクチンがRSV-G単独より良く効くということもなかったが、これは比較的大きい投与量(10μg)を使用したことが理由である可能性がある。
第0日目に、後ろ足の足蹠にRSVナノ粒子構築物を注射することにより、マウスを免疫した。免疫原としては、ACT-1023(RSV-M2)、ACT-1042(RSV-G)+ ACT-1023(RSV-M2)、ACT-1086(単一層におけるRSV-M2+Gの融合ペプチド)、およびACT-1077(異なる層におけるRSV-Gペプチド+ RSV-M2ペプチド)を含ませた。マウスは、第7日目に、RSV-M2を含みCFSE標識された標的細胞による攻撃にさらした。翌日、CFSE標識標的細胞の生存を検出するために、フローサイトメトリーで脾臓細胞を解析した。結果は(表18および19)、RSV-M2エピトープを含有するいずれのナノ粒子で免疫しても、RSV-M2特異的エフェクター細胞が誘発されたが、一価(RSV-M2)構築物で免疫したマウスよりも多価(RSV-M2+G)構築物で免疫したマウスの方が、応答が強力であったことを示している。

前記1つ以上の多層フィルムは、それぞれ、2層以上の高分子電解質を含み、ここで、隣接する層は反対に荷電した高分子電解質を含み、前記多層フィルムの少なくとも1つの高分子電解質が、設計されたポリペプチドを含み、
前記設計されたポリペプチドは、反対に荷電した表面に安定的に結合するために十分な電荷を有し、前記RSV由来の第1のポリペプチドエピトープ、前記RSV由来の第2のポリペプチドエピトープ、またはその両方を含み、
前記設計されたポリペプチド以外の高分子電解質は、1,000より大きい分子量、および、分子あたり少なくとも5個の電荷を有する、ポリカチオン性材料またはポリアニオン性材料を含み、

- 前記RSV由来の第1のポリペプチドエピトープを含む第1の設計されたポリペプチド、および、前記RSV由来の第2のポリペプチドエピトープを含む第2の設計されたポリペプチドが、同一の多層フィルム中に存在する、請求項20に記載の組成物。
- 前記RSV由来の第1のポリペプチドエピトープを含む第1の設計されたポリペプチドが、第1の多層フィルム中に存在し、前記RSV由来の第2のポリペプチドエピトープを含む第2の設計されたポリペプチドが、第2の多層フィルム中に存在する、請求項20に記載の組成物。
- 前記RSV由来の第1のポリペプチドエピトープは、特異的細胞傷害性応答またはヘルパーT細胞応答を誘発し、前記RSV由来の第2のポリペプチドエピトープは、特異的抗体応答を誘発する、請求項17に記載の組成物。
- 前記RSV由来の第1のポリペプチドエピトープは、RSV-M2タンパク質のエピトープであり、前記RSV由来の第2のポリペプチドエピトープは、RSV-Gタンパク質のエピトープである、請求項23に記載の組成物。
- 前記RSV由来の第1のポリペプチドエピトープは、配列番号7を含み、前記RSV由来の第2のポリペプチドエピトープは、配列番号4を含む、請求項24に記載の組成物。
- 前記RSV由来の第1のポリペプチドエピトープは、RSV-Fタンパク質のエピトープであり、前記RSV由来の第2のポリペプチドエピトープは、RSV-Gタンパク質のエピトープである、請求項17に記載の組成物。
- 前記フィルムが、コア粒子上に堆積されている、請求項17に記載の組成物。
