Description
固体マトリックスはWhatman社製のFTA Elute 903マトリックスであった。
1)グアニジンチオシアネートの4M原液を調製し、様々な濃度の試薬を用いて2Mに希釈した。
2)グアニジンチオシアネート/試薬混合物を入れたトレーに903マトリックス(2.25インチ×2.25インチ)を入れ、10秒間穏やかに振盪した。
3)2つのヘアドライヤー(Simply Basic DS−727)を用いて、金属製ラック上でマトリックスを10分間乾燥させた(2つのヘアドライヤーとマトリックスが一直線に並ぶように、一方をマトリックスの15cm上に30°の角度で置き、他方をマトリックスの25cm下に30°の角度で置いた)。さらに、気流なしで21±2℃で一晩乾燥させた。
4)使用時までマトリックスをデシケーター内で室温で保存した。
ヒトDNA(Roche)を160ng/μlの濃度で試験固体マトリックスにスポットした。通常、予めくり抜いておいた直径5mmディスクのマトリックスを使用した。ディスクは室温で最低3時間乾燥させた。
1)各乾燥ディスクを1.5ml滅菌マイクロチューブに入れ、500μlのdH2Oで計5秒間の3回のパルスボルテックスによって洗浄した。
2)ディスクを、100μlのdH2Oが入った0.5mlのマイクロチューブに移し、ディスクが完全に水に浸るようにした。
3)マイクロチューブを98℃のヒートブロックに30分間置いた。
4)マイクロチューブを60秒間パルスボルテックスし、短く遠心した。
5)ディスクを残して、溶出液を新たな0.5mlチューブに移した。溶出液は定量までの間4℃で保存した。
7900HT Thermal Cycler(Applied Biosystems)を用いたQPCRによって、溶出液中のDNAの定量を行った。反応は、RNase Pアッセイ及びTAQMAN(商標)Universal PCR Master Mix(Applied Biosystems)を用いて設定した。実験に用いたものと同じRoche社製DNAの10〜0.01ng/μlの逐次希釈液を用いて4点標準曲線を作製した。初期QPCR定量は96ウェルプレートで行い、手動で設定した。液体ハンドリングロボットの導入及び検証後、384ウェルプレートを用してその後の定量を行った。96及び384ウェルプレートの両方を検証し、互いに対比して方法間の整合性をチェックした。
結果(表1)は、イヌリン又はPEGを含浸したマトリックスは、FTA Eluteだけの場合と比較して、DNA回収率が増大していることを示す。さらに、10%のPEGを含有するスポット用緩衝液を使用すると、DNA収率がさらに増大した(FTA Elute+20%イヌリンと併用すると85%)。
Claims (6)
- 精製DNAの保存に適した固体マトリックスであって、当該マトリックスがイヌリンを含む溶液で処理されている、固体マトリックス。
- PEGでも処理されている、請求項1記載の固体マトリックス。
- イヌリン処理が20%以下の濃度である、請求項1又は請求項2記載の固体マトリックス。
- 10%以下のPEGも含む、請求項1乃至請求項3記載の固体マトリックス。
- 核酸の添加前にイヌリンを含む溶液で固体マトリックスを処理することによって、固体マトリックスから溶出される精製DNAの収率を増大させる方法。
- 固体マトリックスがPEGでも処理されているか、或いはPEGを含む緩衝液中でDNAを適用する、請求項5記載の方法。
