溶血性補体アッセイ
C5およびC5Δ17(エキソン17を欠く)のcDNAを合成し、ベクターpcDNA3.1(−)に結紮する。ヒト胚性腎細胞(HEK293)に、C5またはC5Δ17のいずれかを含む発現ベクターをトランスフェクトし、G−418を含む培地で培養しポジティブコロニーを選択する。その後、ポジティブコロニーを無血清条件にて培養し、培養培地を回収し、精製し、ウエスタンブロット法によりC5およびC5Δ17の存在を試験する。その後、精製した上清を、無処置のC5bが存在するかどうか示す溶血性補体アッセイに供する。この機能アッセイにおいて、両方の上清を、C5を欠く血清で処理し、この溶液を赤血球とともにインキュベートする。C5b−9を介した溶解の程度を、415nmにてODを測定することにより決定する。
AONの設計および化学
AONは、Aartsma-Rus et al., 2009にて以前に議論された基準に基づいて設計した(Aartsma-Rus et al., Guidelines for antisense oligonucleotide design and insight in splice modulating mechanisms, Mol Ther. 2009 Mar; 17(3):548-53)。潜在的なエキソンスプライスエンハンサー(ESE)の位置、または5’/3’スプライスサイト配列に対する位置は、ESEファインダー3.0を使用して予測した(http://rulai.cshl.edu/tools/ESE/)。M−FOLD(http://frontend.bioinfo.rpi.edu/applications/mfold/)によって予測されるmRNA前駆体の二次構造を、さらに考慮に入れた。AONは、2’O−メチルリボース分子および完全なホスホロチオネート骨格(Prosensa)を備えた21−25merだった。AONの配列は配列表にて提供される。5’FAM標識コントロールAONを、標的細胞へのトランスフェクションの効率を試験するために使用した。さらに、標的に対するそれらの効率を増加させるために、それらの長さを25merに増加させることによっておよび/またはいくつかのロックされた核酸(LNA)の改変の組み込みによって、選択したAONを改変した。これらのAONの設計のために、http://www.exiqon.com/custom-antisense-oligonucleotidesの基準を使用した。

培養培地を細胞から取り除き、1mlの冷却したRIPAバッファー(Rocheの完全ミニプロテアーゼ阻害剤を含む)を、75cm2フラスコにて5x106細胞に対して添加し、揺らしながら5分間氷上に維持した。細胞溶解物を、細胞スクレーパーを使用して集め、マイクロ遠心分離チューブに移した。サンプルは15分間14.000xgで遠心分離し、細胞デブリスを沈殿させた。上清を新しいチューブに移し、タンパク質濃度うぃQuanti−itタンパク質アッセイキットを使用して決定した。
7.9mlの水、5mlの1.5MTris−Hcl(pH8.8)、6.70mlのアクリルアミド、200μlの10%SDS、8μlのTEMEDおよび200μlの10%APSを混合して、10%ゲルの分離ゲルを作製した。TEMEDおよびAPSを添加した直後、ゲルを注ぎ(7.2ml/ゲル)、1mlのイソプロピルアルコール/Miliq(50/50)をゲルの上部に置き、重合させた。ゲルは約30分間重合させた。その後、5.5mlの水、1mlの1MのTris−HCl(pH6.8)、1.33mlのアクリルアミド、80μlの10%SDS、8μlのTEMEDおよび80μlの10%APSを混合して、スタッキングゲルを準備し、分離ゲルの上にピペットで移し、コームを設置し、重合させた。タンパク質サンプルを準備し(フード下において、15μlサンプル+5μlローディングバッファー)、サンプルを5分間95℃で煮沸し、氷上で冷却した。スタッキングゲルの準備を整えて、ランニングバッファーをゲルユニットのチャンバーの上部および下部に添加し、それぞれのスロットに15μl充填した。サンプルが分離ゲルに達するまで、ゲルに80Vの電圧をかけ、その後、青い色がゲルを離れるまで120Vの電圧をかけた。
2つの繊維パッドおよび3枚のプリカットされたWhatman3MM紙を用意し、トランスファーバッファーに浸した。PVDF膜を100%メタノールに短時間浸し、蒸留水で2度洗浄し、PBSにてインキュベートした。システムをカセットにて以下の順番で組み立てた:1つの繊維パッド、3つのWhatman紙、ゲル、PVDF膜、3つのWhatman紙、1つの繊維パッド。カセットを電極モジュールに挿入し、膜がゲルと陽極との間に位置するようにトランスファータンクに入れ、冷却室(4℃)にて1時間、サンプルに100Vの電圧をかけた。緩やかに振動する振動プラットフォームにて、室温で1時間または4℃で一晩、膜をOdysseyブロッキングバッファーにてブロッキングした。一次抗体をOdysseyブロッキングバッファーにて希釈し、バックグラウンドを低下させるために、0.2%のTween−20を希釈した抗体に添加し、振動プラットフォームにて一晩穏やかに揺らしながらインキュベートした(最適なインキュベーション時間は、一次抗体ごとに異なる)。ウサギ抗ヒトIL−1RAcP抗体(AbD serotec、製品番号AHP549)およびチキンポリクローナルベータアクチン抗体(Abcam、製品番号Ab 13822)を使用した。膜を、穏やかに振動させながらPBS+0.1%Tween−20で室温にて5分間の洗浄を4回行い、Odysseyブロッキングバッファー(0.2%のTween−20および0.01%−0.02%のSDSを添加)に希釈した蛍光標識二次抗体とともに1時間室温で、光を遮りながらインキュベートした。二次抗体は、ヤギ抗ウサギAb(Licor、製品番号926−32211)およびロバ抗チキンAb(Licor、製品番号926−32228)を使用した。膜に対して、穏やかに振動しながら(光を遮断)、PBS+0.1%のTween−20にて室温における5分間の洗浄を4回行い、その後、膜を、Li−cor Odysseyイメージングシステムにてスキャンした。
NIH−3T3細胞を、24ウェルプレートに40.000細胞/ウェルで播種した。翌日、細胞に、上記の手順によってコントロールおよび試験されるAONをトランスフェクトした。細胞を一晩培養し、リポフェクタミン2000によりpNFκB−LucおよびpRL−CMVプラスミドをトランスフェクトした。翌朝、細胞を4時間、mIL−1βで刺激した。デュアルルシフェラーゼリポーターアッセイ(Promega)の試薬を、製造者の手順にそって準備した。培養培地を除去し、細胞を1mlのPBSで洗浄した。250μlの1X Passive Lysisバッファーを細胞に添加し、細胞を採取した。細胞溶解物をマイクロ遠心分離チューブに移し、4℃で、13,000gにて5分間遠心分離することで、デブリスを除去した。30μlの溶解物を、96ウェルプレートのウェルの各々に添加した。100μlのLARIIをウェルの各々に添加し、ホタルルシフェラーゼ活性をルミノメーターで測定した。その後、100μlの1XStop&Glo試薬をウェルの各々に添加し、レニラルシフェラーゼ活性を測定した。pNFκB−Luc活性はpRL−CMVにて標準化した。
NIH−3T3細胞を、10%FBSを補ったDMEMにおいて、6ウェル培養ディッシュにて、0、5x106細胞/ウェルの密度で、培養した。細胞に対して、先に記載するように、50nMのAON PS−300LNAをトランスフェクトした。16時間後、細胞を1ng/mlのマウスIL−1βで4−5時間刺激した。その後、先に記述するように、全RNAの単離およびcDNA合成を行った。IL−1に誘導される、IL−6およびICAM−1といったサイトカインの誘導を、以下のプライマーセットを使用して、先に記述されるようなqPCRアッセイを行なうことで決定した:マウスIL−6フォワードプライマーCCGGAGAGGAGACTTCACAG、IL−6リバースプライマーTCCACGATTTCCCAGAGAAC;マウスICAM−1フォワードプライマーGGCATTGTTCTCTAATGTCTCCGを進める、ICAM−1リバースプライマーGCTCCAGGTATATCCGAGCTTC。すべてのデータは、ハウスキーピング遺伝子ベータアクチンの発現で標準化した;マウスのベータアクチンフォワードプライマーACTGGGACGACATGGAGAAG、リバースプライマーGGTCATCTTTTCACGGTTGG。これらのプライマーは配列番号66−71として配列表に示される。
炎症性疾患を治療するためのC5aタンパク質のエキソン17のスキッピング
目的は、C5aの量を減少させ、C5bをそのままの状態に維持することである。C5は肝臓によって生産されるため、C5特異的AONの標的器官は肝臓である。
目的は、可溶性IL−1RAcPの量について、その膜結合形態から可溶性形態に変化させることで増大させることである。それは肝臓によって生産されるため、可溶性IL−1RAcPに特異的なAONのための標的器官は肝臓である。
結果:
C5エキソン−17スキッピング
C5aの濃度を減少させるために、一連のAONをマウスまたはヒトエキソン17を標的化するように設計した(例1を参照)。それらは、L929およびHepG2細胞株それぞれでスクリーニングした。細胞株はリポフェクタミン2000にてAONをトランスフェクトし、平均トランスフェクション効率を、蛍光性AONを使用して決定した。マウス標的エキソンについての11AONおよびヒト標的エキソンについての3AONのスクリーニングから、AON PS329およびPS377は、スキッピングエキソン17において最も成功したAONとして選択された。有効なAONは、マウスおよびヒト標的について、それぞれ約178bpおよび約155bpのサイズで、より短い転写物断片を作ると予想された(図2および7)。配列分析は、さらにより短い断片は欠いているエキソン17であることを示した。AON PS−329,329−25mer,354,329−LNAは、異なる濃度において広範囲に試験し、スキッピングにおける効率と比較した。より短い断片のアガロースゲルにおけるバンドの密度の比較に基づいて、PS−329−LNAはその他のものよりも少し有効であった(図8、31−55)。AON PS−329も試験し、パイロット実験において機能的インビボを見出した。100mg/kgのAONを、IVで3または4日間注射し、肝臓試料をRT−PCRで分析し、エキソンスキッピングを検出した。スキップされた産物は正確なサイズを有しており、配列分析から、それが正確な産物であることが確認された。
可溶性IL−1RAcPの産生(IL−1RAcPΔ9)を誘導するために、一連のAONをマウスおよびヒトエキソン9を標的化するように設計した(例2を参照)。それらは、NIH−3T3およびHepG2細胞株にてそれぞれスクリーニングした。細胞株はリポフェクタミン2000にてAONをトランスフェクトし、平均トランスフェクション効率を、蛍光性AONを使用して決定した。マウス標的エキソンについての13AONおよびヒト標的エキソンについての5AONのスクリーニングから、AON PS300およびPS373は、スキッピングエキソン9において最も成功したAONとして選択された。有効なAONは、マウスおよびヒト標的について、それぞれ約150bpおよび約200bpのサイズで、より短い転写物断片を作ると予想された(図4および7)。配列分析は、さらにより短い破片がエキソン9を欠くことを示した。AON PS−300,300−25mer,300−LNA,327および327−25merは、異なる濃度において広範囲に試験し、スキッピングにおける効率と比較した。PS−300−LNAは、特に50nMの濃度での他のAONよりも、スキッピングにおいてはるかに有効だった(約85%)(図8、1−31)。qPCR分析によって結果を確認した(図9)。AON PS−300も、パイロット実験においてインビボで試験した。50−100mg/kgのAONを3または4日間IPまたはIVで注射し、肝臓試料を、エキソンスキッピングのためにRT−PCRで分析した。スキップされた産物は正確なサイズを有していることが、配列分析で確認された。インビボスキッピング効率は、インビトロスキッピング効率と比較可能だった(図10)。
本発明のオリゴヌクレオチドの有効性を、さらに、ウエスタンブロッティング法により、sIL−1RAcP、IL−1、C5aまたはC5bを定量することによって、タンパク質レベルで試験してよい:例えば、可溶性sIL−1RAcPの増加および/または遊離型IL−1の減少および/またはC5aの減少。本発明のオリゴヌクレオチドの有効性は、さらに、分子の活性の存在もしくは濃度またはIL−1によって誘導または活性化された経路を評価することによる、Il−1RAcPのための機能アッセイによって試験してよい。Il−1はNF−κB活性化を誘導する。したがって、本発明のオリゴヌクレオチドの有効性は、さらに、NF−Κbの活性化、NF−Κbの活性の低下を評価することで試験してよい。

Claims (19)

- 前記オリゴヌクレオチドは、配列番号1または2に示されるC5 mRNA前駆体のエキソンおよび/または配列番号3または4に示されるIL−1RAcPのエキソンに相補的であるか、または結合する、請求項7または9に記載のオリゴヌクレオチド。
- 前記オリゴヌクレオチドは、配列番号11−42の配列を含むか、またはそれから成る、請求項1から10の何れか1項に記載のオリゴヌクレオチド。
- 前記オリゴヌクレオチドは、RNAベースのオリゴヌクレオチドと比較して、骨格、糖および/または塩基の修飾を有し、好ましくは、前記オリゴヌクレオチドは、1以上の2’−O−メチルホスホロチオネートを含むか、またはそれから成る、請求項1から11の何れか1項に記載のオリゴヌクレオチド。
- 前記オリゴヌクレオチドは、2’−O−メチルホスホロチオネートオリゴリボヌクレオチドの修飾およびロックされた核酸モノマーを含む、請求項12に記載のオリゴヌクレオチド。
- 前記オリゴヌクレオチドは、ゆらぎ塩基対を形成することができる、少なくとも1つのイノシンおよび/または塩基を含む、請求項1から13の何れか1項に記載のオリゴヌクレオチド。
- 薬物として使用するための、好ましくは、個体における炎症性障害を予防または治療するための、より好ましくは、リウマチ様関節炎(RA)、若年性リウマチ様関節炎、乾癬、乾癬性関節炎、強直性脊椎炎、クローン病または潰瘍性大腸炎を含む炎症性腸疾患、肝炎、敗血症、アルコール性肝疾患および非アルコール性脂肪症を予防または治療するための、請求項1から14の何れか1項に記載のオリゴヌクレオチド。
- 請求項1から15の何れか1項に少なくとも1つの分子を含む組成物であって、前記組成物は好ましくは医薬組成物であり、前記医薬組成物は、医薬的に許容可能な担体、アジュバント、希釈剤および/または賦形剤を含む組成物。
- 少なくとも2つのオリゴヌクレオチドが存在し、1つ目は請求項1から3の何れか1項に記載のものであり、2つ目は請求項4または5に記載されたものである、請求項16に記載の組成物。
- 個体における炎症性障害を予防または治療するための薬物の製造のための、請求項1から15の何れか1項に記載の前記分子の、好ましくはオリゴヌクレオチドの、または請求項16から17の何れか1項に記載の組成物の使用。
- 個体における、1以上の症状および/または特徴を軽減するための、および/または炎症性障害のパラメータを改善するための方法であって、前記方法は、請求項1から15の何れか1項に記載の分子、好ましくはオリゴヌクレオチド、または請求項16から18の何れか1項に記載の組成物を前記個体に投与することを含む方法。
