Abstract
Description

本発明は、2種またはそれ以上の活性物質を含む粒子状薬物送達組成物に関する。
複数の薬物または薬物候補を特定の病んだ細胞および組織に、例えば、癌細胞に、例えば、特定の器官または組織の癌細胞に、時間空間的に(tempospatially)制御された方法で有効に送達するためのナノ技術の発展は、今日までに直面した治療的挑戦を潜在的に克服する。本発明は、送達組成物の形成と、送達組成物、例えば粒子からの、活性物質の放出を制御するための方法および組成物を提供する。ある実施態様では、本発明は、薬物送達組成物、例えば、ペンダント(pendant)官能基を有するポリマーなどのポリマーに結合した活性物質を含む粒子の製造方法を提供する。本発明の1つの利点は、薬物で官能化した、または、リガンドで官能化した別個のポリマーを設計し、配合することにより、2種、3種またはそれ以上の薬物を送達できる粒子を再現可能に設計し、特徴付けられることである。加えて、これらの方法は、例えば、それぞれの物質のタイプについて別々に、例えば、活性物質の特徴、例えば、溶解性、電荷、分子量、半減期および/または生体分布プロフィールに関係なく、薬物放出および薬物動態の特徴を調整することを可能にする。さらに、薬物を負荷した粒子を特定の組織または細胞、例えば癌細胞に標的化することにより、活性物質の生体分布を変えられる相乗作用的な薬物の効果が達成できる。これは、より良好な効力および耐容性と言い換えることができ、活性物質を可能性のある臨床開発に適合させる。

本願は、2種またはそれ以上の活性物質を含み、少なくともその一方が生物分解性ポリマー、例えば、ペンダント官能基を有する生物分解性ポリマーに結合している薬物送達組成物を提供する。送達組成物は、複数の薬物、例えば、異なる特徴を有するもの、例えば、親水性および疎水性の薬物を、各薬物の放出パラメーターを独立して制御する能力をもって、同時送達することを可能にする。加えて、送達組成物は、システムを所望の細胞標的に送達するために、標的化物質を含み得る。一例として、プラチナで修飾されたPLAポリマーの内側にドセタキセルを組合せ、適度に高い負荷量(loading)のシスプラチンおよびドセタキセルを同時に前立腺癌細胞に送達するのに適するサイズの粒子を得ることにより、PLA−官能化デュアル薬物送達組成物が開発された。
ある態様では、送達組成物は、2種またはそれ以上の活性物質を含む粒子である。活性物質は、生物分解性ポリマー、例えば、ペンダント官能基を有する生物分解性ポリマーに結合していることができる。粒子は、また、複数の両親媒性分子、例えば、両親媒性脂質または両親媒性コポリマーを含有する疎水性コアを囲む両親媒性の殻を含み得る。粒子は、さらに、粒子の表面に標的化物質を含み得る。

ある種の実施態様では、本発明の結合体を、本発明の結合体を特定の細胞タイプ、細胞の集合体または組織に導く標的化物質を含むように改変できる。好ましくは、標的化物質は、粒子の表面に結合している。様々な適する標的化物質が当分野で知られている(Cotten et al., Methods Enzym. 217:618, 1993; Torchilin, Eur. J. Pharm. Sci. 11:881, 2000; Garnett, Adv. Drug Deliv. Rev. 53:171, 2001)。例えば、標的細胞の表面の抗原に結合する任意の数の異なる物質を用いることができる。細胞表面抗原を標的とする抗体は、一般的に、必要な標的への特異性を示す。抗体に加えて、Fab、Fab'またはF(ab')2などの適する免疫反応性断片も用いることができる。標的化メカニズムの形成において使用するのに適する多くの抗体断片は、当分野で既に利用可能である。同様に、標的細胞の表面にある任意の受容体のリガンドは、標的化物質として適当に用いることができる。これらには、任意の天然または合成の低分子または生体分子が含まれ、それは、所望の標的細胞の表面に見出される細胞表面の受容体、タンパク質または糖タンパク質に特異的に結合する。
本明細書に記載の送達組成物は、当分野で知られている任意の方法、例えば、ナノ沈降およびエマルジョン方法により製造できる。加えて、新しい送達組成物を製造するために、マイクロフルイディクス法を使用できる。
本発明は、さらに、本明細書に記載の任意の組成物を含む調製物、製剤、キットなどを含む。いくつかの場合では、疾患の処置(例えば、癌)は、本明細書に記載の組成物または「物質」の使用を伴い得る。即ち、本発明の1つの態様は、疾患(例えば、癌または腫瘍)の処置に有用な一連の組成物(例えば、医薬組成物)または物質を含む。これらの組成物は、場合によりそのような症状の処置用の組成物の使用のための指示書を含むキットに包装することもできる。これらおよび他の本発明の実施態様は、本明細書に記載の任意の技法および組成物および組成物の組み合わせに従う、疾患(例えば、癌または腫瘍)の処置の進行も伴い得る。
特許請求の範囲に記載の本発明の範囲を限定しない以下の実施例で、本発明をさらに説明する。
実施例1. PLA−Ptの合成
薬物で官能化されたポリマーを開発するための1つの戦略は、アミノ酸をそれらの対応するα−ヒドロキシル酸(i)に単純に変換することに基づく(図11)。第1段階は、酸の存在下で6時間にわたり亜硝酸ナトリウムを使用するジアゾ化反応による、アミンのヒドロキシルへの変換を伴う。これは、得られたモノマーを乳酸との縮合重合に直接使用し、ポリラクチドコポリマー、ポリ−OBnを得る高収量の反応である。150℃、3時間、連続的アルゴンパージでバルクの反応条件を使用し、続いて、さらに3時間減圧下で、縮合重合を実施した。1%パラ−トルエンスルホン酸を含むトルエン中の非常に希釈された反応条件下でのα−ヒドロキシル酸の脱水反応により製造した環状ラクチドモノマー(ii)の開環重合(ROP)を使用して、同じポリマーを製造した。
ポリ(乳酸−コ−グリコール酸)−ポリエチレングリコール(PLGA−PEG)を有するナノ粒子を合成することにより、PLA−OHポリマーのインビトロの毒性を試験した。粒子は、比較的非毒性であり、せいぜい27mg/mLのIC50値であった(図3)。マイクロフルイディクスのアプローチを使用して、2種の薬物を送達できるナノ粒子をナノ沈降により製造した(図4B)。本発明では、我々は、ポリラクチドに沿ってカルボキシルなどの官能基を有する標準的なPLGA−PEGを使用し、それは、親水性薬物をペンダントとして含有し、疎水性薬物の存在下でナノ沈降に使用された。一度ナノ粒子を製造したら、粒子をカップリングにより標的化リガンドで官能化した。シスプラチンプロドラッグを親水性薬物として、ドセタキセルを疎水性薬物として、A10−アプタマーを、前立腺特異的膜抗原(PSMA)を過剰発現する前立腺癌細胞株に標的化する標的化リガンドとして使用した(図13)。
25℃で、EG&G PAR Model 263 ポテンショスタット(Potentiostat)/ガルバノスタット(Galvanostat)において、電気化学分析ソフトウェア270、並びに、ガラス状炭素作用電極、白金ワイヤー補助電極およびAg/AgCl基準電極を含む3電極設定を用いて、電気化学的測定を行った。電気化学的データは、接合部電位について補正しなかった。KClを支持電解質として使用した。ポリ−Ptは、酸化還元活性を有し、pH7.4でPt(IV)/Pt(II)カップルに−0.801V vs Ag/AgCl付近で非可逆的サイクリックボルタンメトリー(cyclic voltammetric)応答を示し、同じ条件下で、白金一コハク酸塩プロドラッグの値は、−0.850 vs.Ag/AgClであった(図6A−6D)。これらの還元電位は、ポリマー性主鎖の存在が白金中心の電気的または立体的環境に影響を与えず、この構築物が効果的に白金の有効用量を放出して抗癌活性を増強することを示唆する。ポリ−Ptの還元電位の値は、それが血中での早まった還元を回避し、細胞内で還元されることを示す。
ナノ粒子からの白金(IV)およびドセタキセルの制御放出の速度論を研究した。候補薬物のナノ粒子からの制御放出は、薬物が送達媒体から放出された後で初めて活性になるので、新しい粒子の重要な利点である。放出研究のために、我々は、白金およびドセタキセルのナノ粒子を、PBS(pH7.4)20lで、37℃で透析し、生理的条件を模倣した。PolyPt−Doce−NPの水中の懸濁液を、いくつかの半透性小型透析チューブ(分子量カットオフ100kDa、Pierce)に分注し(各100μL)、リン酸緩衝塩水(PBS)(pH7.4)20lで、37℃で透析した。予め定めた時間で、ナノ粒子懸濁液のアリコートを取り出し、アセトニトリルに溶解した。放出された白金含有量を、原子吸光分析により測定し、購入できるタキソールで得られた標準較正曲線を使用して、ドセタキセル放出をHPLCにより定量した。これらのナノ粒子からの白金(図7A)およびドセタキセル(図7B)の両方の制御放出が観察された。結果は、このシステムが、共有結合した白金中心よりもドセタキセルが早く放出される時間的様式で薬物を放出できることを示した。放出のタイミングは、ポリマー主鎖の長さを伸ばすことにより、さらに延長できる。
標的化薬物放出構築物のナノ粒子が細胞死を促進する能力を、ヒト前立腺癌LNCaPおよびPC3細胞を使用するMTTアッセイを使用して測定し、標準化合物のシスプラチンおよびPt(IV)一コハク酸塩プロドラッグと比較した。ヒト前立腺癌LNCaPおよびPC3細胞をATCCから入手した。LNCaP細胞はPSMAを過剰発現し、PC3細胞はしない。細胞を37℃、5%CO2、10%ウシ胎児血清および1%ペニシリン/ストレプトマイシンを添加したRPMI増殖培地でインキュベートした。細胞を3ないし4日毎に植え継ぎ、植え継ぎ回数が20回に達したら、凍結ストックから再開した。MTTアッセイを使用して構築物の細胞毒性活性を評価した。異なる構築物の溶液を、滅菌PBSで使用前に新しく調製した。すべての白金調製物を、原子吸光分析により定量した。LNCaPおよびPC3細胞を100μLRPMI培地中で96ウェルプレートに播き、24時間インキュベートした。
ポリマー性ナノ粒子は、ファゴサイトーシスおよびエンドサイトーシスを含む様々な過程で細胞に取り込まれ得る。PolyPt−NP−Aptの内在化を研究するために、緑色のフルオレセインをベースとするコレステロールである22−NBD−コレステロールを粒子に含めた。22−NBD−コレステロールを含有する非標的化ポリ−Pt−NPを対照として使用した。LNCaP細胞を顕微鏡のカバースリップ(1.0cm)に、カバースリップ当たり1×105細胞のコンフルエンスで播き、終夜、加湿インキュベーターで、5%CO2で、37℃で、RPMI中で増殖させた。培地を交換し、NBD−コレステロール−PolyPt−NP−Aptの懸濁液を最終フルオロフォア濃度10μMで添加した。次いで、細胞を2.0時間37℃でインキュベートした。培地を除去し、2%パラホルムアルデヒド溶液を使用して、1時間室温で細胞を固定した。細胞をPBS(pH7.4)で3回洗浄した。次いで、細胞をPBS中の0.1%TRITON−X100で10分間透過処理し、続いて、PBSを使用して5回洗浄した。次いで、ブロッキングバッファー(PBS、0.1%ヤギ血清、0.075%グリシン)で、30分間室温(RT)で細胞をブロックした。製造業者の推奨する手順に従い、細胞を初期エンドソームマーカーであるマウスモノクローナルEEA−1と、湿った箱の中で、1時間37℃でインキュベートした。PBSで2回洗浄した後、細胞をブロッキングバッファーで、30分間RTでブロックし、次いで、二次Cy5ヤギ抗マウス抗体と1時間37℃でインキュベートした。PBSで4回洗浄し、水で2回洗浄した後、画像化のためにマウント液[20mM Tris(pH8.0)、0.5%没食子酸N−プロピルおよび70%グリセロール]を使用して細胞を顕微鏡のスライドに乗せた。FITCとローダミンのチャンネルの両方につき、500msecで画像を集めた。
シスプラチンの抗癌活性は、核DNAにおける白金化(platination)生成物の形成に基づく。これらの付加体のいくつかは構造的に同定され、グアニン−グアニン鎖内架橋cis−Pt(NH3)2d(GpG)(図10A)であり、総DNA白金化の>75%を占める。細胞内の白金1,2−d(GpG)付加体の検出は、この付加体に特異的な抗体の使用により実施した。簡潔に述べると、LNCaP細胞を、RPMI培地を使用して6ウェルプレートに播き、終夜37℃でインキュベートした。様々な構築物を最終濃度20μMで添加し、37℃で12時間インキュベートした。次いで、細胞をトリプシン処理し、PBSで洗浄し、HAES−STERIL−PBSに1×106/mLの密度で再懸濁し、予め被覆したスライド(ImmunoSelect, Squarix)に乗せ、空気乾燥させた。細胞の固定を、−20℃で、メタノール中、45分間実施した。5分間、0℃のアルカリ(70mM NaOH、140mM NaCl、40%メタノールv/v)処理で核DNAを変性させ、2段階のタンパク分解手順により細胞のタンパク質を除去した。
マイクロフルイディクスのシステムを使用して、PSMA−アプタマー標的化ナノ粒子を含有する約500の薬物のライブラリーを製造し、所望の特徴についてスクリーニングした。前駆物質の水混和性溶液(コ−ブロックポリマー、薬物など)を水に混合することを含むナノ沈降法により、ナノ粒子を作成する。水は薬物およびポリマーには不十分な溶媒であるので、ナノ粒子の沈降が起こる。(a)前駆物質溶液の組成を制御すること、および、(b)混合時間、温度および水流の前駆物質の流れに対する流速の比率などの混合条件を制御することにより、ナノ粒子の特性を制御する。次いで、ナノ粒子に取り込まれた蛍光染料を使用して、PSMA−陰性前立腺癌(PC3)および非前立腺(HeLa)細胞株に対して、PSMAを発現する前立腺癌細胞株(LNCaP)による優先的な取り込みについて、これらの製剤を試験する。このアプローチは、全身投与後にマクロファージを回避でき、かつ、前立腺癌細胞により差次的に取り込まれ得る最適化されたナノ粒子を提供できる。
本明細書に記載の送達組成物を、前立腺癌の動物モデルで評価する。重症複合免疫不全(SCID)マウス(Taconic, Germantown, NY, USA)に、マトリゲル(Matrigel)100μlと混合した1×105LNCaP細胞を両後肢に皮下注射する。平均腫瘍体積が約150mm3に達する6日後に、各グループの平均腫瘍体積が匹敵するように、マウスをグループに分類する(グループ当たり5匹のマウス)。6、9および14日目に、動物をPolyPt−Doce−NP、PolyPt−NP、Doce−NPs、Pt−一コハク酸塩、シスプラチン(5mg/kg)、PolyPt−NP−Apt、PolyPt−Doce−NP−AptおよびDoce−NP−Aptで、I.P.またはi.v.注射により処置する。腫瘍体積の測定を1日目(腫瘍接種)に開始し、腫瘍の体積が身体のサイズの10%を超えるまで、週に2回継続する。腫瘍の増殖に対する最大の効果は、両方の薬物を含有し、LNCaP細胞上のPSMAに標的化されるPolyPt−Doce−NP−Aptで観察される。
用量増大研究を実施し、異種移植片および遺伝子改変マウスモデル(GEMM)における腫瘍の減少および/または腫瘍増殖の減速に必要とされる組合せの薬物用量を決定する。同所性腫瘍サイズ比較および効力測定のために、バイオコンジュゲート(bioconjugate)処置の前およびその後に定期的に、マウス前立腺のMRI画像化を実施する。一度有効な用量を決定したら、比較効力研究を実施し、異種移植モデルおよびGEMMで、ナノ粒子−アプタマーバイオコンジュゲートを含有する薬物を、アプタマーを標的化する基を欠く同様のナノ粒子;並びに、遊離の薬物およびプラセボと比較する。111In−標識化バイオコンジュゲートを使用して、腫瘍を有するマウスで生体分布研究も実施し、腫瘍組織におけるバイオコンジュゲートの存在および相対的濃度を評価する。
白金を基礎とする薬物、即ち、シスプラチンおよびオキサリプラチンの作用に対する様々な代謝活性化因子の相乗効果を評価するために、官能化PLA誘導体を使用することにより、数々の組合せを調査した(表3)。我々は、インビボ研究を使用して、ロニダミン、ジクロロ酢酸と組み合わせたときの、オキサリプラチンおよびシスプラチンの効果を比較した。
−COOH官能基を有する新しいラクチド誘導体を、PLA−OHと無水コハク酸の反応により合成した(図15A)。この新しいPLA誘導体は、−OH、−NH2官能基を有する様々な薬物を結合する機会を提供した。1H分光法を使用して官能化ラクチドPLA−COOHを特徴解析した(図15B)。
ロニダミンは、ミトコンドリアのヘキソキナーゼを阻害する化合物であり、癌細胞の変化した代謝を選択的に攻撃する。研究は、シスプラチンを含むいくつかの他の化学療法薬との組合せでそれが相乗効果を有することを示した。PLA−ロニダミン(PLA−Loni)結合体(図16A)を合成することを目的とした。PLGA−PEG−COOHの存在下でのナノ沈降により、Pt(IV)−ヘキサノエートをPLA−ロニダミンポリマーコアの内側に封入した。ロニダミンを結合したPLAを、1H NMRにより特徴解析した(図16B)。
ジクロロ酢酸(DCA)は、ミトコンドリアのピルビン酸デヒドロゲナーゼキナーゼ(PDK)を阻害し、かくして解糖によるグルコース酸化を促進する小型有機分子である。この現象は、腫瘍細胞のアポトーシスに貢献する。高い割合のDCAを含むシスプラチン−DCAの組合せのために、我々はPLA−DCA誘導体を合成した(図17A)。PLGA−PEG−COOHの存在下でのPLA−DCAにおけるPt(IV)−ヘキサノエートの共封入は、白金とDCAの両方を有するナノ粒子をもたらす。PLA−DCA結合体を、1H分光法により特徴解析した(図17B)。
上記の研究に加えて、官能化ポリマー(PLA−OBn)の分子量を変化させる様々な研究を実施した。この目的で、3つの異なる変数、希釈、触媒濃度およびイニシエーター濃度を使用した。下表は、研究から得られた結果をまとめる。これらの結果から、触媒濃度の変更は、得られる分子量に有意に影響するが、モノマーに関するイニシエーター濃度の変動は有意な変動を示した。典型的な開環重合の速度論に従って、イニシエーターの濃度を下げることにより、得られるポリマーの分子量は増加した(表5)。一方、重合の溶媒を変えることにより、分子量はわずかに変化したが、いかなる傾向にも従わなかった。
本発明の数々の実施態様を記載した。それでもなお、本発明の精神と範囲から逸脱せずに、様々な改変を行い得ることが理解される。従って、他の実施態様は、以下の特許請求の範囲の範囲内にある。
Claims (20)
- 第1の活性物質および第2の活性物質を含むポリマーマトリックスを含む粒子であって、該ポリマーマトリックスが、約37℃で中性の水性溶液に粒子を懸濁してから最初の2時間の内に第1および第2の活性物質の各々の50%未満が放出される第1および第2の活性物質の各々の放出速度をもたらすように形成されている、粒子。
- ポリマーマトリックスが1種またはそれ以上のさらなる活性物質をさらに含む、請求項1に記載の粒子。
- 第1の活性物質および第2の活性物質を含むポリマーマトリックスを含む粒子であって、該ポリマーマトリックスが、第1および第2の活性物質の各々が少なくとも2時間の対象の循環中での半減期を有するように形成されている、粒子。
- ポリマーマトリックスが1種またはそれ以上のさらなる活性物質をさらに含む、請求項3に記載の粒子。
- 第1の活性物質が生物分解性ポリマーに結合している、請求項1ないし請求項4のいずれかに記載の粒子。
- 第1の活性物質がペンダント官能基を有する生物分解性ポリマーに結合している、請求項1ないし請求項4のいずれかに記載の粒子。
- 第2の活性物質が生物分解性ポリマーに結合している、請求項6または請求項7に記載の粒子。
- 第2の活性物質がペンダント官能基を有する生物分解性ポリマーに結合している、請求項6または請求項7に記載の粒子。
- 複数の活性物質を有する粒子であって、
(a)
(i)生物分解性ポリマーに結合した第1の活性物質、および
(ii)第2の活性物質
を含む、疎水性ポリマーコア、および、
(b)表面に露出した親水層
を含む粒子。 - 第1の活性物質が、生物分解性ポリマーに、ポリマー上のペンダント官能基を介して結合している、請求項9に記載の粒子。
- 生物分解性ポリマーが、比較的疎水性の第1末端および比較的親水性の第2末端を有するブロックコポリマーであり、疎水性コアが、第1の活性物質に結合したブロックコポリマーの第1末端を含み、親水層が、ブロックコポリマーの第2末端を含む、請求項9または請求項10に記載の粒子。
- 親水層が、疎水性ポリマーコアと相互作用する相対的に疎水性の末端および表面に露出した相対的に親水性の末端を各々含む複数の両親媒性ブロックコポリマーを含む、請求項9または請求項10に記載の粒子。
- 標的化物質をさらに含む、請求項1ないし請求項12のいずれかに記載の粒子。
- 第1の活性物質および第2の活性物質が、生体分子、生体活性物質、低分子、薬物、プロドラッグ、薬物誘導体、タンパク質、ペプチド、ワクチン、アジュバント、蛍光分子またはポリヌクレオチドから独立して選択される、請求項1ないし請求項13のいずれかに記載の粒子。
- 第1および第2の活性物質が、各々、パクリタキセルまたはドセタキセルおよびゲフィチニブ;ゲフィチニブおよびパクリタキセルまたはドセタキセル;オキサリプラチン(またはオキサリプラチンプロドラッグ)およびイリノテカン;イリノテカンおよびオキサリプラチン(またはオキサリプラチンプロドラッグ);パクリタキセルおよびツバシン;ツバシンおよびパクリタキセル;ロニダミン、ジクロロ酢酸、アルファ−コハク酸トコフェロール、ベツリン酸またはレスベラトロールおよびPt(IV)ヘキサノエート;Pt(IV)ヘキサノエートおよびロニダミン、ジクロロ酢酸、アルファ−コハク酸トコフェロール、ベツリン酸またはレスベラトロール;アルファ−コハク酸トコフェロールまたはジャスモン酸メチルおよびドセタキセル;または、ドセタキセルおよびアルファ−コハク酸トコフェロールまたはジャスモン酸メチルである、請求項1ないし請求項14のいずれかに記載の粒子。
- 少なくとも2種の活性物質を含む粒子の製造方法であって、
第1および第2の活性物質を提供すること;
第1の活性物質またはそのプロドラッグもしくは誘導体を、ペンダント官能基を有する生物分解性ポリマーに結合させること;および、
結合した第1の活性物質および第2の活性物質を含む粒子を製造すること、
を含む方法。 - 第2の活性物質またはそのプロドラッグもしくは誘導体を、ペンダント官能基を有する生物分解性ポリマーに結合させることをさらに含む、請求項16に記載の方法。
- 第1の活性物質および/または第2の活性物質をペンダント官能基を有する生物分解性ポリマーに結合させることが、第1および第2の活性物質の粒子形成への適合性を与える、請求項16に記載の方法。
- 2種またはそれ以上の活性物質を時間空間的に制御して対象に投与する方法であって、
第1の活性物質および第2の活性物質を含むポリマーマトリックスを含む粒子を提供すること、ここで、ポリマーマトリックスは、第1および第2の活性物質の各々の所望の時間空間的放出速度を提供するように形成されている;および、
第1および第2の活性物質が所望の時間空間的放出速度で粒子から放出されるように、粒子を対象に投与すること、
を含む方法。 - 請求項1ないし請求項15のいずれかに記載の複数の粒子を含む、静脈内に、動脈内に、経口で、経皮で、経粘膜で、腹腔内に、頭蓋内に、眼内に、硬膜外に、くも膜下腔内に、局所に、浣腸により、注射により、肺経路により、または、点滴により送達するための医薬組成物。
