Description
１．サンプル調製
診断用のポリペプチドマーカーを検出するために、尿を使用した。健康なドナー（対照群）および血管疾患に罹患している患者から尿を採取した。その後のＣＥ−ＭＳ測定のために、限外ろ過によって、患者の尿に高濃度で含まれるタンパク質（アルブミンおよび免疫グロブリンなど）を分離する必要があった。そこで、７００μＬの尿を採取し、７００μｌのろ過バッファー（２Ｍ尿素、１０ｍＭアンモニア、０．０２％ＳＤＳ）と混合した。この体積１．４ｍＬのサンプルを限外ろ過した（２０ｋＤａ、ザルトリウス社、ドイツ、ゲッティンゲン）。１．１ｍＬの限外ろ液が得られるまで、遠心機を用いて３０００ｒｐｍで限外ろ過を行った。
