Description
HLA-G5タンパク質のアイソフォームは、α1、α2及びα3ドメイン並びに(転写産物のスプライシング及びRNA成熟化の後にイントロン4を保持する結果として)イントロン4によりコードされる21アミノ酸残基の余剰なC末端ペプチド配列を含む。
よって、ILT2及びILT4は、古典的なHLAクラスI分子よりもHLA-Gに強く結合する([20,21]を参照されたい)。
1. その他のHLAクラスI分子よりも良いILT結合能力を付与するそのα3ドメインのユニークな配列。HLA-Gのα3ドメインのこのユニークな配列は、Shiroishiら[21]の図3から理解されるように、より大きく、より疎水性で、かつより強いILT結合領域の創造をもたらす。
2. 二量体化するそのユニークな能力。HLA-Gの二量体化は、2つのHLA-G分子の42位(α1ドメイン)のシステイン間のジスルフィドブリッジの創造を介して起こり(Gonen-Grossら[24]の図8を参照されたい)、HLA-Gの機能に決定的である。事実、42位のシステインを欠き、二量体を形成しない変異型HLA-G分子は、抑制機能も欠くことが示された[24]。Shiroishiら[20]の図4は、ジスルフィドで結合したHLA-G二量体の構造を提供する。
以下の多量体もまた、本発明に含まれる:
全ての単量体が同一である、α3単量体(n個のP1単量体)のホモ多量体;
α3単量体のヘテロ多量体(n個のP1単量体、ここで種々の単量体のα3ドメインが異なる)、nが2〜1000の間又はさらに多くを含む整数である。
‐ペプチドP1はまた、X2のC末端及び/又はN末端部に、20未満、より好ましくは15未満及び最も好ましくは10未満又は5未満の、天然のHLA-Gアイソフォームのα3ドメインの側部にあるさらなるアミノ酸を含んでいてもよい。
‐HLA-Gのα1ドメインは、エキソン2によりコードされ、配列番号6のヒトHLA-Gのアミノ酸25-114に相当する。
‐「機能的フラグメント」は、インビボにおける移植片免疫寛容及び/又はインビトロにおけるアロ増殖の阻害を誘導する能力を保持するフラグメントを指す。より好ましくは、α3ペプチドのいずれかの機能的フラグメントは、α3ドメインの少なくとも20、より好ましくは少なくとも30、少なくとも40又は少なくとも50の連続アミノ酸を含む。
α1ペプチドは、成熟HLA-G抗原のアミノ酸1-90又はその機能的フラグメントから本質的になる。
好ましいα3ペプチドの配列は、配列番号1に提供される。
α1ペプチドの配列は、配列番号2に提供される。
本発明の好ましい実施態様においては、上記に定義されるα3単量体は、共有結合的に又は親和性相互作用を介して結合している。
本発明の多量体の具体的な例は、P1-P1二量体である。
本発明の多量体は、様々な技術によって製造することができる。上記に論じたように、単量体は、種々のカップリング技術、例えば共有結合(例えばジスルフィドブリッジ、二官能基など)又は親和性反応によりカップリングされ得る。
第1の工程において、(ペプチドリンカーX1により提供される)SH側基(lateral SH group)を含む2つのα3単量体P1を、溶液中で、ジスルフィド結合の形成を許容する条件下に接触させる;第2の工程において、好ましくは、二量体又は多量体を分離する。多量体は、例えばその分子量に基づいて、例えば(PAGEのような)ゲル電気泳動により単量体から分離してもよい。適切な多量体の形成はまた、アリコートのサンプルについてこのような方法を用いて、溶液中に存在する多量体の相対量を測定するために確かめてもよく、必要であれば、反応条件を調整してもよい。ジスルフィド結合の形成を許容する条件は、例えば2〜24時間10〜30℃の温度を含む。
α3多量体は、天然アミノ酸又は非天然若しくは改変アミノ酸残基のいずれかを含んでいてもよい。それらは、L及び/又はDのコンホメーションにあり得る。ペプチドは、アミン結合及び/又は改変ペプチド模倣結合のいずれかを含んでいてもよい。また、ペプチドは、例えば側基(lateral group)の化学的又は物理的改変により、末端部を保護及び/又は改変されていてもよい。
- ジスルフィドブリッジを介して結合した配列番号3のα3単量体の多量体;
- マイクロビーズのような担体に結合した配列番号3のα3単量体の多量体
である。
さらに、配列番号3のα3単量体の二量体はまた、本発明の具体物の代表例でもある。本発明は、事実、二量体が移植片拒絶を治療するための相当なインビボ活性を有し、非常に活性な多量体を調製するのに用いられ得ることを示す。
本発明のさらなる物は、配列番号3の2つの単量体と、好ましくは、少なくとも1つの医薬的に許容されるビヒクル又は担体とを有するα3二量体を含む医薬組成物である。
本発明はまた、移植片拒絶を治療するための上記に定義される多量体又は組成物に関する。
配列番号1〜5のペプチドは、ペプチドシンセサイザーを用いて合成した。
種々の二量体を産生した:
- (α3-L1)x2 (配列番号3)、ここで、L1は、本発明による、すなわち主にグリシン及びセリンアミノ酸残基を含む、可撓性のペプチドリンカーに相当する;
- (α3-L2)x2 (配列番号5)、ここで、L2は、前記配列番号5のアミノ酸1-18に相当し、HLA-G(配列番号6を参照されたい)のα1アミノ酸残基42-30(N末端からC末端まで)に由来する;
- α1-α3単量体に相当する配列番号4に相当する(α1-α3)x2。
図6、7及び8は、合成されたペプチドの概括的な構造を示す。
1) α3二量体
配列番号3又は配列番号5のα3単量体を化学的に合成した。単量体をまず合成し、次いでα3ドメインの2つのシステイン(配列番号3のシステイン35及び91、配列番号5のシステイン41及び97)間にジスルフィド結合を生成させることによってリフォールドした。次いで二量体化を、2つの単量体のリンカーX1のシステイン(配列番号3のシステイン2、配列番号5のシステイン1)間にジスルフィドブリッジを生成させることによって行った。合成された生成物の純度を質量分析により確かめた。
配列番号4のα1-α3単量体を化学的に合成した。単量体をまず合成し、次いでα3ドメインの2つのシステイン(配列番号4のシステイン111及び167)間にジスルフィド結合を生成させることによってリフォールドした。次いで二量体化を、2つの単量体のα1ドメインの2つのシステイン(配列番号4のシステイン42)間にジスルフィドブリッジを生成させることによって行った。合成された生成物の純度を質量分析により確かめた。
α1+α3ポリペプチドの配列を配列番号4に示す。
HLA-G受容体ILT2、ILT4及びKIR2DL4への結合を評価するために、実施例2に従って得られた12μgの二量体で、製造者の推奨に従って、Bio-Plex-COOHポリスチレンビーズ(Bio-Rad)を共有結合的にコートした。ビーズを次いで1x Luminexアッセイ緩衝液(Interchim)に、50μl当たり2000ビーズの濃度に再懸濁した。ヒトIgG(ILT2-Fc、ILT4-Fc、R&D Biosystems)のFc部位に融合した組換え受容体を、次いで2μg/mlで加えた。200μlの1x PBS, 0.05% Tweenで2回洗浄した後、ビーズ及び受容体を次いで90分間暗中にて振盪機でインキュベートした。ビーズを次いで2μg/mlのフィコエリスリン標識ヤギ抗ヒトIgG抗体(Sigma)を含む50μlのPBS Luminexアッセイ緩衝液に、30分間暗中にて回転振盪機で再懸濁した。ビーズを次いで200μlの1x PBS, 0.05% Tweenで2回洗浄し、300μlの1xPBSに再懸濁した。
図3は結果を示しており、α3ドメインを含む全てのペプチドが実際にILT4受容体に特異的に結合することを明確に示す。
実施例1の合成(α3-L1)x2二量体の細胞毒性を、200μlウェル中に100,000細胞/mlにて新しく単離された末梢血単核球(PBMC)について、0〜100μg/mlまでの二量体の投与量及び37℃にて0〜24時間の範囲のインキュベーション時間で評価した。毒性は、死細胞におけるトリパンブルー取込み及び生細胞 対 死細胞の比の計算を用いて細胞生存能を分析することにより評価した。
結果(図4)は、前記二量体がPBMCに対して毒性でないことを示す。
健常なボランティアドナーのヘパリン全血からの末梢血単核球(PBMC)を、Ficoll-histopaque 1077 (Sigma-Aldrich)を用いる濃度勾配遠心分離により得た。PBMCを、レスポンダー又はγ-照射刺激細胞(25 Gy)として用いた。アロ刺激アッセイにおいて合成ペプチドの機能を評価するために、100μl中の105のレスポンダー細胞を100μg/mlの濃度のペプチドと共に6時間インキュベートし、次いで100μl中の105のγ-照射同種異系刺激PBMC(25Gy)と混合し、ウェル当たり200μlの最終体積中の50μg/mlの最終ペプチド濃度を得た。全てのサンプルについて3連(in triplicate)で行い、各々の同種異系の組合せについては、レスポンダー細胞単独、照射刺激細胞単独、自己コントロール及び未処理コントロールを含めた。給湿したインキュベーター中で37℃及び5%CO2にて5日間インキュベーション後、培養物をトリチウム化チミジン(ウェル当たり1μCi、Amersham Biosciences)でパルスした。DNAへの3Hチミジンの組込みを、18時間後にβ-カウンター(Wallac 1450, Amersham Biosciences)で定量した。

Claims (9)
- 少なくとも2つの単量体を含み、各々の単量体が式X1-X2(式中X1はシステインアミノ酸を含む可撓性のペプチドリンカーを表し、X2はHLA-Gのα3ドメインを表す)のペプチドP1からなる群において選択されることを特徴とする多量体。
- ペプチドP1が、X2のC末端及び/又はN末端部に20未満、より好ましくは15未満、最も好ましくは10未満又は5未満のさらなるアミノ酸を含み、該アミノ酸が天然HLA-Gアイソフォームのα3ドメインの側部にあることを特徴とする請求項1に記載の多量体。
- 前記可撓性のペプチドリンカーX1が、少なくとも10〜30アミノ酸であって最大100のアミノ酸を含み、システインをそのN末端部に、好ましくはN末端から1位、2位、3位又は4位に含むことを特徴とする請求項1又は2に記載の多量体。
- 前記可撓性のペプチドリンカーが、グリシン及びセリンアミノ酸残基を本質的に含むことを特徴とする請求項1〜3のいずれか1項に記載の多量体。
- α3単量体が配列番号3に与えられていることを特徴とする請求項1〜4のいずれか1項に記載の多量体。
- リンカーX1のN末端部に存在するシステイン残基間のジスルフィドブリッジを介して結合する配列番号3の2つの単量体を有する、α3二量体の形態にあることを特徴とする請求項1〜5のいずれか1項に記載の多量体。
- 請求項1〜6のいずれか1項に記載の多量体と、少なくとも1つの医薬的に許容されるビヒクル又は担体とを含む医薬組成物。
- 移植片拒絶を治療するための、請求項1〜6のいずれか1項に記載の多量体又は請求項7に記載の医薬組成物。
- 炎症性疾患又は自己免疫疾患を治療するための、請求項1〜6のいずれか1項に記載の多量体又は請求項7に記載の医薬組成物。
