Abstract

本出願は、2008年8月19日に出願された米国仮特許出願第61/089,972号の恩典を主張し、その内容の全体は参照により本明細書に組み入れられる。
本発明は、生物科学の分野、より具体的にはがん治療の分野に関連する。特に本発明は、がんワクチンとして極めて有効な新規ペプチド、ならびに腫瘍を治療および予防するための薬物に関連する。

周知の技法を用いて、本発明のペプチドを調製することができる。例えば、組換えDNA技術または化学合成を用いて、ペプチドを合成的に調製することができる。本発明のペプチドは、個々に、または2つもしくはそれ以上のペプチドから構成されるより長いポリペプチドとして、合成することができる。その後ペプチドを単離すること、すなわち他の天然の宿主細胞タンパク質およびそれらの断片、または他の任意の化学物質を実質的に含まないように、精製または単離することができる。

本発明はまた、前述の本発明のペプチドをコードするポリヌクレオチドを提供する。これらは、天然のHIG2もしくはURLC10遺伝子(SEQ ID NO:3または5、GenBankアクセッション番号NM_013332またはNM_017527)由来のポリヌクレオチド、およびその保存的に改変されたヌクレオチド配列を有するポリヌクレオチドを含む。本明細書において「保存的に改変されたヌクレオチド配列」という語句は、同一のまたは本質的に同一のアミノ酸配列をコードする配列を指す。遺伝暗号の縮重のため、数多くの機能的に同一な核酸が任意の特定のタンパク質をコードする。例えば、コドンGCA、GCC、GCG、およびGCUはすべて、アミノ酸のアラニンをコードする。従って、あるコドンによってアラニンが指定されるあらゆる位置において、コードされるポリペプチドを変化させることなく、該コドンを記載された対応するコドンのいずれかに変更することができる。このような核酸の変異は「サイレント変異」であり、保存的に改変された変異の一種である。ペプチドをコードする本明細書中のあらゆる核酸配列は、該核酸の可能性のあるあらゆるサイレント変異をも表す。核酸中の各コドン(通常メチオニンに対する唯一のコドンであるAUG、および通常トリプトファンに対する唯一のコドンであるTGGを除く)を改変して、機能的に同一な分子を得ることができることを、当業者は認識するであろう。従って、ペプチドをコードする核酸の各サイレント変異は、公開した各配列において非明示的に記載されている。
本発明は、本発明のペプチドとHLA抗原との間で形成された複合体を自身の表面上に提示する、エキソソームと称される細胞内小胞をさらに提供する。エキソソームは、例えば公表特許公報 特表平11−510507号およびWO99/03499に詳述されている方法を用いることによって調製することができ、治療および/または予防の対象となる患者から得られたAPCを用いて調製することができる。本発明のエキソソームは、本発明のペプチドと同様の様式で、ワクチンとして接種することができる。
本発明はまた、HLA―A0206抗原と本発明のペプチドとの間に形成される複合体を自身の表面上に提示する単離されたAPCを提供する。本発明のペプチドを接触させることによって、または本発明のペプチドをコードするヌクレオチドを発現可能な形態で導入することによって得られるAPCは、治療および/または予防の対象となる患者に由来してよく、かつ、単独で、または本発明のペプチド、エキソソーム、もしくは細胞傷害性T細胞を含む他の薬物と組み合わせて、ワクチンとして投与することができる。

本発明のペプチドのいずれかに対して誘導された細胞傷害性T細胞は、インビボで腫瘍関連内皮を標的とする免疫応答を増強させるため、ペプチド自体と同様の様式でワクチンとして用いることができる。従って本発明はまた、本ペプチドのいずれかよって特異的に誘導または活性化された、単離された細胞傷害性T細胞を提供する。
本発明はまた、T細胞受容体(TCR)のサブユニットを形成し得るポリペプチドをコードする核酸配列から構成されるポリヌクレオチド、およびそれを用いる方法を提供する。TCRサブユニットは、HLA−A0206抗原を伴ってHIG2ペプチドまたはURLC10ペプチドを提示する腫瘍細胞に対する特異性をT細胞に付与するTCRを形成する能力を有する。当技術分野における公知の方法を用いることによって、本発明のペプチドで誘導されたCTLにおいて発現されるTCRのα鎖およびβ鎖の核酸配列を同定することができる(WO2007/032255、およびMorgan et al., J Immunol, 171, 3288 (2003))。TCR誘導体は、標的細胞上に提示されるHIG2またはURLC10ペプチドと高い結合力で結合することができ、かつ任意で、HLA−A0206抗原を伴ってHIG2またはURLC10ペプチドを提示する標的細胞の効率的な殺傷をインビボおよびインビトロで媒介することができる。
「予防(preventionおよびprophylaxis)」という用語は、本明細書で互換的に用いられ、疾患による死亡率または罹患率の負荷を軽減させる任意の行為を指す。予防(preventionおよびprophylaxis)は、「第一次、第二次、および第三次の予防レベルで」行われ得る。第一次の予防(preventionおよびprophylaxis)は、疾患の発生を回避するのに対し、第二次および第三次レベルの予防(preventionおよびprophylaxis)は、疾患の進行および症状の出現を予防(preventionおよびprophylaxis)することに加え、機能を回復させ、かつ疾患関連の合併症を減少させることによって、既存の疾患の悪影響を低下させることを目的とした行為を包含する。あるいは、予防(preventionおよびprophylaxis)は、特定の障害の重症度を軽減すること、例えば腫瘍の増殖および転移を減少させることを目的とした広範囲の予防的治療を含み得る。

本発明のペプチドは、薬学的な剤もしくは組成物として直接投与することができ、または必要であれば、従来の製剤方法によって製剤化される。後者の場合、本発明のペプチドに加えて、薬物に通常用いられる担体、賦形剤等が特に制限なく適宜含まれ得る。そのような担体の例は、滅菌水、生理食塩水、リン酸緩衝液、培養液等である。さらに、薬学的な剤または組成物は、必要に応じて、安定剤、懸濁液、保存剤、界面活性剤等を含み得る。本発明の薬学的な剤または組成物は、抗がん目的に用いることができる。

細胞株
PSCCA0922(HLA−A0206)は、Pharma SNP Consortium;PSCから購入した。ヒトB−リンパ芽球様細胞株、およびCOS7は、ATCCから購入した。
HIG2またはURLC10由来の9−merおよび10−merのペプチドを、標準的な固相合成法に従ってSigma(札幌、日本)またはBiosynthesis Inc.(Lewisville,TX)によって合成し、逆相高速液体クロマトグラフィー(HPLC)によって精製した。該ペプチドの純度(>90%)および同一性を、それぞれ分析用HPLCおよび質量分析によって測定した。ペプチドをジメチルスルホキシド(DMSO)に20 mg/mlで溶解し、−80℃で保存した。
単球由来の樹状細胞(DC)を抗原提示細胞(APC)として用いて、ヒト白血球抗原(HLA)上に提示されたペプチドに対する細胞傷害性Tリンパ球(CTL)応答を誘導した。他所に記載されているように、DCをインビトロで作製した(Nakahara S et al., Cancer Res 2003 Jul 15, 63(14): 4112-8)。具体的には、Ficoll−Plaque(Pharmacia)溶液によって健常なボランティア(HLA−A0206陽性)から単離した末梢血単核細胞(PBMC)を、プラスチック製の組織培養ディッシュ(Becton Dickinson)へ付着させることによって分離し、それらを単球画分として濃縮した。2%の加熱非働化した自己血清(AS)を含むAIM−V培地(Invitrogen)中で、1000 U/mlの顆粒球マクロファージコロニー刺激因子(GM−CSF)(R&D System)および1000 U/mlのインターロイキン(IL)−4(R&D System)の存在下で、単球が濃縮した集団を培養した。培養7日後、サイトカインで誘導したDCに、AIM−V培地中で3時間、37℃にて、3 マイクログラム/mlのβ2−ミクログロブリンの存在下で20 マイクログラム/mlの各合成ペプチドをパルスした。作製した細胞は、その細胞表面上に、CD80、CD83、CD86、およびHLAクラスIIなどのDC関連分子を発現しているようであった(データは示さず)。次いで、ペプチドパルスしたこれらのDCを、マイトマイシンC(MMC)で不活化し(30 マイクログラム/mlで30分間)、CD8 Positive Isolation Kit(Dynal)を用いた陽性選択によって得られた自己CD8+ T細胞と1:20の比率で混合した。これらの培養物を48ウェルプレート(Corning)中に準備し;各ウェルは、0.5 mlのAIM−V/2% AS培地中に、1.5×104個のペプチドパルスしたDC、3×105個のCD8+ T細胞、および10 ng/mlのIL−7(R&D System)を含んだ。3日後、これらの培養物に、IL−2(CHIRON)を最終濃度20 IU/mlまで補充した。7日目および14日目に、ペプチドパルスした自己DCでT細胞をさらに刺激した。該DCは上記と同じ方法によって毎回調製した。21日目に、3回目のペプチド刺激後、ペプチドパルスしたPSCCA0922細胞に対してCTLを試験した(Tanaka H et al., Br J Cancer 2001 Jan 5, 84(1): 94-9; Umano Y et al., Br J Cancer 2001 Apr 20, 84(8): 1052-7; Uchida N et al., Clin Cancer Res 2004 Dec 15, 10(24): 8577-86; Suda T et al., Cancer Sci 2006 May, 97(5): 411-9; Watanabe T et al., Cancer Sci 2005 Aug, 96(8): 498-506)。
Riddellら(Walter EA et al., N Engl J Med 1995 Oct 19, 333(16): 1038-44;Riddell SR et al., Nat Med 1996 Feb, 2(2): 216-23)によって記載されている方法と類似の方法を用いて、CTLを培養下で増殖させた。40 ng/mlの抗CD3モノクローナル抗体(Pharmingen)の存在下で、MMCによって不活化した2種類のヒトBリンパ芽球様細胞株と共に、合計5×104個のCTLを25 mlのAIM−V/5% AS培地中に懸濁した。培養開始1日後に、120 IU/mlのIL−2を該培養物に添加した。5、8、および11日目に、30 IU/mlのIL−2を含む新たなAIM−V/5% AS培地を、該培養物に供給した(Tanaka H et al., Br J Cancer 2001 Jan 5, 84(1): 94-9;Umano Y et al., Br J Cancer 2001 Apr 20, 84(8): 1052-7;Uchida N et al., Clin Cancer Res 2004 Dec 15, 10(24): 8577-86;Suda T et al., Cancer Sci 2006 May, 97(5): 411-9;Watanabe T et al., Cancer Sci 2005 Aug, 96(8): 498-506)。
96丸底マイクロタイタープレート(Nalge Nunc International)において0.3個、1個、および3個のCTL/ウェルとなるように、希釈を行った。CTLを、7×104個細胞/ウェルの2種類のヒトBリンパ芽球様細胞株、30 ng/mlの抗CD3抗体、および125 U/mlのIL−2と共に、合計150μl/ウェルの5%AS含有AIM−V中で培養した。10日後、50μl/ウェルのIL−2を、最終濃度でIL−2が125 U/mlに到達するように培地に添加した。14日目にCTLのCTL活性を試験し、上記と同じ方法を用いてCTLクローンを増殖させた。
特異的CTL活性を調べるために、インターフェロン(IFN)−γ酵素結合免疫スポット(ELISPOT)アッセイおよびIFN−γ酵素結合免疫吸着アッセイ(ELISA)を行った。具体的には、ペプチドパルスしたPSCCA0922(1×104個/ウェル)を刺激細胞として調製した。48ウェル中の培養細胞を応答細胞として使用した。IFN−γ ELISPOTアッセイおよびIFN−γ ELISAアッセイは、製造元の手順に従って行った。
標的遺伝子(HIG2;SEQ ID NO: 3およびURLC10;SEQ ID NO: 5)またはHLA−A0206(SEQ ID NO: 7)のオープンリーディングフレームをコードするcDNAをPCRによって増幅した。PCR増幅産物をpcDNA3.1 myc−Hisベクター(Invitrogen)にクローニングした。製造業者の推奨する手順に従ってリポフェクタミン(Invitrogen)を用いて、標的遺伝子およびHLA−A0206のいずれか一方または両方を含むプラスミドをCOS7にトランスフェクトした。簡潔に説明すると、2.5×106個のCOS7細胞に、140 Vおよび1000マイクロFで10マイクログラムのプラスミドをパルスした。トランスフェクションから2日後に、トランスフェクトした細胞を細胞解離溶液で処理し、CTL活性アッセイの標的細胞として使用した。
がんにおけるHIG2およびURLC10発現の増強
cDNAマイクロアレイを用いて様々ながんから得られた包括的遺伝子発現プロファイルデータから、HIG2(GenBankアクセッション番号NM_013332;SEQ ID NO: 3)およびURLC10(GenBankアクセッション番号NM_017527;SEQ ID NO: 5)の発現が上昇していることが明らかになった。HIG2発現は、対応する正常組織と比較して、腎癌20例中19例、および軟部組織腫瘍9例中7例において確かに上昇していた。URLC10発現は、対応する正常組織と比較して、膀胱癌29例中29例、子宮頸癌16例中15例、胆管細胞癌7例中7例、食道癌19例中7例、胃癌3例中3例、NSCLC 27例中24例、骨肉腫19例中15例、膵癌5例中4例、および軟部組織腫瘍43例中33例において確かに上昇していた。
HIG2由来のペプチドに対するCTLを、「材料および方法」に記載したプロトコールに従って作製した。IFN−γ ELISPOTアッセイによって、ペプチド特異的なCTL活性を測定した(図1)。本明細書における結果から、HIG2−A0206−9−4(SEQ ID NO: 1)が、対照ウェルと比較して強力なIFN−γ産生を示すことが示される。さらに、SEQ ID NO: 1で刺激した陽性ウェル番号1、2、5、7、8、10、13、および14中の細胞を増殖させて、CTL株を樹立した。それらのCTL株のCTL活性を、IFN−γ ELISAアッセイによって測定した(図2)。本明細書における結果から、すべてのCTL株が、ペプチドをパルスしなかった標的細胞と比較して、対応するペプチドをパルスした標的細胞に対して強力なIFN−γ産生を示すことが示される。したがって、HIG2−A0206−9−4は、HLA−A0206を発現する標的細胞に対する強力なCTL株を誘導し得る。
上記の「材料および方法」の項に記載のプロトコールに従って、これらのCTL株から限界希釈を行った。HIG2−A0206−9−4(SEQ ID NO: 1)CTL株からのCTLクローンの樹立を図3に示す。これらのCTLクローンは、ペプチドをパルスしなかった標的に対する活性と比較して、ペプチドをパルスした標的に対して強力かつ特異的なCTL活性を有した。
HIG2−A0206−9−4(SEQ ID NO: 1)に対して産生された樹立CTLクローンを、HIG2およびHLA−A0206を発現する標的細胞を認識する能力について、試験した。HIG2およびHLA−A0206を内因的に発現する標的細胞の特異的モデルとして役立つ、全長HIG2遺伝子およびHLA−A0206分子の両方をトランスフェクトしたCOS7に対する特異的CTL活性を、HIG2−A0206−9−4(SEQ ID NO: 1)によって産生されたCTLクローンをエフェクター細胞として用いて試験した。全長HIG2をトランスフェクトしたがHLA−A0206をトランスフェクトせず、他のペプチド(HIG2−9−8:YLLGVVLTL)をパルスしたCOS7、およびHLA−A0206をトランスフェクトしたが全長HIG2をトランスフェクトしなかったCOS7を対照として調製した。COS7に対して最も高い特異的CTL活性を示すCTLクローンは、HIG2およびHLA−A0206の両方をトランスフェクトしたものであった(図4)。
URLC10由来のペプチドに対するCTLを、「材料および方法」に記載したプロトコールに従って作製した。IFN−γ ELISPOTアッセイによって、ペプチド特異的なCTL活性を測定した(図5)。本明細書における結果から、URLC10−A0206−10−211(SEQ ID NO: 2)が、対照ウェルと比較して強力なIFN−γ産生を示すことが示される。さらに、SEQ ID NO: 2で刺激した陽性ウェル番号7中の細胞を増殖させ、CTL株を樹立した。CTL株のCTL活性を、IFN−γ ELISAアッセイによって測定した(図6)。本明細書における結果から、CTL株が、ペプチドをパルスしなかった標的細胞と比較して、対応するペプチドをパルスした標的細胞に対して強力なIFN−γ産生を示すことが示される。したがって、URLC10−A0206−10−211は強力なCTL株を誘導し得る。
上記の「材料および方法」の項に記載のプロトコールに従って、これらのCTL株から限界希釈を行った。URLC10−A0206−10−211(SEQ ID NO: 2)CTL株からのCTLクローンの樹立を図7に示す。このCTLクローンは、ペプチドをパルスしなかった標的に対する活性と比較して、ペプチドをパルスした標的に対して強力かつ特異的なCTL活性を有する。
URLC10−A0206−10−211(SEQ ID NO: 2)に対して産生された樹立CTLクローンを、URLC10およびHLA−A0206を発現する標的細胞を認識する能力について、試験した。URLC10およびHLA−A0206を内因的に発現する標的細胞の特異的モデルとして役立つ、全長URLC10遺伝子およびHLA−A0206分子の両方をトランスフェクトしたCOS7に対する特異的CTL活性を、URLC10−A0206−10−211(SEQ ID NO: 2)によって産生されたCTLクローンをエフェクター細胞として用いて試験した。全長URLC10をトランスフェクトしたがHLA−A0206をトランスフェクトしなかったCOS7、およびHLA−A0206をトランスフェクトしたが全長URLC10をトランスフェクトしなかったCOS7を対照として調製した。COS7に対して最も高い特異的CTL活性を示すCTLクローンは、URLC10およびHLA−A0206の両方をトランスフェクトしたものであった(図8)。
本発明は、強力かつ特異的な抗腫瘍免疫応答を誘導し、幅広いがん型に対する適用性を有する新規TAA、詳細にはHIG2またはURLC10由来の新規TAAについて記載する。このようなTAAは、HIG2またはURLC10に関連した疾患、例えば膀胱癌、子宮頸癌、胆管細胞癌、食道癌、胃癌、非小細胞肺癌(NSCLC)、骨肉腫、膵癌、腎癌、および軟部組織腫瘍などのがんに対するペプチドワクチンとしてのさらなる発展を保証する。

- HLA−A0206抗原と、SEQ ID NO: 1および2からなる群より選択されるアミノ酸配列を含むペプチド、または少なくとも1つのアミノ酸が付加、挿入、欠失、および/もしくは別のアミノ酸と置換されている当該ペプチドとの複合体を認識する、単離されたCTL。
- 高いCTL誘導能を有する抗原提示細胞を誘導するための剤であって、
(a) CTL誘導能を有し、
(i) SEQ ID NO: 1および2;ならびに
(ii) 1個、2個、または数個のアミノ酸が置換、挿入、欠失、および/または付加されているSEQ ID NO: 1および2
からなる群より選択されるアミノ酸配列を含む1つまたは複数のペプチド;
(b) 当該ペプチドをコードする1つまたは複数のポリヌクレオチド
を含む、剤。 - HLA抗原がHLA−A0206である対象においてがんに対する免疫応答を誘導する方法であって、CTL誘導能ならびにSEQ ID NO: 1および2からなる群より選択されるアミノ酸配列を有するペプチド、少なくとも1つのアミノ酸が付加、挿入、欠失、および/もしくは別のアミノ酸と置換されている当該ペプチド、前述のペプチドをコードするポリヌクレオチド、請求項6記載の単離された抗原提示細胞、または請求項7記載の単離されたCTLを含む剤を該対象に投与する段階を含む、方法。
- がんが、膀胱癌、子宮頸癌、胆管細胞癌、食道癌、胃癌、NSCLC、骨肉腫、膵癌、腎癌、および軟部組織腫瘍からなる群より選択される、請求項9記載の方法。
- HLA−A抗原がHLA−A0206である対象においてがんに対する免疫応答を誘導するための剤であって、薬理学的に許容される担体と組み合わされた、
(a) CTL誘導能を有し、
(i) SEQ ID NO: 1および2;ならびに
(ii) 1個、2個、もしくは数個のアミノ酸が置換、挿入、欠失、および/もしくは付加されているSEQ ID NO: 1および2
からなる群より選択されるアミノ酸配列を含む1つもしくは複数のペプチド;
(b) 当該ペプチドをコードする1つもしくは複数のポリヌクレオチド;
(c) 請求項6記載の1つもしくは複数の単離された抗原提示細胞;
(d) 請求項7記載の1つもしくは複数の単離されたCTL;または
(e) これらの組み合わせ
を含む、剤。
