水安定化活性一時的コーティング材料
酵素の調製
酵素のPEG化
精製した酵素(α−アミラーゼ、サーモリシン、又はリパーゼ)をスクシンイミジルエステルにより誘導化されたPEG(単官能性直鎖PEG10000、PEG12000、PEG20000、又は組み合わせ)と1:5の酵素:PEGのモル比で0.05Mリン酸ナトリウム緩衝液pH7.5中において混合し、穏やかに撹拌しながら室温において200rpmで60分間振盪する。未反応PEGを含む沈殿を適当な分子量カットオフのフィルターでろ過して除く。
例1の市販ワックス1−7及び濃厚なPEG化酵素(未反応PEGを除去しないでPEG化前に200mg/ml)を1000rpmで4:1〜12:1のベース:酵素の重量比で激しく撹拌し、冷1のベース組成物を用いて水安定化一時的コーティング材料調製液を調製する。調製液A〜KKKを表1に示す。
コートしたアルミニウムパネルを1.2cm×1.9cmのテストサイズに切り、室温においてDI水に0分、30分、又は90分撹拌しながら浸すことにより耐候試験を行う。テストパネルを取り出し、DI水で20秒洗浄し、次いで残っている酵素活性を調べる。
この調製にあたって、60gの凍結乾燥したコオロギをブレンダー(Oster、600ワット)により10分間粉砕することによって粉末にする。2gのコオロギ粉末を6mLのDI水と激しく混合することにより汚染溶液を調製する。均一な間隔のテンプレートを用いてコーティング表面上に汚染を適用する。このコオロギの汚染を40℃で5分間乾燥させる。各テストパネルをガラス皿に載せ、RTにおいて300rpmで振盪させながら200mLのDI水で数回洗浄する。汚染除去の時間を記録する。誤差を小さくするため、最初と最後の時間を含めない。8つの汚染スポットの平均洗浄時間を汚染除去時間として平均する。PEG化サーモリシン含有コーティングでコートしたテストパネルは、ベース材料のみでコートしたパネルと比較して、穏やかな洗浄による改良された汚染除去を示す。
