本発明のさらなる目的は、上記遺伝子を含む組み換えベクターを提供する。本発明のさらなる目的は、上記組み換えベクターを用いて得られる形質転換体を提供する。
本発明のさらに別の目的は、抗CPAA抗体および/または抗CPAAペプチドを用いた、CPAAの存在と関連する動物細胞、組織または病理の試験管内(インビトロ)、その場(インシトゥー)および/または生体内(インビボ)診断および/または処置に有用性のある方法を提供する。本発明はまた、CPAA関連の病的状態を診断および/または処置するための本発明の診断および/または治療方法に有用な、医薬のおよび/または診断の化合物および/または組成物の形式の抗CPAA抗体およびペプチドを提供する。
モノクローナル抗体またはその部分により特定されるCPAAペプチドを特性評価することは、本発明のさらなる目的である。そのような抗原ペプチドは、さらなる抗原結合性リガンドを創製する際に有用でありうるし、あるいはワクチンまたは他の免疫賦活手法として用いられうる。
本発明が、本明細書に記載される特定の方法論、プロトコルおよび試薬、その他のものに限定されることはなく、またそれらが改変されてもよいことは理解されるべきである。本明細書中で用いられる専門用語は、特定の実施形態を記載することのみを目的とするものであって、本発明の範囲を限定する意図はなく、それは特許請求の範囲によってのみ規定される。
特に定義されない限り、本明細書記載の抗体に関連して用いられる科学用語および技術用語は、当業者により一般に理解される意味を有するものである。さらに、文脈により特に要求されない限り、単数形の用語は複数形を含みうるし、複数形の用語は単数形を含みうる。一般に、本明細書記載の、細胞培養および組織培養、分子生物学ならびに蛋白質およびオリゴヌクレオチドまたはポリヌクレオチド化学およびハイブリッド形成に関連して用いられる命名法ならびに技術は、当分野で周知であり、通常用いられるものである。

抗原結合領域を与えるH鎖またはL鎖の可変領域中に、種々の特異性を有する抗体間での極めて大きな多様性のために「超可変」と称される小さい配列が存在する。この超可変領域はまた、「相補性決定領域」または「CDR」領域と称されることもある。これらのCDR領域が、特有の抗原決定構造に関する抗体の基本的な特異性の主たる原因となる。
抗体のCDR領域により認識される抗原決定に関して、これは「エピトープ」とも称される。換言すれば、エピトープは、抗体により認識され、結合されうるいずれかの分子の部分を意味する(相対する抗体の結合領域は、パラトープと称されることもある)。一般に、エピトープは、分子の化学的に活性な表面配置、例えばアミノ酸または糖側鎖からなり、特異的な三次元構造上の特性ならび特異的な荷電特性を有する。
かくして、用語「抗体」は、完全免疫グロブリン分子、ならびにその部分、フラグメント、ペプチドおよび誘導体、例えば抗原もしくはエピトープと結合できる抗体のFab、Fab’、F(ab’)2、Fv、Fsc、CDR領域、パラトープまたはいずれかの部分もしくはペプチド配列、の両方を含むことを意味する。抗体は、それが分子と特異的に反応し、それによって分子を抗体に束縛できる場合に、分子を「結合できる」という。
本発明は、CPAAと特異的に結合する新規のモノクローナル抗体を提供する。このモノクローナル抗体は、「16C3」と称され、それは、ハイブリドーマ・クローンに割り当てられた数字を意味する。本明細書において、16C3はまた、特異的に16C3抗体と結合するその能力のために16C3と特定されたCPAAエピトープと特異的に結合するモノクローナル抗体の部分、パラトープまたはCDRも意味する。本明細書記載の16C3の組み換え体およびヒト型のいくつかは、同じ名前により示されることがありうる。
本発明は、その範囲内に、本発明の抗CPAA抗体の軽鎖および重鎖の可変領域をコードするDNA配列を含む。16C3抗体の軽鎖の可変領域をコードする核酸配列は、図2に示される。16C3抗体の重鎖の可変領域をコードする核酸配列は、図3に示される。
同様に、アミノ酸配列における付加および置換そして変異ならびに上記のモディファイは、CPAA抗原および/またはそのエピトープもしくはペプチドのアミノ酸配列に同じように適用でき、かくして、それらは本発明に包含される。上記のように、本発明に関するモノクローナル抗体をコードする遺伝子は、CPAAの認識に特に効果的である。
モノクローナル抗体は、典型的には、ネズミ・ハイブリドーマ系において天然分子として産生される。その技術に加えて、以下に概説されるように、本発明は、モノクローナル抗体の組み換えDNA発現を提供する。これは、選択された宿主種におけるヒト化抗体ならびに広範囲の抗体誘導体および融合タンパク質の産生を可能にする。最近では、細菌、酵母、トランスジェニック動物および鶏卵における抗体の産生が、ハイブリドーマに基づく生産系の有望な代替手段として出現してきた。トランスジェニック動物の主な利点には、再生可能な資源からの潜在的に高い収量がある。
昆虫における抗CPAA抗体またはそのペプチドもしくは機能的誘導体の産生は、例えば昆虫宿主を当業者に既知の方法により膜貫通ポリペプチドを発現するよう改変されたバキュロウイルスを用いて感染させることによって、実施することができる。Ausubelら、1987, 1993を参照のこと。
一部のcDNAおよび一部のゲノムDNAからなる免疫グロブリン遺伝子については(Whittleら、1 Protein Engin. 499 (1987))、転写プロモーターはヒト・サイトメガロウイルスであってよく、プロモーター・エンハンサーはサイトメガロウイルスおよびマウス/ヒト免疫グロブリンであってよく、そしてmRNAスプライシングおよびポリアデニル化領域は本来の染色体免疫グロブリン配列であってよい。
各ベクターは、2個の選択可能な遺伝子、すなわち、細菌系における選択のために設計された第1の選択可能な遺伝子、そして真核系における選択のために設計された第2の選択可能な遺伝子を含んでいてよく、各ベクターは異なる遺伝子ペアを有する。このストラテジーは、結果として、細菌系において融合遺伝子の産生を最初に導き、そして増幅を可能にするベクターを生じる。このように細菌宿主において産生され増幅される遺伝子は、続いて、真核細胞を同時トランスフェクトするために用いられ、所望のトランスフェクト遺伝子を保有する同時トランスフェクトされた細胞の選択を可能にする。
gptを発現する細胞の選択は、この遺伝子にコードされた酵素がプリンヌクレオチド合成の基質としてキサンチンを利用することができるが、類似の内因性の酵素は利用することがでないという事実に基づいている。(1)イノシン一リン酸塩のキサチン一リン酸塩への転換をブロックするミコフェノール酸と(2)キサンチンを含む培地においては、gpt遺伝子を発現する細胞のみが生存できる。neo産物は、抗生物質G418およびネオマイシン類の他の抗生物質による蛋白質合成の阻害をブロックする。
あるいは、キメラH鎖およびL鎖をコードする融合遺伝子は、同じ発現ベクター上で組み合わされてよい。
抗体タンパク質産生に関し、宿主として有用でありうる哺乳動物細胞は、上記のリンパ球起源の細胞に加えて、線維芽細胞起源の細胞、例えばベロ(Vero)細胞(ATCC CRL 81)またはCHO−Kl細胞(ATCC CRL 61)を含む。
本発明は、CPAAに対する高い特異性と親和性を有するモノクローナル抗体を産生するハイブリドーマ細胞系を提供する。本発明はまた、上記で詳細に特性評価されたハイブリドーマ細胞系のバリアントおよび変異体に関し、それらは、自然に生じたものであってもあるいは既知方法を用いて人工的に作製されたものであってもよいが、出発材料の特徴的な性質をいまだに有しているもの、すなわち、本発明に関する抗体またはその誘導体をいまだに産生でき、そしてそれらを周囲の培地中に分泌できるバリアントおよび変異体に関する。
より詳細には、本発明の核酸、タンパク質またはペプチド分子は、CPAAを結合するモノクローナルまたはポリクローナル抗体を開発するのに利用できる。本発明のCPAAを結合する抗体の調製のために、培養中の連続継代性細胞系による抗体分子の産生を提供するいずれかの技術が、用いられてよい。例えば、KohlerおよびMilstein(256 Nature 495-497 (1975))により最初に開発されたハイブリドーマ技術が用いられてもよい。また、米国特許第4,376,110号;Ausubelら、1988;CURR. PROT. IMMUNOL.(Colliganら、編、Greene Pub. Assoc. & Wiley Interscience NY, 1992-1996)を参照のこと。
本発明の抗体は、ヒト抗体由来の定常領域がマウス由来の軽鎖および重鎖の可変領域にクローン化された、ヒト抗体の一部とマウス抗体の一部を含むキメラ抗体を包含する。いくつかの例において、ヒト配列の70%が保たれている。ヒト化抗体は、ヒト抗体の枠組み構造の約90%が保たれており、マウスの相補決定領域とのみ組み合わせられた、キメラ抗体である。完全ヒト化抗体もまた、本発明において考慮される。
CPAAのアミノ酸残基中に含まれる16C3エピトープを結合する組み換えネズミ抗体またはキメラ・ネズミ−ヒト抗体もしくはヒト−ヒト抗体が、本明細書中で提供され、本明細書中で提供される教示に基づき既知の技術を用いて産生されうる。例えば、Ausubelら、1987、1992および1993;Sambrookら、を参照のこと。例えば、抗体は、EP0239400に従い、所望のCDRをヒト枠組み構造と接ぎ合わせることによってヒト化されてもよい。
図2および図3で示されるCPAA結合性抗体の軽鎖および重鎖を表すオリゴヌクレオチドは、相同遺伝子の存在に関するスクリーニングに、そして抗CPAA抗体の可変領域もしくは定常領域をコードする該遺伝子のクローニングに有用である。そのようなプローブは、CPAAのエピトープと結合する軽鎖もしくは重鎖のCDR領域についての図4および図5中で下線付きで示されたアミノ酸配列をコードするDNA配列(cDNA、ゲノムDNAまたは他のいずれかのDNA)と通常結合する。そのようなオリゴヌクレオチドを合成する技術は、周知である。例えば、Wuら、21 Prog. Nucl. Acids Res. Molec. Biol. 101 41 (1978);Ausubelら、1987, 1993を参照のこと。
このように、1つの実施形態において、融合キメラ遺伝子は、少なくともヒト起源以外の抗原結合領域をコードする第1のDNA断片、例えばジョイニング(J)断片と機能的に再編成されたV領域を含み、少なくともヒトC領域の一部をコードする第2のDNA断片に連結されて、作り出される。
1.抗CPAA抗体を産生する細胞系からの、そして重鎖定常領域および軽鎖定常領域を供給する任意の付加的な抗体からのメッセンジャーRNA(mRNA)の単離;クローニングおよびcDNA作製;
2.L鎖遺伝子およびH鎖遺伝子の適当なV領域および/またはC領域が、(i)適当なプローブを用いて特定され、(ii)配列決定され、そして(iii)他の抗体由来のC遺伝子断片またはV遺伝子断片を用いてキメラ抗体に適合される、精製されたmRNAからの全長cDNAライブラリーの調製;
3.上記のように、クローン化された特定のV領域遺伝子断片をクローン化されたC領域遺伝子に連結することによる、完全なH鎖またはL鎖のコード配列の構築;
4.ネズミ−ネズミ抗体、ヒト−ネズミ抗体、ヒト−ヒト抗体またはヒト−ネズミ抗体を提供するため、原核生物および真核生物細胞を含む選択された宿主におけるL鎖およびH鎖の発現および産生。
発現ビヒクルは、プラスミドまたは他のベクターを含む。とりわけ、適当な付着末端を有するいずれかのVH鎖もしくはVL鎖配列が容易に挿入されるような適当な制限酵素部位を有する、機能的に完全なヒトCH鎖もしくはCL鎖配列を運ぶビヒクルがある。かくして、ヒトCH鎖もしくはCL鎖の配列を含むビヒクルは、いずれかの適当な宿主中で所望の完全なH鎖またはL鎖のいずれかを発現する中間物質として提供される。
1.コンピュータプログラムを用いて、すべての利用可能なプロテイン(およびDNA)データベースを、ネズミ抗体の可変ドメインに対して最も相同性があるヒト抗体の可変ドメイン配列を探索する。適切なプログラムの出力は、ネズミ抗体に対して最も相同な配列の列記と、各配列に対する相同性パーセント、そして各配列のそのネズミ配列に対するアライメントである。これは、重鎖および軽鎖の可変ドメイン配列の両方に対して独立して行われる。ヒト免疫グロブリン配列が含まれていさえすれば、上記分析はさらに容易に達成される。
2.ヒト抗体の可変ドメイン配列を列記し、相同性を比較する。主として、この比較は、極めて可変的な重鎖のCDR3を除き、CDRの長さにおいて行われる。ヒト重鎖およびカッパ軽鎖そしてラムダ軽鎖は、サブグループ、すなわち重鎖の3つのサブグループ、カッパ鎖の4つのサブグループ、ラムダ鎖の6つのサブグループに分けられる。各サブグループ内のCDRの大きさは類似するが、サブグループ間では異なる。相同性の第一近似として、ネズミ抗体のCDRをヒト・サブグループのいずれかとマッチすることが通常行われる。次いで、類似の長さのCDRを有する抗体は、アミノ酸配列の相同性について、特にCDR内の相同性について比較されるが、周辺の枠組み構造領域についても比較される。最も相同性があるヒト可変ドメインが、ヒト化のための枠組み構造に選ばれる。
ヒト可変部分ドメイン枠組み構造に所望の残基を含めるための突然変異を導入するために用いられうるオリゴヌクレオチドは、合成される。それらのオリゴヌクレオチドは、いずれかの好都合なサイズであってよい。それは、通常、利用することができる特定の合成装置の能力によってのみ長さが制限される。オリゴヌクレオチドに直接的に試験管内で変異導入する方法は、当分野で周知である。
最適化されたヒト化16C3抗体(H16C3−Abb*と称される)は、図12に示されるアミノ酸残基を含む。その抗体のCDR中のアミノ酸残基のいくつかが、元のネズミCDRに存在したものとは変わっている点に注意されたい。異なるCDRは、機能的に同等であるため、本発明の範囲内では相互のバリアントの例と考えられる。
(a)少なくともIgの重鎖または軽鎖の可変ドメインであって、ヒト抗体からの枠組み構造領域および本発明のヒト化抗体に必要とされるCDRを含む可変ドメインをコードするDNA配列と作動可能に連結された適切なプロモーターを含む、第一の複製可能な発現ベクターを調製すること;
(b)少なくとも相補的なIg軽鎖または重鎖それぞれの可変ドメインをコードするDNA配列と作動可能に連結された適切なプロモーターを含む、第二の複製可能な発現ベクターを調製すること;
(c)第一のまたは両方の調製されたベクターを用いて細胞系を形質転換すること;および
(d)該形質転換された細胞系を、該改変された抗体を産生させるために培養すること、
により調製されうる。
上記の生産技術に加えて、試験管内の系、例えば完全ヒト抗体および抗体ペプチドのファージディスプレイ法、診断法および治療法としてのヒト抗体の多くの利点が、現在認められている。
本発明の組み換え抗体およびその配列は、抗PCAA結合活性を有する無数の誘導体およびリガンド結合分子の構築を可能にする。例えば、そのCDRは、ヒト抗体ライブラリー、例えばn−CoDeRヒトscFvライブラリーと組み合わせて、高特異性および高機能性の抗体フラグメントを作り出すことができる。Moore, 426 Nature, 725-31 (2003)を参照のこと。
完全なヒト抗体またはその部分のライブラリーもまた、米国特許出願公開番号第20030232333に記載されるように、一本鎖DNAの部位特異的な切断に基づくクローニング法に従って作り出すことができる。
別の関連する誘導体は、抗原と抗体のエフェクター分子(Fc領域の分子)を架橋できる細菌で産生された抗体フラグメント(PEPBODIES(登録商標)抗体フラグメントと称される)を提供するため、新しい技術の利点を有する。米国特許出願公開番号第20040101905号を参照のこと。それゆえ、抗PCAA部位の抗原結合部位を含む結合分子は、概して、Fc領域と関連する1つまたは複数のエフェクター機能を示すペプチドと融合され、そして細胞受容体との相互作用および補体活性化などの機能を提供する。
結論として、本明細書中で提供されるヌクレオチド配列およびアミノ酸配列は、無数の生じうる分子がCPAA結合活性を有することを可能にし、本発明の範囲はこれらの物質を得る方法により制限されない。
抗体の「誘導体」は、付加的な化学部分を含むが、通常はタンパク質の一部は含まない。タンパク質の共有結合修飾は、本発明の範囲内に含まれる。そのような修飾(モディフィケーション)は、標的とする抗体のアミノ酸残基を、選択された側鎖もしくは末端の残基と反応できる有機誘導体化剤と反応させることによって、その分子に導入することができる。例えば、当分野で周知の、二官能性物質による修飾は、抗体またはフラグメントを、水に不溶性のサポート・マトリックスにまたは他の高分子担体に架橋するのに有用である。
モノクローナルもしくはキメラ抗PCAA抗体に加えて、本発明はまた、本発明の抗CPAA抗体に特異的な抗イディオタイプ(抗Id)抗体にも関する。抗Id抗体は、一般に、他の抗体の抗原結合領域に関連する固有の決定基を認識する抗体である。CPAAに特異的な抗体は、イディオタイプ抗体もしくはId抗体と称される。抗Idは、Id抗体の供給源と同じ種でかつ同じ抗原型の動物(例えばマウス株)を、Id抗体またはその抗原結合領域を用いて免疫することによって調製されうる。免疫された動物は、免疫した抗体のイディオタイプ決定基を認識して応答し、抗Id抗体を産生することとなろう。この抗Id抗体はまた、さらに別の動物において免疫応答を引き起こす「免疫原」として用いられ、いわゆる抗−抗Id抗体を産生することもできる。この抗−抗Id抗体は、抗Idを誘導した元の抗体と、エピトープの点で同一でありうる。したがって、あるmAbのイディオタイプ決定基に対する抗体を用いることによって、同一の特異性を有する抗体を発現する別のクローンを特定することが可能である。
加えて、CPAAに対する抗体、その類似体、それらの部分、フラグメント、ペプチドまたは誘導体は、適切な動物、例えば、BALB/cマウスにおいて抗Id抗体を誘導するために用いることができる。そのように免疫されたマウスからの脾臓細胞は、抗Idモノクローナル抗体を分泌する抗Idハイブリドーマを作製するために用いられる。さらに、抗Id抗体は、担体、例えばキーホールリンペットヘモシアニン(KLH)にカップリングされ、さらなるBALB/cマウスを免疫するために用いることができる。これらのマウスからの血清は、CPAAのエピトープに特異的な元のモノクローナル抗体の結合特性を有する抗−抗Id抗体、またはその類似体、フラグメントおよび誘導体を含むであろう。かくして、抗Id抗体は、それ自身がイディオタイプエピトープを有しており、また、評価されるべきエピトープと構造的に類似する「イディオタイプ」を有する。
本発明の抗CPAA抗体およびペプチドの構造類似体は、本明細書で示される教示およびガイダンスに基づいて既知の方法工程により提供される。
タンパク質の三次元構造に関する知識は、それらが機能する様式を理解する際に決定的に重要である。タンパク質構造データベースにおいて(配列データベースの既知のタンパク質配列の数千と対照的に)、何百ものタンパク質の三次元構造が現在利用可能である。これらの構造の解析は、それらが、認識できる種類のモチーフに折り畳まれることを示す。このように、タンパク質の相同性に基づき関連するタンパク質の既知構造に対し、あるタンパク質の三次元構造のモデルを作ることが可能である。比較的低い配列相同性を有する2つのタンパク質が、類似性の高い三次元構造またはモチーフを有しうる例も多く知られている。
本発明はまた、膵臓癌または結腸癌であることが分かっている患者もしくは罹患していることが疑われる患者においてCPAAを検出するための診断方法での使用のための、上記の抗CPAA抗体およびペプチドを提供する。本発明の他の態様において、本抗体は、形態学的に正常な細胞において分子マーカーを検出することができ、無症状の個体の早期発見スクリーニングを提供する。
本発明の抗CPAA抗体および/またはペプチドは、試料中のCPAAまたは抗CPAA抗体を検出するもしくは定量する免疫アッセイに有用である。CPAAの免疫アッセイは、典型的には、臨床試料または生物試料を、選択的にCPAAと結合できる本発明の検出可能に標識化された高親和性(もしくは高い結合活性)の抗CPAA抗体またはポリペプチドの存在下でインキュベートすること、および試料中で結合される標識化されたペプチドまたは抗体を検出すること、を含む。さまざまな臨床アッセイ手順が当分野で周知である。例えば、IMMUNOASSAYS FOR THE 80’S (Vollerら、編、Univ. Park, 1981)を参照のこと。そのような試料は、生検、血液、血清および糞試料、または、浣腸、結腸内視術もしくは経口緩下剤の液剤後の結腸直腸トラック(colorectal track)から回収される液体を含み、後述するELISA分析を受ける。
CPAA特異ペプチドおよび/または抗体の検出可能な標識化は、酵素免疫定量法(EIA)または酵素免疫吸着測定法(ELISA)での使用のための酵素と連結することによって達成することができる。その連結された酵素は、曝された基質と反応し、例えば分光光度法、蛍光分光法により、または可視化手段によって検出できる化学部分を生成する。本発明のCPAA−特異抗体を検出可能に標識化するために用いられうる酵素は、限定するものではないが、リンゴ酸脱水素酵素、ブドウ球菌のヌクレアーゼ、デルタ−5−ステロイドイソメラーゼ、酵母アルコール脱水素酵素、アルファ−グリセロリン酸脱水素酵素、トリオースリン酸イソメラーゼ、西洋わさびペルオキシダーゼ、アルカリホスファターゼ、アスパラギナーゼ、グルコースオキシダーゼ、ベータ−ガラクトシダーゼ、リボヌクレアーゼ、ウレアーゼ、カタラーゼ、グルコース−6−リン酸脱水素酵素、グルコアミラーゼおよびアセチルコリンエステラーゼを含む。
CPAA特異抗体を、蛍光化合物を用いて標識することも可能である。蛍光標識された抗体は、適切な波長の光に曝された場合、次いで、その存在は蛍光によって検出できる。なかでも最も一般的に用いられる蛍光標識化合物は、フルオレセイン・イソチオシアネート、ローダミン、フィコエリトリン、フィコシアニン、アロフィコシアニン、O−フタルアルデヒドおよびフルオレサミンである。
CPAA特異抗体はまた、化学発光化合物とカップリングすることにより検出可能に標識化できる。次いで、化学発光で標識化された抗体の存在は、化学反応の間に生じる発光の存在を検出することにより決定される。有用な化学発光標識化合物の例は、ルミノール、イソルミノール、テロマティック・アクリディニウムエステル(theromatic acridinium ester)、イミダゾール、アクリジニウム塩およびシュウ酸エステルである。
CPAA特異抗体、部分、フラグメント、ポリペプチドまたは誘導体の検出は、例えば検出可能な標識が放射性ガンマ放射体の場合にはシンチレーション計数器により、例えば標識が蛍光物質の場合には蛍光光度計により、達成することができる。酵素標識の場合には、検出は、その酵素に対する基質を用いる比色法により達成することができる。検出はまた、同様にして調製された標準に対して、基質の酵素反応の程度を視覚的に比較することにより、達成することができる。
本発明の抗体、フラグメントまたは誘導体は、「トゥー・サイト(two-site)」または「サンドイッチ」アッセイとしても知られている、免疫測定法での利用のために用いることができる。典型的な免疫測定法においては、ある量の未標識の抗体(または抗体のフラグメント)が試験される液体中で不溶性の固体サポートに結合され、ある量の検出可能に標識化された可溶性抗体が加えられ、固相の抗体、抗原そして標識化抗体の間で形成される三者複合体を検出および/または定量化が可能となる。
「リバース」アッセイにおいては、液体試料に標識化抗体溶液を最初に添加し、続いて適切なインキュベーション時間後に、固体サポートに結合した未標識の抗体を添加する、段階的な添加が用いられる。第2のインキュベーション後に、固相は、試験される試料中のおよび未反応の標識化抗体溶液中の残留物をそこから取り除くために、通常の様式で洗浄される。次いで、固体サポートと会合した標識化抗体の決定は、「同時」アッセイおよび「フォワード」アッセイのように決定される。1つの実施形態において、別々のエピトープに特異的な本発明の抗体の組み合わせは、センシティブ・スリー−サイト免疫放射定量測定法(sensitive three-site immunoradiometric assay)を実施するために用いることができる。
重要なことに、本発明の抗体は、ある種の大腸癌および膵臓癌の侵襲性を診断するのに有効でありうる。より詳細には、本発明の抗体は、非常に急速に進行する悪性の癌と対照的に、幾年もかけて成長する緩徐型の癌(slow cancer)の患者に存在するCPAAを特定しうる。このように、本発明の抗体は、重要な免疫組織学的ツールを提供しうる。
キットもまた、被検抗体を細胞活性に対する保護もしくは検出に、または選択された抗原の存在に関して使用するために供給されてよい。かくして、本発明の抗体は、通常、単独あるいは所望の細胞型に対して特異的なさらなる抗体と組み合わされ、容器中で凍結乾燥形式にて提供されうる。抗体は、ラベルもしくは毒素とコンジュゲートされていてもコンジュゲートされていなくてもよく、それらは、緩衝液、例えばトリス溶液、リン酸溶液、炭酸溶液、その類似の溶液、安定剤、殺生物剤、不活性なタンパク質、例えば血清アルブミンまたはその類似物を含むキットに、含まれる。該して、これらの物質は、活性な抗体量に基づいて5%wt未満存在してよく、通常、抗体濃度に対して全量の少なくとも約0.001%wtで存在してよい。しばしば、活性成分を希釈するために、不活性な増量剤または賦形剤を含んでいることが望ましく、賦形剤は全組成物の約1%〜99%wtまで存在してもよい。第1の抗体に結合できる第2の抗体がアッセイにおいて用いられる場合、それは、通常、別のバイアル中に入っているであろう。第2の抗体は、典型的にはラベルとコンジュゲートされており、そして上記の抗体製剤と類似する様式で製剤化される。キットは、一般に一組の使用説明書もまた含みうる。
本発明の抗CPAA抗体またはペプチドは、例えば癌腫および関連する状態の処置に用いられうる。より詳細には、本発明は、医薬的に許容される担体もしくは希釈剤および活性成分として本発明に関する抗体もしくはペプチドを含む、医薬組成物を提供する。免疫毒素の送達成分は、本発明に関するヒト化抗体である。完全免疫グロブリンまたはそれらの結合フラグメント、例えばFabもまた想定される。典型的には、免疫毒素における抗体は、ヒトIgA、IgMまたはIgGアイソタイプのものであってよいが、望ましい場合には別の哺乳動物の定常領域が用いられてもよい。この成分はまた、本発明に関する免疫毒素を含んでいてよい。本発明のヒト化抗体、免疫毒素および医薬成分は、非経口投与、例えば皮下、筋肉内もしくは静脈内投与に有用である。
本発明の抗CPAA抗体および/またはペプチドは、個別の治療剤として、または別の治療物質との組み合わせのいずれかにおいて、投与されてよい。それらは、単独で投与されてもよいが、一般的には、選らばれた投与経路および標準的な薬務に基づいて選択された医薬担体と一緒に投与される。
本発明の抗体は、貯蔵のために凍結乾燥されてよく、使用前に適切な担体中で再構成されてよい。この技術は、通常の免疫グロブリンに対して有効であることが示されている。いずれかの適切な凍結乾燥技術および再構成技術が、行われうる。当業者であれば、凍結乾燥および再構成が、異なる程度の抗体活性の喪失を導きうること(例えば、一般的な免疫グロブリン、IgM抗体は、IgG抗体よりも大きく活性を損なう傾向にある)、ならびに埋め合わせのために使用レベルが調整されなければならないであろうことは、認められよう。
非制限的な例として、CPAA関連病状のヒトもしくは動物の処置は、本発明の抗CPAAペプチド、モノクローナルキメラ抗体および/またはネズミ抗体の1日投与量、週投与量または隔週投与量として、1日あたり、1週あたりまたは隔週あたり0.1mg/kg〜100mg/kgの範囲の投与量で提供されてよい。
内部への投与に適した投薬製剤(組成物)は、一般には、ユニットあたり約0.1mg〜約500mgの活性成分を含む。これらの医薬組成物において、活性成分は、組成物の全重量に基づく重量あたり約0.5%〜95%の量で通常存在するであろう。
組成物の単回投与または複数回投与は、処置を施す医師により選択される服用レベルとパターンで行われうる。いずれにしても、医薬製剤は、患者を効果的に処置するのに十分な量の本発明の抗体(類)を提供すべきである。
抗体はまた、化学療法剤または免疫抑制剤と併用される場合、別個に投与される組成物として用いられてもよい。典型的には、その試薬は、サイクロスポリンAまたはプリン類似体(例えばメトトレキセート、6−メルカプトプリンまたはその類似物)を含んでいてよいが、当業者に周知の多くのさらなる試薬(例えばシクロホスファミド、プレドニゾン、その類似物)が利用されてもよい。
本発明の抗CPAA抗体および/またはペプチドは、他のモノクローナル抗体またはネズミ抗体およびキメラ抗体、フラグメントおよび領域と組み合わせて、あるいは抗体と相互作用するエフェクター細胞の数および活性を増大させるリンフォカインまたは造血成長因子、その他の類似物と組み合わせて、有利に利用することができる。例えば、本発明の抗体は、疾患に対応する細胞上の別のマーカーと反応性があるヒトモノクローナル抗体と同時に投与されてよい。例えば、適切なT細胞マーカーは、白血球タイピングにおける第1回国際白血球分化に関するワークショップ(Bernardら、編、Springer Verlag, NY, 1984)により名付けられるように、いわゆる「分化抗原群(Clusters of Differentiation)」に分類されるものを含みうる。
16C3抗体が結合する抗原は、全てではないがいくつかの培養ヒト腫瘍細胞系において、全てではないがいくつかの正常なヒト胎児消化管組織において、そして大部分の結腸と膵臓の腫瘍組織で発現されているようである。以下の実施例において詳細に記載されるように、さまざまな腫瘍試料のウェスタンブロットにおいて、16C3抗体は、〜200kDaのペプチド種および〜110kDaのペプチド種の両方を認識した。特定の試料の各種のタンパク質種の量を反映しうる染色強度は、大腸腫瘍試料と膵臓腫瘍試料の間で異なっているようである。16C3抗原は、大部分の大腸と膵臓の腫瘍組織で発現されているようであり、腫瘍胎児抗原(oncofetal origin)であるかもしれない。16C3抗原は、細胞表面に発現されており、糖タンパク質である。16C3抗体が認識する同じ抗原の関連する2種類のものである可能性があり、各種類の相対的な量は、大腸腫瘍組織と膵臓腫瘍組織との間で異なっている。
本発明の他の態様は、癌ワクチンを提供する。「ワクチン」により、生体組織の免疫系を刺激するために用いられる試薬を意味する。これに関して、免疫応答は、予防を提供することができるか、あるいは病的な組織に、例えば既存の状態を軽減することにより良い影響を提供することができる。詳細には、癌ワクチンは、既存の悪性腫瘍を治療的に処置することおよび/または既存の悪性腫瘍の進行または転移を妨げることが意図される。
本発明は、次いで、非制限的な実施例によりさらに記載される。
実施例1.ヒト腫瘍試料からの膵臓癌および結腸直腸癌−関連抗原(CPAA)の調製。
免疫原性の腫瘍関連抗原調製物は、Hollinsheadら、56 Cancer 480 (1985);米国特許第5,688,657号に記載の方法に従って、プールされた結腸直腸癌メンバーから得られた。この抗原性物質は、そのメンバーの分画がHL−A抗原を含まず、多くの非免疫原性糖タンパク質分画から分離される程度に精製された。
ヒト膵臓癌および結腸直腸癌−関連抗原に対するモノクローナル抗体は、マウス骨髄腫細胞Sp2/0−Ag14と上記のCPAAを用いて免疫されたBALB/cマウスからの脾臓細胞とを融合させた結果得られる、ハイブリドーマの作製およびクローニングによって得られる。ハイブリッド・クローンが樹立され、そしてELISAにより評価された場合にCPAAおよび結腸癌腫細胞系(LS174T)と強く反応した。
ネズミ免疫グロブリンは、重鎖(55kD)および軽鎖(25kD〜29kD)をコードする別個の遺伝子から発現される。ネズミ免疫グロブリンのレパートリーを与えるために再編成可能な、4個の重鎖IgGサブクラス(IgG1、IgG2a、IgG2b、IgG3)および2個の軽鎖(カッパ、ラムダ)がある。
16C3mAbのアイソタイプは、サザンバイオテクノロジー社(Southern Biotechnology, Inc.)のマウス・アイソタイピング・キットを用いて決定された。16C3mAbは、IgG1重鎖およびカッパ軽鎖であると決定された。
既述の他のすべてのmAbの既知のアミノ酸配列と比較して、このmAbの直線状のアミノ酸配列によりその特異性が認められる。直線状のアミノ酸配列は、そのポリペプチド分子をコードするDNAの直線配列を最初に決めることにより決定できる。16C3mAbをコードするDNA配列が決定されると、次に、その読み枠は、共通の哺乳動物コドン使用頻度表を用いてアミノ酸配列に翻訳され、このようにして16C3分子の直線配列のアイデンティティーが表されることとなる。
ネズミIgG1重鎖、カッパ軽鎖クローニングに用いたオリゴヌクレオチドプライマーは、文献のRapid cloning of any rearranged mouse immunoglobulin variable genes, Dattamajumdarら、43 Immunogenetics 141-51 (1996)に基づいて得られた。
16C3抗体の配列は、国立衛生研究所の国立医学図書館(National Library of Medicine)の一部門の国立バイオテクノロジー情報センター(NCBI)のタンパク質と核酸のデータベースに対して、BLASTサーチ(Basic Local Alignment Search Tool)にかけた。16C3mAb重鎖または16C3mAb軽鎖のクエリー配列のいずれかを用いたBLASTサーチにより見出された類似の配列の調査によって、16C3重鎖と軽鎖の両方の可変領域が特有な配列であることが示された。かくして、16C3mAb重鎖配列および軽鎖配列は、新規かつ特有な抗体分子であることを示す。
ハイブリドーマにより産生される16C3mAbは、プロテインL−アガロース・マトリクスを用いた親和性カラムクロマトグラフィーにより精製された。精製された16C3mAbは、以下の表1で特定されるように、様々な腫瘍細胞を用いた間接免疫蛍光法により特性評価された。腫瘍細胞系はすべて、ATCCから入手した。細胞は、リン酸緩衝生理食塩水(PBS)中に希釈された精製された16C3mAbと一緒に、4℃で1時間インキュベートされた。細胞を洗浄した後、蛍光標識ヤギ抗マウスmAbと一緒にインキュベートされた。次いで、これらの細胞をPBSで3回洗浄し、ベックマン−ディキソン(Becton-Dickinson)のファックスキャリバー(FACSCalibur)(登録商標)およびセルクエスト(CellQuest)解析ソフトウェアを用いたフローサイトメトリーにより試験された。その結果は、表1に示される(FACSデータ)。このデータは、16C3mAbが大腸腫瘍細胞系および膵臓腫瘍細胞系に特異的に結合するが、前立腺腫瘍細胞系および扁平上皮癌細胞系には結合しないことを示す。
免疫原性腫瘍抗原に特異的な、治療上有効なmAbは、少なくとも以下の特徴;(a)高い腫瘍組織特異性、(b)正常なヒト組織への交差反応がないこと、および(c)腫瘍の破壊に関連する生物活性、例えば、抗体依存性細胞障害(ADCC)、の1つを備えているであろう。16C3mAbのADCC活性は、標的細胞として結腸癌系SW1463および膵臓癌系CFPAC−1とAsPC−1において試験された。メラノーマ細胞系、SK−MELは、特異な対照(specificity control)として提供された。ADCCは、正常なヒトPBMCをエフェクター細胞として用い、一般的な4時間の111In放出アッセイを用いて評価され、その結果は、表2(ADCCデータ)にてアイソタイプ放出パーセント(溶解%)として示される。陰性対照抗体であるUPC−10と比べて、データは、ネズミIgG1抗体の適度な殺活性を示すが、重要なことは、その殺活性が大腸腫瘍系および膵臓腫瘍系に明らかに特異的な点である。この抗体の殺活性は、このADCCアッセイにおいてヒト・エフェクター細胞と相互作用するFc領域を含むヒト枠組み構造配列を有するヒト化抗体またはキメラ化抗体により、増強しうる。
111In標識された標的細胞、5μg/ウェルで用いられた抗体、エフェクター細胞として用いられたIL−2活性化ヒトPBMC、回収前に37℃で4時間のインキュベーション。
ネズミ免疫グロブリンガンマ(IgG1)の本来の立体構造は、重鎖と軽鎖のポリペプチドをそれぞれ2個含む、4個のポリペプチドからなる。1個の重鎖(55kDa)は1個の軽鎖(25kDa〜29kDa)と会合し、その2量体がジスルフィド結合により同じ2量体を連結して機能的な4量体高分子を完成させる。IgG分子は、変性剤(SDS、ドデシル硫酸ナトリウム)と2個のヘテロダイマーを連結するジスルフィド架橋を還元する試薬(DTT、ジチオトレイトール)の存在下で、その成分の重鎖と軽鎖に脱会合され、ポリアクリルアミドゲル・マトリクス上で大きさによって分離されうる。ゲル電気泳動は、抗体などのタンパク質性物質の分子量を決定するために用いられる一般的な分析方法である。
16C3mAbにより示される抗原結合の特異性は、さまざまなヒト組織試料、癌および正常標本の両方の免疫組織学的染色により、測定された。パラフィンおよび新鮮凍結ヒト組織試料は、精製マウス16C3Mab(IgG1)を5μg/mlで染色し、次いで、ペルオキシダーゼ・コンジュゲート抗マウスIgG二次抗体を用いて検出された。その染色強度は表3に示され、0は、所定の標本における抗原もしくは高発現の抗原との交差反応がないことを示し;4は、非常に強い交差反応があること示す、0〜4の評価系を反映する。
ヒト悪性腫瘍を処置するための治療薬としての有用性を改善するため、その薬物が、繰り返しのそして低毒性の両方で投与されうるよう、マウスモノクローナル抗体は、キメラ化抗体またはヒト化抗体に変換されてよい。当分野では、ネズミタンパク質の投与は、ときどき、その外来タンパク質に対する重度の免疫応答および毒性反応の結果を伴いうることが知られている。したがって、ヒト化抗体が、ヒト治療により効果的であることは明らかであろう。
ハイブリドーマにより産生されるマウス16C3mAbは、プロテインLアガロース・マトリクスを用いる親和性クロマトグラフィにより精製された。チャイニーズハムスター卵巣細胞(CHO)により産生された組み換えマウス16C3mAbは、プロテインAセファロース(Sepharose)マトリクスによる親和性クロマトグラフィにより精製された。この精製された16C3調製物またはトランスフェクトされた細胞上清は、以下に示されるようにヒト大腸腫瘍細胞LS174Tまたはヒト膵臓腫瘍細胞AsPC−1を用いた間接免疫蛍光法により、特性評価された。細胞は、リン酸緩衝生理食塩水(PBS)中に希釈された精製16C3(マウスもしくはヒト化)とともに、4℃で1時間インキュベートされた。その細胞は洗浄され、そして蛍光標識化ヤギ抗マウス免疫グロブリン抗体とともにインキュベートされた。次いで、細胞は、PBSにより洗浄され、そしてベクトン−ディッキンソン・ファックスキャリバー(Becton-Dickinson FACScalibur)およびセルクエスト(CellQuest)分析ソフトウェアを用いたフローサイトメトリーにより試験された。そのデータは、ハイブリドーマ由来のマウス16C3と組み換えCHO由来のマウス16C3の両方の大腸腫瘍細胞系および膵臓腫瘍細胞系に対する類似した結合性を示すが、前立腺腫瘍または扁平上皮癌細胞系に結合しないことを示した。
16C3mAbにより示される抗原結合の特異性は、さまざまなヒト組織試料、癌および正常標本の両方の免疫組織学的染色により、測定された。組織マイクロアレイ、パラフィン包埋組織および新鮮凍結ヒト組織試料は、精製マウス16C3抗体(IgG1)を5μg/mlで用いて染色し、次いで、ペルオキシダーゼ・コンジュゲート抗マウスIgG二次抗体を用いて検出された。その染色強度は0〜4の評価系を用いて示され、0は、所定の標本における抗原もしくは高発現の抗原との交差反応がないことを示し、そして4は、非常に強い交差反応があること示す。ハイブリドーマ由来16C3抗体および組み換え16C3抗体のいずれも試験された。この結果を、表5および6にまとめた。
ネズミ16C3抗体を治療上有効なヒト化抗体に変換するための5つの異なる設計物が、図5および図6に示される。293T細胞において発現される組み換えヒト化16C3、マトリクス実験の5つのバリアントの同時トランスフェクション。一過性トランスフェクションからの上清を、2ng/mlに標準化し、次いで、希釈し、精製された組み換えマウス16C3と比較される親和性を評価し、次に、FACSによりAsPC−1膵臓腫瘍細胞に対する潜在的な抗原結合性について試験し、そして組み換えマウス16C3抗体と比較した。表7に示されたデータは、重鎖の5個のバリアントが、4個の軽鎖バリアントのそれぞれと共に正確に折りたたまれ、その結果抗原結合活性を得ることができたことを実際に示す。ヒト化重鎖および軽鎖のそれぞれの組み合わせの結合は、元のマウス16C3抗体による結合性と同等であった。データは、16C3免疫グロブリンを5つの異なる方法のいずれでヒト化しても、その結果得られる抗体の抗原認識部位を変化させないことを示す。それぞれの結合性は、各組み合わせでの抗体量を用いて滴定し、それらは類似の滴定プロファイルを実際に示した。
加えて、コンピューター内(インシリコ)分析ツールは、タンパク質中の潜在的なT細胞エピトープを予測する際に有用である。そのようなアルゴリズムは、ヒトにおける免疫原性が低下する公算の高い改良型組み換えタンパク質を設計するのに有用である。この予測技術の利点を用いて、5つのヒト化16C3重鎖および軽鎖遺伝子バリアントが、分析され、そして特定のHLAハロタイプを持つヒトにおける免疫原性反応を誘導する可能性がある1つまたは複数のT細胞エピトープを有することが示された。そのような分析は、ソフトウェア、例えばエピマー(Epimer)またはエピマトリックス(EpiMatrix)、インシリコ・エピトープマッピング・ツール(in silico epitope-mapping tool)を用いて行われてもよいし、あるいは商業メーカー、例えばアンチトープ社(Antitope, Ltd.(Cambridge, UK))により行われてもよい。潜在的な治療上の16C3抗体の起こりうる免疫原性を低下させるためにこれらのT細胞エピトープを取り除く試みにおいては、結果的に完全に機能的であるが免疫原性が低い抗体分子となるような、特定の部位特異的変異が、T細胞エピトープを取り除き、そして特定のアミノ酸を置換するために行われた。最適化されたヒト化16C3抗体のタンパク質配列は、図12に示され、太字のアミノ酸はCDRを示し、「/」はリーダーペプチド/成熟N末端の連結部、および可変ドメイン/定常ドメインの連結部を示す。

16C3抗体が結合する抗原のいくつかの特性評価は、さまざまな処理をおこなったウェスタンブロットにより試験された。表10のデータは、16C3抗原が、全部ではないが培養された大腸腫瘍細胞および膵臓腫瘍細胞のいくつかに存在することを実際に示す。重要なことは、16C3抗原が、消化管および腸から得られた胎児組織抽出物中に存在することである。分画Iの陽性標本は、膜タンパク質抽出および精製のホリンシェッド法(hollinshead method)により切断され、処理された胚組織を用いた、セファデックスG−200カラムクロマトグラフィー試行からの溶出液(分画I)を表す。大腸腫瘍標本は、外科的処置から得られた。その組織を細分化し、洗浄剤を用いて全タンパク質抽出物とした。表10は、これらの細胞抽出物のウェスタンブロットによる種々の腫瘍細胞における16C3腫瘍抗原の発現に関するデータを示す。これらのデータは、16C3腫瘍抗原が、生体の胚形成期の間に、ならびに癌において発現されうることを示唆する。したがって、16C3抗原の発現は、胚組織発生中、そして癌進行中に再び発現されるように発生上制御される可能性がある。
