Abstract
【解決手段】有効成分として緑茶多糖体(green tea polysaccharide)及び松茸抽出物(Tricholoma matsutake extract)を含有し、皮膚炎症及び酸化現象を抑制することによって、紫外線、環境汚染など老化を促進させる外部有害環境因子に対応して皮膚老化現象を抑制することができる化粧料組成物である。

Description
本発明の有効成分として使用される緑茶多糖体は、緑茶から由来する効能成分であって、緑茶カテキンの陰で注目を受けなかったが、最近、新しく浮び上がっている。他の植物で発見される多糖体とは異なって、酸性多糖体群であって、光合性を通じて作られた糖成分とアミノ酸が結合されて生成される。緑茶多糖体は、外部有害環境によって発生することができる炎症を抑制し、刺激を緩和させる効果がある。
1−1.タバコサンプルの製造
喫煙後、タバコ吸穀を集めて、フィルタ部分を除いて除去した。手入れされたフィルタ1個当たりDMSO 1mLを入れてシェーカ(shaker)利用して13時間反応を行った。このサンプルをボルテクサ(vortexer)利用して10分間振とうした後、フィルタは除去し、遠心分離機を用いて300rpm及び5分間の条件で遠心分離し、沈殿物を除去し、タバコサンプルを得た。
5%の牛胎児血清(FBS)及び100IUのペニシリンGを含むDMEM培地に、8ウェルチャンバースライド(well chamber slide)で5,000細胞数/ウェルとなるように、HaCaT細胞を分株した(HaCaT cells are seeded)。翌日、緑茶多糖体(購入先:バイオランド)50ppm、松茸抽出物(購入先:バイオランド)100ppmを単独又は一緒に24時間処理した。対照群は、DMSO単独で培地に1/1000で処理し、これは、緑茶多糖体と松茸抽出物の試料処理時に使用されたDMSOの濃度と同一である。
2−1.自動車煤煙サンプルの製造
耳拭きを利用してディーゼル自動車の排気筒に付いた煤煙残渣を拭きだし後、耳拭きの煤煙が付いた綿部位だけをDMSOに浸漬し、13時間反応を行った。その後、10分間ボルテクシング(vortexing)をした後、綿を除去し、自動車煤煙サンプルを得た。
5%の牛胎児血清(FBS)及び100IUのペニシリンGを含むDMEM培地に、8ウェルチャンバースライドで5,000細胞数/ウェルとなるように、繊維芽細胞を分株した(Fibroblasts cells are seeded)。翌日、各ウェルは、FBS及びフェノールレッドを含まないDMEM培地に置換され、さらに24時間後に、全体3個の試験群(表2に示すサンプルの各々)で処理された。陰性対照群については、各ウェルは煤煙/排気ガスでのみ処理され、また、対照群については、各ウェルは0.1% DMSOで処理された。試験群については、各ウェルは、自動車煤煙200ppmと、緑茶多糖体50ppm及び松茸抽出物10ppmの一方又は双方とで処理した後、37℃温度で2日間培養した。
結果を下記の表2に示す。
3−1.プロスタグランジンE2(Prostaglandin E2;PGE2)生成抑制確認
DMEM培地に5%牛胎児血清、100IUのペニシリンGを添加した培地に繊維芽細胞を24ウェルプレートに104細胞数/ウェルとなるように分株した。
5%の牛胎児血清(FBS)及び100IUのペニシリンGを含むDMEM培地に、24ウェルプレートで104細胞数/ウェルとなるように、繊維芽細胞を分株した(Fibroblasts cells are seeded)。翌日、各ウェルは、FBS及びフェノールレッドを含まないDMEM培地に置換され、さらに24時間後に下記の表3に示すサンプルの各々で処理された。陰性対照群については、各ウェルは煤煙/排気ガスでのみ処理され、また、対照群については、各ウェルは0.1% DMSOで処理された。試験群については、各ウェルは、自動車煤煙400ppmと、緑茶多糖体50ppm及び松茸抽出物10ppmの一方又は双方とで処理した後、37℃温度で2日間培養した。
結果を下記の表3に示す。
Claims (7)
- 緑茶多糖体(Green tea polysaccharide)及び松茸(Tricholoma matsutake)抽出物を有効成分として含有する化粧料組成物。
- 上記緑茶多糖体及び松茸抽出物を1.0:10.0〜10.0:1.0の混合比で含有することを特徴とする請求項1に記載の化粧料組成物。
- 上記緑茶多糖体及び松茸抽出物を各々組成物の全体重量に対して0.01〜20.0重量%で含有することを特徴とする請求項1に記載の化粧料組成物。
- 上記組成物が抗酸化効果を示すことを特徴とする請求項1乃至3のいずれかに記載の化粧料組成物。
- 上記組成物が抗炎症効果を示すことを特徴とする請求項1乃至3のいずれかに記載の化粧料組成物。
- 上記組成物が外部有害環境から皮膚を保護することを特徴とする請求項1乃至3のいずれかに記載の化粧料組成物。
- 上記組成物が抗老化効果を示すことを特徴とする請求項1乃至3のいずれかに記載の化粧料組成物。
