Abstract
Description
化学薬品
TLC(Sigma−Aldrich Sweden AB、Stockholm、Sweden)のテトラデカノイルホルボール13−アセテート(TPA)、CAS 16561−29−8、純度約99%を凍結乾燥粉末として保存した。ジメチルスルホキシド(DMSO)、CAS 67−68−5、無菌濾過、Hybri−Max(登録商標)(Sigma−Aldrich Sweden AB、Stockholm、Sweden)。アセトン、分析グレード、CAS 67−64−1、Labora AB、Sollentuna、Sweden。
合計で、15種の化合物を、TPAおよび/またはAA実験において試験した。表1〜3を参照されたい。オリゴヌクレオチドは、biomers.net GmbH、Ulm、Germanyによって合成され、−20℃で凍結保存された。
化合物は、室温で、PBS(Fluka、Sigma)を用いて使用濃度まで希釈した。濃度は、UV分光光度法(SmartSpec(登録商標)3000、BIO−RAID、Hercules、USA)を用いて95%の精度まで調整した。
動物
メスおよびオスのSPF NMRI/KSマウス(InDex Pharmaceutical AB所有の種畜のSPF NMRIマウス、MTC、Karolinska Institutet、Stockholm、Sweden)またはメスのBALB/cJマウス(Jackson Laboratory、Bar Harbor、Maine、USA)。動物は、グループ化し、実験に入る前に少なくとも1週間、順応させた。
動物を、21℃±3℃および55%±15%の相対湿度の部屋で飼育した。換気システムを、1時間当たり10回、空気交換するように設計した。部屋を、12時間明および12時間暗のサイクルとなるように照明した。照明を06:00〜18:00の時間つけた。
ケージ中の敷床は、Beekay敷床(B&K、Sollentuna、Sweden)とした。
環境強化のために、動物に、Sizzele NestまたはHappy−Mat(Scanbur A.S、Lellinger、Denmark)を供給した。
十分なペレットげっ歯動物用食餌、R36(Laktamin AB、Stockholm、Sweden)を適宜、供給した。
動物が国内品質の飲料水を有する動物飲用ボトルに自由にアクセスできるようにした。
それぞれのケージを、試験番号、グループ番号、倫理的な承認番号、性別、および動物耳番号を用いてマークしたケージカードによって識別した。動物は、パーマネントインクフェルトペンを使用して、動物の番号に対応する横線を用いて尾に個々にマークした。異なる治療に使用したプロトコルの例を表4に概説する。化合物を、必要な変更を伴う同じプロトコルに従って試験した。
用量投与
腹の右下腹部中に100μLを施すことによって腹腔内(i.p.)注射を行った。首領域中に100μLを用いて皮下注射を行った。鼻孔の中に液滴、全容量40μLをマウスに吸入させることによって鼻腔内投与を行った。投与をすべて、非麻酔マウスに行った。
非麻酔マウスの両方の外耳を、柔らかい歯ブラシを使用して、低刺激石けん(Palmolive、Sweden)を用いて洗浄し、ティッシュを用いて耳を乾燥させ、アセトンを浸したティッシュを用いてきれいにした。アセトンを蒸発させた後に、TPAまたはAAを、耳全体を覆うように注意して、耳のそれぞれの側に10μL、両側に適用した。溶媒を蒸発させるために、マウスを20秒間保持した。
腹腔内注射を、動物を背位に保持して、腹の右下腹部に施した。皮下(s.c.)注射を、動物の首に施した。鼻腔内投与を、わずかに後にもたれかかる姿勢に保持したマウスに施した。調整可能なマイクロピペット40μLの試験物質を鼻孔上に液滴で施し、マウスに自由意志で吸引させた。
殺すまでそれぞれのマウスを毎日観察した。病気のすべての徴候、健康、およびあらゆる行動の変化を記録した。
非麻酔マウスの耳を、TPAを投与した後の96時間までまたはAAモデルについては24時間までの異なる時点で測定した。第1の測定を、浮腫を誘発する前に行った。耳当たりすべての時点で三通り、スプリング式ダイヤルマイクロメーター(Limit、Thickness Gauge、精度0.01mm、接触域φ6mm、Luna AB、Allingsaes、Sweden)を用いて厚さを記録した。
スチューデントのt検定を使用して統計的有意性を計算した(Excel Statistical Functions、Microsoft(登録商標) Excel 2002、Microsoft Corp.、Redmond、USA)。
実験プロトコルを、表4に示す例によって示す。AUC低下およびP−値をポジティブコントロール(TPAまたはAA+PBS)に対して計算した。
この試験は、浮腫の誘発を0日目に開始して、本発明の化合物をi.p.で施した場合、浮腫の低下を示した。本実験において、コントロール物質IDX0150は、炎症のほぼ十分な寛解を示し、AUCを−38.9%低下させた。
試験Bは、浮腫の誘発の4日前にs.c.を施した場合の化合物の予防効果を調査した。IDX0920、IDX0955、およびIDX0001は、それぞれ−19.3%、−22.7%、および−16.3%のAUCの低下を示したが、統計的に有意ではなかった(NS)。IDX0150は、AUCの低下を示さなかった。測定値の間の大きな差異による不十分な結果のために、この試験を繰り返した(試験C)。
試験Bの繰り返しの試験において、浮腫の誘発の4日前にs.c.を施したODNは、それぞれ、−3%、−22%、−24.3%、および−17.9%のAUCの低下を示し、IDX0150(P<0.01)、IDX0955(P<0.01)、およびIDX0001(P<0.05)について統計的に有意であった。
より短い予防レジメンにおいて、浮腫の誘発の2日前にs.c.を施すと、試験Cとほぼ類似した結果が得られた。IDX0920、IDX0150、IDX0955(P<0.01)について統計的に有意であったが、IDX0001について有意ではなかった。AUCの低下は、それぞれ、−19.1%、−16.5%、−17.8%、および−12.4%であった。
この試験において、浮腫を誘発したのと同じ日に化合物を施した。誘発は、それぞれのマウスについて約20分間の待ち時間を考慮に入れるためにずらした。この試験において、AUCの比較的一様な低下、IDX0920 −19.4%、IDX0150 −27.3%、IDX0955 −17.7%、およびIDX0001 −16.7%があった。すべての化合物は、統計的に有意な低下をもたらした(P<0.01)。
この試験において、それぞれの動物に対して等用量(50μg)で、膨張を誘発したのと同じ日に化合物を施した。これもまた、AUCの低下、IDX0920 −17%、IDX0150 −31.1%、IDX0955 −18.3%、およびIDX0001 −13.2%をもたらした。下方への傾斜の遅れを示したIDX0920を除いて、すべての他の化合物は、膨張の有意な低下を示し、IDX0150およびIDX0955はP<0.01およびIDX0001はP<0.05であった。
治療レジメンを調査するために、化合物を浮腫の誘発の2時間後に与えた。これは、AUCの有意な低下、IDX0920 −43.8%(P<0.01)、IDX0150 −42.3%(P<0.01)、およびIDX0955 −40.7%(P<0.05)をもたらした。IDX0001は、統計的に有意ではない−17.9%の低下を示した。
代替投与経路を検討するために、鼻腔内投与を調査した。s.c.投与に対する比較において、類似した時点および用量を選んだ。試験Hにおいて、鼻腔内投与を、膨張を誘発する4日前に予防プロトコルにおいて施した。これは、s.c.投与を用いて得たものよりも膨張の有効な低下をもたらした。AUCの低下は、IDX0920 −33.8%。(P<0.01)、IDX0150 −25.7%(P<0.01)、IDX0955 −17.4%(P<0.05)、およびIDX0001 −25.7%(P<0.05)であった。
鼻腔内投与を、浮腫を誘発したのと同じ日に、0日目に施した場合、膨張の有効な低下が見られた。AUCの低下は、IDX0920 −27.3%。(P<0.01)、IDX0150 −28.4%(P<0.01)、IDX0955 −28.8%(P<0.05)、およびIDX0001 −6.4%(NS)であった。
試験Gにおいて使用したような治療プロトコルにおいて、鼻腔内投与もまた膨張の好適な低下をもたらした。AUCは、IDX0920で−28.7%(P<0.05)、IDX0150で−28.8%(P<0.01)、IDX0955で−17.2%(P<0.05)、およびIDX0001で−25.2%(P<0.05)低下することが示された。
用量応答を試験K〜Oにおいて調査した。試験Kにおいて、0.1、1、10、および100μgのIDX0150を与えた。結果は、用量依存的に膨張の低下を示した。AUCの低下は、0.1μgで−13.9%(P<0.05)、1μgで−16.3%(P<0.05)、10μgで−27.9%(P<0.05)、および100μgで−45%(P<0.01)であった。
GCを用いて置換したCpGモチーフを有するネガティブコントロールIDX0526を、試験Kに類似した用量応答プロトコルにおいて調査した。IDX0526を使用すると、抗膨張活性がなかったことが分かった。AUCにおける差異は、0.1μgで+1.24%、1μgで−1.24%、10μgで−0.26%、および100μgで+6.18%であり、統計的に有意な変化は全くなかった。
オリゴヌクレオチドの骨格におけるホスホロチオエート修飾の程度の有意性を調査するために、完全にホスホロチオエート化したオリゴヌクレオチド、IDX0500を使用した。この化合物は、すべての試験した用量において統計的に有意な抗膨張特性を示し、AUCの低下は、0.1μg −46.9%、1μg −41.3%、10μg −40.9%、および100μg −59.1%(すべてP<0.01)であった。
IDX0500の治療濃度域をさらに検討するために、試験Nを試みた。用量は、0.005μg、0.01μg、0.1μg、および1μgとした。化合物は、試験Mと一致して、用量0.1および1μgにおいて統計的に有意な効果を示した。AUCの低下は、0.005μgについて−15%(NS)、0.01μgについて−15.3%(NS)、0.1μgについて−18.2%(P<0.01)、および1μgについて−54.1%(P<0.01)であった。
本実験において、IDX0500およびIDX0150を、0日目にs.c.を施した、10および100μgの等用量で比較した。類似した結果が用量および2つの化合物の間で得られ、AUCの統計的に有意な低下を示し、IDX0500については、10μgで−25.5%、100μgで−39.9%、IDX0150については、10μgで−25.5%、100μgで−39.9%であった(すべての用量でP<0.01)。
ピーク値およびAUC値もまた、異なる治療計画の抗膨張有効性を階層化するために比較した。
本実験により、腹腔内、皮下、および鼻腔内経路を使用して、広範な予防的なおよび治療上の治療レジメンを検討した。さらに、用量応答を、化合物IDX0150およびIDX0500(それぞれ配列番号11および12)を使用して調査した。
試験A〜Oは、まとめると、マウスにおけるTPA誘発性の耳浮腫モデルにおける本発明の化合物の抗膨張効果を支持する。これらの化合物は、腹腔内、皮下、および鼻腔内投与経路によって与えた場合、有効な免疫調節剤となり、浮腫を低下させるためのそれらの効能はまた、図1〜3におけるIDX0150によって例示されるように、異なる予防および治療計画においても証明された。化合物の抗膨張効果は、一般的な炎症経路に限定されないが、たとえばプロスタグランジン系を活性化するAAによって誘発される炎症をダウンレギュレートすることもできる。試験化合物配列番号1〜10はすべて、TPAおよび/またはAAモデルにおいて示されるように浮腫を低下させることができた。
動物
B&K(Sollentuna、Stockholm、Sweden)から得たメスのBalb/cマウス(8週齢)を実験において使用した。マウスに、十分なペレットげっ歯動物用食餌、R36(Laktamin AB、Stockholm、Sweden)および国内の飲用に適した品質の水を適宜与えた。動物は、21℃±3℃および55%±15%の相対湿度の動物室で飼育した。換気システムは、1時間当たり10回、空気交換するように設計した。部屋は、12時間明および12時間暗のサイクルとなるように照明した。照明は07:00〜19:00の時間、つける。マウスは、それぞれのケージに5匹、透明なポリカーボネート(MacroloneタイプIII)ケージ(床面積:820cm2)において飼育した。ケージ中の敷床は4HVポプラ敷床(Tapvei、Finland)とした。それぞれのケージを、試験番号、グループ番号、性別、および動物番号を用いてマークしたケージカードによって識別した。
マウスは、0および12日目に、200μL OVA/水酸化アルミニウムゲル(1:3)を用いて腹腔内感作した(図6を参照されたい)。OVA(ニワトリ卵アルブミングレードV、Sigma、St.Louis、MO)を生理食塩水中に溶解し、3時間4℃での回転流動によって50μg/mLの濃度まで水酸化アルミニウムゲルと混合した。23、26、30、および33日目に(図6を参照されたい)、Batelle曝露チャンバーを使用して、30分間、エアロゾル化OVAの吸入によってマウスの肺中で誘発した。エアロゾルを、PBS中に溶解した10mg/mL OVA(Sigma、St Louis、MO、USA)の噴霧器濃度を使用して、気流7.4L/分で圧縮空気噴霧器(Collison 6−jet)によって生成した。非感作動物を有するコントロールグループは、23、26、30、および33日目のエアロゾル化OVA以外に治療を受けなかった。エアロゾル誘発を受けなかった感作マウスのコントロールグループもまた用意した。
このOVAモデルにおいて(図6)、合計4つのオリゴヌクレオチドを試験した、IDX9025、IDX9038、IDX9053(表1、配列番号2〜4))、およびIDX0150(表3、配列番号11)。オリゴヌクレオチドは、biomers.net GmbH、Ulm、Germanyによって合成され、−20℃で凍結保存された。オリゴヌクレオチドは、使用濃度(1,247μg/μL)にPBSを用いて希釈し、滴下の1日目まで−20℃に保ち、その後、それらを4℃にした。
第1の実験において、予防および治療プロトコルの両方を試験した(図6を参照されたい)。薬剤治療は、16および21日目(予防プロトコル)、30および33日目(治療プロトコルA)、または30および34日目(治療プロトコルB)のIDX0150(InDex Pharmaceuticals AB、Stockholm、Swedenによって提供される1,247μg/μL)の鼻腔内滴下からなった。30および33日目の治療プロトコル(A&B)における滴下は、エアロゾル誘発の4時間後に施した。34日目のプロトコルBにおける第2の滴下は、最後のエアロゾル誘発の24時間後に施した。薬剤は、40μL PBS中で投与し、約50μg/マウス(49.88μg/マウス)の用量を施した。2つの感作偽治療グループに、予防プロトコル(16および21日目)または治療プロトコルA(30および33日目)による治療グループと同じ全容量のPBSを滴下した。
最後のOVAエアロゾル誘発の42時間後に頚椎脱臼によってマウスを殺した。気管に、ポリエチレン管(Becton Dickinson、Sparks、MD、USA)を挿入し、気管支肺胞洗浄(BAL)を、4×1mL分割量の氷冷ハンクス平衡塩類溶液(HBSS)(Sigma、St Louis、MO、USA)を使用して行った。BAL体液を遠心分離し(400g、10分、4℃)、BAL体液細胞を0.4mL PBS中に再懸濁した。白血球の総数を、Buerkerチャンバー中で、トリパンブルー排除法を使用して数えた。BAL体液細胞の二連のCytospin(Cytospin3、Shandon、Runcorn、UK)調製物を、標準的な形態学的基準を使用して、白血球百分率について、May Gruenewald Giemsaを用いて染色した。
統計比較は、感作PBS治療コントロールマウスと比較するために、ダネットの事後補正を使用する分散(ANOVA)の一元配置分析を使用して行った(GraphPad Prism 3)。データは平均±標準偏差として示す。P<0.05は、有意であると考えた。
オボアルブミン誘発性のアレルギー喘息モデルは、喘息の間に見られる肺炎症を再現するために広く使用されるモデルである。ここで得られた結果は、気道の粘膜の膨張に相互に関連し、結果は、したがって、浮腫の低下の妥当な測定値である。このモデルの分析は、多形核細胞(PMN)などの浸潤炎症細胞のタイプおよび量が同定され、数えられるBAL分析などの喘息の一般的な指標に依存する。
本インビボ試験は、以下の結論を出す:BAL体液に浸潤する白血球、好酸球、およびリンパ球の数の統計的に有意な低下は、本発明の化合物を用いて治療した場合、動物において観察された。本モデルにおいて、炎症を測定することは、より適切であったが、結果は、浮腫に対して等しく適用可能である。さらに、試験は、有効な経路としての経鼻投与の適合性を確認した。
動物モデルは、細胞遊走および血管透過性に対する本発明の実施形態によるオリゴヌクレオチドの効果を検討するために設定した。
チオグリコレート誘発性の胸膜炎モデルは、技術的に複雑になり、時に個々の異なる値を示し得るが、開発中の新薬の実践的なスクリーニングに最適なモデルの1つである。しかしながら、このモデルは、同時に試験することができる動物の数が制限される。
胸膜炎動物モデルにおけるこれらの結果は、化合物が、さらなる試験および薬剤開発に適した好ましい治療プロファイルを有するという事実を指す。
