Abstract
Description
本発明者らは、M−HBsAgを検出する抗HBsのみ、またはS−HBsAgを検出抗HBsとの組合せが、HBV感染を診断する、より高感度かつ正確な手段を提供するという驚くべき発見をなした。M−HBsAgはカプシド全タンパク質のごく一部を構成するに過ぎないため、これは特に重要である。HBVを検出するための現在利用可能な方法は、より多量のS−HBsAgのみに対する抗HBsを利用することにより達成される。しかし、これらの抗HBs抗体試薬を使用する幾つかの商業的に入手可能なアッセイは、他の手段により検出可能なHBV感染が判明しているにもかかわらず、患者によってはS−HBsAgを検出し得ないことが見出されている。この偽陰性結果は、S−HBsAg配列を改変する1つの突然変異または一連の突然変異をそのHBV株が含有するため、該キット試薬がS−HBsAgを検出し得ないことによるものであろう。これらの突然変異は、抗HBsモノクローナル抗体試薬が、もはや、HBV感染を検出できなくなるよう、S−HBsAgエピトープに影響を及ぼしうる(Colemanら,J Med Virol.59:19−24(1999))。エピトープの改変はS−HBsAgに局在化しており、M−HBsAgドメインに有意には伸長していない。
本明細書中で用いる用語は、単に特定の実施形態を説明するためのものであるに過ぎず、限定的なものではない。本明細書および添付の特許請求の範囲において用いる単数形表現は、文脈に明らかに矛盾しない限り、複数形の対象物を含む。
本明細書中で用いる「抗体」は、クラスIgG、IgM、IgA、IgDもしくはIgFの抗体またはそれらのフラグメントもしくは誘導体、例えばFab、F(ab’)2、Fdおよび一本鎖抗体、ジアボディ、二重特異性抗体、二官能性抗体ならびにそれらの誘導体を意味しうる。抗体は、所望のエピトープまたはそれに由来する配列に対する十分な結合特異性を示すモノクローナル抗体、ポリクローナル抗体、アフィニティ精製抗体またはそれらの混合物でありうる。ポリクローナル抗体は、哺乳動物、例えばヒト、ヤギ、ウサギまたはヒツジに由来するものでありうる。抗体はまた、キメラ抗体でありうる。抗体は、当技術分野で公知の1以上の化学的部分、ペプチド部分またはポリペプチド部分の結合により誘導体化されうる。抗体は化学的部分にコンジュゲート化されうる。抗体は特異的結合メンバーでありうる。
ポリペプチドおよび固体支持体に言及するために本明細書中で用いる「結合」または「固定化」は、該ポリペプチドおよび該固体支持体の間の結合が、結合、洗浄、分析および除去の条件下で安定であるために十分なものであることを意味しうる。該結合は共有結合または非共有結合でありうる。共有結合は該ポリペプチドと該固体支持体との間で直接的に形成されることが可能であり、あるいは架橋剤により、または固体支持体もしくはプローブもしくはその両方の分子上に特異的反応性基を含有させることにより形成されうる。非共有結合は静電性、親水性および疎水性相互作用の1以上でありうる。非共有結合に含まれるのは、該支持体への例えばストレプトアビジンのような分子の共有結合、および該ストレプトアビジンへのビオチン化ポリペプチドの非共有結合である。固定化は共有的相互作用と非共有的相互作用との組合せを含みうる。
本明細書中で用いる「エピトープ」または「抗原」はポリペプチドの抗原決定基を意味しうる。エピトープは、該エピトープに特有の空間的コンホメーションにおける3アミノ酸を含みうる。エピトープは少なくとも5、6、7、8,9または10アミノ酸を含みうる。空間的コンホメーションの検査方法は当技術分野で公知であり、X線結晶学および二次元核磁気共鳴を包含する。
本明細書中で用いる「断片」は、言及されているペプチドまたはポリペプチドの一部分を意味しうる。
2以上のポリペプチド配列の文脈において本明細書中で用いる「同一」または「同一性」は、それらの配列が、特定された領域にわたって同一である特定された割合(%)の残基を有することを意味しうる。該割合は、2つの配列を最適に整列させ、特定された領域にわたってそれらの2つの配列を比較し、同一残基が両方の配列に存在する位置の数を決定して、一致した位置の数を得、一致した位置の数を、特定された領域における位置の総数で割り算し、その結果に100を掛け算して、配列同一性の割合(%)を得ることにより計算されうる。それらの2つの配列が、異なる長さのものである場合、または該整列(アライメント)が1以上の食い違った末端を与え、特定された比較領域が単一の配列のみを含む場合には、単一配列の残基を該計算の分母には含め分子には含めない。

本発明は、サンプル中のHBVタンパク質のレベルを決定するための方法を提供する。
HBVタンパク質はHBV表面抗原タンパク質でありうる。HBV表面抗原タンパク質は、HBV粒子の表面上に該HBVタンパク質を露出しうるHBVエンベロープの一部を形成する能力を有しうる。HBV表面抗原タンパク質は、抗原性でありうる又は免疫監視の標的でありうるエピトープを含みうる。エピトープは突然変異エピトープでありうる。HBV表面抗原タンパク質はグリコシル化されていることも可能である。
HBV表面抗原タンパク質はM−HBsAgでありうる。M−HBsAgは第1部分および第2部分を含むことが可能であり、約281アミノ酸の全長を有しうる。第1部分はプレS2領域を含むことが可能であり、M−HBsAgの最初の55アミノ酸でありうる。第2部分は226アミノ酸長であることが可能であり、S−HBsAgの配列を含みうる。M−HBsAgは、表1に記載されている配列またはその変異体を含みうる。M−HBsAgの第1部分はエピトープをも含みうる。エピトープは抗体により結合されうる。該抗体は抗M−HBsAg特異的抗体でありうる。
HBV表面タンパク質はまた、S−HBsAgでありうる。S−HBsAgは約226アミノ酸長であることが可能であり、S領域を含みうる。S−HBsAgは野生型S−HBsAgでありうる。S−HBsAgは、表2に記載されている配列またはその変異体を含みうる。S−HBsAgは、米国特許第5,925,512号または第7,141,242号(それらの内容を参照により本明細書に組み入れることとする)に開示されている「a」決定基の部分でありうるエピトープをも含みうる。S−HBsAgのアミノ酸100−160もエピトープを含みうる。
S−HBsAgは突然変異体であることも可能であり、これは、野生型S−HBsAgとは少なくとも1アミノ酸異なる配列を含みうる。該突然変異はS−HBsAgの抗原性に影響を及ぼしうる。該抗原性は「a」決定基の破壊により影響を受けうる。突然変異S−HBsAgの抗原性は軽減されることが可能であり、これにより、該突然変異S−HBsAgエピトープは免疫監視を逃れうる。該突然変異S−HBsAgは、野生型S−HBsAgと比べて、抗HBs抗体により検出される可能性が低いかもしれない。
本発明はまた、HBVタンパク質への結合能を有しうる抗体を提供する。該抗体は標識に結合していることが可能であり、固相に結合していることが可能である。該抗体はまた、第1抗体、第2抗体または第3抗HBV抗体でありうる。第1抗体、第2抗体および第3抗HBV抗体は全て、異なる位置またはエピトープにおいてHBVタンパク質に結合する能力を有しうる。第1抗体、第2抗体および第3抗HBV抗体はまた、お互いから識別されうる標識に結合することが可能である。本明細書中で用いる「第1抗体」、「第2抗体」および「第3抗体」は単に例示を目的としたものであるに過ぎない。本明細書に記載されている特性を有する他の抗体も、本明細書に記載されている方法のために使用されうる。
第1抗体は、M−HBsAgに結合する能力を有しうる。第1抗体は、M−HBsAgのエピトープに結合する能力を有しうる。第1抗体はまた、M−HBsAgのプレS2領域内のエピトープに結合する能力を有しうる。第1抗体は更に、M−HBsAgとS−HBsAgとを識別する能力を有しうる。第1抗体はS−HBsAgと交差反応し得ない。第1抗体はまた、S−HBsAgに対するより高いアビディティでM−HBsAgに対して結合する能力を有しうる。第1抗体は50−80または116−34モノクローナル抗HBV表面タンパク質抗体または類似抗体でありうる。
第2抗体は、S−HBsAgに結合する能力を有しうる。第2抗体はまた、M−HBsAgのS領域に結合する能力を有しうる。第2抗体はH166、H57、H40、H53もしくはH35モノクローナル抗HBs抗体または類似抗体でありうる。
HBsAgのレベルは、サンプルを第1抗体と接触させることにより決定されうる。第1抗体を、タンパク質/抗体複合体の形成に十分な時間及び条件下、該サンプルと接触させることが可能である。
該方法はまた、サンプル中のS−HBsAgのレベルを決定することを含みうる。これは、サンプルをモノクローナル抗体と接触させることによるものでありうる。S−HBsAgのレベルは、該標識により生成された測定可能なシグナルを検出することにより決定されうる。サンプル中のS−HBsAgの量は、生成したシグナルに正比例しうる。該方法は、HBsAgのレベルを決定するための前記のものに類似した工程を含みうる。

本発明においては、HBVタンパク質を含みうるサンプルが提供される。該サンプルはHBsAgを含むことが可能であり、S−HBsAg、M−HBsAgまたはL−HBsAgを含むことも可能である。該サンプルは更に、M−HBsAgおよびS−HBsAgを、1000:1および1:1000の範囲内でありうる既知比率で含みうる。
本発明は、HBVタンパク質、例えばS−HBsAg、M−HBsAgまたはL−HBsAgのレベルを決定することによるものでありうる、HBVの検出方法を提供する。S−HBsAg、M−HBsAgまたはL−HBsAgのレベルを所定のS−HBsAg、M−HBsAgまたはL−HBsAg値と比較することが可能であり、これはサンプル中のHBVの指標となりうる。2つの異なるHBVタンパク質のレベルの比を計算することが可能である。所定の比の値と比較した該比における相違はサンプル中のHBVタンパク質の指標となりうる。
所定のM−HBsAg値は陽性M−HBsAg対照サンプル中のM−HBsAgのレベルでありうる。該M−HBsAg対照サンプルは既知量のM−HBsAgを含みうる。該M−HBsAg対照サンプルはHBVをも含みうる。所定のM−HBsAg値はまた、陰性M−HBsAg対照サンプル中のM−HBsAgのレベルでありうる。
本発明はまた、HBVを検出することによるものでありうる、突然変異S−HBsAgを検出するための方法を提供する。所定の値の又は所定の値を超えるM−HBsAgのレベル、ならびに所定のS−HBsAg値より小さいS−HBsAgのレベルは、突然変異S−HBsAgの存在の指標となりうる。
所定のS−HBsAg値は、S−HBsAgを含むことが判明している又は既知量のS−HBsAgを含む陽性S−HBsAg対照サンプルにおけるS−HBsAgのレベルでありうる。そのような所定のS−HBsAg値はまた、その値より小さいS−HBsAgのレベルが突然変異S−HBsAgの存在を示すような値でありうる。所定のS−HBsAg値は1×10−12〜1×10−2gmsの範囲内でありうる。
所定のL−HBsAg値は、L−HBsAgを含むことが判明している又は既知量のL−HBsAgを含む、陽性L−HBsAg対照サンプルにおけるL−HBsAgのレベルでありうる。
本発明は、HBVを検出するために使用されうるキットを提供する。該キットは第1抗体を含むことが可能であり、第2抗体をも含みうる。該キットは、サンプルからのタンパク質に結合するのに適した固体支持体をも含みうる。該キットは更に、陽性対照として使用するための既知濃度のHBVタンパク質を含むHBV組成物を含みうる。該陽性対照は、所定の値を測定するために使用されうる。
材料および方法:
以下の方法を用いて、モノクローナル抗HBs抗体(H53または116−34)をポリスチレン微粒子に結合させた。抗体を15mM MESバッファー(pH4.7)中で約400μg/mlに希釈した。ポリスチレン微粒子を、0.01% Tween20を含有する15mM MESバッファー(pH4.7)中で懸濁させ、洗浄した。1% 固体の微粒子を15mM MESバッファー(pH4.7)中の0.04mg/ml EDACで活性化し、抗HBs抗体を200μg/mlの最終濃度で加えた。該混合物を30分間インキュベートし、ついでリン酸緩衝食塩水(pH7.4)で洗浄した。該粒子をイムノアッセイのために0.1% 固体の最終濃度に調節した。
参照IMx HBsAg、微粒子捕捉相上に116−34またはH53を含有する2つの試験アッセイ条件で、臨床サンプルを試験した。臨床サンプルを、より低い抗原濃度に、正常ヒト血清中で1:10希釈した。アッセイ結果を図5に示す。116−34試験形態に関するs/co値は、HBsAgの免疫捕捉のためにH166を使用する参照アッセイまたはHBsAgの免疫捕捉のためにH53を使用する試験アッセイに関する値より有意に高かった。このデータからの結論は、プレS2モノクローナル捕捉形態がHBsAgの野生型形態に対する微粒子アッセイ感度を増加させうることである。
材料および方法:
以下の方法を用いて、モノクローナル抗HBs抗体(H166、H57、H35、H53および116−34)をポリスチレン微粒子に結合させた。抗体を15mM MESバッファー(pH4.7)中で約400μg/mlに希釈した。ポリスチレン微粒子を、0.01% Tween20を含有する15mM MESバッファー(pH4.7)中で懸濁させ、洗浄した。1% 固体の微粒子を15mM MESバッファー(pH4.7)中の0.04mg/ml EDACで活性化し、抗HBs抗体を200μg/mlの最終濃度で加えた。該混合物を30分間インキュベートし、ついでリン酸緩衝食塩水(pH7.4)で洗浄した。該粒子をイムノアッセイのために0.1% 固体の最終濃度に調節した。
プレS2抗HBsモノクローナル116−34またはS−HBsAgモノクローナル(H166、H57、H35、H53)を捕捉試薬として使用するIMx HBsAgアッセイ形態において、サンプルを試験した。アッセイ結果を図6に示す。全体的に、116−34試験形態に関するs/co値は、HBsAgの突然変異形態の免疫捕捉のためのS−HBsAgモノクローナルに関する値より高かった。このデータからの結論は、プレS2モノクローナル捕捉形態がHBsAgの突然変異形態に対する微粒子アッセイ感度を増加させうることである。
材料および方法:
既に記載されているとおりに(Colemanら,J Med Virol.59:19−24(1999))、HBsAg突然変異体を製造した。簡潔に説明すると、同じ出発野生型配列内に導入された種々の一定の突然変異を含有する表面抗原遺伝子配列を、所有権を有する発現ベクターのXhoIおよびHpaI制限部位内にクローニングした。挿入された遺伝子を配列決定により確認し、確認された発現ベクターを使用して、リポフェクタミンの存在下、85%のコンフルエンシーでマウスL細胞を一過性にトランスフェクトした。組換え抗原の発現に関して、細胞培養上清を第3日にモニターした。Abbott Ausriaアッセイを用いて、初期定量を行った。なぜなら、このアッセイのポリクローナル捕捉およびポリクローナル検出試薬形態は全ての突然変異体を検出することが可能だったからである。細胞培養上清を正常ヒト血漿中に希釈し、HBsAgの存在に関して試験した。
2つ組換えHBsAg突然変異体(アミノ酸122位にAsn Thrの挿入を含有するHBsAg突然変異体1208およびアミノ酸123位にThrからAlaへの置換を含有するHBsAg突然変異体T123A)を、この研究において評価した。また、野生型HBsAgを含有するAuszyme陽性対照を2つの濃度で試験した。そのAuszymeの結果および抗HBsモノクローナル反応性の結果を図7に示す。該抗HBsコート化ビーズの全てが該野生型陽性対照を認識したが、それらは2つのHBsAg突然変異体に対しては異なる反応性を示した。両方の突然変異体に関して最高のA(492nm)読取値を有する抗HBsモノクローナル抗体はM−HBsAgのプレS2領域に対する116−34であった。該116−34シグナルは、その他の抗HBsモノクローナルまたはAuszymeキットシグナルより、有意に、より高感度であった。H53と116−34とのモノクローナル組合せはHBsAg突然変異体1208を最も良く検出し、一方、H57と116−34とのモノクローナル組合せはHBsAg突然変異体T123Aを最も良く検出するであろう。このデータからの結論は、プレS2モノクローナル捕捉形態がHBsAgの突然変異形態に対するアッセイ感度を増加させることである。
Claims (17)
- (a)試験サンプルを準備し、
(b)中B型肝炎表面タンパク質(M−HBsAg)/第1抗体複合体を形成するのに十分な時間及び条件下、該試験サンプルを第1抗体と接触させることを含んでなり、該複合体の存在が、該サンプル中に存在するB型肝炎の検出を示す、試験サンプル中のB型肝炎を検出するための方法。 - M−HBsAgのアミノ酸1−55が、第1抗体により認識されるエピトープを含む、請求項1記載の方法。
- 第1抗体が小B型肝炎表面タンパク質(S−HBsAg)と交差反応しない、請求項2記載の方法。
- 小B型肝炎表面タンパク質(S−HBsAg)/第2抗体複合体を形成するのに十分な時間及び条件下、該試験サンプルを第2抗体と接触させる工程を更に含み、該複合体の存在が、該試験サンプル中に存在するB型肝炎の検出を示す、請求項1記載の方法。
- S−HBsAgのアミノ酸100−160が、第2抗体により認識されるエピトープを含む、請求項4記載の方法。
- (a)試験サンプルを準備し、
(b)中B型肝炎表面タンパク質(M−HBsAg)/第1抗体複合体を形成するのに十分な時間及び条件下、該試験サンプルを第1抗体と接触させ、
(c)小B型肝炎表面タンパク質(S−HBsAg)/第2抗体複合体を形成するのに十分な時間及び条件下、該試験サンプルを第2抗体と接触させ、
(d)M−HBsAg/第1抗体複合体のレベルをS−HBsAg/第2抗体複合体のレベルと比較することを含んでなり、M−HBsAg/第1抗体複合体と比べて低い、S−HBsAg/第2抗体複合体のレベルが、該試験サンプル中の突然変異S−HBsAgの検出を示す、試験サンプル中の突然変異S−HBsAgを検出するための方法。 - M−HBsAgのアミノ酸1−55が、第1抗体により認識されるエピトープを含む、請求項6記載の方法。
- 第1抗体が小B型肝炎表面タンパク質(S−HBsAg)と交差反応しない、請求項6記載の方法。
- S−HBsAgのアミノ酸100−160が、第2抗体により認識されるエピトープを含む、請求項6記載の方法。
- (a)試験サンプルを準備し、
(b)小B型肝炎表面タンパク質(S−HBsAg)/第1抗体複合体を形成するのに十分な時間及び条件下、該試験サンプルを第1抗体と接触させることを含んでなり、対照と比べて低い、S−HBsAg/抗体複合体のレベルが、該試験サンプル中の突然変異S−HBsAgの検出を示す、試験サンプル中の突然変異S−HBsAgを検出するための方法。 - 第1抗体が突然変異小B型肝炎表面タンパク質(S−HBsAg)と交差反応しない、請求項10記載の方法。
- (a)小B型肝炎表面タンパク質(S−HBsAg)/第2抗体複合体を形成するのに十分な時間及び条件下、該試験サンプルを第2抗体と接触させ、
(b)S−HBsAg/第1抗体複合体のレベルをS−HBsAg/第2抗体複合体のレベルと比較する工程を更に含み、第2抗体により検出されたS−HBsAgのレベルに対する、第1抗体により検出されたS−HBsAgのレベルの比における、所定の比と比較した場合の相違が、該試験サンプル中の突然変異S−HBsAgの検出を示しうる、請求項10記載の方法。 - (a)M−HBsAgに結合する少なくとも1つの抗体、および
(b)S−HBsAgに結合する少なくとも1つの抗体を含んでなる、B型肝炎を検出するためのキット。 - M−HBsAgのアミノ酸1−55が、(a)の少なくとも1つの抗体により認識されるエピトープを含む、請求項13記載のキット。
- 前記の少なくとも1つの抗体がS−HBsAgと交差反応しない、請求項13記載のキット。
- S−HBsAgのアミノ酸100−160が、(b)の少なくとも1つの抗体により認識されるエピトープを含む、請求項13記載のキット。
- 該B型肝炎の検出のための説明を更に含む、請求項13記載のキット。
