最近、合成ペプチドと組み合わせた組換えMHC分子の可溶性複合体を開発する技術が使用し始めている[Kern,F.et al.,(1998)Nature Medicine 4:975−978;Kwok,W.W.et al.,(2001) Trends in Immunol.,22:583−588]。これらの試薬及び手順は、特定のMHCペプチド複合体を結合することができ及び広い多様なMHCアロタイプに対するスクリーニング多重可能性エピトープに適合しないヒト又は実験動物被検体由来の末梢血試料からT細胞クローンの存在を同定するのに使用される。
本明細書に記載の毒素分子は、組換え方法の使用を含め幾つかの方法のいずれかで調製できる。本明細書において提供されるタンパク質配列及び情報は、アミノ酸配列をコードするポリヌクレオチド(DNA)を推定するのに使用できる。これは、例えば、DNSstar software suite[DNAstar Inc,Madison、Wis.,USA]などのコンピューターソフトウエアツールを使用して達成できる。ポリペプチドをコードするこのようなポリヌクレオチド又はその重要なホモローグ、バリアント、切断、伸長もしくは別の修飾が、本明細書において意図される。
i)ナイーブT細胞活性化アッセイ及び関心のタンパク質配列を集合的に包含する合成ペプチドを使用して、T細胞を活性化することができるエピトープ領域を同定する工程と、
ii)ペプチドリガンドと1つ又はそれ以上のMHCアロタイプとの結合をシミュレートする計算スキームを使用して、工程(i)で同定されたエピトープ領域を解析し、それによってエピトープ領域内にあるMHCクラスIIリガンドを同定する工程と、
iii)ペプチドリガンドと1つ又はそれ以上のMHCアロタイプとの結合をシミュレートする計算スキームを使用して、もはやMHCクラスIIを結合しないか又はより少ない数のMHCアロタイプに対して低い親和性をもって結合するエピトープ領域内に含まれるMHCリガンドの配列類縁体を同定する工程と、及び場合により
iv)関心のタンパク質内の同定されたエピトープ領域を完全に包含するか又は収集において包含し、ナイーブT細胞活性化アッセイにおける配列類縁体を野生型(親)配列と同時に試験するナイーブT細胞活性化アッセイ及び合成ペプチドを使用する工程と、
からなる。
DT T細胞エピトープ
また、1個又はそれ以上のT細胞エピトープに少なくとも1個の修飾を有するDTバリアントが、本明細書において提供される。7個のT細胞エピトープが、本明細書に記載の方法及びさらに実施例9に記載の方法によってDT内で同定された。7個のT細胞エピトープは、配列番号:181、184、187、190、193、196及び199に示すようなジフテリア毒素アミノ酸配列を含有する。本明細書に記載のように、配列番号:181は、配列番号:2又は200のアミノ酸残基1−27に対応し、配列番号:184は、配列番号:2のアミノ酸残基85−117に対応し、配列番号:187は、配列番号:2又は200のアミノ酸残基95−127に対応し、配列番号:190は、配列番号:2又は200のアミノ酸残基104−136に対応し、配列番号:193は、配列番号:2又は200のアミノ酸残基112−144に対応し、配列番号:196は、配列番号:2又は200のアミノ酸残基136−168に対応し及び配列番号:199は、配列番号:2又は200のアミノ酸残基286−318に対応する。これらのエピトープは、さらに、エピトープ(9量体結合レジスター)及び隣接アミノ酸残基のMHCクラスII結合のための最も好ましい結合レジスターであるコア9量体アミノ酸配列を含有する。コア9量体結合レジスターは、一次エピトープであると考えられるが、9量体結合レジスターの外側に配置された残基は、MHCクラスII分子と相互作用し且つペプチド/MHCクラスII複合体の安定性を支えることが明らかにされている。7個の同定されたDT T細胞エピトープのコア9量体結合レジスターは、配列番号:161、164、167、169、173、175及び178のアミノ酸配列を有する。
抗原内に認められる個々のエピトープは、特異的MHCクラスIIアロタイプによって優先的に提示されることができ、同様に同じ抗原内の他の特異的エピトープは、MHCクラスII分子上に提示されないかもしれない。特定のエピトープと特異的MCHクラスII分子とのこのような会合は、個々のMHCクラスIIアロタイプに依存することが明らかにされている。特異的エピトープと特異的アロタイプとの会合もまた、T細胞エピトープの除去のためにDTを修飾する場合に認めることができる。このような考察は、特異的アロタイプについて(例えば、ある種のMHCクラスIIアロタイプを有する被検体の特異的集団について)DT分子の極めて特異的な修飾を可能にすることができる。1人又は複数の被検体のMHCクラスIIアロタイプは、当該技術において知られているゲノタイピング法で容易に決定でき、従ってDT T細胞エピトープと所定のアロタイプの会合は、そのアロタイプに適合した毒素修飾において考察するために、容易に同定される。DT T細胞エピトープとMHCクラスIIアロタイプとの間の会合の同定は、実施例9、表5及び図12に示す。本明細書において、毒素の所定のエピトープについて同定されたMHCクラスII会合に適合したT細胞エピトープ修飾を有する修飾毒素が意図される。
本明細書に記載のようなT細胞エピトープの同定及び修飾の他に、B細胞エピトープの同定及び修飾は、毒素の免疫原性をさらに低下させることができる。血清学的方法、コンピューター法、又は両方の方法の組み合わせを、毒素又は修飾毒素内のB細胞エピトープを同定するのに使用できる。血清学的方法は、被検体の免疫原分子、例えばタンパク質、ペプチド及びポリペプチドに対して抗体を生じる能力を用いる。毒素又は修飾毒素が、例えばワクチン接種用のように集団に予め投与されているものである場合には、被検体において生じた抗体反応は、B細胞エピトープについて毒素又は修飾毒素を選別するのに使用できる。一つの限定されない例において、本明細書に開示されているような修飾されたそのT細胞エピトープを既に有している毒素(例えば、ジフテリア毒素)は、B細胞エピトープを同定するのにさらに選別することができる。修飾毒素の配列に基づいたペプチドライブラリーが合成でき、毒素に対する抗体を含有する毒素ワクチン接種ドナー由来の血清が、ペプチドライブラリーのペプチドに結合する能力について試験することができる。種々の方法及びアッセイ、例えば、以下に限定されないが、ELISA、RIA及びウェスタンブロッティング法は、当該技術において周知であり、毒素ワクチン接種ドナーの血清由来の抗体に結合する1個又はそれ以上のペプチドを同定するのに使用できる。ドナー血清中で抗体を結合するこれら1個又はそれ以上のペプチドは、毒素配列内でB細胞エピトープを提示する。修飾毒素B細胞エピトープの同定の後に、B細胞エピトープに対応するアミノ酸残基は、修飾することができ、修飾毒素はドナー血清に対して再選別される。このプロセスは、修飾毒素の免疫原性をさらに低下させるために多数回行うことができる。本明細書においてT細胞エピトープの同定について認められるように、公知のB細胞エピトープデータベース及び予測タンパク質モデル化プログラムを利用するコンピューター法を、毒素又は修飾毒素内のB細胞エピトープを同定するのに用いることができる。このようなエピトープがコンピューター法で同定され、修飾されると、修飾毒素は、本明細書に記載のようなワクチン接種ドナー血清に対して選別することができる。コンピューターB細胞エピトープ選別法は、単独で使用できるし、又は毒素内のB細胞エピトープの同定及び修飾のためのペプチドライブラリーB細胞エピトープ選別と組み合わせて使用できる。場合により、修飾毒素は、B細胞エピトープ選別に先立って及び又はそれに続いて選別することができる。また、本明細書において、B細胞エピトープの修飾中に生じているかもしれない任意のT細胞エピトープの存在についてのB細胞エピトープ修飾毒素の選別も意図される。
細胞損傷、特に内皮細胞損傷は、ヘビに咬まれたことなどによる毒素又は敗血症性ショックを生じる分子、あるいは治療薬、例えば免疫毒素又はインターロイキンによって生じるか否かに関わらず、患者にとって未だに問題である。
V.T細胞エピトープ、B細胞エピトープ及びVLSモチーフの修飾を有する毒素
T細胞及びB細胞エピトープの修飾によって免疫原性を低下させる方法並びに本明細書に記載のようなVLSモチーフの修飾によって血管漏出症候群を軽減させる方法の他に、毒素及び修飾毒素は、低下した免疫原性(T細胞、B細胞、又はこれら両方)及び血管漏出症候群を引きこす能力の低下(すなわち、内皮細胞及び血管内皮細胞に対して低下した結合並びに内皮細胞間結合の崩壊の減少及び本明細書に記載のような血管漏出症候群の他の適応症)を示すポリペプチドを生成するために両方の修飾を含むことができる。
本明細書で使用する「毒素」という用語は、任意の抗細胞薬を指し、以下に限定されないが、細胞毒素及び/又は抗細胞薬の任意の組み合わせを包含する。ある態様において、毒素は、例えば、植物毒素、真菌毒素、細菌毒素、リボソーム不活性化タンパク質(RIP)又はこれらの組み合わせである。毒素としては、以下に限定されないが、アブリンA鎖、ジフテリア毒素(DT)A鎖、シュードモナス菌外毒素、RTA、志賀毒素A鎖、志賀様毒素、ゲロニン、モモルジン、ポークウィード抗ウイルスタンパク質、サポリン、トリコサンチン、オオムギ毒素及び当該技術において公知の種々のその他の毒素が挙げられる。本明細書で使用される毒素は、具体的にはブドウ球菌エンテロトキシン毒素B(SEB)、ヒルジン及びボウガニンタンパク質を除外する。
本発明はまた、修飾毒素融合タンパク質を提供する。修飾毒素ポリペプチドは、例えば、非毒素ポリペプチドに融合させることができる。一つの実施形態において、非毒素ポリペプチドは、細胞特異的結合リガンドである。本発明において使用される特異的結合リガンドは、リガンド全体を含有するか、又はリガンドの結合ドメイン全体を含むリガンドの部分を含有するか、又は結合ドメインの有効部分を含有することができる。リガンド分子の結合ドメインの全部又は大部分を含有することが最も望ましい。一つの限定されない例において、DT融合タンパク質は、DT関連ポリペプチド(例えば、本明細書に記載の修飾DT)及び相互にインフレームで融合された非DTポリペプチドを含有する。DT関連ポリペプチドは、低下した免疫原性又は修飾T細胞エピトープ、ヒト血管内皮細胞に対する低下した結合、又はこれらの組み合わせを有するDTバリアントの全体又は部分に対応する。一つの実施形態において、DT融合タンパク質は、配列番号:4−147の一つに示される修飾DT配列の少なくとも1つの部分を含有する。別の実施形態において、DT融合タンパク質は、少なくとも1個のT細胞エピトープ修飾を含有する。さらに別の実施形態において、DT融合タンパク質は、少なくとも1個のT細胞エピトープの修飾と、配列番号:4−147の1つ又はそれ以上に示す少なくとも1個の修飾とを含有する。さらに別の実施形態、DT融合タンパク質は、少なくとも1個のT細胞エピトープの修飾と、配列番号:4−147の1つ又はそれ以上に示す少なくとも1個の修飾と、少なくとも1個のB細胞エピトープ修飾を含有する。
本発明の別の態様は、修飾されたDTバリアント又はその融合タンパク質をコードするポリヌクレオチド(核酸、DNA)を含有するベクターに関する。
本発明は、標的エピトープ、例えば罹患組織又は疾患を引き起こす細胞で発現されたエピトープ(例えば、癌細胞表面のIL−2受容体)に対する融合タンパク質を提供する。ある実施形態において、融合タンパク質は、本明細書に記載の修飾されたDTを含有する。別の実施形態において、融合タンパク質は、さらに第二の物質を含有する。このような物質は、例えば、リポーター分子又は検出可能な標識などの分子又は部分であり得る。リポーター分子は、アッセイを使用して検出できる任意の部分である。ポリペプチドに複合されているリポーター分子の限定されない例としては、酵素、放射標識、ハプテン、蛍光標識、リン光分子、化学発光分子、発色団、発光分子、光親和性分子、着色粒子又はリガンド、例えばビオチンが挙げられる。検出可能な標識としては、その特異的な機能特性、及び/又は化学的特性によって検出できる化合物及び/又は要素が挙げられ、これらの使用は、これらが結合されたポリペプチドを検出すること及び/又は所望ならば定量することを可能にする。多数の適切な検出可能な(造影)物質が、ポリペプチドにこれらを付着させる方法があることから、当該技術において知られている(例えば、米国特許第5,021,236号、同第4,938,948号及び同第4,472,509号公報参照、それぞれは、参照することにより本明細書に組み込まれる)。使用される造影部分は、常磁性イオン、放射性同位元素、蛍光色素、NMR検出可能物質、X線造影部分であり得る。アジド基を含有する分子も、低強度紫外線によって生じる反応ナイトレン中間体によってタンパク質に対して共有結合を形成するのに使用できる(Potter & Haley、1983)。特に、プリンヌクレオチドの2−及び8−アジド類縁体が、粗細胞抽出物中のタンパク質を結合するヌクレオチドを同定するために、部位特異的光プローブとして使用されている(Owens & Haley,1987;Atherton et al.,1985)。2−及び8−アジドヌクレオチドもまた、精製タンパク質のヌクレオチド結合ドメインを位置決めするために使用されており(Khatoon et al.,1989;King et al.,1989;及びDholakia et al.,1989)、ポリペプチド結合剤として使用できる。
本明細書に記載の化合物のそれぞれは、許容し得る担体又は賦形剤と組み合わせた場合には組成物として使用できる。このような組成物は、生体外での分析に有用であるし、あるいは開示化合物を用いて被検体を治療するために生体内で又は生体外で被検体に投与するのに有用である。
XI.パッケージ及びキット
さらに別の実施形態において、本出願は、前記の化合物を使用するためのキットに関する。低下した免疫原性、VLS促進又は毒性効果、あるいはこれらの組み合わせを示す毒素はキットで提供できる。このようなキットは、すぐ使用でき且つ保存できる容器において、細胞表面の特定の受容体(例えば、癌細胞表面のIL−2受容体)を標的とする融合タンパク質を生成させるために、毒素を特異的細胞結合リガンドと組み合わせるのに使用し得る。従って、キットは、適当な容器手段に、低下した免疫原性、VLS促進又は毒性効果、あるいはこれらの組み合わせを有する組成物を含有するであろう。キットは、適当な容器手段に、修飾DT又はその融合タンパク質を含有し得る。
また、本明細書において提供されるパッケージ又はキットは、さらに、本明細書において提供される他の部分のいずれか、例えば1個又はそれ以上のリポーター分子及び/又は1個又はそれ以上の検出可能な部分/物質を含むことができる。
DTバリアント及びDT融合タンパク質の組み立て
N末端のメチオニン残基と、未変性DTのアミノ酸残基1−386(配列番号:2)(ここでは先端切断担毒体の残基2−387)とを含有する切断DT型担毒体を、DT387又はDT387の残基1−382として組み立てる。また、DT型担毒体は、N末端のメチオニン残基と、未変性DTのアミノ酸残基1−386(配列番号:2)(ここでは先端切断担毒体の残基2−387)と、未変性DTの残基484−485とを含有することができ、DT389として組み立てられる。DT387及びDT389は、残基7−9(VDS)、残基29−31(VDS)及び残基290−292(IDS)に3つの(x)D(y)モチーフを含有する。DT382は、DT387又はDT389の残基1−382を含有する。本明細書に記載のような他のC末端切断DT構造体が、DTバリアントの機能性を試験するために本明細書において提供されるアッセイで使用できる。DT389に行った修飾は切断構造体(例えば、DT382)でも行うことができ、機能性について試験できることが理解されるであろう。図31に、野生型DT382、DT382バリアント及びヌル構造体DT382(G53E)のアミノ酸配列を示す。下線を付した配列は、ベクター/標識配列であり、エンテロキナーゼ切断部位をイタリック体で強調し、及びWT配列由来の突然変異をボールド体で示す。
切断及び/又は修飾DTタンパク質、修飾DT突然変異体及び修飾DT融合タンパク質をコードするプラスミド構造体を、大腸菌HMS174(DE3)細胞内に形質転換する。大腸菌HMS174は、組換えタンパク質の過剰発現を達成できるプロテアーゼ欠失株である。組換えタンパク質の発現の誘導は、大腸菌HMS174にイソプロピルチオガラクトシダーゼ(IPTG)を加えることによって得られる。インキュベーションの後に、細菌細胞を遠心分離によって収穫し、溶解し、組換えタンパク質を、MurphyとvanderSpekによって報告された封入体製剤(Methods in Molecular Biology,Bacterial Toxins:methods and protocols,145:89−99 Humana press,Totowa,N.J.(2000))からさらに精製する。HUVECに対する効果がVLSに由来し、内毒素が存在することによらないことを確実にするためにタンパク質製剤から内毒素を除去することが必要であり得る。内毒素は、イオン交換樹脂を通すことによって<250 EU/mlまで除去される。全長物質から分解産物の分離が、イオン交換クロマトグラフィー中に生じる。イオン交換樹脂を用いた別の最終精製の後に、内毒素は、<25 EU/mlに減少し、担毒体は、VLSについて生体外でのHUVEC細胞結合の関数として試験される。DT387又はDT389担毒体の分析は、前記の方法からの試料が本明細書に記載の及び当該技術で公知の慣用の方法を使用してSDSポリアクリルアミドゲル電気泳動(PAGE)で分離される場合には、クーマシーブルー染色及びウェスタンブロットを使用して行うことができる。
ヒト血管内皮細胞をEGM培地(Cambrex,Walkersville,Mdから得られる)中に保持する。サブコンフルエント初期継代細胞を、プラスチック製カバースリップ上に、同等のT細胞数で接種する。精製した、内毒素を含有していない野生型DT担毒体及び突然変異体を、化学的結合によって蛍光標識F−150(Molecular Probes,Eugene,Oreg)で標識する。HUVECを、等量の標識担毒体と共にインキュベートする。次いで、培地を吸引し、次いで細胞を洗浄し、固定し、分析のために調製した。異なる処理群からのカバースリップ上の細胞の検査は、蛍光担毒体で標識された細胞の数の分析を可能にする。標的リガンドは担毒体表面に存在せず、従ってHUVEC相互作用のレベルは、HUVECに対する担毒体親和性にのみ比例する。比較を、蛍光顕微鏡を使用し、少なくとも10個の独立した領域、異なるカバースリップ又は異なるスライドから標識された細胞の数を比較することによって行った。特に突然変異体構造体の場合には、細胞標識は容易に見えないので、DAPI染色剤を使用して細胞を局在化する。4’−6−ジアミノ−2−フェニルインドール(DAPI)は、天然二本鎖DNAと蛍光複合体を形成することが知られている。それ自体DAPIは、細胞化学的検討における有用なツールである。DAPIがDNAに結合すると、その蛍光は強く増強される。従って、DAPIは細胞核を標識する方法として役立つ。それに対して、F−150DT担毒体で処理された細胞は、容易に観察される。突然変異体DT担毒体構造体の定量化を促進するために、シグナル強度及びバックグラウンドシグナルの変化もまた増大させる。
DT387又はDT389構造体を最初に使用して、修飾担毒体が多数の標的リガンドに化学的に連結することができ、機能的標的毒素を生成することを実証する。DT387リンカーIL−2又はDT389リンカーIL−2によって代表されるような一本鎖 融合毒素は、複合体毒素の発生において典型的に遭遇するスケールアップ精製の問題を回避する。操作された変化の効果を確認するために、多数の修飾DT387 IL−2融合毒素又はDT389 IL−2融合毒素を産生させ、細胞毒性アッセイで試験する。
細胞毒性アッセイを、HUT102/6TG細胞、すなわち高親和性インターロイキン−2受容体を発現するヒトHTLVl形質転換T細胞株を使用して行う。HUT102/6TG細胞を、10%ウシ胎児血清、2mMグルタミン、50IU/mlのペニシリン及び50μg/mlのストレプトマイシンを補足したRPMI1640(Gibco)培地に保持する。細胞を、5×l04/ウエルの密度で96ウエルのVマイクロタイタープレートに接種する。融合タンパク質毒素を、典型的にはウエルに10−7Mから10−12Mに至るまでの範囲のモル濃度で加える。ウエルの最終容量は、200μlである。プレートを、5%CO2環境で、37℃で18時間インキュベートする。このプレートを遠心分離に供して細胞をペレットし、培地を除去し、1.0μCi/ml[14C]ロイシン(<280mCi/ミリモル、Amersham−Pharmacia)及び21mMグルタミン、50IU/mlのペニシリン及び50μg/mlのストレプトマイシンを含有する200μlのロイシン無含有最小必須培地に置き換える。細胞を90分間振盪し、次いでプレートを遠心分離に供して細胞をペレット化する。上清を除去し、細胞を60μlの0.4M KOHに溶解し、次いで室温で10分間インキュベーションする。次いで、140μlの10%TCAを各ウエルに加え、さらに10分、室温インキュベーションを行う。沈降したタンパク質を、PHD細胞ハーベスターを使用してガラス繊維フィルターで収集し、取り込まれた放射能を、標準方法を使用して測定する。結果を、対照(タンパク質合成を阻害するために加えられた融合タンパク質はない)[14C]−ロイシン取り込みの割合として報告する。Toxilight(商標)、Vialight(商標)及びALAMARBLUE(商標)キットは、バリアントを評価するために使用できる非放射性市販アッセイである。アッセイは96ウエルプレートフォーマットで行い、感受性及び耐性細胞株を使用して時間にわたって毒素(10−7M〜10−12M)を滴定する。
種々の修飾DT融合タンパク質の細胞毒性活性は、CD22+Daudi細胞及び先に記載のような[3H]−ロイシン取り込みを使用して測定される(Ghetie et al.,1988)。未処理対照培養物に対して50%まで[3H]−ロイシン取り込みを減少させる融合タンパク質の濃度は、IC50として定義される。
修飾DTを用いて調製された融合タンパク質の血管損傷を誘発させる能力を評価する最初の段階として、HUVECを使用する一連の生体外試験を行うことができる。生体外アッセイについて、HUVEC単層の形態に対する修飾DT融合タンパク質の効果を、先に記載のようにして試験する(Baluna et al.,1996)。
修飾DT融合タンパク質の効果は、SCID/Daudi腫瘍モデルで調べることができる(Ghetie et al.,1992)。簡潔に言えば、雌性SCIDマウスに、当日に5×106個のDaudi細胞を静脈内注射する(I.V.、側尾静脈)。融合タンパク質を、1日目、2日目、3日目及び4日目にI.V.注射する。マウス5匹の群を、それぞれの処理に使用し、試験を反復する。処理群には、(1)未処理(対照)、(2)未修飾DT融合タンパク質、又は(3)未修飾DT融合タンパク質を受けさせる。マウスを追跡し,その後肢の麻痺が生じたときに犠牲死させる。SCIDマウスの肺血管漏出を記載のようにして評価する(Baluna et al.,1999)。肺の含水量を、10μg修飾DT融合タンパク質/マウス体重を注射したマウスから取り出した肺の湿潤/乾燥重量比として算出する。
段階2−VLS及びDT活性についてのアッセイ;
段階3−多数のバリアント(約100−250個のバリアント)をスクリーニングするのに適したフォーマットで、大腸菌での全DTの遺伝子合成、発現及び精製;
段階4−低下したVLS(HUVEC結合アッセイ)についてDTバリアントの生成及び試験−バリアントを2回試験する(単一遺伝子座/多重遺伝子座);
段階5−T細胞エピトープを除去した後のDTバリアントの生成及び試験−バリアントを複数回試験する(単一エピトープ、多重エピトープ、最適化)、第2回目の試験はVLSバリアントとの組み合わせを含む(段階4);
段階6−段階5からのリードDTバリアントと、タンパク質リガンド、例えばヒトIL−2との融合によるリードDT−融合バリアントの生成及びタンパク質精製/試験(この段階は任意である);並びに
段階7−EpiScreen(商標)(野生型先端切断DT(ΔR)の対照)による、リードDT1−389(「先端切断DT(ΔR)」)バリアントの免疫原性試験。
[EpiScreen用のドナー選択]
Addenbrooke病院地域研究倫理委員会(Addenbrooke’s Hospital Local Research Ethics Committee)によって許可された承認に従って英国国立血液サービス(UK National Blood Tranfusion Service)(Addenbrooke病院、Cambridge、UK)から得られた健康な地域社会ドナーの軟膜(24時間以内に採取した血液から)から、末梢血単核細胞(PBMC)を単離する。PBMCを、軟膜からLymphoprep(Axis−shield、Dundee、Scotland)密度勾配遠心分離によって単離し、CD8+T細胞を、CD8+RossetteSep(商標)(StemCell Technologies,Inc)を使用して欠乏させる。ドナーを、Biotest HLA SSP−PCRに基づいた組織適合キット(Biotest,Landsteinerstraβe、Denmark)を使用してHLA−DRハプロタイプを同定することによって特定する。また、対照抗原、キーホール・リンペット・ヘモシアニン(KLH)(Pierce,Rockford,USA)に対するT細胞応答も調べた。ドナー54人の集団を、社会集団(world population)において発現されるHLA−DRアロタイプの数及び頻度を最も良く表すために選択する(表1及び図1)。社会集団において発現されるアロタイプに対して、前記集団で発現されるアロタイプの分析により、>80%の範囲が達成されること及び全ての主要HLA−DR対立遺伝子(社会集団において発現される頻度>5%を有する個々のアロタイプ)が十分に提示されることが明らかにされた。表1は、ドナーのハプロタイプ、及びドナーPBMCの処理及び単離中に得られるKLHに対する応答(試験1)と、この試験中の再試験(ANG01)の後に得られるドナー応答との比較を示す。ドナーハプロタイプの概要を表2に示し、試験で使用したドナー アロタイプの頻度と、社会集団において存在する頻度との比較を図1に示す。
EpiScreen(商標)を使用して、C末端にヒトIL−2の10個のアミノ酸を有するDTのC及びIドメインを含むDT1−389の配列から誘導されるオーバーラップペプチドを試験した。C末端のヒトIL−2の10個のアミノ酸(DT1−389/IL−2 2−10)と一緒に、DT−Iの残基1−389に及ぶオーバーラップペプチドを設計した。12個のアミノ酸まで重複する一連の128×15量体ペプチドを、1×l4量体及び1×11量体と一緒に合成し、51人の健康なドナーの集団からEpiScreen(商標)T細胞エピトープマッピングを使用して誘導された末梢血単核細胞(PBMC)を刺激するのに使用した。個々のペプチドを、反復培養物中で試験し、T細胞増殖アッセイを使用して応答を評価して、エピトープの正確な位置を同定した。各ドナー由来のPBMCを解凍し、計数し、生存率について評価した。細胞を室温のAIM V培地(Invitrogen、Carlsbad,California)中で生き返らせ、その後に細胞密度を2.5×106PBMC/ml(増殖細胞ストック)に調整した。ペプチドを、10mMの最終濃度までDMSO(Sigma−Aldrich,St Louis,MO,USA)に溶解した。次いで、ペプチド培養ストックを、AIM V培地に5μMの最終濃度まで希釈することによって調製した。それぞれのペプチド及びそれぞれのドナーについて、6重反復培養物を、平底96ウエルプレートで100μlのペプチド培養ストックを100μlの増殖細胞ストックに加えることによって確立した。陽性対照培養物及び陰性対照培養物の両方も、6重反復で確立した。9×96ウエルプレート全部をそれぞれのドナーについて使用し、それぞれのプレートは、15個のペプチドを1つの陰性対照(担体単独)と共に6重反復で試験するのに十分であった。最終プレートに、陽性対照KLHを加えた。
増殖アッセイで陽性であったペプチドの配列を、Antitopeの予測iTope(商標)ソフトウエアを使用して解析した。このソフトウエアは、ペプチドのアミノ酸側鎖とMHCクラスII結合溝内の特異的結合ポケットとの間の好ましい相互作用を予測する。重要な結合残基の配置は、長いペプチド配列の上に及ぶする1個のアミノ酸によって重ね合わされた10量体ペプチドを生成することによって調べた。それぞれの10量体は、MHCクラスIIアロタイプ(合計32)のAntitopeのデータベースと対照して試験し、そのMHCクラスII分子との適合及び相互作用に基づいて採点した。多数の対立遺伝子に対して高い結合スコアを生じたペプチドは、コア9量体を含有するとみなした。このような試験は、(a)DT1−389/IL−2 2−10 DT1−389/IL−2 2−10の完全T細胞エピトープマップ、(b)個々のT細胞エピトープの効力及びDTから除去するための優先順位付けの評価、及び(c)MHCクラスIIアロタイプとのT細胞エピトープ関連性の評価を含めDT1−389/IL−2 2−10の包括的なT細胞エピトープ分析を提供した。
前記のEpiScreen(商標)分析を使用して同定された130のペプチド全部を、51人の健康なドナー(54人のドナーが最初に選択されたが、ドナー13、20及び38は、低い基礎cpmである、すなわち150cpmのカットオフ値よりも低いcpmであるために分析から除外した)に対して試験するために首尾よく合成した。陽性応答は、所定のペプチドに対してSI≧2を有する有意な(p<0.05)応答を生じたドナーによって定義される。ボーダーラインの応答(SI≧1.90を有する有意な(p<0.05)応答)もまた含めた。非調整データ解析の結果と調製データ解析の結果とを、アッセイ内変動性が低いこと及び陽性応答は個々のウエルでの偽性増殖の結果ではないことを確証するために比較した。それぞれの解析の結果は、方法の間にほとんど相違がないことを示した。従ってT細胞エピトープマップは、調整データ解析を使用して編集した。非調整解析と調製解析の両方からのドナー刺激指数を図12及び13それぞれに示す。T細胞エピトープは、試験での全てのペプチドに対する応答の平均頻度プラス標準偏差の2倍(「バックグラウンド応答率」と呼ぶ)を算出することによって同定した。これは、5.6%であると算出され、3人以上のドナーで陽性応答を誘導することについて同等であった。この閾値を上回る増殖反応を誘導するペプチドは、T細胞エピトープを含有するとみなした。
(a)エピトープ1−ペプチド2
ペプチド2(DDVVDSSKSFVMENF;配列番号:159)は、表2及び図2に示すようなT細胞エピトープを含有する。合計3人のドナー(30、36及び47)がペプチド2に応答したが、ドナー30の応答はボーダーラインであった(1.92のSI)。前記の明細全体を通じて考察するように、T細胞エピトープの排除は、同定されたエピトープのアミノ酸の修飾によって達成できる。エピトープのアミノ酸、例えばMHCクラスII結合裂隙のアンカーポケット(p1及びp9)を結合することに関連するアミノ酸残基の修飾は、MHCクラスII分子上のエピトープの首尾よい提示に影響を及ぼすことができる及び/又は防止する(すなわち、エピトープを排除する)ことができる。同様に、MHCクラスII結合裂隙の内部ポケットを結合することに関連するアミノ酸残基、及び/又はMHCクラスII分子に影響を及ぼすか又はそれと相互作用するコア9量体エピトープの外側の残基の修飾は、エピトープを排除できる。T細胞エピトープの排除のためのアミノ酸残基修飾の種々の組み合わせが、本明細書に記載の方法で調製でき、試験できる。
また、VDSSKSFVMコア9量体(配列番号:161)も、ペプチド2とオバーラップするペプチド1及び3の両方に存在する。興味深いことに、ドナー36は、ペプチド2及び3の両方に応答し(それぞれ2.13及び2.02のSI)、また統計学的に有意であった(p<0.05)ペプチド1(SI=1.88)に対する高い応答を有していて、VDSSKSFVM(配列番号:161)がコア9量体結合レジスターであることを示す。同様に、ドナー30は、1.64のSIでペプチド1に応答し、これは1.9〜2.0のカットオフよりも明らかに低かったが、統計学的に有意であり(p<0.05)、全体的なバックグラウンドSIよりも高かった。ドナー47は、ペプチド1及び3に対する陽性応答を開始しなかった。これは、おそらくはペプチド内のコア9量体の配置によるものと思われる。9量体結合レジスターの外側の残基の相互作用がペプチド:MHCクラスII複合体の安定性を裏付けることが十分に実証されている(Engelhard et al.,1994)。従って、ペプチド2は、コア9量体をMHCクラスIIの結合に最適な立体配置で含有していると思われる。DT−1の結晶構造の精査(Steere et al.,2000)により、p1バリン残基(V8)が部分的に露出した位置内にあることが明らかにされた。従って、極性置換残基は、極性部分が表面露出され且つ疎水性領域が覆い隠されるように選択できる。T細胞エピトープに対するこのような修飾は、毒素の免疫原性を低下させることができる。
表3及び図2は、4人のドナー(ドナー12、23、30及び36)がこの領域にT細胞エピトープの存在を示すペプチド31(KVLALKVDNAETIKK;配列番号:162)に応答したことを示す。また、オーバーラップペプチドの分析により、ドナー23がペプチド32に対してボーダーライン応答を生じ、ドナー36もペプチド32に応答し、その応答は、閾値下(SI=1.81)であるが統計学的に有意(p<0.05)であることが明らかにされた。iTope(商標)コンピューター解析を使用すると、このペプチドのコア9量体は、バリン(V97)を主要なp1アンカー残基として有するVDNAETIK(配列番号:164)であると予測された(図4)。これは、ドナー23のペプチド32に対する応答及びドナー36の同じコア9量体を含有していたペプチド32に対する閾値下の応答によって裏付けられた(図5)。この立体配座において、解析は、32個のMHCクラスII対立遺伝子の中の28個の結合を示した。構造及び相同性モデリングは、V97の配置が分子の表面に十分に露出されており且つ活性部位に関連づけられないことを明らかにした。
ペプチド35(配列番号:166)は、段階1において前記のように刺激の後に陽性増殖応答を生じた3人のドナー(ドナー1、2及び35)によって示されるようなT細胞エピトープを含有する(図2及び表3)。iTope(商標)による解析により、このペプチドについて最も好ましい結合レジスターは、ロイシン(L107)を主要なアンカー残基、p1として有するLGLSLTEPL(配列番号:167)であることが明らかにされた(図5)。この9量体は、32個のMHCクラスII対立遺伝子の中の13個を結合する可能性を有していた。ドナー35もまた、同じコア9量体を含有していたペプチド34に対して増殖応答(SI=1.83)を有していた。これは、陽性応答についてカットオフ1.9〜2.0より低かったが、統計学的に有意(p<0.05)であり、且つ配列LGLSLTEPL(配列番号:167)がこの領域内のエピトープであることを示す。
3人のドナー(ドナー5、15及び50)が、ペプチド39(LMEQVGTEEFIKRFG;配列番号:168)を用いた刺激の後に陽性増殖応答を生じたが、ドナー50の応答はボーダーライン(1.94のSI)であった(図2及び表3)。このペプチド内のT細胞エピトープは、iTope(商標)解析後にMEQVGTEEF(配列番号:169)をコア9量体MHCクラスII結合レジスターとして含有すると予測された(図6)。この立体配座において、位置116のメチオニンはp1アンカー残基を形成し、このエピトープは、32個のMHCクラスII対立遺伝子の中の19個を結合すると予測される。このMHCクラスII結合レジスターの位置1及び9は、毒素の触媒ドメインのコアの中に覆い隠され、互いに満たされる。従って、これらの残基は、タンパク質の全体的な安定性にとって重要であると考えられる。エピトープ3と同様に、コア9量体の他のポケット、例えばp6及びp7(これらは十分に露出されている)と相互作用する残基が、置換のために考慮される。
有力なT細胞エピトープは、ペプチド40、41及び42(配列番号:170−172)内にあると思われる。検出されたエピトープ全部について、この領域のエピトープは、試験集団の15.7%に相当する合計8人の異なるドナーにおいて応答を誘導するその能力によって示されるように、最も免疫原性である(表3及び図2)。6人のドナーが、ペプチド40(ドナー15、36、46、47、49及び50)、ペプチド41(ドナー35、36、40、46、49、50)及び42(ドナー35、36、40、47、49及び50)に応答し、3つのオーバーラップペプチド全部に対する応答がドナー49及び50について観察され、一方ドナー30、40、46及び47は、2つのオーバーラップペプチドに応答した。iTope(商標)コンピューター解析により、このペプチドについての結合モチーフはフェニルアラニン(F124)をP1アンカー残基として有するFIKRFGDGA(配列番号:173)であると予測された(図7)。この9量体は、3つのオーバーラップペプチド全部に存在しており、32個のMHCクラスII対立遺伝子の中の23個を結合すると予測された。フェニルアラニン124は、触媒ドメインのコア内に実質的に覆い隠され且つM115及びV118に対してパックし、従って潜在的に構造上重要であると考えられる。しかし、アンカーの位置4、6、7及び9は、種々の程度に露出されており、エピトープを除去するために標的として向けることができる。
また、ペプチド49(SSSVEYINNWEQAKA;配列番号:174)も、T細胞エピトープを含有する。図2及び表3は、3人のドナー(ドナー30、39及び49)がペプチド49による刺激の後に陽性増殖応答を生じたことを示す。iTope(商標)コンピューター解析を使用すると、このペプチドに最も好ましい結合レジスターは、32個のMHCクラスII対立遺伝子の中の22個を結合する可能性を有するVEYINNWEQ(配列番号:175、図8)であると予測された。この結合レジスターは、バリン(V148)をp1アンカー残基として有していた。ペプチド49とオーバーラップしたペプチド48及び50も同じコア9量体を含有していたが、ドナー30、39及び49は、オーバーラップペプチに応答せず、コア9量体の外側の残基もまたMHCクラスII分子に対するペプチドの結合に重要であることを実証している。バリン148は部分的に表面露出され、従って極性置換残基は、親水性部分が表面露出され且つ疎水性領域が覆い隠されるように選択できる。
エピトープ7は、ペプチド100(LEKTTAALSILPGIG、配列番号:177)内に見出された。4人のドナー(ドナー1、15、30及び35)がペプチド100に応答し、ドナー15の応答はボーダーラインであった。iTope(商標)解析により、このペプチドのコア9量体結合レジスターは、ロイシン(L298)をp1アンカー残基として有するLEKTTAALS(配列番号:178)であると予測された(図9)。この9量体は、32個の可能なMHCクラスII対立遺伝子の中の24個を結合すると予測された。この9量体もペプチド99内に存在し、ドナー30はこれに対する統計学的に有意である(p<0.05)閾値下応答(SI=I.84)を開始し、従ってこれらのペプチドの結合モチーフとしてLEKTTAALS(配列番号:178)を支持する。ロイシン298は、トランスロケーションドメインの表面に十分に露出されており且つこのドメインの活性に関連づけられない。従って、それは置換について明らかな問題を有していない。従って、残基は、T細胞エピトープを修飾/排除することを目的として容易に置換できる。
表4は、DT−1配列内に見出される7個のT細胞エピトープの詳細を要約し、図10は、タンパク質配列内のエピトープの配置を表す。DT−1とIL−2の間の接合部内にエピトープは検出されなかった。

種々のアッセイを使用して、本明細書に記載のDTバリアントのVLS活性を評価できる。
DT特異抗体を使用して、VLSモチーフによって内皮細胞表面に結合されたDTを検出できる。共通エピトープを、バリアント同士の間の結合の相違を検出するために直接に標識した。ELISAを使用して、利用できる抗DT抗体の特異性/親和性を確認した。ここに記載した方法の他に、His標識Abは、DTバリアントを選択できる別の手段を提供する。
蛍光色素(Alexa488)に対するDT及び毒素突然変異体の直接複合体を使用して、本明細書に記載のDTバリアントを評価できる。Alexa色素は、高安定性であり、明るい。少量タンパク質標識キット(Microscale Protein Labelling Kit(Molecular Probes/Invitrogen:A30006)は、タンパク質(20〜100μg)の微小規模の標識を可能にする。標識の度合い(DOL)は最適化することができ、色素:タンパク質(D:P)の比は、分光光度計で測定され、再現可能である。FACSを使用して蛍光を測定する。
細胞膜完全性を評価して、毒素に露出後の細胞膜の完全性の喪失を測定できる。VLSモチーフを包含するペプチドを、Balunaら(PNAS USA,96:3957(1999))に記載されているような方法を使用して蛍光色素に直接に結合させた。あるいは、本明細書に記載されているか又は当該技術で知られている他のアッセイを利用できる。
DT活性の測定に適したアッセイは、すでに確立されている。細胞毒性アッセイを利用して、DT−IL2T細胞エピトープ及びIVTTアッセイで選択されたVLSバリアントリードの毒素活性を確認する。
DT活性を測定するために生体外転写/翻訳(IVTT)アッセイを使用すると、DT遺伝子の直接的転写/翻訳が、ウサギ網状赤血球溶菌液系を使用して試験された。典型的には、これは、標的プラスミド(T7−ルシフェラーゼ)のIVTTを測定する化学発光アッセイによるルシフェラーゼ遺伝子の連結転写/翻訳を含み、活性毒素は、ルシフェラーゼシグナルを抑制し、減少する。滴定曲線によるメディアムスループットスクリーニング(MTS)を可能にするPCR生成物を利用し、全てのバリアントを同じ96ウエルアッセイプレートでのWT DTと比較した。代用IC50は、阻害曲線から測定できる。DTは結合し、伸長因子−2(EF−2)の共有結合修飾を生じることから、これは、活性DTバリアントについてルシフェラーゼ産生の阻害を生じるであろう。連結転写/翻訳アッセイを、リボソーム阻害タンパク質の分析に使用して、DTのCドメインの活性に関する情報を得ることができる。例えば米国特許第5,976,806号及び同第5,695,983号明細書(これらのそれぞれは、その全体を参照することにより本明細書に組み込まれる)に記載されているようなこのような連結転写/翻訳アッセイ反応を実施するのに有用な種々の方法が、当業者に知られている。
Toxilight(商標)、Vialight(商標)及びALAMARBLUE(商標)キットは、細胞毒性を測定するのに使用できる非放射性の市販アッセイである。アッセイは、96ウエルプレートフォーマットで行い、感受性及び耐性T細胞系を使用して時間と共に毒素(10−7〜10−12M)を滴定する。ヒトT細胞株に対するONTAKとIL−2の細胞毒性を、Toxilightキットを使用して48時間評価した。1秒当たりの蛍光カウント(LCPS)は、アデニレートキナーゼ放出の度合いを反映する(図19)。
連結転写/翻訳アッセイの他に、リボシルトランスフェラーゼアッセイ(例えば、実施例3に記載)が96ウエルフォーマットで確立された。このアッセイは、大腸菌でのDT遺伝子の発現(段階3)からのDTの試料を使用し、前記の連結転写/翻訳アッセイと同時に試験する。ADPリボシル化を測定する従来の方法は、二本鎖(ds)活性化因子DNAオリゴヌクレオチドで処理した透過性細胞を使用し、その後の放射標識NAD+の測定は、酸不溶性物質に組み込まれる。
DT及びヒトIL−2遺伝子は、段階3の初めに、大腸菌中で発現させるのに最適化させたコドンを使用して、当該技術において知られている慣用の技法を使用して合成される。細胞毒性及びHUVEC結合アッセイ(段階2参照)の分析において使用すべき全長DT及びDT(ΔR)(CドメインとIドメインのみを含有する)を生成させるために、大腸菌の細胞周辺腔内へのDTの搬出のための分泌リーダー配列を含むベクター系を使用する。プラスミド及び大腸菌株は、可溶性生成物を生成させるのに最適化させる。His標識DT生成物は、Ni−IDAカラムで精製し、スピンカラムは、リードの並行精製を可能にする。DTの精製方法としては、例えば、DTに融合させた親和性標識(例えば、His6標識)による精製が挙げられる。段階3で開発された方法は、以下のバリアントの活性を段階4及び5でのリード候補を同定することを目的として正確に比較できるような同様の品質を有する多数のDTバリアントの信頼性のある製造を提供する。
VLSを誘導する可能性の低下のためのDTのバリアントを、2回の突然変異、すなわち、1回目のDTの3つの(x)D(y)モチーフそれぞれでの別個の突然変異、及び2回目のリード突然変異と任意の追加の突然変異の組み合わせで生じさせる。それぞれのDTバリアントを、HUVEC結合アッセイ(段階2)で試験し、2回目の突然変異の後に選択された最適突然変異を、前記プロジェクトの段階5の中間段階のT細胞エピトープ突然変異と組み合わせる。これらのアッセイのためのDTバリアントの生成は、大腸菌での先端切断DT(ΔR)の発現によるものである(段階3)。
免疫原性を低下させるためにT細胞エピトープを排除するためのDTのバリアントを、CドメインとIドメインでの2回の置換によって生成させる。1回目のバリアントは、単一のエピトープ遺伝子座に別個の置換を含み、次いでこれらを2回目のバリアント中で組み合わせて、2つ、3つ又はそれ以上のバリアント遺伝子座の組み合わせを生成させる(T細胞エピトープの数及び優先順位に依存する)。2回目のバリアントは、段階4からのVLSバリアントとの組み合わせを含む。VLS突然変異体をT細胞エピトープ置換と組み合わせる追加の工程により、リードDTバリアントのさらなる最適化が必要とされる場合には、任意の3回目のDTバリアントが含有される。
DTエピトープバリアントはタンパク質合成を阻害する
DT382構造体が使用され、DTの配列番号:2又は149のアミノ酸残基1−382及びIL2を含有していた。制限酵素部位が、Rドメイン又はIL2部分のいずれかでのクローニングのためにアミノ酸残基382で操作された。修飾は、以下に記載のようにして組み込まれる。
認識されたx(D)yモチーフが破壊されるように、VLSモチーフが突然変異するDT382遺伝子のバリアントを生成させた。単一及び多重遺伝子座でのバリアントの活性を、PCR産物を使用する生体外転写/翻訳アッセイでの活性について評価した(実施例1に記載のように)。
ヒト血管内皮細胞(HUVEC)を、EBM(CC−3124 Lonza,Basel,Switzerland)に保持した。使用前に、細胞を、酵素無含有解離用緩衝液(C5914 Sigma,Poole,UK)を使用してプラスチック培養基板から分離し、1%BSAと0.05%NaN3とを含有するリン酸緩衝生理食塩水に再懸濁した。次いで、細胞を、5%正常ヒト血清を含有する上記と同じ緩衝液中で20分間インキュベートし、その後にAlexa488蛍光色素(A30006 Invitrogen、Carlsbad CA)に複合されている精製DT382タンパク質又はDT382 VLSバリアントの滴定液を、製造業者の取扱い説明書に従って加えた。細胞を、前記標識タンパク質と共に30分間インキュベートし、その後に洗浄し、PBS+1%BSA+0.05%NaN3緩衝液に再懸濁した。標識DT−389−IL2融合物を、陽性対照として使用し、標識BSAを陰性対照として使用した。次いで、細胞をFACSCaliburフローサイトメーター(Becton Dickinson、Franklin Lakes、NJ)で分析し、細胞集団の蛍光染色を測定した。次いで、バックグラウンドを超える染色を示した細胞%を、使用した標識タンパク質の濃度に対してプロットした。
段階6において、1個又はそれ以上のリードDT−IL2バリアントが、段階5からのリードDTバリアントと、段階3からのヒトIL2(2−133)遺伝子との融合によって生成される。大腸菌での野生型及びリードDT−IL2バリアントの発現は、例えば、封入体中のタンパク質凝集体の貯留及び再生を含むDT−IL2の従来の方法に従う。次いで、野生型及び1個又はそれ以上のリードDT−IL2バリアントを、段階2に記載のようにして、細胞毒性及びVLS−関連アッセイで試験する。
段階5からのリードDT(ΔR)バリアントを精製し、EpiScreen(商標)経時T細胞アッセイを使用して野生型DT(ΔR)と比較する。HLAアロタイプの発現に社会集団を代表する多数の健康なドナーが、前記のドナーライブラリーから選択される。ドナーを、2〜4×106 CD8+T細胞欠乏PBMCを含有する別個のバルク培養物中のそれぞれのタンパク質を用いて刺激する。重複試料(Tブラストの試料)を5〜8日目にバルク培養物から取り出し、及び増殖をIL−2分泌(ELISPOT)と共に評価する。野生型とDT(ΔR)バリアントの間の評価をさらに確証するために、試験集団に、段階1の間に行ったEpiScreen(商標)T細胞エピトープマッピング試験からの応答ドナー(提供された十分な数のCD8+T細胞欠乏PBMCが残る)を補充した。
(ii)各ドナーを、陽性対照抗原、例えばキーホール・リンペット・ヘモシアニン(可能な新抗原)又は破傷風トキソイド(想起抗原)に対して試験する;
(iii)MHCクラスI制限T細胞応答の検出を除外するために、CD8+T細胞を欠乏させる;
(iv)リードDT(ΔR)バリアント及び野生型DT(ΔR)を、互いに対して比較して、T細胞CD4+T細胞を活性化する相対能力を評価する;
(v)データを、統計学的及び頻度分析を含む追加の情報によって裏付けられたSI>2の陽性応答を有する先に実証したアッセイパラメーターを使用して解析する;
(vi)EpiScreen(商標)経時T細胞アッセイからのデータは、個々のDT分子に対するT細胞の応答の大きさ及び反応速度論に関する情報を提供する;
(vii)陽性応答を生じるドナーからの残存PBMCが、保存され、反復試験研究で使用するために利用できる;及び
(viii)ドナーアロタイプ並びにDT(ΔR)に対する応答及びバリアントDT(ΔR)リードに対する応答の関連性について評価を行う。
臨床試験デザイン及び患者適格性
患者の治療は、治験審査委員会(Institutional Review Board)及び米国食品医薬品局によって承認されたプロトコールの一部として書面によるインフォームドコンセントに従って行われる。組織学的に確認された転移性RCCを有する患者が、試験に適任である。全患者は、適切な肝臓、腎臓及び神経機能、6ヶ月を超える寿命及び70%以上のカルノフスキーパフォーマンス状況を有することを要求される。患者は、任意の以前の治療に関連したあらゆる毒性から正常な状態に戻っていなければならず、しかも試験に入る前の少なくとも6週間は任意の化学療法、放射線療法又は免疫療法を受けていてはならない。CNS転移を有する患者、自己免疫疾患の履歴を有する患者及び重篤な併発性の慢性又は急性疾患を有する患者は、この試験から除外される。免疫抑制剤を受けている患者も除外される。適格対象は、DTバリアント−IL2融合タンパク質(18μg/kg)の単回投与、次いで腫瘍RNA移入DCによる免疫処置を受けるか又はDTバリアント−IL2融合タンパク質単独を受ける同じ確率で無作為に選ばれる。全ての被検体は、腫瘍RNA移入DCの皮内注射を3回受ける。この注射は、2週間の間隔で皮内投与され及びそれぞれの注射で200μlの0.9%塩化ナトリウムに懸濁された1×107細胞からなる。処置後に、被検体は、臨床毒性並びに免疫応答及び臨床応答について評価される。調節制限及びある被検体においては腫瘍組織に対する制限されたアクセスにより、腫瘍生検は行われない。
DTバリアント−IL2融合タンパク質を、2mlの1回使用バイアル中のクエン酸緩衝液に150μg/mlの濃度で配合された凍結滅菌溶液として提供する。解凍後に、DTバリアント−IL2融合タンパク質を、滅菌標準食塩水で最終濃度15μg/mlに希釈し、30分間にわたる静脈内注入によって送達させる。患者は、注入の30〜60分前にアセトアミノフェン(600mg)及び抗ヒスタミン剤を受けることを許される。DC培養のために、濃縮白血球画分を、白血球分離により収集する。PBMCを、白血球分離生成物から密度勾配遠心分離で単離する(Histopaque;Sigma−Aldrich)。半接着性細胞画分を、組換えヒトIL−4(500U/ml;R&D Systems)及び組換えヒトGM−CSF(rhGM−CSF、800U/ml;Immunex Corp.)を補足した血清無含有X−VIVO15培地(Cambrex Corp.)中でのDC培養に使用する。7日後に、未成熟DCを、収集し、これに明細胞癌として組織学的に分類される腫瘍組織から抽出した全RNAを移入する。免疫学的モニタリング研究に使用される対照RNAを、自己良性腎臓組織(RE)又はPBMCから単離する。未成熟DCの移入は、電気穿孔によって行う。DCを、PBS中で洗浄し、ViaSpan(Barr Laboratories)中に4×107細胞/mlの濃度で再懸濁する。次いで、細胞を、1×106細胞当たり5μgのRNAと共に5分間同時インキュベートし、0.4cmキュベット中で300V及び150μFでの指数関数的減衰送達によって電気穿孔する(Gene Pulser II;Bio−Rad)。電気穿孔後に、細胞を、X−VIVO15培地に再懸濁し、10ng/mlのTNF−α、10ng/mlのIL−1β、150ng/mlのIL−6(R&D Systems)及び1μg/mlのプロスタグランジンE2(PGE2;Cayman Chemical Co.)の存在下で、20時間で成熟させる。投与の前に、完全成熟DC:Linneg、HLAクラスI及びIIhigh、CD86high及びCD83highの典型的な表現型を満たしていることを確実にするために、細胞を特性決定する。
IFN−γ及びIL−4ELISPOTの分析を、免疫処置の前、間及び後に得られたPBMCを使用して行う。PBMCを、完全RPMI 1640培地中で一夜培養する。PBMCから、CD4+T細胞及びCD8+T細胞を、ネガティブデプレション(negative depletion)によって単離する(Miltenyi Biotec)。ブロッキング後に、1×105T細胞及び1×104RNA移入DCを、2μg/mlのIFN−γ捕捉抗体(Pierce Biotechonology Inc.)又はIL−4捕捉抗体(BD Biosciences Pharmingen)で予め被覆された96ウエルニトロセルロースプレート(Multiscreen−IP、Milhpore)のそれぞれのウエルに加える。プレートを、37℃で20時間インキュベートし、それぞれのウエルに、ビオチン化IFN−γ検出抗体(Pierce Biotechonology Inc.)又はビオチン化IL−4抗体(BD Biosciences Pharmingen)を加える。次いで、細胞を、室温でさらに2時間インキュベートし、次いでストレプトアビジン−アルカリホスファターゼ(1μg/ml、Sigma−Aldrich)を加え、プレートを、基質(KPL)と共に展開した。洗浄後に、スポットを、自動ELISPOTリーダー(Zeiss)を使用して計数した。
Claims (28)
- 少なくとも1個のT細胞エピトープに少なくとも1個のアミノ酸残基修飾を有する毒素からなる修飾毒素であって、前記修飾毒素が未修飾毒素と比べて低下した免疫原性を示すことを特徴とする修飾毒素。
- 前記毒素がジフテリア毒素又はその断片である、請求項1に記載の修飾毒素。
- ジフテリア毒素の少なくとも1個のT細胞エピトープに少なくとも1個のアミノ酸残基修飾を含有する修飾ジフテリア毒素であって、前記少なくとも1個のT細胞エピトープが配列番号:181、184、187、190、193、196及び199から選択され並びに前記修飾ジフテリア毒素が未修飾ジフテリア毒素と比べて低下した免疫原性を示すことを特徴とする修飾ジフテリア毒素。
- 前記少なくとも1個のアミノ酸残基修飾が、前記少なくとも1個のT細胞エピトープのエピトープコア、N末端、C末端、N末端及びC末端、又はこれらの任意の組み合わせにおいて行われる、請求項3に記載の修飾ジフテリア毒素。
- 前記毒素がジフテリア融合毒素である、請求項1〜4のいずれか1項に記載の修飾毒素。
- 前記ジフテリア融合毒素が、ジフテリア毒素と、非ジフテリア毒素ポリペプチド由来の少なくとも1個の細胞結合ドメインとを含有してなる、請求項5に記載の修飾毒素。
- 非ジフテリア毒素ポリペプチドが細胞結合リガンドである、請求項6に記載の修飾毒素。
- 細胞結合リガンドが、抗体もしくはその抗原結合断片、サイトカイン、ポリペプチド、ホルモン、増殖因子又はインスリンである、請求項7に記載の修飾毒素。
- サイトカインがIL−2又はIL−3である、請求項8に記載の修飾毒素。
- 抗体が、モノクロナール抗体、ポリクロナール抗体、ヒト化抗体、遺伝子組換え抗体、又は移植抗体である、請求項8に記載の修飾毒素。
- 抗原結合断片が、Fab、Fab2、F(ab’)2、ScFv、(ScFv)2、一本鎖結合ポリペプチド、VH又はVLである、請求項8に記載の修飾毒素。
- 前記修飾ジフテリア毒素が、V7S、V7T、V7N、V7D、D8E、S9A、S9T、V29S、V29T、V29N、V29D、D30E、S31N、I290T、D291E、S292A、S292T、V97A、V97T、V97D、L107N、M116A、M116Q、M116N、F124H、V148A、V148T、L298A及びL298Nの中から選択される1個又はそれ以上の修飾を含有する、請求項2に記載の修飾ジフテリア毒素。
- 前記修飾ジフテリア毒素が、V7N、V7T、V29N、V29T及びV29Dの中から選択される2個の修飾を含有する、請求項2に記載の修飾ジフテリア毒素。
- 前記修飾ジフテリア毒素が、V7D、V7N、V7T、L107A、L107N及びF124Hの中から選択される3個の修飾を含有する、請求項2に記載の修飾ジフテリア毒素。
- さらに(x)D/E(y)モチーフ中に1個又はそれ以上のアミノ酸修飾を含有する修飾毒素であって、前記修飾毒素が未修飾毒素に匹敵する細胞毒性を有し且つ未修飾毒素と比べて低下した免疫原性及び内皮細胞に対する結合の減少を示す、請求項1に記載の修飾毒素。
- さらにその中に1個又はそれ以上のアミノ酸修飾を含有し、少なくとも1個のアミノ酸修飾が(x)D/E(y)モチーフ内部で行われ、前記修飾ジフテリア毒素が、未修飾ジフテリア毒素に匹敵する細胞毒性を有し且つ未修飾毒ジフテリア素と比べて低下した免疫原性及び内皮細胞に対する結合の減少を示す、請求項2〜7のいずれか1項に記載の修飾ジフテリア毒素。
- 少なくとも1個のアミノ酸修飾が(x)D/E(y)モチーフ内で行われ;及び位置(x)での修飾が、A、S、E、F、C、M、T、W、Y、P、H、Q、D、N、K、R、G、L及び明細書の表1の修飾又は異常アミノ酸の中から選択されるアミノ酸残基によるV又はIの置換であるか;位置D/Eでの修飾が、A、S、E、I、V、L、F、C、M、G、T、W、Y、P、H、Q、N、K、R及び明細書の表1の修飾又は異常アミノ酸の中から選択されるアミノ酸残基によるD又はEの置換であるか;位置(y)での修飾が、I、F、C、M、A、G、T、W、Y、P、H、E、Q、D、N、K、R、S、L、V及び明細書の表1の修飾又は異常アミノ酸の中から選択されるアミノ酸残基による置換であるか;又はこれらの組み合わせである、請求項15又は16に記載の修飾毒素。
- 前記修飾ジフテリア毒素が、V7T、V7N、V7D、D8N、S9A、S9T、S9G、V29N、V29D、V29T、D30N、S31G、S31N、I290T、D291E、S292A、S292G及びS292Tから選択される1個又はそれ以上の修飾を含有する、請求項16に記載の組成物。
- 前記修飾ジフテリア毒素が、V7N V29N、V7N V29T、V7N V29D、V7T V29N、V7T V29T又はV7T V29Dから選択される2個の修飾を含有する、請求項16に記載の組成物。
- 前記修飾ジフテリア毒素が、V7N V29N I290N、V7N V29N I290T、V7N V29N S292A、V7N V29N S292T、V7N V29T I290N、V7N V29T I290T、V7N V29T S292A、V7N V29T S292T及びV7T V29T I290Tの中から選択される3個の修飾を含有する、請求項16に記載の組成物。
- 請求項1〜20のいずれか1項に記載の前記修飾毒素と製薬学的に許容し得る担体とを含有してなる組成物。
- 被検体の悪性疾患を治療する方法であって、有効量の請求項21に記載の医薬組成物を前記被検体に投与することからなり、前記悪性疾患が血液癌、固形腫瘍又は転移であることを特徴とする方法。
- 被検体の悪性疾患の治療薬を製剤するための請求項21に記載の医薬組成物の使用であって、前記悪性疾患が血液癌、固形腫瘍又は転移であることを特徴とする使用。
- 被検体の非悪性疾患を治療する方法であって、治療有効量の請求項21に記載の医薬組成物を前記被検体に投与することからなり、前記非悪性疾患が移植片対宿主病又は乾癬であることを特徴とする方法。
- 非悪性疾患の治療薬を製剤するための請求項21に記載の医薬組成物の使用であって、前記非悪性疾患が移植片対宿主病又は乾癬であることを特徴とする使用。
- 抗癌剤と請求項1〜21のいずれか1項に記載の組成物とを被検体に投与することからなる抗癌剤の活性を高める方法。
- 被検体の悪性疾患を治療する方法であって、有効量の請求項1〜21のいずれか1項に記載の組成物と抗癌剤とを前記被検体に投与することからなり、前記悪性疾患が血液癌、固形腫瘍又は転移であることを特徴とする方法。
- 被検体の悪性疾患の治療薬を製剤するための請求項21に記載の医薬組成物と抗癌剤の使用であって、前記悪性疾患が血液癌、固形腫瘍又は転移であることを特徴とする使用。
