いくつかの実施形態では、特異的結合ペアの結合要素、例えば、モノクローナル抗体などの抗体は、ナノ粒子の表面またはナノ粒子をカプセルに封入するシェルの外面に直接付着してもよい。例示的な一実施形態では、結合ペアの特異的結合要素、例えば、モノクローナル抗体は、リンカー、例えば、ポリエチレングリコール(PEG)で処理され、PEGリンカーを通じてナノ粒子に直接付着できる。PEGリンカーなどのリンカーを使用すると、特異的結合要素の固有の特性および構造を保持することができ、またそのようなリンカーを使用すると、ナノ粒子への他の化学種の非特異的結合に立体障害を発生させることによって官能化ナノ粒子の特異性が高まる。PEGリンカーを通じて特異的結合要素が付着しているSERS活性ナノ粒子について、ここでさらに詳しく説明する(以下のセクションI.D.を参照)。

SERS活性金属ナノ粒子は、水溶液中で凝集する傾向を有し、いったん凝集すると、再度分散することが困難である。さらに、いくつかのラマン活性分子の化学組成は、タンパク質などの他の分子を金属ナノ粒子に付着させるために使用される化学作用と相容れない。これらの特性は、ラマン活性分子、付着化学作用、および金属ナノ粒子に付着される他の分子の選択を制限することがある。

直径60nmの球状金ナノ粒子をTed Pella,Inc.(米国カリフォルニア州レディング所在)社から購入した。透過電子顕微鏡法と光散乱法を用いて、メーカーによって報告されているナノ粒子のサイズを確認した。ラマン活性種、例えば、非蛍光分子および本明細書で開示されているNIR色素を金ナノ粒子に付着させるために、最終混合物中のラマンレポーターの濃度が10μMになるように化学物質を金ナノ粒子と混合した。バルク溶液から金ナノ粒子を5倍に濃縮し、520nmで5に等しい最終光学密度を得た。振盪器上で一晩かけて反応を進行させた。
近赤外色素のSERSスペクトル
本明細書で開示されているSERS活性色素のSERS強度をテストするために使用される計測器は、660nmレーザー励起を使用するCentice(米国ノースカロライナ州モリスビル所在)スペクトロメーターであった。一般に使用されている非蛍光ラマン活性分子、例えば、トランス−1,2−ビス(4−ピリジル)エチレン(BPE)および本明細書で開示されているNIR色素、例えば、クマリンピコリニウム色素(CoPic)に対する実験結果が図1に示されている。BPEは、2つの大きなピークを呈示し、1つ目のピークは約1200cm-1で、2つ目のピークは1600から1650cm-1の間である。同様に、CoPicは、約1150および1550cm-1で2つの大きなピークを呈示する。BPEスペクトル中の最も顕著なピークの最大強度は、約47,000であるが、CoPicに対する最大強度は、約160,000である。この実施例では、CoPicに対するピーク最大値は、BPEからの強度に比べて約3倍である。
磁気捕捉液体ベースSERSアッセイにおける非特異的結合の低減
ポリエチレングリコール(PEG)リンカー分子を通じてDNAオリゴヌクレオチドをSERS活性ナノ粒子(SERS活性レポーター分子、例えば、さまざまなビピリジル色素でコーティングされ、チオール官能化ガラスコーティングによってカプセル封入された金粒子)の表面に付着させた。本明細書で開示されている発明対象の実施形態の代表的な一例が、図5に示されており、これは、5nmの異種二官能性PEGリンカー分子を介してSERS活性ナノ粒子および磁気捕捉粒子の表面にDNAを固定化するのを示している。

- 前記リンカーは、ポリエチレングリコール(PEG)およびチオール含有部分からなる群から選択されることを特徴とする請求項17に記載の方法。
- 前記ポリエチレングリコールは、少なくとも12個のエチレングリコールサブユニットを含むことを特徴とする請求項18に記載の方法。
- 前記ポリエチレングリコールは、異種二官能性ポリエチレングリコール(PEG)分子を含み、前記異種二官能性PEG分子は、異種二官能性PEG分子の第1の末端に第1の官能基を、異種二官能性PEG分子の第2の末端に第2の官能基を備えることを特徴とする請求項18に記載の方法。
- 前記第1の官能基は、N−ヒドロキシスクシンイミド(NHS)エステルを含み、前記第2の官能基は、マレイミドを含むことを特徴とする請求項20に記載の方法。
- 前記異種二官能性PEG分子は、前記SERS活性ナノ粒子上のチオール基および前記異種二官能性PEG分子のマレイミド基を通じて前記SERS活性ナノ粒子に共有結合することを特徴とする請求項21に記載の方法。
- 前記特異的結合要素は、ポリヌクレオチドを含み、前記ポリヌクレオチドは、前記ポリヌクレオチドの5’または3’末端のアミン基および異種二官能性PEG分子のNHSエステルを介して異種二官能性PEG分子に共有結合することを特徴とする請求項22に記載の方法。
- 生体試料中の1つまたは複数の分析対象の存在または量を検出するための方法であって、
(a)1つまたは複数の分析対象を含むことが疑われる生体試料を提供するステップと、

(d)前記SERSシグナルを測定して、前記生体試料中の1つまたは複数の分析対象の存在または量を検出するステップとを含むことを特徴とする方法。 - 前記生体試料は、全血試料、血清、血漿、腹水、尿、唾液、汗、ミルク、滑液、腹膜液、羊水、経脳脊髄液、リンパ液、肺塞栓症、脳脊髄液、心嚢液、頚腟部試料、組織抽出物、細胞抽出物、およびこれらの組合せからなる群から選択されることを特徴とする請求項24に記載の方法。
- 前記1つまたは複数の分析対象は、核酸、DNAフラグメント、ヌクレオチド、ポリヌクレオチド、およびオリゴヌクレオチドからなる群から選択されることを特徴とする請求項24に記載の方法。
- 前記分析対象は、細胞内の標的構造を含むことを特徴とする請求項24に記載の方法。
- 前記試薬は、式A−Yの複数のSERS活性レポーター分子が会合しているナノ粒子を含み、前記1つまたは複数のSERS活性レポーター分子はそれぞれ、測定可能な別個のSERSシグナルを呈示することを特徴とする請求項24に記載の方法。
- 前記1つまたは複数の分析対象に対する親和性を有する前記特異的結合要素は、前記ナノ粒子の外面に直接付着することを特徴とする請求項24に記載の方法。
- 前記特異的結合要素は、リンカーを通じて前記ナノ粒子に共有結合することを特徴とする請求項24に記載の方法。
- 前記リンカーは、ポリエチレングリコール(PEG)を含むことを特徴とする請求項30に記載の方法。
- 前記ポリエチレングリコールは、少なくとも12個のエチレングリコールサブユニットを含むことを特徴とする請求項31に記載の方法。
- 前記ポリエチレングリコールは、異種二官能性ポリエチレングリコール(PEG)分子を含み、前記異種二官能性PEG分子は、異種二官能性PEG分子の第1の末端に第1の官能基を、異種二官能性PEG分子の第2の末端に第2の官能基を備えることを特徴とする請求項31に記載の方法。
- 前記第1の官能基は、N−ヒドロキシスクシンイミド(NHS)エステルを含み、前記第2の官能基は、マレイミドを含むことを特徴とする請求項33に記載の方法。
- 前記異種二官能性PEG分子は、前記SERS活性ナノ粒子上のチオール基および前記異種二官能性PEG分子のマレイミド基を通じて前記SERS活性ナノ粒子に共有結合することを特徴とする請求項34に記載の方法。
- 前記特異的結合要素は、ポリヌクレオチドを含み、前記ポリヌクレオチドは前記ポリヌクレオチドの5’または3’末端のアミン基および異種二官能性PEG分子のNHSエステルを介して異種二官能性PEG分子に共有結合することを特徴とする請求項35に記載の方法。
- 前記1つまたは複数の分析対象に対する親和性を有する前記特異的結合要素は、式A−Yの前記SERS活性レポーター分子とコンジュゲートすることを特徴とする請求項24に記載の方法。
- 式A−Yの前記SERS活性レポーター分子は、前記ナノ粒子の外面上に1つの層を形成し、前記層は前記ナノ粒子の前記外面を少なくとも部分的に覆い、かつ内面と外面とにより定められることを特徴とする請求項24に記載の方法。
- 前記ナノ粒子は、さらに、カプセル材料を含み、前記カプセル材料は、前記ナノ粒子コアの前記外面および式A−Yの前記SERS活性レポーター分子の前記層の前記外面のうちの少なくとも一方に配置されることを特徴とする請求項38に記載の方法。
- さらに、複数の磁気捕捉粒子を前記生体試料と接触させるステップを含み、それぞれの磁気捕捉粒子には、前記1つまたは複数の分析対象に対し親和性を有する少なくとも1つの結合要素が付着しており、前記磁気捕捉粒子は、式A−YのSERS活性レポーター分子が会合している前記SERS活性ナノ粒子と複合体を形成することを特徴とする請求項24に記載の方法。
- さらに、前記生体試料を磁場に曝すことで、前記磁気捕捉粒子とSERS活性ナノ粒子との複合体を試料収集デバイスの局在領域に移動させるステップを含むことを特徴とする請求項40に記載の方法。
- さらに、入射光を前記試料収集デバイスの前記局在領域に照射して前記磁気捕捉粒子およびSERS活性ナノ粒子と会合する前記1つまたは複数の分析対象の存在または量を検出するステップを含むことを特徴とする請求項41に記載の方法。
- 前記1つまたは複数の分析対象は、固相担体上に固定化されることを特徴とする請求項24に記載の方法。
- 前記1つまたは複数の分析対象に対し親和性を有する少なくとも1つの特異的結合要素が会合している前記1つまたは複数のSERS活性ナノ粒子および式A−Yの少なくとも1つのSERS活性レポーター分子は、固相担体上に固定化されることを特徴とする請求項24に記載の方法。
