Abstract
Description
以下、本発明を実施例を挙げて詳述する。これらの実施例は単に本発明をより具体的に説明するためのものであり、本発明の範囲がこれらの実施例に制限されないことは当業界において通常の知識を持った者にとって自明である。
実施例1−1(製造例1)
肥満患者から抽出されて廃棄された脂肪組織を粉砕した後、蒸留水を入れて60〜80℃、110Wにおいて10分間10〜15回に亘って超音波により処理して脂質を脱離させた。前記脱離された脂質と脂質未脱離の脂肪組織をフィルターを用いてろ過するか、あるいは、3500rpmの条件下で5分間遠心分離し、70%エタノールにより洗浄した後、蒸留水により洗浄した。前記脂質の除去された脂肪組織を凍結乾燥または空気乾燥の方法により乾燥した後、放射線を用いて滅菌することにより脂質の除去された粉末を製造した。
肥満患者から抽出されて廃棄された脂肪組織を粉砕した後、蒸留水を入れて加圧ノズル噴射して脂質を脱離させた。前記脱離された脂質と脂質未脱離の脂肪組織をフィルターを用いてろ過するか、あるいは、3500rpmの条件下で5分間遠心分離し、70%エタノールにより洗浄した後、蒸留水により洗浄した。前記脂質の除去された脂肪組織を凍結乾燥または空気乾燥の方法により乾燥した後、放射線を用いて滅菌することにより脂質の除去された粉末を製造した。
肥満患者から抽出されて廃棄された脂肪組織を粉砕した後、蒸留水とヒアルロニダーゼを1:1:0.1の割合にて入れ、60〜80℃、110Wにおいて10分間10〜15回超音波により処理して脂質を脱離させた。前記脱離された脂質と脂質未脱離の脂肪組織をフィルターを用いてろ過するか、あるいは、3500rpmの条件下で5分間遠心分離した後、70%エタノールにより洗浄し、次いで、蒸留水により洗浄した。前記脂質の除去された脂肪組織を凍結乾燥または空気乾燥の方法により乾燥した後、放射線を用いて滅菌することにより脂質の除去された粉末を製造した。

実施例1−1において製造された粉末をオイルレッドOで染色し、対照群としては上記の方法により粉末化させる前の脂肪組織を使用した。その結果、対照群には脂質が存在するのに対し(図1)、前記実施例1−1の方法に従い製造された粉末には脂質が存在しないことを確認することができた(図2)。

(a)脂肪組織を粉砕した後、超音波により処理して脂質を脱離させるステップと、
(b)前記脱離された脂質と脂質未脱離の脂肪組織を除去するステップと、
(c)前記脂質の除去された脂肪組織を滅菌して脂質の除去された支持体を得るステップ。 - 前記(a)ステップは、タンパク質分解酵素を使用しないことを特徴とする請求項5に記載の方法。
- 前記(a)ステップは、粉砕した脂肪組織をヒアルロニダーゼにより処理した後、超音波処理することを特徴とする請求項5に記載の方法。
- (d)前記得られた支持体を乾燥した粉末化するスタップをさらに含むことを特徴とする請求項5に記載の方法。
- 前記脂質は脂肪小滴であることを特徴とする請求項5に記載の方法。
- 次のステップを含む人体体積置換用または細胞培養用支持体の製造方法:
(a)脂肪組織を粉砕した後、加圧ノズル噴射を通じて脂質を脱離させるステップと、
(b)前記脱離された脂質と脂質未脱離の脂肪組織を除去するステップと、
(c)前記脂質の除去された脂肪組織を滅菌して脂質の除去された支持体を得るステップ。 - 前記(a)ステップは、タンパク質分解酵素を使用しないことを特徴とする請求項10に記載の方法。
- 前記(a)ステップは、粉砕した脂肪組織をヒアルロニダーゼにより処理した後、超音波処理することを特徴とする請求項10に記載の方法。
- (d)前記得られた支持体を乾燥した粉末化するスタップをさらに含むことを特徴とする請求項10に記載の方法。
- 前記(b)ステップは、ろ過または遠心分離により行うことを特徴とする請求項5または請求項10に記載の方法。
- 前記遠心分離後、エタノール洗浄または蒸留水洗浄をさらに行うことを特徴とする請求項14に記載の方法。
- 前記滅菌は放射線またはEOガスを用いて行うことを特徴とする請求項5または請求項10に記載の方法。
