Abstract
【解決手段】医療機器は、固定化された線維素溶解酵素及び硫酸デキストランを有する基材を含む。硫酸デキストランは40キロダルトン(kDA)未満の分子量を有する。医療機器は少なくとも1つの基材から形成される。基材上には線維素溶解酵素が固定化される。この線維素溶解酵素が、40(kDa)未満の分子量を有する硫酸デキストランで安定化される。

Claims (34)
- 固定化された線維素溶解酵素を含む基材と、40キロダルトン(kDa)未満の分子量を有する硫酸デキストランと、
を含むことを特徴とする医療機器。 - 前記線維素溶解酵素が前記基材に共有結合する、
ことを特徴とする請求項1に記載の医療機器。 - 前記線維素溶解酵素がウロキナーゼ型プラスミノゲン活性化因子(uPA)を含む、
ことを特徴とする請求項1に記載の医療機器。 - 前記基材に含浸させる2又はそれ以上の抗菌剤の組み合わせをさらに含む、
ことを特徴とする請求項1に記載の医療機器。 - 前記2又はそれ以上の抗菌剤の組み合わせが、クロルヘキシジンベースの及びこの薬学的に容認できる塩を含む、
ことを特徴とする請求項4に記載の医療機器。 - 前記基材に含浸させる抗菌剤をさらに含む、
ことを特徴とする請求項1に記載の医療機器。 - 前記抗菌剤が、クロルヘキシジンベースの又はこの薬学的に容認できる塩を含む、
ことを特徴とする請求項6に記載の医療機器。 - 前記抗菌剤が二酢酸クロルヘキシジンを含む、
ことを特徴とする請求項6に記載の医療機器。 - 前記抗菌剤がクロルヘキシジンドデカノエートを含む、
ことを特徴とする請求項6に記載の医療機器。 - 前記抗菌剤がゲンチアナバイオレットを含む、
ことを特徴とする請求項6に記載の医療機器。 - 前記硫酸デキストランが約8kDaの分子量を有する、
ことを特徴とする請求項1に記載の医療機器。 - 前記基材を含む管状構造をさらに含む、
ことを特徴とする請求項1に記載の医療機器。 - 前記基材が重合体である、
ことを特徴とする請求項1に記載の医療機器。 - 前記基材がポリウレタンである、
ことを特徴とする請求項13に記載の医療機器。 - ポリウレタン基材と、
患者内の血餅の形成を減少させるのに十分な、前記基材内で固定化された線維素溶解酵素の量と、
40キロダルトン未満の分子量を有する、前記線維素溶解酵素を安定化するための低分子量硫酸デキストランと、
前記ポリウレタン基材内に微生物増殖を低減させるのに十分な量で処理された、クロルヘキシジンベースの及び/又はこの薬学的に容認できる塩と、
を含むことを特徴とする医療機器。 - ポリウレタン基材と、
患者内の血餅の形成を減少させるのに十分な、前記基材に共有結合で固定化されたウロキナーゼ型プラスミノゲン活性化因子(uPA)の量と、
40キロダルトン(kDa)未満の分子量を有する、前記線維素溶解酵素を安定化させるための低分子量硫酸デキストランと、
前記ポリウレタン基材内に微生物増殖を低減させるのに十分な量で処理された、クロルヘキシジンベースの及び/又はこの薬学的に容認できる塩と、
を含むことを特徴とする医療機器。 - 医療機器を作製する方法であって、
少なくとも1つの基材から前記医療機器を形成するステップと、
前記基材上に線維素溶解酵素を固定化するステップと、
前記線維素溶解酵素を40キロダルトン(kDa)未満の分子量を有する硫酸デキストランで安定化するステップと、
を含むことを特徴とする方法。 - 前記基材を、前記線維素溶解酵素及び硫酸デキストランからなる線維素溶解溶液中で培養するステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - マイクロリットル当たり約30単位の前記線維素溶解酵素を酢酸ナトリウム緩衝液中に溶解させることにより前記線維素溶解溶液を調製するステップをさらに含み、前記線維素溶解酵素がウロキナーゼ型プラスミノゲン活性化因子(uPA)を含む、
ことを特徴とする請求項16に記載の方法。 - 約1%の前記硫酸デキストランを前記線維素溶解溶液に添加するステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - 約8kDaの分子量を有する約1%の前記硫酸デキストランを前記線維素溶解溶液に添加するステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - 前記線維素溶解酵素を前記基材に共有結合させるための結合部位を生成するように前記基材を処理するステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - 前記基材に2又はそれ以上の抗菌剤の組み合わせを含浸させるステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - 前記基材に、クロルヘキシジンベースの及びこの薬学的に容認できる塩の組み合わせを含浸させるステップをさらに含む、
ことを特徴とする請求項23に記載の方法。 - 前記基材に抗菌剤を含浸させるステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - 前記基材に、クロルヘキシジンベースの又はこの薬学的に容認できる塩を含浸させるステップをさらに含む、
ことを特徴とする請求項25に記載の方法。 - 前記基材に二酢酸クロルヘキシジンを含浸させるステップをさらに含む、
ことを特徴とする請求項25に記載の方法。 - 前記基材にクロルヘキシジンドデカノエートを含浸させるステップをさらに含む、
ことを特徴とする請求項25に記載の方法。 - 前記基材にゲンチアナバイオレットを含浸させるステップをさらに含む、
ことを特徴とする請求項25に記載の方法。 - 前記基材にブリリアントグリーンを含浸するステップをさらに含む、
ことを特徴とする請求項25に記載の方法。 - 前記基材から管状構造を形成するステップをさらに含む、
ことを特徴とする請求項17に記載の方法。 - 前記基材が重合体を含む、
