Description
実験1:ポリウレタンカテーテルセグメント上で固定化されたウロキナーゼ型プラスミノゲン活性化因子(uPA)に関する線維素溶解活性の持続時間の判定
Claims (31)
- 基材と共有結合されて固定化された線維素溶解酵素と、硫酸デキストランとを含む基材を有する医療機器であって、
前記医療機器が、患者における血管埋め込みのためのものであり、且つ
前記硫酸デキストランが、8キロダルトン(kDa)の分子量を有し、これにより前記患者の血液と接触したときに前記繊維素溶解酵素を安定化させる、前記医療機器。 - 前記線維素溶解酵素がウロキナーゼ型プラスミノゲン活性化因子(uPA)を含む、請求項1に記載の医療機器。
- 前記基材に含浸させた2又はそれ以上の抗菌剤の組み合わせをさらに含む、請求項1に記載の医療機器。
- 前記2又はそれ以上の抗菌剤の組み合わせが、クロルヘキシジンベース及びこの薬学的に容認できる塩を含む、請求項3に記載の医療機器。
- 前記基材に含浸させた抗菌剤をさらに含む、請求項1に記載の医療機器。
- 前記抗菌剤が、クロルヘキシジンベース又はこの薬学的に容認できる塩を含む、請求項5に記載の医療機器。
- 前記抗菌剤が二酢酸クロルヘキシジンを含む、請求項5に記載の医療機器。
- 前記抗菌剤がクロルヘキシジンドデカノエートを含む、請求項5に記載の医療機器。
- 前記抗菌剤がゲンチアナバイオレットを含む、請求項5に記載の医療機器。
- 前記基材を含む管状構造をさらに含む、請求項1に記載の医療機器。
- 前記基材が重合体である、請求項1に記載の医療機器。
- 前記基材がポリウレタンである、請求項11に記載の医療機器。
- 患者における血管埋め込み用のポリウレタン基材と、
患者内の血餅の形成を減少させるのに十分な量の線維素溶解酵素であって、前記基材に共有結合されることによって固定化されている、前記繊維素溶解酵素と、
患者の血液と接触したときに前記繊維素溶解酵素を安定化させるための低分子量硫酸デキストランであって、8キロダルトンの分子量を有する、前記低分子量硫酸デキストランと、
前記ポリウレタン基材内に微生物増殖を低減させるのに十分な量で配置されている、クロルヘキシジンベース及び/又はこの薬学的に容認できる塩と、
を含むことを特徴とする医療機器。 - 患者における血管埋め込み用のポリウレタン基材と、
患者内の血餅の形成を減少させるのに十分な量の、ウロキナーゼ型プラスミノゲン活性化因子(uPA)であって、前記基材に共有結合により固定化されている、前記ウロキナーゼ型プラスミノゲン活性化因子(uPA)と、
患者の血液と接触したときに繊維素溶解酵素を安定化させるための低分子量硫酸デキストランであって、8キロダルトン(kDa)の分子量を有する、前記低分子量硫酸デキストランと、
前記ポリウレタン基材内に微生物増殖を低減させるのに十分な量で配置されている、クロルヘキシジンベース及び/又はこの薬学的に容認できる塩と、
を含むことを特徴とする医療機器。 - 医療機器を作製する方法であって、
少なくとも1つの基材から、患者における血管埋め込み用の医療機器を形成するステップと、
前記基材上に繊維素溶解酵素を共有結合させることにより該線維素溶解酵素を固定化するステップと、
8キロダルトン(kDa)の分子量を有する硫酸デキストランを用いて、患者の血液と接触したときにおける前記線維素溶解酵素を安定化させるステップと、
を含むことを特徴とする方法。 - 前記医療機器を、前記線維素溶解酵素及び硫酸デキストランからなる線維素溶解溶液中で培養するステップをさらに含む、請求項15に記載の方法。
- マイクロリットル当たり30単位の前記線維素溶解酵素を酢酸ナトリウム緩衝液中に溶解させることにより線維素溶解溶液を調製するステップをさらに含み、前記線維素溶解酵素がウロキナーゼ型プラスミノゲン活性化因子(uPA)を含む、請求項15に記載の方法。
- 前記硫酸デキストランが、線維素溶解溶液の1%重量/容量である、請求項15に記載の方法。
- 前記線維素溶解酵素を前記基材に共有結合させるための結合部位を生成するように前記基材を処理するステップをさらに含む、請求項15に記載の方法。
- 前記基材に2又はそれ以上の抗菌剤の組み合わせを含浸させるステップをさらに含む、請求項15に記載の方法。
- 前記2又はそれ以上の抗菌剤の組み合わせが、クロルヘキシジンベース及びこの薬学的に容認できる塩の組み合わせを含む、請求項20に記載の方法。
- 前記基材に抗菌剤を含浸させるステップをさらに含む、請求項15に記載の方法。
- 前記抗菌剤が、クロルヘキシジンベース又はこの薬学的に容認できる塩を含む、請求項22に記載の方法。
- 前記抗菌剤が、二酢酸クロルヘキシジンを含む、請求項22に記載の方法。
- 前記抗菌剤が、クロルヘキシジンドデカノエートを含む、請求項22に記載の方法。
- 前記抗菌剤が、ゲンチアナバイオレットを含む、請求項22に記載の方法。
- 前記抗菌剤が、ブリリアントグリーンを含む、請求項22に記載の方法。
- 前記基材から管状構造を形成するステップをさらに含む、請求項15に記載の方法。
- 前記基材が重合体を含む、請求項15に記載の方法。
