前記基質抱合体又は前記生成物抱合体の拘束が、前記第1の検出ゾーン内に第1の検出信号を発生させ、酵素又は酵素阻害剤の存在又は量を前記第1の検出信号から求めることができるようになる、
ことを特徴とする診断キット。 - 前記レポーターが、前記第1の検出信号を直接発生させることができる検出可能物質を含むことを特徴とする請求項1に記載の診断試験キット。
- 前記レポーターが、特定の結合メンバーを含むことを特徴とする請求項1又は請求項2に記載の診断試験キット。
- 特定の結合メンバーに抱合されたプローブを更に含み、該プローブが、前記検出信号を直接生成することができる検出可能物質を更に含むことを特徴とする請求項1−3のいずれか1項に記載の診断試験キット。
- 前記プローブの前記特定の結合メンバーが、前記レポーターの前記特定の結合メンバーに対して特定の結合親和性を有することを特徴とする請求項4に記載の診断試験キット。
- 受容物質が、前記第1の検出ゾーン内に固定され、前記生成物抱合体が、前記受容物質に対して特定の結合親和性を示すことを特徴とする請求項1から請求項5のいずれか1項に記載の診断試験キット。
- 前記受容物質が、モノクローナル抗体であることを特徴とする請求項6に記載の診断試験キット。
- 前記クロマトグラフ培地は、前記基質抱合体をその中に優先的に拘束する第2の検出ゾーンを更に含み、前記第2の検出ゾーン内の前記基質抱合体の拘束が、第2の検出信号を発生させることを特徴とする請求項1から請求項7のいずれか1項に記載の診断試験キット。
- 第2の受容物質が、前記第2の検出ゾーン内に固定され、前記基質抱合体が、前記第2の受容物質に対して特定の結合親和性を示すことを特徴とする請求項8に記載の診断試験キット。
- 前記試験サンプル中の酵素の量が、前記第2の検出信号の強度に反比例することを特徴とする請求項8又は請求項9に記載の診断試験キット。
- 前記試験サンプル中の酵素の量が、前記第1の検出信号の強度対前記第2の検出信号の強度の比率に正比例することを特徴とする請求項8又は請求項9に記載の診断試験キット。
- 前記試験サンプル中の酵素の量が、前記第1の検出信号の強度に正比例することを特徴とする請求項1−10のいずれか1項に記載の診断試験キット。
- 前記試験サンプル中の酵素阻害剤の量が、前記第1の検出信号の強度に反比例することを特徴とする請求項1−10のいずれか1項に記載の診断試験キット。
- 前記クロマトグラフ培地が、較正ゾーンを更に形成することを特徴とする請求項1から請求項13のいずれか1項に記載の診断試験キット。
- 前記クロマトグラフ培地が、制御ゾーンを更に形成することを特徴とする請求項1から請求項14のいずれか1項に記載の診断試験キット。
- 請求項1から請求項15のいずれか1項の前記診断試験キットを利用する、試験サンプル中の酵素又は酵素阻害剤を検出するための方法であって、
培養混合物を形成するように、複数の基質抱合体と試験サンプルを接触させる段階と、
前記クロマトグラフ培地に前記培養混合物を付加する段階と、
前記第1の検出ゾーン内の前記検出信号の存在又は強度を求める段階と、
を含むことを特徴とする方法。 - 前記方法が、前記第2の検出ゾーン内の前記検出信号の存在又は強度を求める段階を更に含むことを特徴とする請求項16に記載の方法。
- 前記酵素が、加水分解酵素であることを特徴とする請求項16に記載の方法。
- 前記酵素が、転移酵素、リガーゼ、又はポリメラーゼであることを特徴とする請求項16に記載の方法。
- 前記転移酵素が、キナーゼ又はメチラーゼであることを特徴とする請求項19に記載の方法。
- 前記基質が、タンパク質、糖タンパク質、ペプチド、核酸、炭水化物、脂質、エステル、抗体、抗原、又はこれらの誘導体であることを特徴とする請求項16に記載の方法。
- 前記基質が、カゼイン、アルブミン、ヘモグロビン、ミオグロビン、ケラチン、ゼラチン、インスリン、プロテオグリカン、フィブロネクチン、ラミニン、コラーゲン、エラスチン、又はこれらの誘導体であることを特徴とする請求項16に記載の方法。
