細胞における野生型Parkinの過発現がプロテアソーム活性を阻害し、そしてParkinタンパク質の大きい不溶性封入体(inclusions)の沈着をもらすことが発見された(参照、下記実施例)。脳組織の分析は、PD患者では、Parkinレベルが健康な脳組織に較べて上昇され、そして不溶性画分において富有化されるように見えることを示した。参照、実施例5。特定のメカニズムに捕らわないことを意図すれば、実施例において記述された実験に一部基づいて、野生型Parkinタンパク質はミスフォールディング(misfolding)を起こしがちであり、ミスフォールドされたParkinの蓄積は、(1)プロテアソーム活性の損傷、(2)Parkinおよび他の細胞タンパク質を含有するaggresomeの生成、(3)細胞の罹病および(4)Parkin活性の喪失をもたらすことが信じられる。Parkin活性の喪失は、PDをもたらす直接的メカニズムであり(Kitada et al.,1998,Nature 392:605−608)、そしてこの研究において記述された点突然変異がまた、疾病をもたらすParkinの機能の喪失に関係しているのであろう(Foroud et al.,2003,Ann.Neurology 60:796−801)。

実施例1〜3に記述されるように、野生型Parkinが過発現される細胞では、aggresome(または「Parkin封入体」)が形成され、そしてプロテアソーム活性が減少される。培養におけるHEK293細胞では、Parkinタンパク質が低レベルで内因的に発現される。この内因性の発現レベルにおいては、タンパク質はその正常なリガーゼ活性の遂行による外には、プロテアソーム経路には検出可能には影響を与えない。しかしながら、Parkinタンパク質が、異種のプロモーターによって作動されるcDNAから組み換え技術によって発現される(すなわち、正常な内因性発現に比較して、細胞において高レベルに発現される)場合には、少なくとも若干のParkinタンパク質はミスフォールドされ、かつ/または不溶性になり、プロテアソーム機能を妨害する。また、Parkinの発現レベルの高い発現のプロテアソーム抑制特性は、正常な発現条件下でははるかに低い、検出不可能なレベルで起きている可能性もある。脳細胞における不溶性画分としてのミスフォールドされたParkinタンパク質の徐々の蓄積が長期間(例えば、40〜80年)にわたって起きて病気をもたらすかも知れない。
細胞におけるプロテアソーム機能に及ぼす作用物の効果は、プロテアソーム機能のすべてのアッセイを使用して検査することができる。主なプロテアソーム機能は細胞内タンパク質の分解である。このアッセイの1つの実施態様では、Parkinは、レポーター−デグロン(degron)融合タンパク質をまた発現する細胞において発現され、そして
レポーターはプロテアソーム活性を測定するために使用される。融合タンパク質は、タンパク質のC末端(またはN末端)に付加された分解シグナル(「デグロン(degron)」)をもつ検出可能なポリペプチド配列を含む。具体的な説明のためには、そして限定されるものではないが、代表的なデグロン配列は、配列番号:9として提供される。分解シグナルは、ポリペプチドが分解されるプロテアソームに対してポリペプチドを標的化させるのに役立つ。プロテアソームの活性が損傷される場合、細胞におけるポリペプチドのレベルは、ポリペプチドが正常に機能するプロテアソームによって分解される細胞に較べて増大する。タンパク質レベルにおける増大は種々の方法において検出することができる。
スミドにより細胞を形質転換することによってde novoで調製されてもよい。HEK293細胞(ATCC CRL−1573)、SHSY−5Y細胞(ATCC−2266)、COS細胞(CRL−1651);CHO細胞(ATCC−CCL−61)または他の哺乳動物細胞系を含む、種々の細胞のいずれが使用されてもよい。細胞は安定にまたは一過性にトランスフェクトすることができる。好ましくは、細胞は多数のアッセイをとおしての首尾一貫性のために安定なトタンスフェクタントである。
好適なプロテアソーム機能アッセイでは、レポーター−デグロン融合タンパク質を発現する細胞は、前記のように、Parkinを発現する発現ベクターによりトランスフェクトされる。1つの実施態様では、発現ベクターは野生型Parkinをコードしている。例えば、ヒトParkin(NM004562)のためのcDNAは、このアッセイにおいて使用するためにベクターpcDNA3.1(Invitrogen,San Diego,CA)のHindIII/Xbal部位中に挿入することができる。
物に曝露し;試験化合物に曝露された細胞におけるプロテアソーム機能と試験化合物に曝露されなかった変異Parkinを発現する細胞のプロテアソーム機能特性とを比較することを伴う。代表的なParkin変異体は、S167N、C212Y、T240M、R275W、C289G、P437Lを含む(参照、表1)。ある実施態様では、使用されるParkin変異体は、R275W、C212YまたはC289Gである。Parkin変異体を使用するアッセイは、野生型Parkinを使用するアッセイの代替法として、またはそのアッセイと組み合わせて使用されてもよい。

1つの実施態様では、アッセイは、(a)野生型または変異Parkinを発現する哺乳動物細胞を得て;(b)細胞を試験作用物に対して曝露し;そして(c)試験作用物に
曝露された細胞におけるParkin凝集を、試験作用物に曝露されなかった対照細胞のParkin凝集特性と比較すること;によるパーキンソン病の処置のための候補化合物のスクリーニングを含む。試験作用物の存在下のParkin凝集の低下されたレベルは、その作用物がパーキンソン病の処置のための候補化合物であることを指示する。1つの実施態様では、哺乳動物細胞は野生型Parkinを発現する。1つの実施態様では、哺乳動物細胞は変異Parkinを発現する。若干の例では、変異Parkinは、S167N、C212Y、T240M、R275W、C289G、P437Lである。好ましくは、R275W、C212YまたはC289Gが使用される。

Parkinはユビキチンリガーゼである(Shimura et al.,2000,Nature Genetics 25:302)。ユビキチンリガーゼ活性は、特異的リガーゼ基質を認識し、そしてE2酵素と相互作用してユビキチン分子をE2から基質に転移するタンパク質の能力によって定義される。リガーゼ活性は、補助タンパク質によって調節されるが、またリガーゼ単独でも生じ得ることが示された(参照、Joazeiro and Weissmen,2000,Cell 102:549−52)。

前記の細胞に基づくおよびタンパク質に基づくアッセイは、独立にまたは種々の組み合わせにおいて使用されて、Parkinタンパク質を発現する細胞におけるプロテアソーム損傷を低下させるパーキンソン病の処置のための候補化合物が同定されてもよい。1つの実施態様では、野生型Parkinを発現する細胞におけるプロテアソーム機能の抑制を改善する作用物の「基本アッセイ」は、さらなるアッセイ、例えば(1)Parkin変異体を使用するプロテアソーム機能アッセイ、(2)Parkin凝集アッセイ、(3)Parkin特異性を確立するプロテアソーム機能アッセイ、(4)Parkin結合アッセイ、(5)インビトロのタンパク質活性アッセイ:との組み合わせにおいて使用されてもよい。組み合わせて使用される場合、これらのアッセイはいかなる順序で実施されてもよい。例えば、最初の高処理スクリーニングがインビトロのタンパク質アッセイを用いて実施され、そして基本の細胞に基づくアッセイが第2の選別法として使用されてもよい。あるいはまた、例えば、細胞に基づくアッセイが最初に実施され、そしてインビトロのタンパク質結合および活性アッセイが第2の選別法として使用されてもよい。アッセイの他の配列および組み合わせも読者には明らかであろう。
野生型および変異Parkinタンパク質は、組み換え哺乳動物細胞(例えば、HEK−293細胞中に安定に組み込まれたpcDNA−Parkin発現ベクター)から発現され、そして精製することができる。あるいはまた、組み換えParkinはBaculoウイルス発現または細菌発現を使用して得られてもよい。しかしながら、今日まで、細菌細胞(例えば、大腸菌)において発現された酵素的に活性のあるParkinの効率的精製をもたらす技術は報告されていない。

B.2 封入体の破壊
種々の方法が、また、封入体の破壊のために知られている。参照、例えば、Current Protocols in Protein Science,前出およびClark,1998,Current Opinion in Biotechnology
9:157−163。本発明の好適な実施態様では、封入体は塩酸グアニジン(例えば、2〜6M GuHCl)および還元剤(例えば、1〜10mM DTT;4mM TCEP[Tris(2−カルボキシエチル)ホスフィン塩酸塩];4〜10mM β−メルカプトエタノールなど)を用いて破壊される。例えば、封入体ペレットが5〜10容量の懸濁バッファー[50mM HEPES,pH8.5,6M GuHCl,10mM β−メルカプトエタノール]と合わせられ、そしてdounceホモジナーザー、音波処理または他の方法を使用して破壊できる。破壊後、何か残っている不溶性材料は遠心分離および/または濾過によって除去されてもよい。得られる可溶性画分はParkinを含有し、そして下記アフィニティークロマトグラフィーのために適当である。
使用されるアフィニティークロマトグラフィーの性質は使用されるタグに応じて異なるであろう。注意すべきは、固相(アフィニティー樹脂)とParkin融合タンパク質の間の親和性相互作用は、高濃度の塩酸グアニジニウム(例えば、2〜6M GuHCl)中で安定でなければならない。1つの実施態様では、固定化金属アフィニティークロマトグラフィー(IMAC)が使用される。IMACは、アミノ酸ヒスチジンがキレート化された遷移金属イオン、例えば、ニッケル(Ni2+)、亜鉛(Zn2+)、銅(Cu2+)またはコバルト(Co2+)を結合する能力を利用する。通常、ニッケルまたはコバルトが使用される。クロマトグラフィーにおいて使用するための固定化されたニッケル生産物は、容易に入手できる(例えば、Ni−NTA樹脂(Qiagen,Inc))。クロマトグラフィーにおいて使用するための固定化されたコバルト生産物は、容易に入手できる(例えば、HIS−SelectTMCobalt Affinity Gel;Sigma−Aldrich Corp.)。
2つの透析段階がGuHCl含有溶液から活性Parkinを回収するために使用される。最初の透析段階では、溶出された材料は、アルギニンおよび還元剤を含有するバッファー溶液に対して透析される。アルギニンは範囲0.1〜1M、例えば範囲0.2〜0.8M、0.3〜0.6Mまたは0.35〜0.5Mにおいて存在してもよい。代表的な還元剤は、DTT(例えば、1〜10mM、例えば10mM);Tris(2−カルボキシエチル)ホスフィン塩酸塩(TCEP、例えば4mM TCEP);β−メルカプトエタノール(例えば、4〜10mM β−メルカプトエタノール)および類似の還元力をもつ薬剤を含む。代表的なバッファーは、0.4Mアルギニン、50mM HEPES,pH8.0、10mM DTTである。
例1:GFPu HEK293細胞
GFPu発現細胞系が次のように作成された:HEK293細胞(ATCCNo.CRL−1573)は、短いデグロンをコードしているオリゴヌクレオチド(Gilon et al.,1998,EMBO Journal 17:2759−66)がGFPのコーディング配列(Heim et al.,1994,Proc.Natl.Acad.Sci.USA 91:12501−504;Accession#P42212)に対してC末端に挿入された構築物を使用して形質転換された。細胞は2ugcDNAによりトランスフェクトされた。細胞は48時間培養され、形質転換株が1000ug/mlのG418(geneticin)を用いて選択された。さらなる7日後、細胞増殖培地(DMEN+1000ug/mlG418)は古い培地を除去し、そして新鮮な培地を添加することによって交換された。細胞は2週間成長を続けられて、G418に耐性である細胞について選択された。次いで、これらの細胞は回収され、そしてFACS技術によって選別されて、単細胞を同定かつ単離した。これらの単細胞は、96穴プレート中に個々に選別され、そして2週間にわたって成長かつ増殖された。次いで、細胞は2並列の96穴プレート中に入れられた。1つのプレートはFacScanによって分析され、他方のプレートは、FacScan分析によってポジティブとして同定されたクローンを拡大するために使用された。

図2は、Parkinタンパク質の発現が、GFPuのような他のプロテアソーム基質の安定化および凝集をもたらすことを具体的に説明するエピ蛍光および免疫蛍光画像を示す。Parkin cDNAは、単独で(パネルAおよびB)またはα−synucleinのcDNAおよびParkincDNAにより(パネルC,DおよびE)実施例1に記述されたようにGFPu 293細胞中にトランスフェクトされた。細胞は48時間後に固定され、そして免疫蛍光検鏡法のために処理された。Parkinタンパク質は、ヒトParkinタンパク質の残基85−96に対する抗体HPA1Aにより染色することによって位置決定され、α−synucleinは、syn−1抗体(Transduction labs,San Jose,CA)により染色することによって位置決定され、そしてGFPuはタンパク質の緑色蛍光に基づいて位置決定された。
異型接合性がPDの発生と相関される(1)野生型Parkinまたは(2)変異Parkinをコードしている発現プラスミドが、HEK293/GFPu細胞中にトランスフェクトされて、プロテアソーム機能および凝集に及ぼす変異Parkinタンパク質の影響が検査された(参照、表1)。
ピ蛍光画像を示す。画像は各サンプルについて同じカメラ設定を使用して記録されて、蛍光強度のレベル、細胞におけるGFPuレベルの直接測定値を反映させた。図において見られるように、Parkin変異体の発現はGFPuをaggresome形成へと促進することができる。図4において示され、そしてArrayScan(R)高処理スクリーニング装置を使用する実験において確認されたように(データ未掲載)、変異体S167N、R275W、C212YおよびC289Gの発現は、GFPレベルを増大した(すなわち、プロテアソーム活性を有意に低下させた)。
散発性PD患者および健全な対照からのヒト脳組織におけるParkinタンパク質の局在場所および特性が、脳抽出物のイムノブロッティングによって決定された。
プロテアソームに標的化される緑色蛍光タンパク質(GFP)を安定して発現するHEK293細胞が、野生型Parkinを発現する発現ベクターまたはベクターのみの対照(pEAK;Edge Biosystems,Mountain View,CA)により一過性にトランスフェクトされた。細胞は5%CO2中37℃で16〜24時間維持された。続いて、細胞は3.7%ホルムアルデヒドで固定され、次いで、PBSにより2回洗浄された。次いで、細胞は1ug/mlのHoechst染料により室温で15分間染色され、次いでPBSにより2回洗浄して、ウェルにPBSの200ulを残した。細胞は、GFPに対して最適化されたXF100フィルターの設定を使用してArrayScanVTIにおいて造影された。データは少なくとも200細胞/ウェルから収集された。Target Activation Bioapplicationプログラムが使用されて、細胞内蛍光(「平均(mean)平均(average)強度」)が分析さ
れたが、ここで、マスクモディファイヤー(mask modifier)は2ピクセルに設定された。
この実施例は、組み換えヒトParkin−オリゴヒスチジン融合タンパク質(すなわち、His6−Parkin)の発現および精製を記述する。実施例8は変性タンパク質をリフォールドさせて酵素的に活性のある生成物を得ることを記述する。高濃度アルギニンおよび強力な還元剤の使用は、変性組み換えParkinをリフォードして、活性酵素を生成することにおいて有効であった。実施例9は、精製において使用されてもよい任意のさらなるクロマトグラフィー段階を記述する。
ヒスチジン・タグに融合された全長ヒトParkinをコードしている配列(参照、配列番号:5)が、細菌発現プラスミドpET30a(Novagen,Madison,WI 53719)中にクローン化されて、pET30a−Parkinが生成された。N末端Hisタグはpet30aベクターによってコードされていて、そして5’および3’末端にそれぞれ付加されたBamHI/HindIII制限部位をもつParkin(NM004562)のPCRフラグメントが、またBamHI/HindIIIにより切断されたpet30aベクター中に挿入される場合に、Parkinに対してフレームN末端において融合される。大腸菌菌株B21(DE3)−pLysSが形質転換され、そして形質転換株がカナマイシンを含むLBプレートにおいて抗生物質耐性に基づいて選択された。
上記サンプルが、硫酸ニッケルを予め負荷された40mlのIMACカラム上に負荷された。クロマトグラフィーバッファーは、次のとおりであった:
バッファーA:50mM HEPES,pH8.0、5.5M GuHCl、10mM β−ME
バッファーB:50mM HEPES,pH8.0、5.5M GuHCl、500mMイミダゾール、10mM β−ME
全てが添加された後(使用直前に新たに添加されるβ−MEを除いて)pHが再チェックされた。サンプルは、サンプルを負荷するために使用される2カラム容量の総量をもつ1%バッファーBを用いて2ml/minにおいて負荷された。カラム容量対して2.5カラム容量の5%Bを用いる4ml/minにおけるさらなる洗浄。10mlフラクションが回収された。
実施例7からのプール3が、(50mM HEPES,pH8.0、10mM DTT)により約1.0mg/mlのタンパク質濃度まで希釈され、そして2X1mlのサンプルが、10kMWCO透析チューブを使用して500ml(1.5M GuHCl、50mM HEPES,pH8.0、10mM DTT)に対して4Cで一夜透析された。翌朝、肉眼的に明らかな沈殿はなかった。サンプルの1つは、(0.4Mアルギニン、50mM HEPES,pH8.0、10mM DTT)に対して4Cで一夜透析された。明らかな沈殿はなかった。透析物が回収され、そして濾過によって清澄化された。
His6−タグされたParkinは、実施例6、節Aに記述されたように、封入体から単離された。GuHCl−可溶化画分は−80℃で保存され、迅速解凍され、合わされた。新鮮DTTが10mMまで添加された。320mlのS200クロマトグラフィーカラムに負荷する前に、タンパク質サンプルは、10k MWCOを用いAmicon Ultra15を使用して濃縮された。最終濃度はSECバッファー(50mM HEPES,pH8.0、3M GuHCl、1mM DTT(実施直前に新しく添加される))を用いて10mg/mlに調節された。
実施例6および7において記述されたように調製されたHis6−Parkinを含有するアッセイ混合液が調製された。50ul容量アッセイは、次のものを含有した:

*1mMメチル化ユビキチン(U−502,Boston Biochem lot#2880574,dH2O中1mMに溶解された)の29.2μlが、ビオチン−ユビキチン(UB−560,Boston Biochem lot#2011584)の50μgを再懸濁するために使用された。これは、17%ビオチン化された1.17mMユビキチンの30μlをもたらす。
Claims (18)
- (a)Parkinを発現する哺乳動物細胞を試験作用物に対して曝露し;
(b)その細胞におけるプロテアソーム機能と、試験化合物に曝露されなかったParkinを発現する対応する哺乳動物細胞のプロテアソーム機能特性とを比較すること;を含み、
ここで、試験作用物に曝露された細胞におけるプロテアソーム機能の増大されたレベルが、その作用物がパーキンソン病の処置のための候補化合物であることを指示する、パーキンソン病の処置のための候補化合物を同定するための細胞に基づくアッセイ。 - (a)Parkinを発現する哺乳動物細胞を得て;
(b)細胞を試験作用物に対して曝露し;
(c)その細胞におけるプロテアソーム機能を、試験作用物に曝露されなかった細胞におけるプロテアソーム機能と比較すること;を含み、
ここで、試験作用物に曝露された細胞におけるプロテアソーム機能の増大されたレベルが、その作用物がパーキンソン病の処置のための候補化合物であることを指示する、パーキンソン病の処置のための候補化合物を同定するための細胞に基づくアッセイ。 - 哺乳動物細胞がGFPuを発現し、そしてプロテアソーム機能がその細胞におけるGFPuの量を測定することによって測定される、請求項1の方法。
- 細胞におけるGFPuの量がGFPu蛍光を測定することによって決定される、請求項3の方法。
- (a)
(i)変異Parkinを発現する哺乳動物細胞を候補化合物に対して曝露し;
(ii)(a)(i)における細胞におけるプロテアソーム機能を、候補化合物に曝露されなかった変異Parkinを発現する細胞のプロテアソーム機能特性と比較すること;を含む、プロテアソーム機能アッセイ、および/または
(b)
(i)Huntingtinを発現する哺乳動物細胞を候補化合物に対して曝露し;
(ii)(b)(i)における細胞におけるプロテアソーム機能と、候補化合物に曝露されなかったHuntingtinを発現する細胞のプロテアソーム機能特性とを比較すること;を含む、プロテアソーム機能アッセイ、および/または
(c)
(i)化合物の存在下で、精製Parkinタンパク質のオートユビキチネーション(autoubiquitination)活性を測定し;そして
(ii)その化合物の存在下での精製Parkinタンパク質のオートユビキチネーション活性を、その化合物の不在下での精製Parkinタンパク質のオートユビキチネーション活性と比較すること;を含む、インビトロの活性アッセイ、および/または
(d)
(i)化合物を精製Parkinタンパク質と接触させ;
(ii)存在する場合には、その化合物とParkinタンパク質の結合を検出すること;を含む、インビトロの活性結合アッセイ、
をさらに含む、請求項1の細胞に基づくスクリーニング方法。 - (i)変異Parkinを発現する哺乳動物細胞を候補化合物に対して曝露し;(ii)その細胞におけるプロテアソーム機能と、変異Parkinを発現し、かつ候補化合物に曝露されなかった細胞のプロテアソーム機能特性とを比較すること;を含むプロテアソーム機能アッセイを含み、ここで、変異ParkinがR42P、S167N、C212Y、T240M、R275W、C289GまたはP437L Parkinである、請求項5の方法。
- 方法が、
(a)封入体を破壊し、そしてヒスチジン・タグされたParkinを含有する可溶性画分を回収し;
(b)(a)からのヒスチジン・タグParkinのアフィニティークロマトグラフィーによってヒスチジン・タグParkinを精製し、該クロマトグラフィーが、グアニジン−HClを含む溶液により結合タンパク質を溶出して、ヒスチジン・タグParkinとグアニジン−HClを含んでなる組成物を生成することを含み;
(c)ヒスチジン・タグParkinとグアニジンを含んでなる組成物を、高濃度のアルギニンおよび還元剤を含有する緩衝水溶液に対して透析して、第1の透析物を生成し;そして
(d)アルギニンを実質的に含まない緩衝水溶液に対して第1の透析物を透析すること;を含む、
Parkinを発現する細菌細胞の封入体からヒスチジン・タグされたParkinを精製する方法。 - 封入体が、グアニジンHClまたはイソチオシアン酸グアニジニウムの存在下で破壊される、請求項7の方法。
- 封入体が2〜6M塩酸グアニジンの存在下で破壊される、請求項8の方法。
- 還元剤が、β−メルカプトエタノール、DTTまたはTCEPである、請求項7の方法。
- 高濃度のアルギニンを含有する緩衝水溶液中の高濃度のアルギニンが、約0.1M〜1Mアルギニンを含有する、請求項7の方法。
- アルギニンを実質的に含まない緩衝水溶液が0.5mM未満のアルギニンを含有する、請求項7の方法。
- アルギニンを実質的に含まない緩衝水溶液が0.1mM未満のアルギニンを含有する、請求項12の方法。
- (b)における溶出溶液が、50mM HEPES,pH8.0、5.5M GuHCl,500mMイミダゾール、10mMβ−ME、0.5mM EDTAであり;(c)における緩衝水溶液が、0.4Mアルギニン、50mM HEPES,pH8.0、10mM DTTであり;そして(d)における緩衝水溶液が、50mM HEPES,pH8.0、0.2M NaCl,10mM DTTである、請求項7の方法。
- ヒスチジン・タグを含む酵素的に活性のある精製組み換えParkinを含んでなる、組成物。
- Parkinが細菌発現系から得られる、請求項15の組成物。
