Description

i)前記GCP−II遺伝子を含む組成物を試験対象物質と接触させる工程;及び
ii)工程i)の反応を確認して前記遺伝子の発現を増進させる活性または抑制する活性を示すかどうかを決定する工程を含む、Aβ脳内蓄積予防及び治療剤スクリーニング方法を提供する。
i)前記GCP−IIタンパク質を含む組成物を試験対象物質と接触させる工程;及び
ii)工程i)の反応を確認して前記遺伝子の発現を増進させる活性または抑制する活性を示すかどうかを決定する工程を含む、Aβ脳内蓄積予防及び治療剤スクリーニング方法を提供する。
<1−1>GCP−IIの発掘
ヒトGCP−IIcDNAを獲得するために、RT−PCRを遂行した。pEF1/Myc(C−末端にMyc−tagを含み、BamH IとNot I制限酵素サイト使用,Invitrogen,米国)ベクター内に挿入するためのプライマーは、BamH I制限酵素を含む配列番号3で表されるフォワードプライマー及びNot I制限酵素を含む配列番号4で表されるリバースプライマーを使用し、下記のようなPCR条件で反応を遂行した。
リバースプライマー:atttgcggccgctggctacttcactcaaagtctc(GCP−II−IRと称する)
フォワードプライマー:cgcagatctaccatgtggaatctccttcacgaaa(GCP−II−2Fと称する)
リバースプライマー:atttagatctttaggctacttcactcaaagtctc(GCP−II−2Rと称する)

GCP−IIのAβ分解程度を測定するために、2ng/ulの合成Aβ40またはAβ42(Biosource international INC.)を、各々GCP−IIが過発現されるHEK293細胞の培養液に入れて20時間後、ELISA分析キット(Biosource international INC.)を使用して培地に残っているAβの量を測定した。
GCP−IIのAβ分解効果が大脳皮質細胞でも起きるのかどうかを調べるための実験として、レンチウイルスを使用して一次大脳皮質細胞にGCP−IIを過発現させた後、Aβが分解される程度を調べた(図2)。皮質細胞は、E16ラット(Sprague−Dawley rat)から分離培養し、培養の10%以下を構成する非神経細胞の拡散を防止するために培養後3〜6日からは、5−フルオロ−2−デオキシウリジン(5−fluoro−2−deoxyuridine)が入った24ウェルプレート(ウェル当り1×105細胞)で培養した。GCP−IIを発現するレンチウイルスを製造するために、pLentiH1.2ベクターにGCP−IIを挿入した後(以下、pLentiH1.2−GCPIIと命名)293T細胞株にpLentiH1.2−GCP−II及びパッケージング構成物(packaging construct)と外皮構成物(envelope construct)を一緒に導入して、48時間後に培地に放出されたウイルスの濃度を計算した。
実験的にGCP−IIを過発現させた細胞だけではなく、生理的水準でGCP−IIが実際に合成Aβを分解することができるのかどうかを確認するために、本発明者等は、HEK293細胞株でsiRNA方法でGCP−IIの発現を遮断した細胞株、GCP−IIを除去した安定的な細胞(knock−down stable cell)を製作して、合成されたAβを添加して16時間後にELISA分析を通じて培地内に残っているAβの量を測定した(図3)。26個の塩基配列からなるGCP−II siRNAが入っているネオマイシン抵抗性の構成物をOpen Biosystems社から購入してHEK293細胞株に形質導入した後、600μg/mLのG418で選別してGCP−IIを除去した安定的な細胞を製作し、RT−PCR方法でsiGCP−II効果を確認した。空ベクターを含むGCP−IIを発現する対照群に比較して、siGCP−IIによるGCP−IIを除去した安定的な細胞(knock−down stable cell)でsiGCP−IIによるGCP−II発現減少を確認し(図3A)、特に、HEK293細胞コロニー中の#4−5と#4−7で顕著に減少した。
<4−1>rGCP−IIの製造
GCP−IIが過発現された細胞でAβの量が減少されることがGCP−IIによる分解作用なのかを確認し、またAβがGCP−IIによって分解されるアミノ酸位置を確認するために、rGCP−IIタンパク質を製作した。
Aβ分解に対するrGCP−IIの効能を確認するのに先立って、37℃、50ng/μlのrGCP−IIによる0.002ないし2μMのN−アセチル−L−アスパルチル−L−[3,4−3H]−グルタメート(glutamate)から遊離された[3H]グルタメートを測定することで、rGCP−IIの酵素活性を示すKm値を分析した。rGCP−IIの酵素作用は、1.0μMのN−アセチル−L−アスパルチル−L−[3,4−3H]から飽和された。その結果、rGCP−IIのKm値は、0.5μMと示された。
GCP−IIがAβを特異的に分解するということを示す実験として、アミルリン、ACTH(adrenocorticotropic hormone)またはインシュリンβ−チェーン(chain)とGCP−IIを37℃で15時間反応させた後、分子量分析装置で各試料の分子量を測定した(図7)。インシュリンβ−チェーン(図7A及び7B)及びACTH(図7C及び7D)は、GCP−IIによって分解されない基質特異性を持っている一方、アミルリンの場合には、GCP−IIと反応させると1500及び2000で二つのピークが生成された(図7E及び7F)。
本発明では、ヒトGCP−IIが実質的に細胞内で形成されたAβに対して分解効果があるのかどうかを確認するために、Aβが過量に形成されるswAPP(Swedish mutant APP)遺伝子を安定に発現する細胞株を製作した。このためにHEK293細胞株にswAPPを形質導入させてG418で選別した後、培地を集めてrGCP−IIタンパク質と反応させて、培地に残っているAβの量をELISAで測定した(図8)。
Aβのモノマー及びマルチマーに対するGCP−IIの分解効果を調べるために、7日間成熟させたAβをrGCP−IIと20時間反応させて、16%トリス−トリシンゲル(tris−tricine gel)で電気泳動した後、抗−Aβ抗体(6E10)でウエスタンブロットを実施した(図9A)。5回以上実施されたブロットティング結果をイメージプログラム(Image J program,NIH,米国)を使用して定量した(図9B)。その結果、GCP−IIの濃度を増加させてAβと反応させると、モノマー形態よりはオリゴマー(またはマルチマー)形態のAβがより早く減少することを観察することができた。このような結果は、GCP−IIが神経細胞に有害なオリゴマー形態のAβをより効率的に分解するということを意味する。
反応溶液内に多様な形態のAβ40が存在するので、本発明者等は前記Aβ40に対するrGCP−IIの親和性を確認した(図10)。
21日間凝集したAβを16時間rGCP−IIタンパク質と反応させた後、変化された凝集Aβの量をコンゴレッド方法を使用して測定することで、rGCP−IIの分解能をもう一度確認した。
本発明のGCP−IIが実質的に生体内で過発現されるAβを分解することができるのかどうかを確認するために、本発明者等は、前記Aβを過発現させた形質転換マウスでGCP−II抑制剤によってAβの分解能が抑制されるのかどうか調査した。具体的には、Aβを遺伝的に過発現させた痴呆症モデルマウス(APP Swedish/presenilin delta9 deltion,12週齢、The Jackson Laboratory,米国)を準備した後、GCP−II抑制剤である2−PMPA(2−phosphonomethyl−pentanedioic acid)を1週間に2回程度、10mg/kgの用量で30日間腹腔内に注入した。ここで、対照群にはリン酸緩衝溶液(PBS)を使用した。前記GCP−II抑制剤を最後に注入した後、マウスの脳組織の中で大脳皮質、海馬及び小脳部分を切除した後、微細秤で各々重さを測定した。続いて、前記組職を室温で6Mグアニジン−ハイドロクロライド(guanidine−hydrochrolide)溶液で4時間程度インキュベートした後、冷たいPBSを1/10の割合で添加して遠心分離させた後、上澄み液を収集した。このように得られた試料は、ELISA測定のために適正な濃度に希釈し、またAβの標準溶液と適正な比率で混合してAβ抗体がコーティングされたプレートに添加した後、3時間反応させた。以後、ホースラディシュペルオキシダーゼ(horseradish peroxidase、HRP)が標識された2次抗体で1時間処理した後、TMB(tetramethylbensidine)で発色させて、前記反応物の吸光度を450nm波長で測定した。また、GCP−II抑制剤が正しく処理されたのかどうか確認するために、前記マウス脳組織の一部分を切開してGCP−II活性を測定した。具体的に、マウス脳組織からGCP−IIが存在する細胞膜層を分離した後、それを50mMのTris−HCl溶液で粉砕した後、超遠心分離器(ultracentrifuge)を使用して1,000,000gで遠心分離した後、その上澄み液を収集した。このように得た細胞膜層で10ugのタンパク質を同位元素(3H)が標識されたNAAG(N−acetyl−aspartydyl−glutamate)と混合して37℃で培養し、これをAG−1X8陰イオン交換樹脂(anion exchange resin)が満たされた96−ウェルスピンカラムを通じて1,000gで遠心分離した後、0.5Mギ酸塩でGCP−IIによって切られたグルタミン酸塩を分離した。このように、分離した試料は、同位元素定量計を使用してGCP−IIの活性度を測定した。
配列番号2は、ヒトGCP−IIをコードするアミノ酸配列である。
配列番号3は、ヒトGCP−IIcDNAをpEF1/MycベクターにクローニングするためにPCR反応に使用したフォワードプライマー配列である。
配列番号4は、ヒトGCP−IIcDNAをpEF1/MycベクターにクローニングするためにPCR反応に使用したリバースプライマー配列である。
配列番号5は、ヒトGCP−IIcDNAをpSG5ベクターにクローニングするためにPCR反応に使用したフォワードプライマー配列である。
配列番号6は、ヒトGCP−IIcDNAをpSG5ベクターにクローニングするためにPCR反応に使用したリバースプライマー配列である。
Claims (16)
- GCP−IIタンパク質を有効成分として含む、Aβの脳内蓄積予防及び治療用薬学的組成物。
- GCP−IIタンパク質が、配列番号2で表されるアミノ酸配列を有することを特徴とする、請求項1に記載のAβの脳内蓄積予防及び治療用薬学的組成物。
- GCP−IIタンパク質が、配列番号1で表される塩基配列を有する遺伝子によってコードされることを特徴とする、請求項2に記載のAβの脳内蓄積予防及び治療用薬学的組成物。
- Aβが、溶解性または不溶性Aβであることを特徴とする、請求項1に記載のAβの脳内蓄積予防及び治療用薬学的組成物。
- ヒトGCP−II遺伝子を含むAβの脳内蓄積予防及び治療剤スクリーニング用組成物。
- ヒトGCP−II遺伝子が、配列番号1で表される塩基配列、配列番号1の多型現象を含む塩基配列及び配列番号1またはその多型現象を含む遺伝子断片塩基配列の中から選択される1種以上である、請求項5に記載のAβの脳内蓄積予防及び治療剤スクリーニング用組成物。
- ヒトGCP−IIタンパク質を含むAβの脳内蓄積予防及び治療剤スクリーニング用組成物。
- ヒトGCP−IIタンパク質が、配列番号2で表されるアミノ酸配列を有するタンパク質、配列番号1の塩基配列または配列番号1の多型現象を含む塩基配列の中から選択された一つから発現されたタンパク質及びGCP−IIと同等な生理的活性を示すGCP−IIポリペプチド断片の中から選択される1種以上である、請求項7に記載のAβ脳内蓄積予防及び/または治療剤スクリーニング用組成物。
- i)請求項5または請求項6の組成物を試験対象物質と接触させる工程;及び
ii)工程i)の反応を確認して前記遺伝子の発現を増進させる活性または抑制する活性を示すかどうかを決定する工程を含む、Aβ脳内蓄積予防及び治療剤スクリーニング方法。 - i)請求項7または請求項8の組成物を試験対象物質と接触させる工程;及び
ii)工程i)の反応を確認して前記遺伝子の発現を増進させる活性または抑制する活性を示すかどうかを決定する工程を含む、Aβ脳内蓄積予防及び治療剤スクリーニング方法。 - GCP−IIが発現される発現ベクターを有効成分として含む、Aβ脳内蓄積予防及び治療剤。
- 発現ベクターが、アデノウイルス(adenovirus)、アデノ随伴ウイルス(adeno−associated virus)、レンチウイルス(lentivirus)、レトロウイルス(retrovirus)及びワクシニアウイルス(vaccinia virus)からなる群から選択される1種以上のウイルス性発現ベクターであることを特徴とする、請求項11に記載のAβ脳内蓄積予防及び治療剤。
- GCP−IIタンパク質または前記タンパク質をコードする遺伝子を含む組換え発現ベクターを治療的有効量で対象に投与する工程を含む、Aβ脳内蓄積予防及び治療方法。
- GCP−IIタンパク質が、配列番号2で表されるアミノ酸配列を有することを特徴とする、請求項13に記載のAβ脳内蓄積予防及び治療方法。
- GCP−IIタンパク質が、配列番号1で表される塩基配列を有する遺伝子によってコードされることを特徴とする、請求項13に記載のAβ脳内蓄積予防及び治療方法。
- 治療的有効量が、0.1μg/kgないし0.1g/kgであることを特徴とする、請求項13に記載のAβ脳内蓄積予防及び治療方法。
