Description
本願は、本明細書における開示と矛盾しない程度までこの全体が参照により本明細書に組み込まれる、2005年7月6日に出願された米国仮出願第60/696,932号への優先権を、米国特許法第119条(e)の下で主張する。
本発明は、電磁放射を用いてサンプルを処理するための方法、デバイスおよびデバイスの構成要素を提供する。本発明は、従来的な病原体減少処理プロセスと比較して、改善された病原体減少の有効性を提供し、かつ処理された生物学的サンプルから誘導される治療剤および再輸液剤の生物学的活性および生存度を最適化する、生物学的サンプル中の病原体の生物学的活性を減少させるための方法およびシステムを提供する。
「クエン酸可塑剤」とは、増強された可撓性、柔軟性、伸長性、耐衝撃性、またはこれらの組み合わせを含む、提供される所望の機械的、物理的、化学的、および光学的特性を提供するために、ポリ(塩化ビニル)などのポリマー材料に加えられる、クエン酸のアルコールエステルなどのクエン酸エステルをいう。治療剤を備える生物学的サンプルを処理するための方法およびデバイスにおいて有用であるクエン酸可塑剤は非毒性である。例示的なクエン酸可塑剤には、クエン酸n−ブチリルトリ−n−ヘキシル、クエン酸トリエチル、クエン酸アセチルトリエチル、クエン酸トリ−n−ブチル、およびクエン酸アセチルトリ−n−ブチルが含まれるがこれらに限定されない。
図1は、ポリ(塩化ビニル)およびクエン酸可塑剤を備える容器(すなわち、クエン酸可塑化PVC容器)中に保持される血液または血液成分中の病原体を減少させる方法を図示する概略図を示す。図1において示されるように、電磁放射(矢印100によって模式的に図示される)が電磁放射の供給源110によって発生され、ポリ(塩化ビニル)およびクエン酸可塑剤を備える容器120に方向付けられる。容器120は、抗凝固剤、増強剤、光増感剤、保存剤または希釈剤の1種以上を選択的に備え得る、病原体減少処理を受けている血液または血液成分サンプル125を保持する。容器120はまた、選択された波長の分布を有する電磁放射(矢印135によって模式的に図示される)、例えば、病原体を直接的に減少させることが可能であり、および/または病原体減少を生じる化学反応を誘導可能である電磁放射を少なくとも部分的に伝達する、少なくとも1つの部分的に透明な表面130を有する。選択された波長の分布を有する電磁放射135は容器120を通して伝達され、そして血液または血液成分サンプル125によって少なくとも部分的に吸収され、これによって、存在する病原体の生物学的活性を減少する。選択的に、電磁放射が、処理を受けるサンプルのすべての構成成分に均一に供給されることを確実にするために、電磁放射への曝露の間、攪拌機160が、血液または血液成分サンプル125を混合するために供給される。攪拌機160は、液体処理の分野において公知である任意の手段を使用して容器120に選択的に接続されてもよい。
背景:
血小板保存の間のいくつかのインビトロ血小板品質パラメーターの変化は、放射性標識された血小板の回収および輸血後の生存によって測定されるインビボ血小板生存度の減少と関連付けられてきた。本研究の目的は、血小板のインビボ回収とのインビトロパラメーターの相関を同定することに焦点を置いた。次いで、本発明者らは、リボフラビンおよび光を使用して、病原体減少プロセスを用いて、血小板のインビボ回収のためのインビトロ細胞品質測定の予測可能性を確証した。
放射性標識血小板を使用する2つの血小板回収臨床研究を、規制調査および認可の下で実施した。最初の研究において、インビトロ細胞品質パラメーターの相関は、種々の線量のUV光で処理し、5日間保存した、トリマアフェレーシス手順によって収集した18例の血小板製剤を使用するインビボ血小板回収を用いて確立した。乳酸産生およびpHに基づくインビボ回収の予測因子を使用して、リボフラビンおよび光(ミラゾール(Mirasol)PRT)(6.2J/ml+50μM リボフラビン)を使用する血小板製剤の病原体減少のために設計された新規なプロセスを開発した。次いで、インビボ回収のための乳酸産生およびpHの予測可能性を、引き続くヒト臨床試験において、PRT処理された血小板を用いる直接的試験を通して確証した。
UV処理は、乳酸産生、グルコース消費およびP−セレクチン発現を増加し、そして保存の間のpH、HSRおよびスワールの減少を生じた。この挙動は、UV線量依存性様式で示された。細胞品質パラメーターにおける変化のすべては、血小板インビボ回収と相関した。これらの中で、乳酸産生およびpHは、血小板インビボ回収と最も強力に相関したパラメーターとして、直線回帰分析によって同定された。乳酸産生およびpHの相関係数は、それぞれ、0.9090および0.8831であり、p値は0.007および0.031であった。乳酸産生およびpHの血小板生存との同様の相関および同じ傾向の予測もまた観察した。これらのアルゴリズムからの予測された5日目の血小板回収値は、ミラゾールPRTを用いて処理して血小板について44〜55%であった。24例の血小板製剤を用いる引き続く臨床研究は、PRT処理した血小板のインビボ回収が51.4+/−18.6であり、十分にこの予測の範囲内にあったことを実証した。
これらの結果は、血小板インビボ回収がインビトロ細胞品質パラメーターから予測できること、ならびにここで利用した条件下では、乳酸産生およびpHが、インビボでのPRT処理血小板のための最も関連性が高いインビトロ指標であることを実証する。
トリマ収集アフェレーシス血小板濃縮調製物
本研究におけるすべての血小板製剤は、トリマ自動血液成分収集システム(Automated Blood Component Collection System)(ガンブロ(Gambro)BCT,レイクウッド(Lakewood),CO)を使用して、地方の血液センターによって、クエン酸n−ブチリルトリ−n−ヘキシル−38%重量%を有する1リットルのクエン酸可塑化ポリ(塩化ビニル)バッグ(「クエン酸化PVC ELP(商標)バッグ」)中に収集された、単一ドナーからのアフェレーシス血小板濃縮物であった。臨床研究においては、標的血小板収率は3.51×1011であった。ところが、標的収率は、2回目の臨床研究においては4.42×1011血小板であった。
本研究は、オレンジ自由州大学の倫理委員会(the Ethics Committee of the University of the Orange Free State)および南アフリカ医学管理審議会(MCC)の検査および認可の下で、オレンジ自由州大学、健康科学部、血液学および細胞生物学部門(the Department of Haematology&Cell Biology,Faculty of Health Sciences,University of the Orange Free State)(ブルームフォンテーン(Bloemfontein)、南アフリカ(South Africa))において実施した。インフォームドコンセントが終了した際に、18歳から65歳の年齢の研究ボランティアを、血小板提供のための地方の判断基準およびAABB要件に基づいて、スクリーニングし、本研究に選択的に含めた。
250mLの体積の血小板濃縮液を、3リットルのポリオレフィンバッグ(センゲバルト、ロードルフ、ドイツ国(Sengewald,Rohrdorf,Germany))に移し、製剤中最終濃度が約50μMであるように、27mLの滅菌500μMリボフラビンの添加を行った。次いで、中程度の線量レベル(7.2J/ml)または高線量レベル(12.4J/ml)のいずれかで、血小板製剤をUV光(リン光体265〜370nm)に曝露した。25〜30℃の温度で攪拌しながら、全体の照射時間は約5〜10分間で変化した。処理後、血小板製剤をクエン酸化ポリ塩化ビニルELP(商標)バッグ(ガンブロ(Gambro)BCT,レイクウッド(Lakewood)に移した。処理したPCおよび対照PCを、標準的な血液バンク条件下で、20〜24℃でさらに5日間保存した。対照血小板製剤は、リボフラビンを添加せず、UV光処理を実施しなかったこと以外は、処理した対応物と同じ様式で調製した。
血小板サンプルは、無菌技術を使用して、血小板保存の0日目、3日目および5日目に研究室試験のために採取し、分析は2時間以内に完了した。血小板計数、スワールスコア、pH、pO2、pCO2、乳酸およびグルコースについてのインビトロ細胞品質試験を、治験実施施設の標準操作手順(SOP)によって実施した。低張ショック応答(HSR)およびP−セレクチン発現は、Ruaneら(Ruane PH,Edrich R,Gampp Dら、リボフラビンおよび光を使用する血小板濃縮物中の選択されたウイルスおよび細菌の光化学的不活化(Photochemical inactivation of selected viruses and bacteria in platelet concentrates using riboflavin and light.)Transfusion 2004;44:877−85.)によって記載されるように測定した。
5日間の保存期間の最後に、研究施設のSOPに従って(ヒト血小板の放射性標識のための地方および国際標準に一致して)、血小板の少量のアリコートを111インジウムで放射性標識した。標識手順は、トロポロンとの塩化111インジウム(アマシャム(Amersham))の混合を通しての111In−トロポロンの形成後に実施した。標識される血小板サンプルのpHが>6.5である場合、このpHは、ペレット化の間の血小板の不可逆的な凝集を妨害するために、6.5まで低下させた。
ミラゾールPRTプロセスを用いて処理した血小板のインビボ回収を確認することに焦点を置いた第2の臨床研究は、ニューハンプシャー(New Hampshire)のダートマス−ヒッチコック医学センター(Dartmouth−Hitchcock Medical Center)およびバージニア(Virginia)のノーフォーク赤十字センター(Norfolk Red Cross Center)において実施した。本研究は、治験用医療機器に対する適用免除(IDE)の下で、両方の研究施設のための施設内審査委員会(IRB)および米国FDAによって審査および認可された。インフォームドコンセントが終了した際に、各々の参加しているボランティアは、血小板供与のためのすべてのFDAおよびAABB判断基準に基づいて、適格性についてスクリーニングし、次いで本研究に選択的に含めた。
血小板アフェレーシス収集の2〜8時間後に、250mLの体積の血小板を、収集ELPバッグから、別の照射および保存ELP容器に重量測定的に移した。28mLの体積のリボフラビン溶液(500μM)を、シリンジを有する滅菌バリアフィルターを通して試験製剤に加えた。この製剤を、ナビガントバイオテクノロジーズ,インク(Navigant Biotechnologies,Inc.)(レイクウッド(Lakewood),CO)によって製造されたミラゾールPRT照射デバイスの中に配置し、6.2J/ml線量の紫外光に曝露した。対照製剤はリボフラビンを加えず、UV光で処理しなかった。ミラゾールPRTプロセス後、製剤(対照および試験)は、5日間、水平方向に攪拌しながら、22±2℃の重要の血液バンク条件下で保存した。
5日間の保存期間の最後に、Holmeらによって特定された手順を使用して、血小板製剤のアリコートを111In−オキシムで放射性標識した。Holmeら(Holme S,Heaton A,Roodt J.111Inおよび51Crを用いる同時標識法は、保存された血小板濃縮物の血小板生存度の正確な評価を可能にする(Concurrent label method with 111In and 51Cr allows accurate evaluation of platelet viability of stored platelet concentrates.)Br J Haematol 1993:84:717−23)によって記載されるように、各放射標識サンプルに対して2時間の放射溶出評価を実施した。
統計学
すべてのインビトロ細胞品質パラメーターについて、平均および標準偏差を計算した。統計学的な比較は、適用可能な場合、反復測定のための共分散分析(ANCOVA)を使用して実施した。この分析は、SAS v8.1の中の「proc mixed」を使用して実施した。配列効果は最初にモデルに含まれたが、有意でない場合には脱落した。
臨床試験1:UV処理は細胞の解糖代謝を加速する
南アフリカの監督官庁であるMCCの倫理委員会認可および届出後、全体で18例の血小板製剤を、トリマアフェレーシス手順を使用して、2.9〜3.8×1011血小板の範囲の収率で収集した。製剤を、収集日に、ポリオレフィン容器、センゲバルド(Sengewald)バッグ中で、50μMリボフラビンの存在下で、中程度UV線量(7.2J/ml;N=5)または高UV線量(12.4J/rhl;N=6)のいずれかで、UVで処理した。さらなる7例の製剤は、UV光で処理しなかった対照として働いた。すべての処理および対照PC製剤は、ELP(商標)バッグに移され、ここで、アフェレーシス収集後5日間、通常の血液バンク条件下で保存した。保存の0日目(処理前)、3日目、および5日目において、すべてのPC製剤からのサンプルを、pH、pO2、pCO2、乳酸およびグルコース濃度、P−セレクチン、HSRならびに血小板スワールについて測定した。表1は、これらの細胞代謝および品質測定の結果を要約する。処理した血小板は、対照血小板と比較したときに、保存の間のサンプルpHの減少に伴う乳酸産生およびグルコース消費の増加を示し、このことは、UV光処理が細胞解糖代謝を増加したことを示した。UV処理はまた、血小板保存の間に、P−セレクチン発現の増加およびHSRおよびスワールスコアの減少を加速した。血小板保存の間のこれらの変化の厳しさは、適用されたUV線量のレベルと直接的に比例していたようである。
5日間の保存の最後に、すべての処理および対照血小板サンプルは、インジウムで放射標識し、続いて、同じドナーへの標識した血小板の注入を行った。インビボで注入した血小板の放射能は7日目まで測定し、注入した血小板のインビボ回収は、複数ヒットモデル分析を使用して計算した。平均インビボ回収は、ゼロ、中程度線量、および高線量のUV光で処理した血小板について、それぞれ、60%(SD=16)、30%(SD=8)および14%(SD=7)であった。図8A〜8Hは、インビボ血小板回収とのインビトロ細胞品質パラメーターの相関を示す。インビボ血小板回収は、乳酸産生(a)、5日目の22℃におけるpH(b)、グルコース消費(c)、5日目のP−セレクチン発現パーセント(d)、5日目のスワールスコア(e)、5日目のHSRパーセント(f)、pO2(g)、およびpCO2(h)の値の関数である。グラフ中で、白丸は対照血小板であり、黒ひし形は中程度線量のUV光処理血小板であり、そして黒四角は高線量のUV光処理血小板である。図8A〜8Hにおいて、保存の間または5日目に測定された各代謝および細胞品質パラメーターは、すべての個々の血小板製剤について、血小板回収に対してプロットする。インビボ回収とのパラメーターの明確な相関を観察した。グラフの中にプロットしたパラメーターの中で、乳酸産生、pH、グルコース消費、およびP−セレクチンは、測定したインビボ回収と良好な相関を有するようである。これらの相関の程度は直線回帰分析によって定量され、表2に要約される。r値とF値の両方の相関係数は、乳酸産生およびpHがインビボ血小板回収と最も顕著に相関されることを同定し、p値はそれぞれ0.007および0.031であった。残りのパラメーターは、インビボ回収との相関のこれらの程度の順に、グルコース消費速度、P−セレクチン発現、HSR、スワール、pCO2およびpO2であった。
南アフリカにおいて実施された最初の臨床研究から得られた情報は、ミラゾールPRT処理手順のために血小板処理条件を設計するために使用した。これらの条件下で、ミラゾールPRT処理は、血小板保存の間の一連のインビトロ細胞品質試験によって評価されるように、血小板の治療剤的価値を損なうことなく、病原体還元能力を最大化した。最初の臨床研究において説明された血小板プロセシングスキームとは異なり、本研究における新鮮な血小板は、クエン酸化塩化ポリビニルELP(商標)バッグの中で、50μMリボフラビンの存在下で、6.2J/mlのUV光にこれらを曝露することによって、ミラゾールPRTでプロセスした。次いで、製剤を、処理後5日間の間、同じバッグの中に保存し、バッグ輸送ステップの必要性を除外した。図4に図示されるように、ELPバッグの使用は、短波長領域(285〜305nm)における光伝達の顕著な減少、および比較的長波長(365〜400nm)における伝達の相加を通して、最初の臨床研究において使用したセンゲバルトバッグを超えた利点を有した。長波長を有する領域は、リボフラビンが最大吸収を有する領域と一致する。インビトロ血小板細胞品質に対する、ならびにウイルスおよび細菌不活化に対する、本研究において使用されるミラゾールPRT処理条件の効果は、Liら(Li J,Xia Y,Bertino AMら 保存の間のヒト血小板のアポトーシスのメカニズム(The mechanism of apoptosis in human platelets during storage.)Transfusion 2000;40:1320−9.)およびRuaneらによって報告されているように、広範に評価されている。
インビトロ細胞品質パラメーター測定から血小板のインビボ生存度を予測するために多くの試みが行われてきた。これらの予測を使用することの成功は、これらのパラメーターと回収のインビボ測定の間の比較的乏しい相関の限界におそらく起因して、わずかに報告されてきたのみであった。難問は、レシピエントの生理学的状態に主として起因する正常ボランティアの中のインビボ回収測定のかなり大きな変動から、および血小板製剤のためのインビトロ試験における生物学的変動性からの両方に由来する。相関の程度はまた、各変数の範囲および分布に依存する。得られる細胞品質パラメーターの値の範囲がより広く、かつこの分布がより均一であるほど、観察することができる相関はより良好である。本研究において観察することができる細胞品質値の範囲を広くするために、血小板は、最初の臨床研究において、3つの異なる線量のUV光で処理し、5日間保存した。結果は、すべての測定したインビトロ細胞品質パラメーターが、用量依存的様式でUV光処理に応答したことを示した。UV処理は、乳酸産生、グルコース消費、およびP−セレクチン発現を増加し、そして保存の間にpH、HSRおよびスワールの減少を生じた。細胞品質パラメーターのこれらの変化のすべては、様々な程度まで血小板インビボ回収と相関した。これらの中で、乳酸産生およびpHを、血小板インビボ回収と最も強力に相関したパラメーターとして、直線回帰分析によって同定した。乳酸産生およびpHの相関係数は0.909および0.883であり、p値は0.007および0.031であった。インビトロ細胞品質の血小板生存との相関の同様のパターンもまた観察した。これらの予測アルゴリズムの値は、引き続く臨床研究を通して首尾よく確認し、この研究は、観察された血小板インビボ回収が予測値の範囲内に十分にあることを実証した。これらの結果は、以前に示唆されてきたように、血小板インビボ回収がインビトロ細胞品質パラメーターから予測可能であること、ならびにここで利用された条件下では、乳酸産生およびpHが、インビトロPRT処理血小板生存度の最も関連性のあるインビトロ指標であることを実証する
ここで報告した本発明者らの観察は以前の研究と一貫している。乳酸塩は、血小板解糖系路の最終代謝生成物であり、乳酸に転換され、保存の間に保存媒体に放出される。乳酸の蓄積は、血小板解糖流量の状態を直接的に反映するのに対して、UV処理は、線量依存的様式で乳酸産生速度を刺激し、高エネルギーUV光が血小板解糖流量を加速することを示す。乳酸の蓄積は、保存の間の血漿pHの減少の原因である。新鮮な血漿は緩衝能を有するので、pHは、乳酸産生が有するのと同程度のインビボ回収との相関を有するとは予測されない。本発明者らの直線回帰分析はこのことを確認した。興味深いことに、乳酸産生の上流前駆体であるグルコース消費は、乳酸産生速度よりもインビボ回収との比較的低い相関係数を実証し、統計学的な有意さはなかった(p>0.05)。この観察のための1つの可能な説明は、グルコース消費が、解糖の最終生成物である乳酸に独占的に導かれる解糖系路には完全には関連付けられていないかもしれないことである。確かに、解糖およびTCAサイクルにおけるグルコール誘導性中間体の多くは、脂肪酸、脂質、アミノ酸およびタンパク質にもまた転換可能である。代替的な説明は、保存の開始時において製剤中に存在するグルコールの残存レベルが、保存の間にグルコース消費の速度を変化させる可能性があることである。解糖の速度はグルコースの濃度に直接的に関連するので、このメカニズムは、応答メカニズムのさらなるバリエーションを導入する際に働き得ることが可能である。
1.緒言
O2およびCO2に関するクエン酸可塑化ポリ(塩化ビニル)容器の透過性は、本発明の方法におけるこれらの有用性を確認するために、電磁放射への曝露の前後で特徴付けした。血液および血液成分をプロセスするために有用である波長、放射エネルギーおよび放射出力を有する電磁放射への曝露の際に、顕著に減少しない、O2およびCO2に関する透過性を示す容器を提供することが本発明の目的である。さらに、不必要かつ資源消費的なさらなるサンプル移動ステップを回避するために、血液および血液成分を保存することと、これを電磁放射を用いて処理することの両方のために有用な多機能容器を提供することが本発明の目的である。
本研究において、1リットルのクエン酸可塑化ポリ(塩化ビニル)ELP血小板保存バッグに、250mLの生理食塩水(血小板製剤体積を模倣するため)および28mLのリボフラビンで満たす。これらのバッグをイルミネーターの中に配置し、UV電磁放射に曝露する。これらのバッグは、0(対照サンプル)または5J/cm2(試験サンプル)に等しい標的エネルギーが送達された後で取り出す。液体を引き続き取り出し、バッグを切り開き、内部を吸い取って乾燥させる。試験品目の3つの複製を2つのエネルギー点で実施する。試験品目の6つを、O2伝達試験のために使用し、残りの6つの試験品目をCO2伝達試験のために使用する。ガス伝達試験を、確立されたプロトコールを使用して、90% RHおよびCO2のために改変されたASTM D3985に従って実施する。表4および5は、本研究のための、試験品目行列および照射条件の要約をそれぞれ提供する。
表6は、計算した平均および標準偏差を含む、O2伝達速度についての結果および要約統計量を列挙する。加えて、平均のt検定(α=0.05)を、試験サンプルおよび対照サンプルの各群について実施した。図9は、各サンプル(各群について3つのバッグサンプル、サンプルあたり2つの複製)についての測定したO2伝達速度を示す。図10は、各群(試験および対照)についてのO2の平均を示し、エラーバーは±1標準偏差を示す。図10において示されるように、試験実験および対照実験についてのO2伝達速度の測定した平均値は、それぞれの標準偏差の中にある。加えて、O2伝達速度の決定した平均は、t検定評価によって、試験サンプルと対照サンプルの間で有意な違いはない。

Claims (32)
- 生物学的サンプル中の病原体を減少させるための方法であって、
容器がポリマー材料およびクエン酸可塑剤を備え、前記容器は波長の分布を有する電磁放射を伝達する、生物学的サンプルを保持する前記容器を供給することと、
前記波長の分布を有する電磁放射が前記容器によって伝達され、前記生物学的サンプルによって少なくとも部分的に吸収され、これによって、前記生物学的サンプル中の病原体を減少する、前記容器を電磁放射に曝露することと、
の各ステップを備え、
前記容器による前記波長の分布を有する電磁放射の伝達が、電磁放射への曝露の間に実質的に一定である方法。 - 前記容器による前記波長の分布を有する電磁照射の伝達は、電磁放射への曝露の間に10%以内まで一定である、請求項1に記載の方法。
- 前記容器による前記波長の分布を有する電磁照射の伝達は、電磁照射への曝露の間に5%以内まで一定である、請求項1に記載の方法。
- 前記波長の分布は、電磁スペクトルの紫外領域、電磁スペクトルの可視領域、または両方にある、請求項1に記載の方法。
- 前記容器は、約0.1分から約30分の範囲から選択される時間、前記電磁放射に曝露される、請求項1に記載の方法。
- 前記容器は、約0.1ジュールcm−2〜約10ジュールcm−2の範囲から選択される正味の放射エネルギーに曝露される、請求項1に記載の方法。
- 前記波長の分布を有する前記電磁放射は、約285ナノメートル〜約365ナノメートルの範囲にわたって選択される波長を有する、請求項1に記載の方法。
- 前記クエン酸可塑剤は、
クエン酸トリエチルと、
クエン酸アセチルトリエチルと、
クエン酸n−ブチリルトリ−n−ヘキシルと
クエン酸アセチルトリ−n−ブチルと
からなる群より選択される、請求項1に記載の方法。 - 前記容器は少なくとも1種のさらなるクエン酸可塑剤をさらに備える、請求項1に記載の方法。
- 前記クエン酸可塑剤の濃度は約25%〜約50%重量の範囲にわたって選択される、請求項1に記載の方法。
- 前記クエン酸可塑剤の濃度は約38%重量である、請求項1に記載の方法。
- 前記ポリマー材料はポリ(塩化ビニル)である、請求項1に記載の方法。
- 前記容器は、可塑剤、光安定剤、熱安定剤、抗酸化剤、難燃剤、離型剤、および核化剤からなる群より選択されるさらなる添加剤をさらに備える、請求項1に記載の方法。
- 前記生物学的サンプルは血液成分である、請求項1に記載の方法。
- 前記生物学的サンプルは体液である、請求項1に記載の方法。
- 前記血液成分は、血小板、血漿、赤血球、白血球、および血漿タンパク質からなる群より選択される、請求項14に記載の方法。
- 前記生物学的サンプルは光増感剤をさらに備える、請求項14に記載の方法。
- 前記光増感剤は7,8−ジメチル−10−リビチルイソアロキサジンである、請求項17に記載の方法。
- 前記生物学的サンプルは、全血、血液成分、赤血球含有血液成分、血漿含有血液成分、血小板含有血液成分、白血球含有血液成分、血液由来の1種以上のタンパク質を含有する溶液、および腹腔溶液からなる群より選択される材料を備える、請求項1に記載の方法。
- 生物学的サンプル中の病原体を減少させるための方法であって、
容器がポリ(塩化ビニル)および少なくとも1種のクエン酸可塑剤を備え、前記容器は選択された波長の分布を有する電磁放射を伝達し、そして前記容器による前記選択された波長の分布を有する電磁放射の伝達が電磁放射への曝露の間に実質的に一定である、前記生物学的サンプルを保持する前記容器を供給することと、
前記選択された波長の分布にわたる波長の関数として、前記容器の伝達のパーセンテージを測定することと、
電磁放射の供給源を使用して電磁放射を発生することと、
前記光源によって発生される前記電磁放射の出力をモニターすることと、
前記容器の前記測定された伝達のパーセンテージを使用して、前記生物学的サンプルに送達された放射出力を計算することと、
所望の程度の病原体減少を提供するために必要とされる前記生物学的サンプルの曝露時間を決定することと、
前記選択された波長の分布を有する電磁放射が前記容器によって伝達され、前記生物学的サンプルによって少なくとも部分的に吸収され、これによって、前記生物学的サンプル中の前記病原体を減少する、前記曝露時間の間、前記容器を電磁放射に曝露することと、
の各ステップを備える方法。 - 前記クエン酸可塑剤は、
クエン酸トリエチルと、
クエン酸アセチルトリエチルと、
クエン酸n−ブチリルトリ−n−ヘキシルと、
クエン酸アセチルトリ−n−ブチルと
からなる群より選択される、請求項20に記載の方法。 - 前記クエン酸可塑剤の濃度が約25%〜約50%重量の範囲にわたって選択される、請求項20に記載の方法。
- 前記クエン酸可塑剤の濃度が約38%重量である、請求項20に記載の方法。
- 前記ポリマー材料はポリ(塩化ビニル)である、請求項1に記載の方法。
- 生物学的サンプル中の病原体を減少させるための方法であって、
容器がポリマーおよび少なくとも1種の光学フィルター添加剤を備え、前記添加剤の組成および濃度が、第1の波長の分布を有する電磁放射が前記容器によって少なくとも部分的に伝達されるように選択され、第2の波長の分布を有する電磁放射の伝達が実質的に妨害され、前記第1の分布を有する電磁放射が前記生物学的サンプルの病原体の減少を開始可能である、前記生物学的サンプルを保持する容器を供給することと、
前記第2の波長の分布の電磁放射の伝達が実質的に妨害され、前記第1の波長の分布を有する電磁放射が前記容器によって伝達され、前記生物学的サンプルによって少なくとも部分的に吸収され、これによって、前記生物学的サンプル中の前記病原体を減少する、前記容器を電磁放射に曝露することと、
の各ステップを備える方法。 - 前記添加剤は前記ポリマーのポリマーネットワーク中に固定化される、請求項25に記載の方法。
- 前記添加剤および前記ポリマーはコポリマーであり、前記添加剤は前記ポリマーに共有結合されている、請求項25に記載の方法。
- 前記添加剤は、1種以上のアミノ酸、1種以上のタンパク質、1種以上のペプチド、1種以上の核酸、および1種以上のオリゴヌクレオチドからなる群より選択される、請求項25に記載の方法。
- 前記添加剤は、クエン酸トリエチル、クエン酸アセチルトリエチル、クエン酸n−ブチリルトリ−n−ヘキシル、およびクエン酸アセチルトリ−n−ブチルからなる群より選択される1種以上のクエン酸可塑剤からなる群より選択される、請求項25に記載の方法。
- 前記添加剤は、チロシン、ヒスチジン、フェニルアラニンおよびトリプトファンからなる群より選択される1種以上のアミノ酸である、請求項25に記載の方法。
- 前記ポリマーはポリ(塩化ビニル)である、請求項25に記載の方法。
