ジノアデノシン、N−エチルカルボキサミドアデノシン、5’−(N−シクロプロピル)カルボキサミドアデノシン、N−[2−(3,5−ジメトキシフェニル)フェニルエチル]アデノシン、N−(2−メチルベンジル)アデノシン、2−オクチニルアデノシン、2−フェニルアミノアデノシン、2−[2−(4−フルオロフェニル)エトキシ]アデノシン、2−[(シクロヘキシルメチレン)ヒドラジノ]アデノシン、2−クロロ−N6−(3−ヨードベンジル)アデノシン−5’−N−メチルウロナミド、N6−(3−イソチオシアナートベンジル)アデノシン−5’−N−メチルウロナミド、2−クロロ−N6−(3−ヨードベンジルアデノシン−5’−N−メチルカルボキサミド、デフィブロチド(短鎖デオキシリボヌクレオチド)、ピナシジル、アデノシン−5’−モノリン酸、アデノシン−5’−モノリン酸メチルエステル、N6−メチルアデノシン−5’−モノリン酸、8−ブチルアミノアデノシン又はこれらの過ヨウ素酸酸化物が好ましく、また、育毛効果の面では、A1及びA2タイプのアデノシン受容体への刺激作用が高い化合物がより好ましい。
R−S−CoA
[式中、Rは分岐しても良い炭素数14〜30のアシル基で、二重結合を0〜3個含む]で示される長鎖アシルコエンザイムA、その中でも特にオレオイル−CoA、パルミトイル−CoA、オレイル−CoA、ミリストイル−CoA、ペンタデカノイル−CoA、ステアリル−CoAが挙げられる。
COS−1細胞(3×105個 /直径35mmシャーレ)を10%牛胎児血清を含んだダルベッコ変性MEM培地にて5%CO2雰囲気下、37℃で培養した。 別に、ヒトアデノシン受容体cDNA(GenBank Accession No.X68485、参考文献:C.A.Kollias-Bakerら、The Journal of Pharmacology and Experimental Therapeutics,281(2),761-768(1997))とラットスルホニルウレア受容体cDNA(GenBank Accession No.L40624、参考文献:Y.Tokuyamaら、Biochemical and Biophysical Research Communications, 220, 532-538 (1996))をpcDNA3.1ベクター(インビトロジェン社、カリフォルニア、米国)とpcDNA3.1/Hygroベクター(インビトロジェン社、カリフォルニア、米国)に常法に従ってそれぞれ導入し、pcDNA3.1hADおよびpcDNA3.1/Hygro raSURを得た。
上記操作によって得られた細胞を用いて、以下の試験を行った。
スルホニルウレア受容体刺激剤として知られているニコランジルを用いて、本発明のスクリーニング法の有効性を確認した。その結果、図2に示す通り、ニコランジル25、100μM添加により細胞内カルシウム濃度が上昇するが、この作用はアデノシン受容体の阻害剤である8−スルホフェニルテオフィリン1μMの前処理により抑制されることが確認された。
アデノシン受容体刺激剤としてアデノシン1.0および10.0μMを添加すると、図3に示す通り、細胞内カルシウム濃度は濃度依存的に上昇した。この作用はアデノシン受容体阻害剤である8−スルホフェニルテオフィリン1μMの前処理により抑制された。

上記成分を撹拌し、均一に溶解させ外用ローション剤を調製した。
表1に示すように、参考例1におけるニコランジルを、ピナシジル(参考例2)、クロマカリム(参考例3)、MnATP(参考例4)、メグリチニド(参考例5)、L−アルギニン(参考例6)に代えた以外は参考例1と同様に外用ローション剤を調製した。

試験法はマウスを1群10匹とし、試料投与群および無投与群の8群に分け、マウスの背部をバリカンおよびカミソリにて剃毛し、それぞれのサンプルを1日1回、剃毛した背部8平方cm当たり100μLずつ塗布した。参考例23〜28の試料投与群、比較例の試料投与群および無投与群の塗布面積に対する毛の再生面積(15日後)を表2に示す。

- プリン誘導体に対する受容体遺伝子がアデノシン受容体遺伝子である請求項1または2記載のスクリーニング方法。
- プリン誘導体に対する受容体遺伝子がATP受容体遺伝子である請求項1または2記載のスクリーニング方法。
- 細胞内へのカルシウム流入量を、蛍光カルシウム指示薬を用いた蛍光強度で測定することを特徴とする請求項1から4のいずれかに記載のスクリーニング方法。
