(1)界面活性剤−カテキン類混合液の調製
界面活性剤は、オートクレーブ処理(121℃、20分間)後、滅菌水で所定の濃度に希釈して使用した。
カテキン類溶液は、カテキン製剤(POLYPHENON 70A(三井農林(株)製))を10000ppmとなるように純水に溶解し、pHを6.5に調整し、所定の濃度に希釈後、ろ過滅菌(0.2μm、関東化学社製)した。
界面活性剤−カテキン類混合液は、9mLの0.1Mリン酸(pH6.5)緩衝溶液に上記滅菌処理した界面活性剤を1mL添加混和し、カテキン類濃度の終濃度が所定の濃度となるようにカテキン類を加えて調製した。なお、pH調整(pH5又は8)をする場合には、リン酸緩衝溶液(pH5〜8付近)を適宜用い、表1に示すような所望のpHに適宜調整した。

対象とする細菌は、LB培地(Becton&Dickinson社製;pH7.0)にて24時間、30℃、振とう培養を行い、107〜108CFU/mL菌液に調製した。
LB培地組成は、1%カゼイン膵液分解物、0.5%酵母エキス、0.5%NaClからなる。
上記界面活性剤−カテキン類混合液10mLに予め培養した107〜108CFU/mLに調製した上記菌液を100μL添加し、終菌数を105CFU/mL程度に調整した。30℃、24時間静置培養し、随時ボルテックスを行った。
24時間後の菌数を界面活性剤未添加カテキン類溶液はプレーティングにより求め、界面活性剤添加カテキン類混合液はLP希釈液後、プレーティングにより細菌の菌の存在を確認した。
プレーティングについては、各希釈液100μLをTSA寒天平板に塗抹して48時間、37℃で静置培養した。培養後のコロニー数から生菌濃度を算出した。
使用培地:TSA寒天平板培地(Tryptic Soy Agar,Becton Dickinson 社):1.7%カゼイン分解物、0.3%大豆酵素分解物、0.25%デキストロース、0.5%塩化ナトリウム、1.5%寒天、pH7。

The number of bacteria after 24 hours was determined by plating for a catechin solution without addition of a surfactant, and the presence of bacteria was confirmed by plating after a LP diluted solution of a surfactant-added catechin mixture.
For plating, 100 μL of each diluted solution was smeared on a TSA agar plate and statically cultured at 37 ° C. for 48 hours. The viable cell concentration was calculated from the number of colonies after culture.
Medium used: TSA agar plate (Tryptic Soy Agar, Becton Dickinson): 1.7% casein degradation product, 0.3% soybean enzyme degradation product, 0.25% dextrose, 0.5% sodium chloride, 1.5 % Agar, pH 7.
