In one aspect, the present specification has an effect of providing an antimicrobial composition comprising a water scouring extract having antibacterial activity against pathogenic bacteria and antibiotic resistant bacteria as an active ingredient. In another aspect, the present specification has an effect of providing an antimicrobial composition comprising an effective water-scarifier extract which is harmless to the human body and has no side effects.
Description
The present invention discloses an antimicrobial composition comprising a water scouring powder extract as an active ingredient.
Since human skin is in direct contact with the surrounding environment, it is easily exposed to bacteria and fungi. Staphylococcus sp. , Streptococcus sp. , Gram-negative bacteria, Pseudomonas aeruginosa , Escherichia coli , and fungi, which are Gram-positive bacteria and Staphylococcus sp. It is known that various bacterial and fungal infections cause various skin diseases.
In general, antibiotics are selective toxic substances that kill microorganisms and are toxic to humans or animals, and are not inactivated by enzymes in the body. This is a destruction of the cell membrane of microorganisms, Inhibition of transcription and translation of information, biosynthesis of cell wall, and the like, thereby exhibiting an effect through the mechanism of inhibiting the proliferation of microorganisms.
Sir Alexander Fleming first discovered penicillin in 1929 and Flory and others first separated and purified penicillin from a culture of Penicillium chrysogenum in 1939, Has been known as a miracle medicine by saving the lives of many patients with. Since then, research has been conducted in earnest until 1959. Benzylpenicillin, streptomycin, tetracyclin and macrolide erythromycin, which have been used so far as antibiotics, have been developed . Penicillin was first developed by the synthetic method in 1965, and semicylline, a semisynthetic penicillin, was developed. In 1973, cephalosporin type cephalosporin was developed.
However, shortly after penicillin was used, bacteria resistant to this began to appear. The first penicillin-resistant strains were reported in Australia in 1967, and bacteria that were resistant to methicillin or cephazoline soon began to appear, causing serious problems. In addition, a number of antibiotics such as quetiapine, carbapenem, monobactam, and glycosides were developed in the 1980s, and many of these antibiotics were first developed in South Africa. Respectively. However, penicillin-resistant Staphylococcus aureus (MRSA) and methicillin-resistant Staphylococcus aureus (MRSA) started to become a problem in the world after this period. In the 1990s, vancomycin-resistant Streptococcus (VRE) It has emerged as a medical concern.
On the other hand, most antibiotics not only have several chiral centers but also have complex structures such as macrolides or peptides, so that their synthesis is difficult. In addition, this synthesis method is also very economical compared with the fermentation method. In addition, in the recent strict safety review process, it is often found that they can not meet the safety of human body 100%. Therefore, the safety of microbicides is emerging as an urgent problem.
Thus, most of the synthetic antibiotics are known to have side effects. Due to excessive abuse of antibiotics, the appearance and resistance rate of resistant bacteria to existing antibiotics are increasing. Especially recently, due to the appearance of super bacteria resistant to antibiotics It is urgently required to develop a new natural antibiotic agent which does not cause resistance by using relatively safe natural materials.
In the case of microbial disinfectants, there is a strong tendency to use natural antimicrobial agents which are not harmful to the human body, and the development of nontoxic natural antimicrobial agents is currently being actively promoted due to the wide range of industrial applications. In addition, modern people are increasingly inclined to prefer natural remedies from natural sources because of their rejection of chemical synthetic products worldwide. There are fields that require the use of natural antimicrobial agents, such as wet tissues and cleansing tissues, which are commonly used in everyday life. Since they are in direct contact with the skin, a harmful processing agent can be absorbed into the human body through the skin This is because there is a risk. As such safety is emphasized, various kinds of safety such as transdermal toxicity, acute toxicity, mutagenicity and the like are required for antimicrobial agents.
At present, representative examples of natural antibiotics that can be used include niacin and polylysine, lactic acid bacteriocins. However, they are required to have a large amount for antibacterial activity, and they are expensive. In addition, chitosan is more effective against Gram-positive bacteria than Gram-negative bacteria, but exhibits relatively weak antibacterial activity against fungi, and has little effect on genus containing chitosan in the cell wall.
In one aspect, the present invention aims to provide an antimicrobial composition comprising, as an active ingredient, a water scouring extract having antimicrobial activity against pathogenic bacteria and antibiotic resistant bacteria.

Test example.
The methanol extract of waterslip using the outpost of Myriophyllum verticillatum was purchased from the Plant Extract Bank of Korea Research Institute of Bioscience and Biotechnology. Each extract was dissolved in dimethyl sulfoxide (DMSO) and used for the experiment.
Staphylococcus aureus , Staphylococcus epidermidis , opportunistic infectious pathogens Pseudomonas aeruginosa and Burkholderia sephacia as strains for the antibacterial activity test of the water scouring extract, ( Burkholderia cepacia ) was purchased from the Korea Genetic Resources Institute (KCTC) and used for the experiment. Methicillin resistant staphylococcus aureus (MRSA) was purchased from Wonju Severance Christian Hospital and used for the experiment.
In order to confirm the antimicrobial activity of Myriophyllum verticillatum extract, a disk diffusion method was performed using the prepared sample and strain.
Specifically, mouse Staphylococcus fringing grown in liquid medium for 24 hours (Staphylococcus aureus), Staphylococcus epidermidis (Staphylococcus epidermidis), opportunity infectious pathogens Rouge Pseudomonas labor (Pseudomonas aeruginosa), Burke holde Ria Sepharose cyano (Burkholderia and methicillin resistant staphylococcus aureus (MRSA) were diluted 1/10 each, and 200 μl of each was diluted in an agar medium. The waterwash extract prepared was dissolved in DMSO to a concentration of 20 mg / ml, and a round filter paper having a diameter of about 8 mm was impregnated with 3 쨉 l of the solution, and the solution was spread on an agar medium to diffuse in an agar medium. The results of the antimicrobial activity measurement are shown in Table 1 below by measuring the clear zone in which bacterial growth was inhibited after incubation at 30 ° C or 37 ° C for 24 hours.
Table 1. Antimicrobial activity of each strain according to the size of the transparent zone
* Control: DMSO
** -: No suppression effect, +: less than 1mm, ++: more than 1mm, less than 2mm, +++: more than 2mm
As a result, it was confirmed that water waxy extract has a antibacterial activity by forming a transparent ring in all the above-mentioned bacteria. That is, when the water scarlet extract was included, it exhibited a wide transparent zone or a high antimicrobial activity, in which bacteria hardly grow. Especially, S. aureus and S. epidermidis showed strong antimicrobial activity. Therefore, it has been found that the water scouring extract disclosed in the present specification is excellent in antimicrobial effect and has no side effects, so that it can be effectively used for the purpose of removing bacteria.
