Abstract
Description
本明細書で使用する時、「イオン性液体」という用語は、約100℃以下、あるいは約60℃以下、又は更に代替的には約40℃以下の融解温度を有する塩を意味する。いくつかのイオン性液体は、識別可能な融点(DSC分析に基づく)を示さないが、約100℃以下の温度で「流動可能」であり、他のイオン性液体は約20〜約80℃の温度にて「流動可能」である。本明細書で使用する時、「流動可能」という用語は、イオン性液体が約100℃以下又は約20〜約80℃の温度にて、約10,000mPa・s未満の粘度を示すことを意味する。このため、イオン性液体の「流体状態」とは、溶融状態及び流動可能な状態を含む、これら全ての実施形態を包含することを意味する。
本発明は、その流体状態であり且つ実質的な水の不存在下のイオン性液体にて、バイオポリマーに溶解させる方法を包含する。イオン性液体の強力な溶媒和力により、有機溶媒又は水に不溶性又は限定された溶解度を有するバイオポリマーを、かなり穏やかな条件下にて溶解させることができる。溶解過程は、ほぼ室温(20℃)〜約100℃の温度にて、大気圧下で行うことができる。幾つかの実施形態では、前記溶解過程は約40〜約90℃の温度にて実施される。所望により、溶解速度を上げるため、つまり、処理時間を短くするために、高温(例えば、約130℃以下)を使用してもよい。
0.12グラムのPHA(約6モル%のヘキサン酸塩を含有するポリ(ヒドロキシ酪酸塩/ヒドロキシヘキサン酸塩)コポリマー)を、室温で液体であるテラセイル(Terrasail)(登録商標)(サケム社(Sachem, Inc.)より入手可能)2gを含むガラス瓶に添加する。バイアル瓶の温度は、115℃にて3分間保たれる。バイアル瓶は、溶解を促進するために攪拌する。粘稠な、視覚的に透明なポリヒドロキシアルカノエート溶液をイオン性液体中に得る。
ケラチン(0.48g)を、室温で液体である1−エチル−3−メチルイミダゾリウムアセテート(BASF社より入手可能)3gを含有するガラス瓶に添加する。バイアル瓶の温度は、油浴中で95℃にて10分間保たれる。バイアル瓶は、溶解を促進するために攪拌する。粘稠な、視覚的に透明なケラチン溶液をイオン性液体中に得る。ケラチン−イオン性液体溶液を150mLの水に注ぎ込む。次に前記混合物のpHを1N HClを使用してpH4.5に調整する。ケラチンが前記混合物から沈殿し、濁った溶液を得る。結果として得られる溶液を濾過し、沈殿したケラチンを回収する。
60gのバイオマス(遺伝子操作された生物試料を使用して発酵プロセスを介して調製された。PHA、6モル%の3−ヒドロキシヘキサン酸塩の繰り返し単位を有するポリ(3−ヒドロキシ酪酸塩−コ−3−ヒドロキシヘキサン酸塩)を含有する。)を、600mLの1−ブチル−メチルイミダゾリウムアセテート(BASF社から入手可能)を充填した密閉容器内に入れ、85℃にて3時間攪拌する。次に、抽出されたPHAを含有する混合物を、不溶性物質(即ち、非PHA細胞物質)から、ワイヤー−メッシュ・フィルターを使用して流出させる。
Claims (9)
- バイオポリマー及び付加的物質を含むバイオマスからバイオポリマーを分離する方法であって、
バイオマスと、流体状態であり且つ実質的に水の不存在下のイオン性液体とを混合して混合物を形成し、
該混合物から付加的物質を除去すること
を含んでなり、
前記バイオポリマーが、キチン、キトサン、エラスチン、コラーゲン、ポリヒドロキシアルカノエート又はケラチンである、方法。 - 前記イオン性液体が、ハロゲン化物、C1〜C6カルボキシレート、C1〜C6アルキルサルフェート、モノ−又はジ−・C1〜C10アルキルスルホサクシネート、モノ−又はジ−・C1〜C10エステルスルホサクシネート、及びこれらの混合物から成る群から選択されるアニオン性構成成分を含有する、請求項1又は2に記載の方法。
- 前記方法が、前記混合物からバイオポリマーを除去するに先だって、回収溶媒を前記混合物に添加することを更に含む、請求項1〜3のいずれか一項に記載の方法。
- 前記回収溶媒が、水、C1〜C6アルコール、C2〜C6エーテル、アセトン、及びこれらの混合物から成る群から選択される、請求項4に記載の方法。
- 回収溶媒と、イオン性液体との重量比が、100:1〜1:2である、請求項4又は5に記載の方法。
- 回収可能組成物からバイオポリマーを分離することを更に含む、請求項4,5又は6に記載の方法。
- 前記分離工程が、遠心分離、沈殿、結晶化、デカンテーション、濾過、及びこれらの組み合わせから成る群から選択される、請求項7に記載の方法。
- 請求項7に従って生成された、回収可能組成物。
