Abstract
Description
に対応する本発明によるペプチドは、有利に、BeclinモチーフとBcl−2タンパク質ファミリーの抗アポトーシスメンバーとの間のタンパク質相互作用を、その相互作用を活性化または阻害することによってモジュレートする化合物のスクリーニングに、用いることができる。
モチーフと、Bcl−2タンパク質ファミリーの抗アポトーシスメンバーとの間の相互作用の工程、次にテストする化合物の存在下または非存在下で相互作用を検出する工程、を含む、プログラム細胞死のモジュレーターを同定する方法に関する。
a)Beclinタンパク質のモチーフ
を蛍光プローブでラベルする工程;
b)前記モチーフへBcl−2タンパク質ファミリーの抗アポトーシスメンバーを加える工程;
c)テストする化合物の存在下でまたは非存在下で、a)およびb)に記述したパートナーをインキュベーションする工程;
d)蛍光偏光度を測定する工程;および
e)テストする化合物の存在下の測定値および非存在下の測定値を比較する工程。
に関する。本発明によるこの核酸配列を、Beclinモチーフのアミノ酸配列およびその変異体から出発して、遺伝子コードを用いて得ることができる。
− 配列番号2の核酸配列と厳格な条件下でハイブリダイズすることができる配列またはそれに相補的な配列であって、配列番号1によってコードされるBeclinモチーフと実質的に同一の特性を有するポリペプチドをコードする配列、または
− ヒトから単離した配列番号2の配列に相同の哺乳動物種の配列、
である。
3つのヒトcDNA(胎座、脳、細胞系統CEMC7)バンクを、酵母中の2−ハイブリッド技術(Fields et al.)により、Legrain et al., Nature Genetics, 1997, vol.16, 277-282 (US 6,187,535) によって記述された接合プロトコルを用いてスクリーニングした。
− LexA DNA結合ドメインへ融合された、Bcl−XLのC末端切断物(1〜209)(受入番号 Z23115);
− LexA DNA結合ドメインへ融合されたBcl−2のC末端切断物(1〜211)(受入番号 XM_008738)。
これらの餌を、Saccharomyces cerevisiae酵母の菌株L40Δgal4(MATa ade2, trp1-901, leu2-3, 112, lys2-801, his3Δ200, LYS2 (lexAop)4-HIS3, ura3-52 ::URA3 (lexAop)8-LacZ, GAL4 ::KanR)中で発現させ、30℃で、トリプトファンを欠く合成培地(DO−Trp)中で、全体として吸光度OD600nm0.1〜0.5を得るまで前培養する。その前培養の稀釈液(OD600nm =0.006)50mlを、30℃で終夜インキュベーションする。
「餌」/「獲物」比:2を用いて、接合を実行する。50単位のOD600nmに対応する量の、工程1)a)中で得られた酵母「餌」細胞を、工程1)b)中で得られた酵母「獲物」と混合する。遠心分離後、沈殿物をYPGlu培地に再懸濁し、YPGlu培養プレート上に広げ、30℃で4時間30分インキュベーションする。互いに相互作用することができる「餌」と「獲物」を含む接合した酵母の選択を、DO−Leu−Trp−His培地中で実行する:ロイシンおよびトリプトファンの欠乏により、2つの型のプラスミド(「餌」/「獲物」)を含む酵母のみの成長を許す淘汰圧を維持することが可能になる;培地からヒスチジンを欠くことにより、互いに相互作用することができる「餌」プラスミドおよび「獲物」プラスミドを含む接合した酵母を選択することが可能になる:この相補性により、ヒスチジンの生合成に関係する酵素をコードするレポーター遺伝子であるHIS3遺伝子を活性化することが可能になるからである。
項2)に記述した接合法により選択した酵母のコロニーの「獲物」断片を、そのコロニーの粗溶解物から出発するPCRにより、「獲物」ベクターの特異的なプライマー:
を用いて増幅する。次に、PCR生成物を配列決定し、得られた配列を、データベースと比較して同定する。
テストされる各「餌」断片について、2−ハイブリッドシステムにより、複数の「獲物」断片の同定が可能である。この同定を、選択された「獲物」の配列をBlastwunのようなソフトウェアプログラムを用いて比較することにより実行する。そのプログラムはワシントン大学のウェブサイト:

で利用可能である。
蛍光偏光を用いるKiの決定は、親アポトーシスBakペプチドとBcl−XL、Bcl−2またはBcl−WのようなBcl−2タンパク質ファミリーの抗アポトーシスメンバーとの間の相互作用に対する、Beclinペプチドの拮抗的効果を測定する工程より成る。
a)最終濃度1nM〜100μMの拮抗ペプチド;
b)最終濃度15nMの蛍光性ペプチドリガンド(Bak BH3 カルボキシフルオレセイン);
c)Bcl−2タンパク質ファミリーの抗アポトーシスメンバーを、Bcl−XLについては最終濃度100nM、ならびにBcl−2およびBcl−Wについては1μM。

Bcl−2/Bak/Beclin Ki=1.8μM
Bcl−W/Bak/Beclin Ki=7.4μM
蛍光偏光を用いるKiの決定は、Beclinタンパク質の完全な配列の位置116がロイシンからアラニンへ突然変異されたBeclinペプチド(L116A)の、親アポトーシスBAKペプチドとBcl−XL、Bcl−2またはBcl−WなどのBcl−2タンパク質ファミリー抗アポトーシスメンバーとの間の相互作用に対する、拮抗的効果を測定する工程より成る。
flagエピトープを所持するBcl−XLタンパク質をコードする発現ベクターおよび野生型または突然変異(L116A)Beclinタンパク質をコードする発現ベクターで、HeLa細胞を共トランスフェクションする(Effecteneキット、 Qiagen)。トランスフェクションの24時間後に、溶解緩衝液(Hepes 10mM pH7.5、KCl 150mM、MgCl2 5mM、EDTA 1mM、Triton 0.4%、抗プロテアーゼ、および抗ホスファターゼ)中に細胞を取り上げて、氷中でインキュベーションし、10000rpmで遠心する。
実施例1において得られた、蛍光性の形(フルオレセインに結合された)であり、Na2HPO4 20mM pH7.4、EDTA 1mM、NaCl 50mM、プルロン酸F−68 0.05%を含む緩衝液中に15nM濃度で溶解されているペプチドを、融合タンパク質GFT−Bcl−XLの増加する濃度(10-9〜10-5M)の存在下でインキュベーションし、次に蛍光偏光度をEn Vision装置(Packard Perkin-Elmer)を用いて測定する。この相互作用のKD 0.2μMを決定することができた。
テストする化合物を最終濃度10μg/mlで、384ウエルプレート(Corning, 平底)に分配する。1つのウエルには、対照として使用するために、テスト化合物を含まない等量の緩衝液/溶媒を入れる。実施例1で得られたフルオレセインでラベルされたペプチドを、1〜100nMの範囲の最終濃度を得るように各ウエルに加える。次に、融合タンパク質GST−Bcl−XL、またはGST−Bcl−2、またはさらにGST−Bcl−Wを、Na2HPO4 20mM pH7.4、EDTA 1mM、NaCl 50mMおよびプルロン酸F−68 0.05%を含む緩衝液中に、0.1〜1μMの最終濃度を得るように加える。次に、En Vision装置(Packard Perkin-Elmer)により、蛍光偏光を測定する。テスト化合物存在下で行なわれたテストで記録された蛍光偏光度の、テスト化合物無し(対照ウエル)で得られた偏光度と比較して、有意な減少によって、その化合物が阻害活性を有するという結論を下すことが可能になる。反対に、対照と比較して、テスト化合物存在下でのテストでの蛍光偏光度が有意に増加すれば、化合物が活性剤活性を有するという結論を下すことができる。

- ベクリンタンパク質のモチーフとBcl−2タンパク質ファミリーの抗アポトーシスメンバーとの間の相互作用のモジュレーターが、螢光偏光度を減少させる阻害剤であることを特徴とする、請求項1または2のいずれか1項記載の方法。
- ベクリンタンパク質のモチーフとBcl−2タンパク質ファミリーの抗アポトーシスメンバーとの間の相互作用のモジュレーターが、螢光偏光度を増加させる活性化剤であることを特徴とする、請求項1または2のいずれか1項記載の方法。
- 螢光プローブがフルオレセインであることを特徴とする、請求項1〜4のいずれか1項記載の方法。
- Bcl−2タンパク質ファミリーの抗アポトーシスメンバーが、タンパク質Bcl−2、Bcl−XLまたはBcl−Wであることを特徴とする、請求項1〜5のいずれか1項記載の方法。
- 抗アポトーシスタンパク質のBcl−2ファミリーと相互作用することを特徴とする、請求項7記載のアミノ酸配列。
- 請求項7記載のアミノ酸配列 配列番号1から遺伝子コードに従って導き出される核酸配列。
- 請求項9および請求項10のいずれか1項記載の核酸配列を含むことを特徴とする、組換えベクター。
- ベクリンタンパク質のモチーフの発現に必要な配列を含むプラスミド、コスミド、バクテリア人工染色体、またはバクテリオファージであることを特徴とする、請求項11記載の組換えベクター。
- ベクリンタンパク質のモチーフの発現に必要な配列が、転写および翻訳のプロモーター配列を含むことを特徴とする、請求項12記載の組換えベクター。
- 請求項11〜13のいずれか1項記載の組換えベクターによって形質転換されたことを特徴とする、ホスト細胞。
- バクテリア細胞または真核細胞であることを特徴とする、請求項14記載のホスト細胞。
- 請求項7または8のいずれか1項記載のアミノ酸配列を含むことを特徴とする、ペプチド。
- 請求項9または10のいずれか1項記載の核酸配列によってコードされることを特徴とする、請求項16記載のペプチド。
- 活性成分として、請求項16または17のいずれか1項記載のペプチドを、1以上の薬学的に許容され得る賦形剤と組み合わせて含む、医薬組成物。
- 癌患者に、有効量の請求項18記載の医薬組成物を投与することを含む、アポトーシス型および/またはオートファジー型のプログラム細胞死をもたらす方法。
- 活性成分として、請求項1〜6のいずれか1項記載の同定方法を用いて得られたモジュレーターを、1以上の薬学的に許容され得る賦形剤と組み合わせて含む、医薬組成物。
- 癌患者に、有効量の請求項20記載の医薬組成物を投与することを含む、アポトーシス型および/またはオートファジー型のプログラム細胞死をもたらす方法。
