Abstract
本发明涉及鉴定程序性细胞死亡的调节剂的方法,其包括Beclin蛋白质基序与BCL-2蛋白质家族的抗细胞凋亡成员发生相互作用及通过荧光偏振方法鉴定所述相互作用。可向癌症患者施用基于该方法鉴定的调节剂以诱导细胞凋亡型和/或自体吞噬型程序性细胞死亡。本发明也涉及与BCL-2蛋白质家族的抗细胞凋亡成员相互作用的Beclin蛋白质基序及其在癌症患者中诱导程序性细胞死亡的用途。
Description
本发明属于搜索和开发用于在癌症患者治疗中调节细胞凋亡型和/或自体吞噬型程序性细胞死亡的新制剂的领域。
本发明涉及鉴定程序性细胞死亡的调节剂的方法,其包括Beclin蛋白质基序与Bcl-2蛋白质家族抗细胞凋亡成员间发生相互作用及通过荧光偏振检测所述相互作用。向癌症患者施用通过上述方法鉴定的调节剂以在那些患者中引起细胞凋亡型和/或自体吞噬型程序性细胞死亡。
本发明尤其涉及能与Bcl-2蛋白质家族抗细胞凋亡成员相互作用的Beclin蛋白质基序并涉及其在癌症患者中引起程序性细胞死亡的用途。
在一个方面,程序性细胞死亡包括细胞凋亡,在另一个方面包括自体吞噬死亡。细胞凋亡是更熟知的现象。这种类型的细胞死亡包括形态学变化,如核凝集和DNA片段化,也包括生化现象,如胱天蛋白酶的活化(其然后降解细胞的关键结构成分从而引起细胞解装配和死亡)。细胞凋亡过程的调节是复杂的并包括多个胞内信号通路的激活或抑制。
自体吞噬死亡是第二位的不太熟知的程序性细胞死亡机制。在细胞水平上,自体吞噬可概括为三个阶段:起始自体吞噬泡(自噬体)的形成、自噬体成熟为降解性泡然后其与溶酶体融合。因此自体吞噬死亡涉及溶酶体降解过程,其特征在于自体吞噬泡的累积并且不依赖胱天蛋白酶型调节通路。
大量病理(如癌症)的开始存在生长、存活和程序性细胞死亡间细胞平衡的失调。
根据其通常的意义,“癌症”在本发明中通过两个主要特征定义:不受外界信号调节的细胞生长和增殖以及侵染组织的能力和一些情况下的通过远位点克隆化形成转移灶的能力。
这些特征是癌性细胞内在特性的结果,也就是其遗传和核型的不稳定性、不受控制的增殖和其转移能力,同时伴随获得新表型与那些癌性细胞中癌基因的激活和去阻遏。在本发明中,“癌症”因此理解为具有上述特征的细胞生长或增殖的任何阶段,尤其是出现原发肿瘤和/或转移肿瘤(继发性肿瘤)之后。
保持细胞存活或使其程序性死亡使得尤其是BCl-2蛋白质家族参与的主要信号通路的调节成为必需。BCL-2蛋白质家族分为三个主要类别。抗细胞凋亡蛋白质(如Bcl-2、Bcl-XL和Bcl-W)在其四个BH结构域中具有高度的同源性。促细胞凋亡蛋白质分成两类:一类是多结构域蛋白质(如BAX和BAK),另一类是促细胞凋亡蛋白质(如BID、NOXA、PUMA、BIK、BIM和BAD),其特征在于存在单一同源结构域-BH3基序(Cory和Adams,The Bcl-2 family:regulators of the cellular life-or-death switchNature reviews,第12卷,2002年9月)。
BH3基序为两亲性α螺旋区域,其在Bcl-2蛋白质家族中的序列同一性相对较低。此外,蛋白质中需要存在BH3基序以允许与Bcl-2蛋白质家族抗细胞凋亡成员相互作用。实际上,Bcl-2蛋白质家族抗细胞凋亡成员的活性受所述家族的促细胞凋亡基因产物调节,两种蛋白质组装成异二聚体。当处于那种状态的时候,Bcl-2蛋白质家族的抗细胞凋亡成员是无活性的并且因此不再具有其抗细胞凋亡活性。此外,BH3基序与Bcl-2蛋白质家族抗细胞凋亡成员的特异相互作用可由调节剂修饰从而以特定方式引起细胞凋亡型程序性细胞死亡。
而且需要注意到细胞凋亡信号通路也可由病毒感染策略调节。实际上,大量病毒蛋白质通过结构同源性、功能模拟或在促细胞凋亡和抗细胞凋亡信号转导水平上干扰细胞凋亡通路。因此,某些病毒编码抑制线粒体通路的Bcl-2的抗细胞凋亡类似物或封闭效应期的胱天蛋白酶抑制剂。受感染细胞的早熟细胞凋亡代表了限制病毒颗粒丰度的宿主防御机制;与之相反,宿主细胞延迟的细胞凋亡允许病毒复制和扩散。
自体吞噬参与细胞的存活机制并且也与程序性细胞死亡的进程相关。大量研究已经确定自体吞噬受肿瘤抑制基因(如Beclin、PTEN和TSC1)产物的控制。那些基因的失活和自体吞噬能力的降低为癌症患者肿瘤进程中的早期事件。Beclin在自体吞噬现象中,尤其在自体吞噬泡或自噬体的形成中发挥早期和核心的作用(Edinger等人,Defective autophagy leads tocancer,Cancer Cell,2003年12月)。
肿瘤对化疗剂的抗性是医学肿瘤学的核心问题。发现了获得抗性的出现,这使其在最初对化疗反应随后或多或少在短期对治疗产生抗性的肿瘤中自我显现。肿瘤细胞中存在的此种抗性通常与依赖胱天蛋白酶的通路或程序性细胞死亡的细胞凋亡通路(主要的当前抗癌治疗剂(细胞毒性剂)作用于此类通路)的抑制有关。为了更有效,因此在抗癌治疗剂方面必需提出治疗剂的备选和/或补充策略,其着眼于至少克服专一作用于细胞凋亡的已知治疗剂的缺陷而作用于程序性细胞死亡的细胞凋亡通路。因此本发明提出了通过二元调节剂作用于自体吞噬型和/或细胞凋亡型通路。
因而,如本发明所进行的,将不依赖胱天蛋白酶的通路或自体吞噬通路的调节作为特异作用于细胞凋亡通路的备选治疗剂。
一方面从Bcl-2蛋白质家族抗细胞凋亡成员在细胞凋亡过程中的作用的角度,另一方面从Beclin蛋白质在自体吞噬现象中的作用以及获得抗性在肿瘤发展中的重要性的角度,可以理解在抗癌治疗中同时作用于程序性细胞死亡的自体吞噬和细胞凋亡通路的价值。因而,必需能够鉴定能同时作用于Bcl-2蛋白质家族抗细胞凋亡成员和Beclin蛋白质的调节剂。为了辅助和加速筛选那些二元调节剂,发明者开发了一种策略,其包括:一方面确定Bcl-2蛋白质家族抗细胞凋亡成员与Beclin蛋白质间结构相互作用,另一方面搜索本来就能作用于Bcl-2蛋白质家族抗细胞凋亡成员和Beclin蛋白质的调节剂。因而,以那种方式选择的二元调节剂对获得对抗使细胞凋亡型和/或自体吞噬型程序性细胞死亡失调的病原体的候选药物是理想的。
用双杂交系统研究了Beclin蛋白质或其特异基序与Bcl-2蛋白质家族抗细胞凋亡成员间蛋白质相互作用的调节剂的选择和鉴定。这种双杂交系统最初由Fields等人描述和开发(US 5,283,173;US 5,468,614;US5,667,973)。
最开始双杂交系统由在酵母中测试两种重组蛋白质组成。第一种蛋白质(称作“饵”)为含有与蛋白质A上游结合的DNA结合结构域(或BD)的融合蛋白。第二种蛋白质也是融合蛋白,通常称作“猎物”,其含有与蛋白质B结合的激活结构域(或AD)。通常使用的结合结构域和激活结构域为Gal4或大肠杆菌(E.Coli)Lex A的结构域。蛋白质A和B分别为Bcl-2蛋白质家族抗细胞凋亡成员和Beclin蛋白质的特异基序。蛋白质A和B通过蛋白质相互作用的结合允许通过互补形成能结合报告基因上游存在的结合位点(或BS)并且确保该报告基因转录的功能性结构域(BD-AD)。
然而,这种常规的双杂交系统具有其局限性。例如,众所周知,此类筛选方法可造成假阳性和/或假阴性并且需要通过生化验证所获得的结果。双杂交系统获得的假阳性尤其频繁并且证明的是功能相互作用而非结构相互作用。
国际专利申请WO 99/42612或专利US 6,187,535中描述了允许假阳性和/或假阴性最小化的更有效技术并且使用含“饵”和“猎物”多肽的重组单倍体酵母。这个系统允许以比本领域所用的其它常规方法更精确、更高重复性和更灵敏的方式用单个的“饵”检测更大数量的“猎物”。
使用双杂交系统,发明者已经确定了Bcl-2蛋白质家族抗细胞凋亡成员和Beclin蛋白质间存在结构相互作用。那些配偶体间的这种蛋白质相互作用类似于Bcl-2蛋白质家族抗细胞凋亡和促细胞凋亡配偶体间的细胞凋亡现象调节中出现的相互作用,该相互作用参与程序性细胞死亡过程的平衡。
具体而言,已经在本发明中鉴定了Beclin蛋白质的最初基序。Beclin蛋白质的这个基序能从用于选择细胞凋亡和/或自体吞噬的特异调节剂的角度以高度特异的方式与Bcl-2蛋白质家族抗细胞凋亡成员相互作用。这种相互作用的特异性与Beclin蛋白质的序列、三维结构和/或原始基序的螺旋性相关。
具有26个氨基酸的Becliln蛋白质的最初基序实际上对应于Bcl-2、Bcl-XL和/或Bcl-W相互作用的精确结构域并具有允许形成同型或异型二聚体的典型结构标准。
这个基序的大小使它成为理想候选者用于开发允许高效筛选能调节Beclin肽与抗细胞凋亡蛋白质间相互作用的化合物的测试法。在文献中可以找到大量筛选蛋白质-蛋白质相互作用的测试法但是它们经常在敏感性和高通量可行性方面具有局限性。通常使用的方法必需使用与高通量筛选不十分相容的复杂工具(融合蛋白、重组蛋白质等等)。它们非常频繁地产生高水平背景噪音并且从定量角度来说可信度低:它们提供减少的阅读窗口不能够最佳筛选所测试化合物。
作为已经可利用方法的备选方法,基于荧光偏振的高效筛选测试法已经在本发明中使用(Owicki等人,Journal of Biomolecular Screening,5,2000,297-306)。这种技术允许例如测量荧光团标记的配体和受体间的相互作用。原理包括测量与游离配体发射的偏振相比结合其受体时配体发射的荧光偏振增加。游离配体的荧光偏振取决于其分子量,并且分子量越高荧光偏振越大。因此,当用高分子量、具有高水平固有荧光偏振的配体进行测试时很难可信地评价游离配体和结合配体的荧光偏振差异。另一方面,使用最小分子量的配体强化了差异并因而使方法的精确度提高。因此可以更好地评价化合物的真正活性并且开展高通量筛选。
对应于Beclin的基序GTMENLSRRLKVTGDLFDIMSGQTDV(SEQID NO.1)的根据本发明的肽可有利地用于筛选通过激活或抑制相互作用来调节Beclin基序与Bcl-2蛋白质家族抗细胞凋亡成员间的相互作用的化合物。
本发明涉及鉴定程序性细胞死亡的调节剂的方法,其包括Beclin蛋白质的GTMENLSRRLKVTGDLFDIMSGQTDV基序与Bcl-2蛋白质家族抗细胞凋亡成员间的相互作用的步骤和后来的检测是否存在测试化合物的相互作用的步骤。
鉴定程序性细胞死亡的调节剂的方法有利地包括下列步骤:
a)荧光标记Beclin蛋白质的GTMENLSRRLKVTGDLFDIMSGQTDV基序;
b)向所述基序中加入Bcl-2蛋白质家族的抗细胞凋亡成员;
c)在存在或不存在测试化合物条件下孵育a)和b)中所述的配偶体;
d)测量荧光偏振;和
e)比较含或不含测试化合物的测量值。
“调节剂”理解为任何能增强、阻止或至少限制特异活性(如蛋白质-蛋白质相互作用、酶促活性或结合细胞受体)的化合物。根据本发明,调节剂为配偶体Beclin与Bcl-2蛋白质家族抗细胞凋亡成员间蛋白质相互作用的抑制剂或真正的激活剂。
本发明也涉及鉴定Beclin蛋白质基序与Bcl-2蛋白质家族抗细胞凋亡成员间相互作用的抑制剂的方法,其与不存在调节剂条件下这种相互作用组成的对照相比能降低荧光偏振。
本发明还涉及鉴定Beclin蛋白质基序与Bcl-2蛋白质家族抗细胞凋亡成员间相互作用的激活剂的方法,其与不存在调节剂条件下这种相互作用组成的对照相比能增强荧光偏振。
荧光配体(即荧光Beclin肽)在与Bcl-2蛋白质家族抗细胞凋亡配偶体结合后具有低于相应游离配体的旋转常数,因而,结合的配体发射的荧光变成偏振的。因此,观察到与游离配体相比结合的配体发射的荧光偏振的增加。
在优选的实施方案中,用于根据本发明的筛选方法中的荧光探针为Bodipy、Oregon Green,或优选地为荧光素。
更特别地,在本发明的筛选和鉴定过程中作为相互作用配偶体使用的Bcl-2蛋白质家族抗细胞凋亡成员可为蛋白质Bcl-2、Bcl-XL或Bcl-W。
Bcl-2蛋白质家族抗细胞凋亡成员有利地为融合蛋白。“融合蛋白”理解为是指蛋白质Bcl-2、Bcl-XL或Bcl-W的结构域与蛋白质(如GST,谷胱甘肽S转移酶)的结构域的融合。
本发明涉及具有氨基酸序列GTMENLSRRLKVTGDLFDIMSGQTDV的Beclin基序及其功能性变体。
“氨基酸序列”理解为从天然环境中分离的肽序列,尤其是经分离的、化学合成的和/或纯化的以及(如果可能)通过基因工程修饰的序列。
“功能性变体”理解为Beclin基序的氨基酸序列,其包含保守的取代或保守的点突变并且具有与序列SEQ ID NO.1编码的基序基本相同的特性,或者也就是说与Bcl-2蛋白质家族抗细胞凋亡成员相互作用的能力。氨基酸序列SEQ ID NO.1的保守取代或突变为例如下列取代或突变:丙氨酸取代甘氨酸(G--A)、亮氨酸取代缬氨酸(V--L)、谷氨酸取代天冬氨酸(D--E)、谷胺酰胺取代天冬酰胺(N--Q)、异亮氨酸取代亮氨酸(L--I)、赖氨酸取代精氨酸(R--K)。
发明者观察到具有肽序列SEQ ID NO.1的Beclin基序与Bcl-2抗细胞凋亡家族成员(如Bcl-2、Bcl-XL或Bcl-W)相互作用。
本发明也涉及编码Beclin蛋白质最初基序的核酸序列5’ggcaccatggagaacctcagccgaagactgaaggtcactggggacctttttgacatcatgtcgggccagacagatgtg 3’(SEQ ID NO.2)。根据本发明的这个核酸序列通过从Beclin基序及其变体氨基酸序列开始的遗传密码的途径获得。
该核酸序列的“变体“尤其是:
-能在严谨条件下与核酸序列SEQ ID NO.2或与其互补的序列杂交以及编码具有与序列SEQ ID NO.1编码的Beclin基序基本相同特性的多肽的序列,或者
-与从人类中分离的序列SEQIDNO.2同源的哺乳动物物种的序列。
“严谨条件”理解为在大约65℃,例如在6×SSC溶液、0.5%SDS、5×Denhard’s溶液和100μg非特异载体DNA或任何其它相等离子强度溶液中并且于65℃在至多0.2×SSC和0.1%SDS或任何其它相等离子强度溶液中洗涤之后允许两个单链DNA序列特异杂交的条件。定义严谨条件的参数取决于50%配对链分离的温度(Tm)。对含有多于30个碱基的序列来说,Tm通过如下公式定义:Tm=81.5+0.41(%G+C)+16.6Log(阳离子浓度)-0.63(%甲酰胺)-(600/碱基数量)。对长度低于30个碱基的序列来说,Tm通过如下公式定义:Tm=4(G+C)+2(A+T)。因此严谨条件可由本领域技术人员根据序列大小、GC含量和任何其它参数调整,尤其根据Sambrook等人,2001所述的方法调整(Molecular Cloning:A laboratoryManual,第三版,Cold Spring Harbor,laboratory press,冷泉港,纽约)。
“与序列SEQ ID NO.2同源的哺乳动物物种的序列”理解为与序列SEQ ID NO.2结构相似并编码具有与非人哺乳动物物种、尤其是灵长类、大鼠和小鼠中的特性基本相同的多肽的序列。两种同源序列在功能性区域内的同一性百分比通常高于80%,优选地高于90%。
本发明也涉及含有根据本发明要求专利保护的核酸序列的重组载体。载体理解为允许向宿主细胞中引入核酸序列并且任选地允许在宿主细胞中表达核酸序列所编码的多肽的任何类型载体。
此种载体为例如质粒、粘粒、细菌人工染色体或噬菌体,其含有Beclin蛋白质基序表达所必需的序列。
根据本发明的重组载体优选地含有在宿主细胞中表达要求权利保护的Beclin蛋白质基序所必需的序列。这些序列尤其是在宿主细胞中转录和翻译的启动子序列,还有终止子序列。重组载体也可含有编码分泌信号的序列,其允许将翻译的蛋白质释放到胞外环境中。
本发明也涉及用根据本发明的重组载体转化的宿主细胞。在特别的实施方案中,那些宿主细胞为细菌细胞(如大肠杆菌和链球菌)或真核细胞(如酵母细胞、丝状真菌细胞、昆虫细胞并且优选地为哺乳动物细胞)。
用含有本发明核酸序列的重组载体转化适当宿主细胞允许表达要求专利保护的Beclin蛋白质基序。之后可以用本领域技术人员已知并在现有技术中大量描述的各种方法纯化在那些宿主细胞中表达的蛋白质。需要提到的是,例如通过用硫酸铵沉淀纯化、通过大小排阻层析纯化和优选地通过亲和层析纯化。
具有氨基酸序列SEQ ID NO.1的肽也可由Néosystem化学定制合成。通过借助于“Applied Biosystems 430A”肽合成仪用Boc/苯甲基策略在固体支持物上合成进行SEQ ID NO.1及其功能性变体的化学合成。合成基于在树脂上组装所需序列以及随后的N端和C端官能团的去保护。以Boc/苯甲基策略为例,必需在肽合成过程中引入氨基酸Boc-L-Lys(Fmoc)-OH。完整序列组装完毕之后,将氨基官能团去保护并在存在强酸条件下从树脂上切下肽。
本发明也涉及药物组合物,其包含与一种或更多种可药用赋形剂组合的、对应于根据本发明的Beclin基序SEQ ID NO.1的肽作为有效成分。
在本发明中,药物组合物的“赋形剂”理解为任何保证有效成分转运到待治疗患者的内部血管中的药剂。“有效成分”理解为赋予药物组合物的药效特性或治疗特性的任何物质。
通过实例且不意味着任何限制的方式提到的非毒性、可药用赋形剂有稀释剂、溶剂、防腐剂、润湿剂、乳化剂、分散剂、黏合剂、膨胀剂、崩解剂、延缓剂、润滑剂、吸光剂、悬浮剂、着色剂或调味剂。
本发明不仅涉及如此考虑并如上定义的药物组合物,还涉及该组合物在引起根据本发明的细胞凋亡型和/或自体吞噬型程序性细胞死亡的方法中的用途。
本发明也涉及通过给患者(尤其是癌症患者)施用有效量的含Beclin肽(具有序列SEQ ID NO.1)的药物组合物引起细胞凋亡型程序性细胞死亡(即程序性细胞死亡的依赖胱天蛋白酶的通路)和/或自体吞噬型程序性细胞死亡(即程序性细胞死亡的不依赖胱天蛋白酶)的方法。
本发明也涉及药物组合物,其包含与一种或更多种可药用赋形剂组合的、根据本发明的鉴定调节剂的方法鉴定的至少一种调节剂、激活剂或抑制剂作为该组合物的有效成分。
本发明也涉及通过向癌症患者施用有效量上述组合物引起细胞凋亡型(依赖胱天蛋白酶)程序性细胞死亡和/或自体吞噬型(不依赖胱天蛋白酶)程序性细胞死亡的方法。
上述药物组合物适用于通过作用于细胞凋亡型和/或自体吞噬型程序性细胞死亡治疗癌症。
根据本发明的组合物为适于口腔、肠胃外、鼻、经皮肤、直肠、经舌、眼或呼吸施用的形式,尤其为片剂、糖衣丸、舌下片剂、药囊、paquets、胶囊剂、glossettes、锭剂、栓剂、乳膏剂、软膏剂、皮肤用凝胶和可饮用或可注射的安瓿剂。
本发明由下列图和实施例不限制性地描述:
-图1.与Bcl-2蛋白质家族抗细胞凋亡成员相互作用的Beclin基序的氨基酸序列SEQ ID NO.1。
-图2.编码与Bcl-2蛋白质家族抗细胞凋亡成员相互作用的Beclin基序的核酸序列SEQ ID NO.2。
-图3.用荧光偏振测定突变体和非突变体Beclin竞争肽与促细胞凋亡的Bak肽和抗细胞凋亡成员Bcl-2、Bcl-XL和Bcl-W间相互作用相关的Ki。
-图4.Bcl-XL蛋白质和Beclin蛋白质的免疫共沉淀结果。
-图5.测定Bcl-XL和Beclin肽相互作用的KD。
-图6.测定参照Bcl-XL拮抗剂的IC50。
实施例1:通过双杂交系统鉴定图1所述的肽
用Legrain等人(Nature Genetics,1997,第16卷,277-282)(US6,187,535)所述的联合方案在酵母中通过双杂交技术(Fields等人)筛选了三种人cDNA库(胎盘、脑、细胞系CEMC7)。
1)制备“饵”和“猎物”
a)所用的“饵”为:
-与LexA DNA结合结构域融合的Bcl-XL(登录号Z23115)的C端截短物(1-209);
-与LexA DNA结合结构域融合的Bcl-2(登录号XM_008738)的C端截短物(1-211)。
在酿酒酵母(Saccharomyces cerevisiae)菌株L40Δgal4(MATa ade2,trp1-901,leu2-3,112,lys2-801,his3Δ200,LYS2(lexAop)4-HIS3,ura3-52::URA3(lexAop)8-LacZ,GAL4::KanR)中表达这些饵,并于30℃在缺少色氨酸(DO-Trp)的合成培养基中预培养到光密度OD600nm为0.1-0.5(包括)。将50毫升预培养物的稀释液(OD600nm=0.006)在30℃孵育过夜。
b)通过转化获得酿酒酵母菌株YHGX13(MTAα Gal4Δ Gal80Δade2-101::KanR,his3,leu2-3-112,trp1-901,ura3-52URA3::UASGAL1-LacZ,Met),之后在缺少亮氨酸(DO-Leu)的培养基上选择,其中所述的菌株含有与Gal4转录激活结构域融合表达的cDNA库的质粒。将这些酵母分成等份并存于-80℃。
2)结合
用2的“饵”/“猎物”比例进行结合。
将一定数量的在步骤1)a)获得的酵母“饵”细胞(对应于OD600nm的50单位)与步骤1)b)中获得的酵母“猎物”混合。离心之后将沉淀重悬于YPGlu培养基中,涂布到YPGlu培养板上并在30℃孵育4小时30分钟。在DO-Leu-Trp-His培养基中选择含能彼此相互作用的“饵”和“猎物”的结合酵母:亮氨酸和色氨酸的缺少使得维持选择压力只允许含两种类型质粒(“饵”/“猎物”)的酵母生长成为可能;培养基中缺少组氨酸使得选择含能彼此相互作用的“饵”质粒和“猎物”质粒的结合酵母成为可能;所述的互补使得激活作为报告基因的HIS3基因(编码参与组氨酸生物合成的酶)成为可能。
3)鉴定阳性克隆
用“猎物”载体的特异引物从菌落的粗裂解液开始通过PCR扩增根据段落2)所述的结合方法选择的酵母菌落的“猎物”片段,其中所述的引物为:
ABS1 5’-GCTTTGGAATCACTACAGG-3’(SEQ ID NO.3);
ABS2 5’-CACGATGCACGTTGAAGTG-3’(SEQ ID NO.4)。
然后进行PCR产物测序并通过与数据库比较鉴定获得的序列。
4)鉴定图1所述的肽
对每一个所测试的“饵”片段来说,双杂交系统允许鉴定大量“猎物”片段。这种鉴定通过用软件程序(如Blastwun,可从华盛顿大学网站获得,http://bioweb.pasteur.fr/seqanal/interfaces/blastwu.html)比较所选“猎物”的序列进行。
实施例2:验证实施例1所述的肽与Bcl-2、Bcl-XL和/或Bcl-W的相互作用
1)用荧光偏振测定Ki(图3)
用荧光偏振测定Ki包括测量Beclin肽对促细胞凋亡Bak肽和Bcl-2蛋白质家族抗细胞凋亡成员(如Bcl-XL、Bcl-2或Bcl-W)间的相互作用的的竞争作用。
以所述顺序混合下列试剂:
a)终浓度1nM-100μM的竞争肽;
b)终浓度15nM的荧光肽配体(Bak BH3羧基荧光素);
c)终浓度100nM(Bcl-XL)和1μM(Bcl-2和Bcl-W)的Bcl-2蛋白质家族抗细胞凋亡成员。
这些试剂溶解于相互作用缓冲液中(Na2HPO4 20mM pH 7.4,EDTA 1mM,NaCl 50mM和pluronic acid F-68 0.05%)。
然后在室温将混合物孵育30分钟并在Fusion仪器上(Packard)(在485nm激发并在530nm读数)测定荧光偏振。以mP(荧光偏振的单位)表示数值。
这些荧光偏振分析证明Beclin肽对Bak和Bcl-XL、Bcl-2或Bcl-W间肽相互作用的竞争作用。这些荧光偏振测试过程中获得的Ki值如下:
Bcl-XL/Bak/Beclin Ki=1.15Mm
Bcl-2/Bak/Beclin Ki=1.8μM
Bcl-W/Bak/Beclin Ki=7.4μM
这些分析证明了Beclin肽与Bcl-2蛋白质家族抗细胞凋亡成员的高亲和力。
2)用荧光偏振测定突变体Beclin肽(L116A)的Ki
用荧光偏振测定Ki,包括测量在Beclin蛋白质完整序列的116位点已经从亮氨酸突变成丙氨酸的Beclin肽(L116A)对促细胞凋亡BAK肽和Bcl-2蛋白质家族抗细胞凋亡成员(如Bcl-XL、Bcl-2或Bcl-W)间的相互作用的竞争作用。
突变体肽(L116A)序列如下:
GTMENLSRRAKVTGDLFDIMSGQTDV(SEQ ID NO.5)
用荧光偏振测定Ki的方案与上述方案相同。
比较荧光偏振分析的结果表明,与根据本发明的Beclin肽相比突变体Beclin蛋白质(L116A)在促细胞凋亡Bak肽和Bcl-2蛋白质家族抗细胞凋亡成员(如Bcl-XL、Bcl-2或Bcl-W)间的肽相互作用中丧失了的竞争作用。
3)免疫共沉淀(图4)
用编码携带flag表位的Bcl-XL蛋白质的表达载体和编码野生型或突变体(L116A)Beclin蛋白质的表达载体共转染Hela细胞(Effectene试剂盒,Qiagen)。转染之后24小时将细胞放于裂解缓冲液中(Hepes 10mM pH 7.5,KCl 150mM,MgCl2 5mM,EDTA 1mM,Triton 0.4%,抗蛋白酶和抗磷酸酶),于冰上孵育并以1000转/分离心。
然后将上清液(细胞裂解物)在存在与抗Flag抗体缀合的琼脂糖珠(Flag M2琼脂糖,Sigma)的条件下孵育2小时。
然后离心琼脂糖珠并在裂解缓冲液中洗涤,然后收集于Laemmli缓冲液中并通过用抗Beclin抗体的蛋白质印记分析。
通过蛋白质-蛋白质相互作用的抗细胞凋亡蛋白质Bcl-XL与全部Beclin蛋白质的免疫共沉淀验证了Bcl-XL蛋白质和Beclin蛋白质间确实存在结构相互作用。
实施例3:能抑制Bcl-2和/或Bcl-XL与实施例1获得的肽之间相互作用的化合物的筛选测试
1)测定实施例1获得的肽与Bcl-XL的KD(图5)
将实施例获得的肽(与荧光素偶联的荧光形式,以15 nM浓度溶解于含Na2HPO4 20mM pH 7.4、EDTA 1mM、NaCl 50mM、pluronic acid F-680.05%的缓冲液中)在存在增加浓度的融合蛋白GFP-Bcl-XL(10-9-10-5M)的条件下孵育,然后用En Vision仪器(Packard Perkin-Elmer)测量荧光偏振。测定此相互作用的KD为0.2μM。
2)鉴定Bcl-XL-肽相互作用的抑制剂
以10微克/毫升的终浓度将所测试化合物分散于384孔板上(CorningFlat Bottom)。一个孔用不含测试化合物的等量缓冲液/溶剂填充作为对照。将实施例1获得的荧光素标记肽加入到孔中以获得1-100nM的终浓度。然后加入融合蛋白GST-Bcl-XL、GST-Bcl-2或者GST-Bcl-W以使得在含Na2HPO4 20mM pH 7.4、EDTA 1mM、NaCl 50mM和pluronic acid F-680.05%的缓冲液中终浓度为0.1-1μM。然后通过En Vision仪器(PackardPerkin-Elmer)测量荧光偏振。与不合测试化合物(对照孔)获得的荧光偏振相比,用测试化合物进行的测试中记录到荧光偏振显著降低,得出化合物具有抑制活性的结论。相反地,与对照相比,用测试化合物的测试中荧光偏振显著增加,得出化合物具有激活剂活性的结论。
图6显示了用参照化合物(Bcl-XL的拮抗物,Ref 1,A.D.Hamilton等人,J.Am.Chem.Soc.,2002,124,11838-11839中的化合物4)获得的结果。
测定此化合物的IC50为3.4μM。
Claims (22)
1.鉴定程序性细胞死亡的调节剂的方法,其特征在于包括下列步骤:
a)Beclin蛋白质的GTMENLSRRLKVTGDLFDIMSGQTDV基序与Bcl-2蛋白质家族抗细胞凋亡成员间发生相互作用;
b)在存在或不存在测试化合物的条件下检测相互作用。
2.根据权利要求1所述的鉴定程序性细胞死亡的调节剂的方法,其特征在于包括下列步骤:
a)荧光标记Beclin蛋白质的GTMENLSRRLKVTGDLFDIMSGQTDV基序;
b)向所述基序中加入Bcl-2蛋白质家族抗细胞凋亡成员;
c)在存在或不存在测试化合物的条件下孵育该体系;
d)测量荧光偏振;和
e)比较含或不含测试化合物的测量值。
3.根据权利要求1或2中任一项所述的方法,其特征在于Beclin蛋白质基序和Bcl-2蛋白质家族抗细胞凋亡成员间相互作用的调节剂为降低荧光偏振的抑制剂。
4.根据权利要求1或2中任一项所述的方法,其特征在于Beclin蛋白质基序和Bcl-2蛋白质家族抗细胞凋亡成员间相互作用的调节剂为增加荧光偏振的激活剂。
5.根据权利要求1-4中任一项所述的方法,其特征在于荧光探针为荧光素。
6.根据权利要求1-5中任一项所述的方法,其特征在于Bcl-2蛋白质家族抗细胞凋亡成员为蛋白质Bcl-2、Bcl-XL或Bcl-W。
7.Beclin蛋白质的GTMENLSRRLKVTGDLFDIMSGQTDV基序的氨基酸序列SEQ IN NO.1及其功能性变体。
8.根据权利要求7所述的氨基酸序列,其特征在于所述氨基酸序列与Bcl-2蛋白质家族抗细胞凋亡成员相互作用。
9.编码根据权利要求7或8所述Beclin蛋白质基序的核酸序列SEQID NO.2:5’ggcaccatggagaacctcagccgaagactgaaggtcactggggacctttttgacatcatgtcgggccagacagatgtg 3’。
10.从根据权利要求7所述氨基酸序列SEQ ID NO.1根据遗传密码推导出的核酸序列。
11.重组载体,其特征在于含有根据权利要求9或10中任一项所述的核酸序列。
12.根据权利要求11所述的重组载体,其特征在于载体为含有表达Beclin蛋白质基序所必需序列的质粒、粘粒、细菌人工染色体或噬菌体。
13.根据权利要求12所述的重组载体,其特征在于表达Beclin蛋白质基序所必需的序列含有转录和翻译的启动子序列。
14.宿主细胞,其特征在于用根据权利要求11-13中任一项所述的重组载体转化。
15.根据权利要求14所述的宿主细胞,其特征在于所述宿主细胞为细菌或真核细胞。
16.肽,其特征在于含有根据权利要求7或8中任一项所述的氨基酸序列。
17.根据权利要求16所述的肽,其特征在于由根据权利要求9或10中任一项所述的核酸序列编码。
18.药物组合物,其包含与一种或更多种可药用赋形剂组合的、根据权利要求16或17中任一项所述的肽作为有效成分。
19.引起细胞凋亡型和/或自体吞噬型程序性细胞死亡的方法,其包括向癌症患者施用有效量的根据权利要求18所述的药物组合物。
20.药物组合物,其包含与一种或更多种可药用赋形剂组合的、用根据权利要求1-6中任一项所述鉴定方法获得的调节剂作为有效成分。
21.引起细胞凋亡型和/或自体吞噬型程序性细胞死亡的方法,其包括向癌症患者施用有效量的根据权利要求20所述的药物组合物。
22.根据权利要求18或20中任一项所述的药物组合物,其用于治疗癌症。
