Abstract
Description
本発明は、あらかじめ定められた特徴に基づいて微粒子を選別するための方法および装置など、微粒子を処理するための方法および装置に関する。
本発明は、その内容が参照により本明細書に組み入れられる、2004年12月3日に提出された米国特許仮出願第60/633,396号「Unitary Cartridge for Particle Sorting」の優先権を主張する。
微粒子を処理するための従来のシステムは、個別に製造され現場で組み立てられる、個別で非一体式の複数のコンポーネントに依存している。このような従来のシステムは扱いにくく、かつ、潜在的な汚染の問題が生じる可能性もある。
本発明は、マイクロ流体式微粒子処理技術に使用するためすべての流体接触表面が封入された、微粒子選別などの微粒子処理を行うための単一の使い捨てカートリッジを提供する。流体接触表面の封入により、オペレータの隔離および/または製品の隔離が確保、向上、または促進される。カートリッジは微粒子処理のための任意の好適な技法を用いていてもよい。
本発明は、微粒子選別を含む微粒子処理をサンプルに対して行うためのユニット式カートリッジを提供する。以下に、例示的態様に関して本発明を説明する。当業者は、本発明が多数の異なる適用および態様において実施できること、およびその応用において本明細書に記載の特定の態様に具体的に限定されるわけではないことを理解すると思われる。

- サンプルを処理するための微粒子処理コンポーネントと、
ユニット式微粒子処理カートリッジ内に封入された複数の流体接触表面であって、該ユニット式微粒子処理カートリッジのすべての流体接触表面が封入されかつ外部環境から密封されている複数の流体接触表面とがその上に形成されているユニット式微粒子処理カートリッジを含む、サンプルに処理を行うためのユニット式微粒子処理カートリッジ。 - 複数の流体接触表面のうち少なくとも1つが、カートリッジ内で処理されるサンプルを投入するための密封可能なサンプル投入ポートを含む、請求項1記載のユニット式微粒子処理カートリッジ。

- 微粒子処理コンポーネントが、サンプル中のあらかじめ定められた特性を有さない微粒子から、サンプル中のあらかじめ定められた特性を有する微粒子を選別するための微粒子選別コンポーネントを含む、請求項1記載のユニット式微粒子処理カートリッジ。
- 微粒子選別コンポーネントと、
選別対象の微粒子を該微粒子選別コンポーネントに提供するための、該微粒子選別コンポーネントより上流の微粒子供給源と、
該微粒子を懸濁するためのシース流体を提供するためのシース流体リザーバと、
選別された微粒子を回収するための、該微粒子選別コンポーネントより下流の保持チャンバとがその上に形成されているユニット式カートリッジを含む、微粒子選別システム。 - カートリッジが、すべての流体接触表面を環境への曝露から封入する、請求項12記載の微粒子選別システム。
- システムへの流体の流動を促進するため流体経路に圧力を加えるための少なくとも1つの空気ポートをさらに含む、請求項12記載の微粒子選別システム。
- シース流体をシース流体リザーバに提供するためカートリッジに形成された第一の装填ポートと、

- フィルタが、保持チャンバからシース流体を除去するため、保持チャンバより下流に配置されている、請求項16記載の微粒子選別システム。

該カートリッジの該チャンバおよび該流体経路を該マイクロチャネルと流体連通状態にするため該微粒子処理チップを該カートリッジに取り付けるためのホルダと、
処理済サンプルを該マイクロチャネルから受け取りかつ集合させるための複数の集合チューブとを含む微粒子処理システム。 - 微粒子処理チップの各マイクロチャネルが専用のアウトレットを有する、請求項26記載の微粒子処理システム。
- 集合チューブがホルダ内の処理済サンプルを集合させる、請求項26記載の微粒子処理システム。
- 集合チューブがカートリッジ内の処理済サンプルを集合させる、請求項26記載の微粒子処理システム。
- サンプルを処理するための微粒子処理コンポーネントと、
少なくとも1つの流体チャンバおよび少なくとも1つの流体経路を含み、ユニット式微粒子処理カートリッジ内に封入された複数の流体接触表面であって、該ユニット式微粒子処理カートリッジのすべての流体接触表面が封入されかつ外部環境から密封されている複数の流体接触表面と、
該カートリッジのチャンバ内に貯蔵された該サンプルに対して処理を行うための化学試薬とがその上に形成されているユニット式微粒子処理カートリッジを含む、サンプルに処理を行うためのユニット式微粒子処理カートリッジ。 - 化学試薬が、微粒子処理コンポーネントを用いて行われる細胞または微粒子の処理プロトコルに特有である、請求項30記載のカートリッジ。
- 化学試薬が、微粒子処理コンポーネントを用いて行われる細胞または微粒子の処理プロトコルに依存しない、請求項30記載のカートリッジ。
- 化学試薬が、サブセットを濃縮または除去するためのビーズを含む、請求項30記載のカートリッジ。
- サンプルを処理するための微粒子処理コンポーネントと、
少なくとも1つの流体チャンバおよび少なくとも1つの流体経路を含み、ユニット式微粒子処理カートリッジ内に封入された複数の流体接触表面であって、該ユニット式微粒子処理カートリッジのすべての流体接触表面が封入されかつ外部環境から密封されている複数の流体接触表面と、
該カートリッジの第一のチャンバに貯蔵された、該サンプルに対してビーズに基づく除去または検出を行うために用いられるビーズとがその上に形成されているユニット式微粒子処理カートリッジを含む、サンプルに処理を行うためのユニット式微粒子処理カートリッジ。 - ビーズが、CD4発現細胞に結合する抗CD4抗体でコーティングされた磁性ビーズである、請求項34記載のカートリッジ。
- ビーズが、カートリッジの第二のチャンバから第一のチャンバへと供給された細胞懸濁液と該第一のチャンバ内で混合する、請求項34記載のカートリッジ。
- ビーズとの反応後に細胞懸濁液を受け取るための第三のチャンバをさらに含む、請求項36記載のカートリッジ。
- 微粒子処理コンポーネントが、あらかじめ定められた特性に基づいてサンプル中の微粒子の選別を行うためのマイクロ流体式微粒子選別チップである、請求項34記載のカートリッジ。

バイオセーフティフード内で、該密封されたユニット式微粒子処理カートリッジから処理済サンプルを取り出す段階。 - 処理が、あらかじめ定められた特性に基づいて微粒子を選別する段階を含む、請求項39記載の方法。
- 密封されたユニット式微粒子処理カートリッジから廃棄物を除去する段階をさらに含む、請求項39記載の方法。
- サンプルを装填する前に、密封されたユニット式微粒子処理カートリッジに試薬が装填される、請求項39記載の方法。
