Abstract
Description
本発明は日本脳炎ウイルスおよび西ナイルウイルスに対するワクチンに関する。
フラビウイルス(Flaviviridae)科のフラビウイルス(Flavivirus)属の中には、およそ70種のウイルス、大部分はアルボウイルスがあり、黄熱病(YF)ウイルス、デング(DEN)ウイルス、日本脳炎(JE)ウイルス、およびダニ媒介性脳炎(TBE)ウイルスなどの、その多くが主要なヒト病原体である(rev. in Burke and Monath, Fields Virology, 第4版: 1043-1126, 2001)。例えば、日本脳炎はアジアにおけるウイルス脳炎の主な原因であり、そこでは毎年30,000〜50,000件の新たな症例が報告されている。別の例として、西ナイルウイルスは、最初の症例が1999年にニューヨーク地域で診断されて以来、北米中に急速に広がり続けている。このウイルスが南米に移動するばかりでなく、発展途上国での蔓延の危険性も極めて高い。これらのウイルスによる感染を阻止するのに効果的な方法が必要とされており、その結果、ワクチン接種が最も費用効果的な手段になっている。
本発明は、フラビウイルスを弱毒化させる突然変異(例えば、フラビウイルスの内臓向性/ウイルス血症を減少させる突然変異)、細胞培養での増殖(例えば、無血清培養での製造)の間のフラビウイルスの遺伝的安定性を増大させる突然変異、および/またはワクチンウイルスの収量を増大させる突然変異などの、一つまたは複数の膜(M)タンパク質突然変異(例えば、置換、欠失、または挿入)を含む組換えフラビウイルスを提供する。本発明のフラビウイルスは、第一のフラビウイルス(例えば、YF 17Dなどの、黄熱病ウイルス)のカプシドおよび非構造タンパク質ならびに第二のフラビウイルス(例えば、日本脳炎ウイルス、西ナイルウイルス、デングウイルス(デング-1、デング-2、デング-3、またはデング-4ウイルス)、セントルイス脳炎ウイルス、マリーバレー脳炎ウイルス、ダニ媒介性脳炎ウイルス、ならびにYF、JE、DEN、およびTBE血清型群由来のヒト/動物病原体であるその他任意のフラビウイルス)の膜および/または外被タンパク質を含むフラビウイルスなどの、キメラフラビウイルスでありうる。
本発明はフラビウイルス(例えば、日本脳炎(JE)または西ナイル(WN)ウイルス)感染の予防および処置で用いるためのワクチンおよび方法を提供する。本発明の方法は全体として、一つまたは複数の構造タンパク質(例えば、膜および/または外被タンパク質)が第二のフラビウイルス(例えば、日本脳炎(JE)および/または西ナイル(WN)ウイルス; 同様に以下参照)のものと交換された第一のフラビウイルス(例えば、黄熱病ウイルス)からなる弱毒化生キメラフラビウイルスを用いた被験者のワクチン接種に関係する。本発明のキメラの膜タンパク質は、以下にさらに記述されるように、一つまたは複数の突然変異を含む。同様に以下に記述されるとおり、その他のフラビウイルスの膜および/または外被タンパク質などの構造タンパク質を、本発明のキメラウイルスにおける日本脳炎ウイルスまたは西ナイルウイルスのものに代えて使用することができる。さらに、本発明の膜タンパク質突然変異を同様に、構造タンパク質の任意の交換を含んでいない、および本明細書において記述されるものなどの、一つまたは複数のさらなる突然変異を任意で有する、無傷の非キメラフラビウイルス(例えば、本明細書において掲載されるもののいずれか)で使用することができる。

実施例1: ChimeriVax(商標)-WN
実験結果
背景と概要
キメラ黄熱病-西ナイル(YF-WN)ウイルスChimeriVax(商標)-WNは、WNウイルス(NY99)の前膜(prM)および外被(E)遺伝子のYF17D骨格への挿入によって産生された。このウイルスは無血清条件下のVero細胞(継代数5回、P5の時点)で産生され、前臨床モデルで安全性、免疫原性、および有効性について評価され、ヒトでの第I相試験において試験されている。ワクチンウイルス(P5)のさらなる弱毒化はEタンパク質中の三つのSA14-14-2特異的な突然変異(残基番号107、316、および440)によって判定される。ワクチンウイルスは、IC経路によりワクチン接種されたマウスおよびサルで試験された場合、YF-VAX (登録商標)よりも神経毒性が低く、単回のワクチン接種によってマウス、ハムスター、およびサルを防御した(Arroyo et al., J. Virol. 78:12497-12507, 2004; Tesh et al., Emer. Infect. Dis. 8:1392-1397, 2002)。ワクチンウイルスには、Mタンパク質中66位のアミノ酸残基(M66)が一つ異なるウイルスの混合群(小型Sおよび大型Lのプラーク表現型を示す)が含まれた。この突然変異は8日齢の授乳マウスに対するウイルスの神経毒性に影響を与えなかった(Arroyo et al., J. Virol. 78:12497-12507, 2004)。本発明において、本発明者らはM66突然変異が宿主内でのウイルス血症を低減し、したがって現行のワクチン(ChimeriVax(商標)-WN02、P5、親および突然変異体ウイルスの混合群)または大型のプラーク変異体(非突然変異体)ウイルスの安全性を向上させるために使用できるという発見について記述する。
は膜領域以外にあると予測され、その一方で残りの35アミノ酸(40〜75)
はウイルス膜領域内にあると予測される。さらに、最初の40アミノ酸残基内には9〜10個の荷電アミノ酸(3〜4個の酸性アミノ酸EまたはD)および6個の塩基性アミノ酸(RまたはK)が存在するのに対し、本明細書で記述される5種全てのフラビウイルス(WNV、SLE、MVE、JE、およびKunjin)の残基番号60には荷電アミノ酸(塩基性)が一つしか存在しない。したがって、M60残基がその隣接するアミノ酸内での相互作用によりウイルスの生物学において決定的な役割を果たすということなのかもしれない。
ChimeriVax(商標)-WN02ワクチン材料(P5)を無血清Vero細胞中で増殖させた:「小型」(S)と同定された10プラークおよび「大型」(L)と同定された10プラークをピックした。次いで、各分離株を無血清Vero細胞にて継代し、1プラークを各分離株からピックした。この手順を計3ラウンドのプラーク精製の間に、もう一度繰り返した。プラーク精製された分離株(P8)を、無血清Vero細胞を含有するT25 cm2フラスコの中で増幅させ(および無血清(SF)培地中で増殖させ)、次いで回収し、-80℃で保存した。分離株を配列決定して、誤った突然変異を含んでいないPMS候補を見出した。2つの分離株は発現された(サイレントではない)突然変異を含んでいないと同定された: 1つの分離株は小型のプラーク(M66プロリン)であると確認された(表1)、およびその他はwt配列(M66ロイシン)を含んでいた(表2)。次いで、これらの2つの分離株を大きいフラスコの中で増殖させ、等分し、LPおよびSP PMS (P10)ウイルスとしてQCインベントリに付した。
Sプラーク表現型が大規模製造過程の間に安定であるかを判定するため、小型プラークのPMSウイルスをVero細胞に感染させ、無血清条件下で増殖させることにより生物反応器の中で2回継代して、P12ウイルスを産生させた。P12ウイルスを回収し、6ウェルプレート中でプラーク形成させた。プラークの大部分は小サイズのものであった。利用可能な最大のプラークのうち20個をピックし、O-Veroにて増幅させ(継代1回)、prME領域をTitan One-Tube RT-PCRキット(Roche)によって転写/増幅させた。M領域を含むcDNA断片を配列決定し、WN特異的なモノクローナル抗体を用いた免疫染色によって分離株の形態を確認した。20プラークのうちの13個にはM66 (SP形態に関与する遺伝子マーカー)だけが含まれ、5個の分離株にはM66に加えてその他の突然変異が含まれていた。分離株#4にはM63 (LP表現型)がふくまれ、分離株#16にはwtとM66の混合群が含まれた。これらのデータから、一部のプラークは大きなサイズのものであるように思われたという事実にもかかわらず、それらがM66突然変異を含んでおり、増幅によってハッキリとSサイズのものであることが実証された。20個のうち1個のプラーク(#4)だけが、明らかにM63でのLからPの突然変異に起因して、Lサイズのものであるように思われた。プラーク#16はM66位でwtのLと突然変異体のPアミノ酸の両方を含む大型と小型のプラークサイズのウイルスの混合群を産生するように思われた(表3)。
ヒト肝細胞がん細胞系HepG2およびTHLE-3細胞に0.005のMOIでChimeriVax(商標)-WN01 (野生型prME)、ChimeriVax(商標)-WN02 P5 (E107、E313、E316、E440の突然変異、M66 L/P混合アミノ酸、SおよびL混合プラークを含む)、ChimeriVax(商標)-WN LP (E107、E313、E316、およびE440、WNL)、ならびにChimeriVax(商標)-WN SP (E107、E313、E316、E440、およびM66P、WNS)を感染させた。上清を毎日回収し、標準的なニュートラルレッド含有アガロース重層法を用いてO-Vero細胞で力価判定した。
WN、JE、およびYFウイルスなどの、フラビウイルスに対する検出可能な抗体を欠いた(プラーク減少中和試験(PRNT)によって判定した場合)実験未使用のカニクイザル計8頭にChimeriVax(商標)-WN02 (P5) (n=4)またはYF-VAX(登録商標) (n=4)のいずれかを皮下経路により接種した。本研究の目的は3日の観察期間の間のChimeriVax(商標)-WN02ワクチンのウイルス血症、生体内分布、および毒性の可能性を評価することであった。接種量はChimeriVax(商標)-WN02およびYF-VAX(登録商標)に対し、それぞれ約1.25×105 PFU/0.5 mLおよび5.5×104 PFU/mLであった。動物から毎日採血し、動物を接種から4日後に殺処理した。Vero細胞での標準的なプラークアッセイを用いてウイルス血症レベルを判定するのに血液を使用し、その一方、回収された組織はウイルス分析のために急速冷凍されたかまたは組織病理学的評価のために保存されたかのいずれかであった。
本研究で使われた動物は、BL2下のマイクロセパレータ内で管理され、研究の間中IACUCにより承認された動物プロトコルにしたがって扱われた。3種のChimeriVax(商標)-WN02ウイルス(SP、PMS、P10; LP、PMS、P10、ならびに混合SPおよびLPワクチンウイルス、P5)を用いて、Harlan Sprague-Dawley系由来の7週齢雌性ゴールデンシリアンハムスター(Mesocricetus auratus)に感染させた。各ウイルスをハムスター15匹の群へ皮下経路を介して鼠径部に注射した。感染量は105 pfuであり、接種量は100 μlであった。動物5匹のさらなる群にシャム対照としてウイルス希釈剤100 μlを同様に注射した。ウイルス感染日(0日目)および感染から5日後までの各後日に、血液サンプルをシャム対照群以外の全動物から後方眼窩採血によって採取した。動物は採血および接種の前にイソフルオランの吸入によって効くまで麻酔させた。試験サンプルのウイルス濃度は、12ウェルプレートの中で増殖されたVero細胞培養物の二重ウェルにおいて10倍希釈済み血清サンプル0.1 mLを直接的にプラーク形成させることにより測定された(図5)。
背景と概要
以下に記述される研究において、本発明者らはウシ伝染性脳症のプリオン因子によるワクチン汚染の可能性に関する懸念を排除するため、無血清(SF)培地中で増殖されたVero細胞を用いて新たなChimeriVax(商標)-JEシードウイルスを調製し、特徴づけた。SF培養での増殖の間、クローニングされていないウイルスは、血清含有培養で以前には見られなかった突然変異を蓄積しており、これはウイルス複製の速度を増大させる、SF増殖条件への適応であるように思われた。これらの突然変異はEまたはMタンパク質中で起こっており(E-107でのFからLまたはM-60でのRからCの突然変異)、ウイルス複製の過程におけるMタンパク質の機能的重要性を示唆するものであった。それらはSF条件でのウイルス増殖の間に顕著になった(実施例1 (ChimeriVax(商標)-WN)に示されるMタンパク質の60位のアミノ酸Rを参照のこと)。ワクチンの生物学的特性に及ぼすMタンパク質内部(M60、ChimeriVax(商標)-JEではM5)またはEタンパク質内部(ChimeriVax(商標)-JEではE-107、ChimeriVax(商標)-DEN1および-DEN3ではE202/204ならびにChimeriVax(商標)-DEN2ではE251)の突然変異の影響が規定された。これらのキメラウイルスの全てが既に治験において試験されている。
細胞と培地
Vero細胞は、もとはアメリカン・タイプ・カルチャー・コレクション(ATCC; Manassas, VA; CCL 81; アフリカミドリザル腎細胞)から受け取った。これらの細胞をSF培地中で増殖するように適合させ、継代数133でBaxter (Orth, Austria)から得た後に、フラスコの中に播いて直接的に用いるかまたは細胞バンクから継代数136で始めて播いた。全ての実験において、Vero細胞の継代レベルは継代数149を超えなかった。細胞およびウイルスを7.5% CO2下、36℃で増殖させた。細胞をSF条件下で増殖させた。
前臨床試験および治験で試験された非SF ChimeriVax(商標)-JEワクチン候補の産生のため以前に使われ(Monath et al., Vaccine 20:1004-1018, 2002)、既報(Chambers et al., J. Virol. 73:3095-3101, 1999)のように調製された、同一のインビトロRNA転写産物(-80℃で保存された)を用いてのSF Vero細胞のエレクトロポレーションにより、ウイルスを始動させた(継代P1)。増幅的継代を通常、0.001 pfu/細胞のMOIで行い、ウイルス収集物を感染後3〜4日で(CPEが約10%であった場合に)回収し、低速の遠心分離によって清澄化し、10%ソルビトールを追加し、-80℃で保存した。クローニングされた変異体は、ガンマ線を照射したFBS (HyClone; SF培地を用いて調製された寒天下で細胞はプラークを形成できなかったので、FBSが使われた)の存在下、標準的な寒天ニュートラルレッド重層法を利用した3回連続のプラーク精製によりBaxter Vero細胞で産生させ、その後SF条件で増幅させた。指定のサンプルでウイルス力価を測定するためのプラークアッセイは、感染後5日でのクリスタルバイオレットによるプラークの可視化とともに単回のメチルセルロース重層法を利用して行った。
ChimeriVax(商標)-DEN1〜4ワクチンウイルスは、ウイルスcDNAから調製されたRNA転写産物を用いてのVero細胞のエレクトロポレーションにより調製された。子孫ウイルスを3ラウンドのプラーク精製に供して、継代数7 (P7)の時点でプレマスターシード(PMS)ウイルスを産生させた。さらに3回の継代を米国の現行の医薬品、医薬部外品の製造管理および品質管理基準(Good Manufacturing Practice)(cGMP)により行って、ワクチンロット(P10ウイルス)を産生した。Vero細胞での複数回の継代後に、いくつかの突然変異がキメラのE遺伝子中に現れた(Guirakhoo et al., J. Virol. 78:4761-4775, 2004)。これらの突然変異のうちの一つ(ChimeriVax(商標)-DEN1中のE204)は、非ヒト霊長類においてウイルスの内臓向性を著しく低下させた(Guirakhoo et al., J. Virol. 78:9998-10008, 2004)。
指示されたウイルスサンプルのコンセンサス配列の決定は、既報(Pugachev et al., Vaccine 20:996-999, 2003)のように行われた。手短に言えば、TRIZOL LS試薬(Life Technologies-Gibco BRL)を用いて抽出されたウイルス粒子RNAをTitan One-Tube RT-PCRキット(Roche)により、長さが約2〜3 kbの重複する5つのcDNA単位複製配列で増幅させた。正と負の両方向のJE特異的なおよびYF特異的なオリゴヌクレオチドプライマーならびにCEQ Dye Terminator Cycle Sequencingキット(Beckman)を用いて単位複製配列を配列決定した。配列決定用の反応産物はCEQ2000XL自動配列決定装置(Beckman Coulter)で分解した。Sequencher 4.1.4 (GeneCodes)ソフトウェアを用いて、データをアライメントし解析した。ヌクレオチド不均質性は、プラス鎖とマイナス鎖の両方の配列決定反応に相当する全てのクロマトグラムで不均質なシグナルが確認された場合に記録された。一部のウイルスについては、構造遺伝子を含む、5個のcDNA単位複製配列のうちの1番目(断片I)だけが配列決定された。
マウスの管理と世話は、実験動物の人道的使用に関する国立衛生研究所のガイドラインに従った。妊娠非近交系ICR雌性マウスはTaconic Farms (Germantown, NY)から購入した。新生マウスを育成し、接種の6日前に新たな群に混ぜ合わせた。8日齢の授乳マウスの群に脳内(IC)経路によって指示されたウイルスサンプル0.02 mlを接種した。接種に使われたウイルスの1:10連続希釈はMEM-10% FBS中で行われた。未希釈の接種材料を逆力価判定し、各希釈液の正確な用量を計算した。死亡率を21日間にわたって記録した。YF 17D対照ウイルスは市販のワクチンバイアルから再構成されたYF-VAX(登録商標) (Aventis Pasteur, Swiftwater, PA)とした。
実験1
YF-VAX(登録商標)対照(YF 17Dワクチンウイルス)と比べたときの、新たなクローンC (M-60) ChimeriVax(商標)-JEのワクチンマスターウイルスバンク(Vaccine Master Viral Bank) (MVB; P11)とプロダクションウイルスバンク(Production Viral Bank) (PVB; P12)ストックの神経毒性と毒性プロファイルをカニクイザルにおいてGLP基準にしたがって研究した。実験未使用の、フラビウイルス血清反応陰性(HAI試験により判定された場合)カニクイザル33頭を表9に示される処置群に割り当てた。全てのサルに単回のIC注射を介して1日目に投薬し、30日間観察し、それから安楽死させて解剖した。サルを臨床的徴候(毎日2回)、ならびに飼料消費(毎日)、体重(毎週)、および臨床病理学的指数の変化について評価した。黄熱病ワクチンに関する世界保健機関(WHO)の必要条件(WHO, Technical Report Series, No. 872, 1998)に基づき、臨床スコアリングシステムにしたがって臨床スコアを割り当てた。血液サンプルを臨床病理学的解析(血液生化学的および血液学的パラメータ)のため、1日目の接種前にならびに3、5、7、15、および31日目に採取した。さらなる血液サンプルをウイルス血症の定量的測定のため1日目(投薬前)および2〜11日目に採取し、中和抗体力価分析のため1日目(投薬前)および31日目に採取した。完全な解剖は31日目に行い、組織を保存用に採取した。組織を肝臓、脾臓、心臓、腎臓、および副腎の病理組織診断用に調製した。Levenbookら(J. Biol. Stand. 15:305, 1987)により記述され、黄熱病ワクチンに関するWHOの必要条件(WHO, 1998)に組み入れられている方法にしたがって、脳および脊髄の病理組織診断を行った。
この実験はカニクイザルにおいて単回皮下(SC)投与後30日間にわたりChimeriVax(商標)-JEワクチン[Eタンパク質の疎水性尾部内のE491でのLからFの変化を除いて突然変異を含有していない、FBS存在下のLS5 Vero細胞で以前に産生された、オリジナルの非クローン化ワクチンP5 (BB-IND #9167, Serial #000)]および新たなクローンC (M-60突然変異体) ChimeriVax(商標)-JE精製ワクチン大量調製物(P13)のウイルス血症、免疫応答、および安全性をGLP基準にしたがって比較するために行われた。実験未使用の、フラビウイルス血清反応陰性(HAI試験による)カニクイザル18頭を表10に示される処置群に割り当てた。全てのサルにSC注射を介して片腕の単一個所に1日目に一度投薬した。サルを毒性の臨床的徴候(毎日2回)、体重の変化(毎週)、ならびに血液生化学的、血液学的、および凝固パラメータについて評価した。血液サンプルを血液生化学的、血液学的、および凝固パラメータ分析のため、1日目(接種前)にならびに4、7、15、および31日目に採取した。さらなる血液サンプルをウイルス血症の定量的測定のため1日目(接種前)および2〜11日目に採取し、日本脳炎ウイルス特異的な血清抗体の力価分析のため1日目(接種前)および31日目に採取した。
低pHへのフラビウイルスの曝露(細胞膜の非存在下で)の結果の一つが、Eタンパク質の不可逆的な立体構造変化の誘導とウイルス不活性化(効力の喪失)である。細胞膜の存在下では、これらの立体構造変化は、細胞膜のものとのウイルス膜の融合に必要であり、宿主細胞へのウイルスゲノムの放出を結果的に引き起こす。蚊細胞系C6/36を用いて、WN、DEN、YF、およびJEなどの蚊媒介性ウイルスに対する融合のpH閾を内部からの融合(FFWI)により測定することができる(Guirakhoo et al, Virology 169(1):90-99, 1989)。しかしながら、本発明者らは、恐らく蚊および蚊細胞系でのウイルスの十分な増殖の欠如によって、本発明者らのChimeriVax(商標)ウイルスの全てでFFWIを実証することができなかった(Johnson et al., Am. J. Trop. Med. Hyg. 70(1):89-97, 2004)。したがって、本発明者らは「間接的融合アッセイ」と本明細書で呼ばれるアッセイにおいて、異なるpHレベルへの曝露後のウイルス効力の喪失を測定することを試みた。このアッセイでは、細胞膜のものとのウイルス膜の融合が起こるpH閾を間接的に測定する。
M-60突然変異(およびE-107突然変異)がSF Vero細胞での侵入を促進することを実証するため、SF Vero細胞に5、10、20、または60分間、SF培地に適切に希釈されたクローンA、C、およびIウイルスを感染させ、その後0.1 Mグリシン、0.1 M NaCl、pH 3.0で3分間処理して、細胞外のウイルスを不活性化させた。ウェルをPBSで2回洗浄し、次いで単層にメチルセルロースを重層した後、クリスタルバイオレットを用いて5日目にプラークを染色した。グリシンの代わりにPBSで処理された対照感染ウェルと比べたときの、グリシン処理後に確認されたプラーク数の割合として侵入率を計算した。
臨床試験(プロトコルH-040-003)が行われた。健常な成人男性および女性被験者に投与されたワクチンは、M60に天然配列(アルギニン)を持っていた。健常な成人被験者/群はChimeriVax(商標)-JEワクチンの段階的用量の皮下投与を受け、各種の対照群が含まれた。1用量群につき被験者11〜33人が試験された。Vero細胞単層でのプラークアッセイによって、ウイルス血症が毎日測定された。同一のアッセイおよび検査室により、両試験でのウイルス血症レベルが測定された。
非クローン化SF ChimeriVax(商標)-JEウイルスの適応突然変異、およびクローン化変異体の調製
本研究で産生されたウイルスサンプルの線図を図6に示す。クローニングされていない継代数2回(P2)の初期サンプル(プレマスターシード候補; PMS)は、以前の研究(Monath et al., Vaccine 20:1004-1018, 2002)のためFBS含有培地中でワクチンを産生するために用いられたインビトロRNA転写産物を細胞にトランスフェクトし、続けてさらに増幅的継代を行うことによりSF培養で得られた。このウイルスの全ゲノムを配列決定し、検出可能な任意の突然変異を含まないことが示された(表7) (コンセンサス配列決定法では微量な亜集団を検出していない; 突然変異の検出限界は約10%であることに留意されたい)。このP2ウイルスから始めてP10レベルまでの小規模継代をT25フラスコ中で行って、SF培養での持続的増殖の間にその遺伝的安定性(g.s.)を分析した(図6; g.s.継代)。g.s. P5およびg.s. P10継代物の全ゲノム配列には、残基M-60でのRからCのアミノ酸置換を引き起こす、ヌクレオチド番号935でのCからTの1ヌクレオチド変化があった(表7)。この突然変異はg.s. P3ではなく、g.s. P4継代物で不均質性として最初に検出可能であった。
SF培養での非突然変異体、M-60突然変異体、およびE-107突然変異体ウイルスの増殖速度を比較するため、細胞に0.001 pfu/mlのMOI (逆力価判定により確認された)で非クローン化P2 PMS、非クローン化P5 g.s.サンプル(M-60突然変異体)、または非クローン化P5ワクチンバルク変異体(E-107突然変異を含有する)、ならびにある割合のE-107突然変異を同様に含有する非クローン化P3マスターシードおよびP4プロダクションシードウイルスを感染させた。ウイルスを含有する培地の日単位の一定分量を回収し、プラークアッセイによって力価判定した。図7に示されるように、M-60ウイルスは非突然変異体P2ウイルスよりも速く増殖し、感染後の3日目および4日目に著しく(10倍を超える)高い力価をもたらした。E-107突然変異もM-60突然変異と同様にウイルス複製を強めた。このように、M-60およびE-107突然変異はどちらもSF培養での増殖優位性を明らかに付与した。この結論の裏付けとして、非突然変異体クローンAおよびM-60突然変異体クローンCの両ウイルスのS、SSS、およびSSF g.s.継代(図6参照)由来の日単位サンプルを回収し力価判定して、増殖速度を分析し、その結果、M-60突然変異体は非突然変異体に比べて最大10倍までの高いピーク時力価(ほぼ8 log10 pfu/ml)を常にもたらした。さらに、この結論は小規模なSF培養でのクローンA、C、およびIウイルスの増殖曲線を比較することで確認された。クローンC (M-60)およびI (E-107)がクローンA (非突然変異体)よりも高い力価にまで常に増殖していたからである。
マウス神経毒性試験を利用して、ChimeriVax(商標)ワクチン候補の神経毒性がYF 17Dベクターのものを超えないことを裏付けた。YF 17DワクチンはIC接種後の全齢のマウスに致死的である。対照的に、ChimeriVax(商標)ワクチンはかなり弱毒化されている。成体マウスは一般に、神経毒性の微妙な相違、例えば、単一のアミノ酸変化によるものを検出するには感度がよくないので、生存率解析を用いたさらに感度のよい授乳マウスモデルをその目的に使うことができる(Guirakhoo et al., Virology 257:363-372, 1999; Guirakhoo et al., Virology 298:146-159, 2002; Monath et al., J. Virol. 76:1932-1943, 2002)。
実験1
この実験では、クローンC (M-60突然変異体) ChimeriVax(商標)-JEワクチンマスターウイルスバンク(MVB)およびプロダクションウイルスバンク(PVB)の神経毒性を、対照としてYF-VAX(登録商標)ウイルスを利用し、カニクイザルへのIC投与後に比較した(表9)。
この実験はカニクイザルでの単回の皮下(SC)投与後のオリジナルの非クローン化P5 ChimeriVax(商標)-JEワクチン[FBS存在下のVero細胞で以前に産生されており、生物学的表現型の点から見ると良性の突然変異であるように思われる、Eタンパク質の疎水性尾部に位置するE491でのLからFの変化を除いては突然変異がなく、治験で既に試験されている(Monath et al., J. Infect. Dis. 188:1213-1230, 2003; Monath et al., Vaccine 20:1004-1018, 2002)]と新しいクローンC (M-60突然変異体)のChimeriVax(商標)-JE精製済みワクチンバルク(P13)のウイルス血症、免疫応答、および安全性を比較するために行われた。ChimeriVax(商標)-JEワクチンは、オリジナルの非クローン化P5 ChimeriVax(商標)-JEワクチンを接種された血清反応陰性のサル5頭のうち5頭(100%)の血清中で検出された。ウイルス血症の期間は2〜5日で、力価が20〜790 PFU/mLの範囲であった。平均のピーク時ウイルス血症(±SD)は244 (±310) PFU/mLであり、平均のウイルス血症日数は3.4 (±1.34)であった(表10)。
ChimeriVax(商標)-JEワクチンは、YF 17Dウイルスの骨格へのJEウイルスのSA14-14-2株由来のprMおよびE遺伝子の挿入によって産生された。SA14-14-2ウイルス(ChimeriVax(商標)-JEの中に存在する)の外被は、その親ウイルスSA14とは10アミノ酸異なっていた: E107でLからF、E138でEからK、E176でIからV、E177でTからA、E227でPからS、E244でEからG、E264でQからH、E279でKからM、E315でAからV、およびE439でKからR (Guirakhoo et al., Virology 257:363-372, 1999)。部位特異的突然変異誘発法により、これらの残基の一部がChimeriVax(商標)-JEウイルスの弱毒化に関与することが明らかになった。ChimeriVax(商標)-JEの突然変異体または復帰突然変異体を選択して、突然変異がこれらのウイルスのpH閾を変化させたかどうかを確認した。M-60、E-107、またはM-5の突然変異がpH依存的にウイルス感染性に影響を及ぼすかどうかを判定するため、感染性のpH閾に対する標準的なアッセイを材料と方法で記述したように行った。以下のウイルスを試験した: (1) ChimeriVax(商標)-JE非突然変異体(Eタンパク質中の10種全てのSA14-14-2突然変異を含有するクローンA、P7); (2) ChimeriVax(商標)-JEのE107でのFからLの復帰突然変異体(9種のEタンパク質突然変異を含有するクローンI P6); (3) ChimeriVax(商標)-JEのM60でのRからCの突然変異体(10種全てのEタンパク質突然変異を含有するクローンC、P10)、および(4) M-5でのQからPの突然変異体(10種全てのEタンパク質突然変異を含有するクローンE、P6) (表12)。
各ChimeriVax(商標)-DENワクチンウイルスの二つの群、つまりEタンパク質突然変異を含有しないChimeriVax(商標)-DEN1〜4 P7およびChimeriVax(商標)-DEN4 P10を除いて、Eタンパク質中に単一の突然変異を含んだChimeriVax(商標)-DEN1〜4 P10を利用し、間接的融合アッセイを行った。ウイルスを室温で10分間、異なるpHの培地を用いてインキュベートした。pHを中性pHに戻した後に、標準的なプラークアッセイを利用して、力価を測定した。表13に示されるように、ウイルス不活性化(融合)の閾値は、ChimeriVax(商標)-DEN2およびDEN4ウイルスのP7とP10との間で類似していた(pH 6.4)。その一方、ChimeriVax(商標)-DEN1 P10のpH閾は、ChimeriVax(商標)-DEN1 P7ウイルスのものよりも0.4単位低かった(pH 6.0 対 pH 6.4)。pH閾の相違はChimeriVax(商標)-DEN3 P10ウイルスの場合にはあまり劇的ではなかった(pH 6.4 対 pH 6.2)。
E単量体のうちの一つの204Kおよび261Hの側鎖が、対向する単量体の、それぞれ、252Vおよび253Lの骨格原子とH結合を作り出すように思われた。204位で、ワクチンウイルス(VL P10)のEタンパク質中のRは、これらの水素(H)結合が失われるようにそれ自身を再配向すると予測される。その代わりに、突然変異体Rの側鎖が261Hおよび257Eと近接する結果、204Rと261Hとの間の新しい分子内H結合の生成と、恐らく204Rと257Eとの間の新しい塩橋の生成を引き起こす。ヒスチジンのpkはおよそ6.0になる可能性があり、これは融合閾値(pH約6.4)をわずかに下回っているので、Guirakhooら(J. Virol. 78:9998-10008, 2004)による初期の仮説は、204Rと261Hとの間の予測される新しいH結合および204Rと257Eとの間の塩橋が融合のpH閾に影響を及ぼす可能性があるというものであった。本明細書に記述される実験は、ChimeriVax(商標)-DEN1の融合の閾値が6.0前後であり、これはそのP7ウイルス(pH 6.4)よりも0.4 pH単位低いことを明らかにしたので、この理論は正しいと判明した。残基番号204のRにより導入される新しい分子間結合は、低pHへの移行がしかるべき残基(例えば、H261)のいっそうのプロトン化を必要とするようにE二量体の会合を強固にするのは明らかである。低い融合閾値はサルにおけるウイルスの内臓向性に影響を及ぼし、i.c.経路により接種された授乳マウスの神経毒性を低減する(Guirakhoo et al., J. Virol. 78:9998-10008, 2004)。
Chambers (Vlaycheva et al., J. Virol. 76:6172-6184, 2002)の方法を用いて、SF Vero細胞へのウイルス侵入に及ぼすM-60 (クローンCウイルス)およびE-107 (クローンIウイルス)突然変異の影響を調べた。この実験では、SF Vero細胞に5、10、20、または60分間、(各時点で約50プラーク/ウェルをもたらすよう)適切に希釈されたウイルスを感染させた。内部移行しなかったウイルスを酸性グリシン溶液の添加によって不活性化し、その一方で対照の並行ウェルをPBS (中性pH)によって処理する。細胞をPBSで洗浄し、これをメチルセルロースの重層で覆い、その後5日目にプラークの可視化と計測を行った。侵入率は、対照のPBS処理ウェルでのプラーク数に対するグリシン処理ウェルでの平均プラーク数の割合として示される。予備的な侵入試験の結果を図9Aに示す。侵入したクローンCとクローンIウイルスの割合が、5および10分の時点で非突然変異体クローンAウイルスよりも高かったことは重要であり、その時点で、侵入に及ぼす突然変異の影響が検出される可能性がいっそう高い。この結果は標準偏差の棒からも明らかなように統計的に有意ではなく、さらなる繰り返し試験で確認される必要がある。それでもなお、この実験によって、M-60とE-107での突然変異のどちらも、SF条件で増殖された細胞とのChimeriVax(商標)-JEウイルスの膜融合の効率を改善できることが示唆された。膜融合の過程に及ぼすM-60およびE-107残基の影響に関して考えられる機構を図9Bに図解する。M-60残基はウイルス膜中に位置しており、その一方E-107残基は細胞膜に挿入されるので、低pH依存的なEタンパク質の再構成に続いてこの二つの膜は強引に融合される(これは本発明者らのデータに基に、Mタンパク質の細胞外ドメインによって促進されうる)。これらの二つの残基のいずれかでのさらに適切なアミノ酸は、膜の融合を促進することができる。
上述の治験から得られた、アルギニンおよびシステインM60残基を有するChimeriVax(商標)-JEのウイルス血症プロファイルを表11AおよびBで比較する。ChimeriVax(商標)-JE M60アルギニンを受けた被験者では、ChimeriVax(商標)-JE M60システインを受けた被験者の場合の29〜50%に比べて、被験者の67〜100%が一日またはそれ以上の日数でウイルス血症であった。ChimeriVax(商標)-JE M60アルギニンを受けた被験者での平均最大ウイルス血症レベルは、ChimeriVax(商標)-JE M60システインの場合の3.5〜6.3 PFU/mlの平均最大ウイルス血症レベルに比べて、13〜40 PFU/mlの範囲であった。ウイルス血症の期間もChimeriVax(商標)-JE M60アルギニンの場合には著しく長かった。

1, 2: この列はC/prMまたはE/NS1接合部に対応するキメラYF/フラビウイルスプライマーを作出するために使われたオリゴヌクレオチドを明示している。(本文参照)。X = YFカプシドのカルボキシル末端のコード配列。下線部はNarI部位(アンチセンス - ccgcgg)のすぐ上流の標的異種配列に相当する。この部位は、完全長の鋳型cDNAを作出するのに必要なYfm5.2 (NarI)プラスミドへのPCR産物の挿入を可能にする。その他のヌクレオチドは異種ウイルスに特異的である。オリゴヌクレオチドプライマーは5'から3'方向に記載されている。
3, 4: 完全長の鋳型キメラcDNAを産生するためにインビトロで単離でき連結できる制限断片を作出するのに使われた唯一の制限部位が記載されている。配列の中には好都合な部位を含んでいないものもあるので、適切な部位の遺伝子工学的な作出が場合によっては必要になる(脚注5)。
5: 括弧内は、効率的なインビトロライゲーションを可能にするためYF骨格または異種ウイルスのいずれかで作出されなければならない制限酵素部位である。括弧内にない部位は除去されなければならない。そのような改変は全て各クローンに対するcDNAのサイレントな突然変異誘発により行われる。空欄はcDNAクローンの改変が必要ないことを示す。
Claims (45)
- 膜タンパク質突然変異を含む組換えフラビウイルス(Flavivirus)。
- 突然変異がフラビウイルスを弱毒化する、請求項1記載のフラビウイルス。
- 突然変異がフラビウイルスの内臓向性/ウイルス血症を減少させる、請求項2記載のフラビウイルス。
- 突然変異が、突然変異のない対応するフラビウイルスに比べて、フラビウイルスの安定性の増大をもたらす、請求項1記載のフラビウイルス。
- 突然変異が、突然変異のない対応するフラビウイルスに比べて、細胞中でのウイルス複製の増大をもたらす、請求項1記載のフラビウイルス。
- フラビウイルスがキメラフラビウイルスである、請求項1記載のフラビウイルス。
- キメラフラビウイルスが第一のフラビウイルスのカプシドおよび非構造タンパク質ならびに第二のフラビウイルスの膜および/または外被タンパク質を含む、請求項6記載のフラビウイルス。
- 第一のフラビウイルスが黄熱病ウイルスである、請求項7記載のフラビウイルス。
- 黄熱病ウイルスがYF-17Dである、請求項8記載のフラビウイルス。

- 突然変異が日本脳炎ウイルスの膜タンパク質のアミノ酸番号60に対応するアミノ酸の置換である、請求項15記載のフラビウイルス。
- 突然変異が膜タンパク質のアミノ酸60位でシステインとのアルギニンの置換をもたらす、請求項16記載のフラビウイルス。
- 突然変異が西ナイルウイルスの膜タンパク質のアミノ酸66位に対応するアミノ酸の置換である、請求項15記載のフラビウイルス。
- 突然変異が膜タンパク質のアミノ酸66位でプロリンとのロイシンの置換をもたらす、請求項18記載のフラビウイルス。
- 突然変異が日本脳炎ウイルスまたは西ナイルウイルスの膜タンパク質の60、61、62、63、64、65、または66位のものに対応する一つまたは複数のアミノ酸にある、請求項15記載のフラビウイルス。
