Description
フレビオプシス ギガンテア(Rotstop、Verdera Oyの商標)を、シリカを基本とした固体成長培地で充填床固体反応装置を用いて培養した。用いた反応装置の見取り図は図1に示した。
P.ギガンテアを図2に示した充填床固体発酵反応装置を用いて生育した。
固体成長培地としてコーン スティープ ソリッド(CSS)0.62kg、ラクトース0.62kg、石灰0.62kg、非晶質シリカ12kgおよび水道水28.8kgを使用し、ストレプトマイセス エスピー.(Mycostop、Verdera Oyの商標)を実施例1および2に記載のように生育した。該バクテリアは28℃で7日間培養した。
グリオクラジウム カテヌラタム(GlioMix、Verdera Oyの商標)は、実施例1に記載のように、固体成長培地として濃縮蒸留かす0.53kg、石灰60g、非晶質シリカ1.5kg、水道水3.4kgを使用して生育した。菌類は22℃で18日間培養した。
P.ギガンテアは、2種の異なる成長容器および接種材料供給注入口を備える図2に示した充填床固体発酵反応装置を用いて生育した。
フレビオプシス ギガンテア(Rotstop、Verdera Oyの商標)は、底から該反応装置スペース内への注入口を有するらせん状のリボンミキサーを備えた円柱状(sylindrical)の充填床固体発酵装置で、実施例1に記載された方法で、シリカを基本とする固体成長培地上で生育した。該反応装置は圧力容器であり、in situで蒸気を用いて121℃で滅菌した。滅菌および冷却後、反応装置の頂上にある反応装置内への注入口を通して成長培地上へ接種材料を流し込むことによって、および同時にミキサーのスイッチを入れることによって該反応装置に植菌した。使用した該成長培地は、混合に対して非常に脆弱であることが観察された。5分後には、植菌の間、緩く軽い構造は失われ、得られた該産物はドウのようになった。
Claims (19)
- 固体発酵(SSF)反応装置内の固体成長培地上の微生物を培養する方法であって、
a.外部振動によって固体成長培地を移動させる工程、および
b.移動中に、純粋培養によって固体成長培地の植菌を均一に行う工程
を含有する方法であることを特徴とする方法。 - 植菌が無菌的に行われることを特徴とする、請求項1に記載の方法。
- 固体成長培地が有機担体および/または無機担体を含有することを特徴とする、請求項1に記載の方法。
- 有機担体が穀物、ふすま、おがくず、ピートまたは木片からなる群から選択されることを特徴とする、請求項3に記載の方法。
- 無機担体がバーミキュライト、パーライト、非晶質シリカまたは粒状粘土からなる群から選択されることを特徴とする、請求項3に記載の方法。
- 培養される微生物が酵母を含む菌類、細菌または線虫であることを特徴とする、請求項1に記載の方法。
- 振動が外部の電気回転バイブレーター、磁気、液圧または空気圧式バイブレーターにより発生することを特徴とする、請求項1に記載の方法。
- 植菌に使用される微生物が液体または固体の形状で固体成長培地上に供給されることを特徴とする、請求項1に記載の方法。
- 微生物の液体接種材料がノズルを用いて懸濁液として噴霧されることを特徴とする、請求項8に記載の方法。
- 微生物の固体接種材料がスクリュー、振動またはベルトコンベヤーを用いて固体成長培地に供給されることを特徴とする、請求項8に記載の方法。
- 固体発酵(SSF)に適した反応装置であって、
a.請求項1に記載のように内容物を移動させる間、該反応装置本体およびその内容物に効果的な振動を与えるように、該反応装置本体外側に取り付けられた外部バイブレーター1、および
b.該反応装置本体に取り付けられた少なくとも1つの植菌供給注入口2
を含むことを特徴とする前記反応装置。 - 発酵が無菌的に行われることを特徴とする、請求項11に記載の反応装置。
- 該反応装置本体が、
a.上部区画4および下部区画3の2つの区画、
b.該2区画を分離する透過性のセパレーター5、および
c.該反応装置本体の外側に取り付けられた外部バイブレーター1、および
d.下部区画に取り付けられた少なくとも1つの植菌供給注入口2、
e.該反応装置本体を上下に反転させる、任意の反転手段
を含むことを特徴とする、請求項11または12のいずれかに記載の反応装置。 - 透過性のセパレーターがふるいまたは穿孔プレートであることを特徴とする、請求項13に記載の反応装置。
- 該反応装置本体が、
a.外部バイブレーター1、
b.脱着可能な固体成長培地滅菌ユニット6、
b.下方管7、
c.成長容器8、および
d.少なくとも1つの植菌供給注入口2
を含むことを特徴とする、請求項11または12のいずれかに記載の反応装置。 - 固体成長培地の移動中に、接種材料を供給するための少なくとも1つの植菌供給注入口が、滅菌ユニット6と成長容器8との間にあることを特徴とする、請求項15に記載の反応装置。
- 脱着可能な固体成長培地滅菌ユニット6が振動らせん供給容器であることを特徴とする、請求項15に記載の反応装置。
- 下方管7が自由に振動できるようにするために可撓性であることを特徴とする、請求項15に記載の反応装置。
- 無菌的に移動させるための、および純粋培養物を成長させるためのSSF反応装置内の固体成長培地の均一な植菌のための外部振動の使用。
