ン測定値を、該細胞を変異原性刺激剤にin vitroで暴露した後に取得し、当該個体の細胞を変異原性刺激剤にin vitroで暴露し、暴露後に、該細胞における成長抑制マーカーのレベルを測定し、そして、癌に対する個体の感受性を決定する。ここで、前記平均ベースライン測定値よりも高いレベルは、癌の危険性がより低いことを示し、前記平均ベースライン測定値よりも低いレベルは、癌の危険性がより高いことを示す。

開示される方法において使用されるmRNA測定プロトコルは、より大量の未調製の全血の分析を可能にし、専ら白血球に由来するmRNAを分析する効率的な手段を提供し、rRNA及びtRNAを除去し、安定したmRNA回収を提供し、容易に自動化に適合できる。リアルタイムPCRを使用した絶対的な定量化及び20〜25%の範囲の変動係数を有する優れた再現性を含む、高感度定量化システムが提供される。

本発明の方法は、個体の全血において成長抑制マーカーのレベルを測定する。好ましい実施形態では、測定されるマーカーのレベルは、mRNAのレベルである。しかしながら、マーカーのレベルはまた、例えば該個体の細胞中に存在するマーカータンパク質のレベルを測定することなどにより、当業者に既知の他の方法で評価されてもよい。本出願で使用される場合、「個体」は、ヒトを含む任意の種の動物であり得る。さらに、本出願で使用される場合、「成長抑制」mRNAは、細胞増殖を停止させる細胞破壊mRNA、及び細胞を死滅させる細胞破壊mRNAの両方を包含する。以下の実施例では、変異原刺激後の成長抑制mRNAのレベルのより大きな増大は癌の危険性がより低いことを示すのに対して、減少は、癌の危険性がより高いことを示す。しかしながら、他のマーカーは反対のパターンを示すかもしれない。すなわち、変異原刺激後のmRNAのレベルの大きな減少が癌の危険性がより低いことを示すのに対して、増加は癌の危険性がより高いことを示すということもあろう。
壊マーカーBAX及びPUMAが測定された。米国仮特許出願第60/653,557号(参照により本明細書に援用される)に開示されるように、PUMAは、ヒト血液中で主要なアポトーシス促進性mRNAであることが、本発明者らにより決定された。しかしながら、Bak、Bok、Bcl−XS、Bid、Bad、Bik、Bim及びNOXAのような当業者に既知の他の細胞破壊マーカーmRNAが測定されてもよい。
この実施例で用いるmRNAレベルを測定するためのプロトコルは以下の通りであった。アッセイ手順は、1)フィルタープレート上での白血球単離及び溶解、2)mRNA単離、逆方向プライマーハイブリッド形成及びオリゴ(dT)固定されたマイクロプレートにおけるcDNA合成、並びに3)リアルタイム定量的PCR、の3つの主要な工程から構成される。簡潔に述べると、フィルタープレートを回収プレート上に配置し、5mmol/Lのトリス(pH7.4)150μLを添加して、フィルタ膜を湿潤させた。フィルタープレートから溶液を除去するため120×g、4℃で1分間遠心分離し、その後に、十分混合した血液サンプル50μLを各ウェルに添加して、直ちに120×g、4℃で2分間遠心分離した後、リン酸緩衝生理食塩水(PBS)300μLを加えて2,000×g、4℃、5分間の遠心分離することにより、各ウェルを一度洗浄した。その後、1%の2−メルカプトエタノール(Bio Rad)、0.5mg/mLのプロテイナーゼK(Pierce)、0.1mg/mLのサケ精子DNA(5 Prime Eppendorf/Brinkmann)、0.1mg/mLのE.coli tRNA(Sigma)、それぞれ10mmol/Lの特異的逆方向プライマー及び107分子/mLの内部標準としての合成RNA34を追加したストック溶解緩衝液(下記を参照)60μLをフィルタープレートに適用した後、37℃で10分間インキュベートした。その後、フィルタープレートをオリゴ(dT)を固定化したマイクロプレート(GenePlate、RNAture)上に配置して、2,000×g、4℃で5分間遠心分離した。4℃で一晩保存した後、マイクロプレートを溶解緩衝液(他の成分を添加していないもの)100μLで3回、その後洗浄緩衝液(0.5mol/LのNaCl、10mmol/Lのトリス(pH7.4)、1mmol/LのEDTA)150μLで3回、4℃で洗浄した。cDNAは、1×RT緩衝液(50mmol/LのKCl、10mmol/Lのトリス−HCl(pH8.3)、5.5mmol/LのMgCl2、ジチオスレイトールなし)、それぞれ1.25mmol/LのdNTP、4ユニットのrRNasin、及び80ユニットのMMLV逆転写酵素(Promega)を含有する緩衝液(プライマーなし)30μLを添加し、37℃で2時間インキュベートすることにより、各ウェル中で直接合成された。得られたcDNA4μLを384ウェルPCRプレートへ直接移して、そこにTaqManユニバーサルマスターミックス(Applied Biosystems)5μL及びオリゴヌクレオチドカクテル(それぞれ15μmol/Lの順方向及び逆方向プライマー、及び3〜6μmol/LのTaqManプローブ)1μLを添加して、95℃で10分間を1サイクル、続いて95℃で30秒間、55℃で30秒間、及び60℃で1分間の45サイクルを用いて、PRISM 7900HT(Applied Biosystems)によりPCRを実施した。各遺伝子は別個のウェルで増幅させた。ある特定量のPCR産物(蛍光)を生成するためのPCRのサイクルであるサイクル閾値(Ct)は、解析用ソフトウェア(SDS、Applied Biosystems)を用いて測定された。PCRはまた、iCycler(BioRad)を用いて、GenePlate中で直接実施された。

上記と同じプロトコルを使用して、細胞破壊マーカーであるBAXのmRNAレベルを測定した。BAX mRNAが、アポトーシスの初期段階で誘導されるため、BAXは、細胞破壊マーカーであるとみなされる。図3に示すように、実施例1の癌患者は、10Gyの放射線による刺激時に、BAX mRNAの減少を示したのに対して、試験した健常な成人の大部分が、放射線刺激後にBAX mRNAの増大を示した。
上記と同じプロトコルを使用して、細胞破壊マーカーであるPUMAのmRNAレベルを測定した。ヒト血液中において、PUMA mRNAは、BAXファミリーの遺伝子の中で最大のアポトーシス促進効果を有することが見出されている。10Gyの放射線で刺激した癌患者の血液は、乏しいPUMA応答を示した一方で、試験した健常な成人の大部分が、放射線刺激後にPUMA mRNAの増大を示した。
興味深いことに、図4の値は、同じ個体(黒菱形、黒三角)内で一定ではなく、時間とともに変動した。これは、癌危険性(DNA損傷応答)が改変され得ることを示す。したがって、この試験は、in vivoで、又はin vitroで候補化合物とともに全血をインキュベートすることにより、各個体に関して癌予防レジメンを同定するのに適用可能であり得る。

Claims (60)
- 個体において癌に対する感受性を決定する方法であって、
前記個体の細胞を、変異原性刺激剤にin vitroで暴露すること、
前記個体の暴露された細胞及び暴露されていない細胞において、成長抑制マーカーのレベルを測定すること、並びに
前記暴露された細胞及び暴露されていない細胞におけるマーカーレベルの差に基づいて、前記個体の癌に対する感受性を決定すること
を含む方法。 - 前記細胞は前記個体の全血に由来する、請求項1に記載の方法。
- 測定されるレベルは、mRNAのレベルである、請求項1に記載の方法。
- 前記個体はヒトである、請求項1に記載の方法。
- 前記成長抑制マーカーは細胞分裂抑制マーカーである、請求項1に記載の方法。

前記個体の細胞を、変異原性刺激剤にin vitroで暴露すること、
前記細胞において、成長抑制マーカーの暴露後のレベルを測定すること、及び
癌に対する前記個体の感受性を決定すること
を含み、前記平均ベースライン測定値に比べて高いレベルは癌の危険性がより低いことを
示し、前記平均ベースライン測定値に比べて低いレベルは癌の危険性がより高いことを示すことを特徴とする方法。 - 前記細胞は前記個体の全血に由来する、請求項16に記載の方法。

前記インキュベートされた細胞及びインキュベートされていない細胞における暴露後の前記成長抑制マーカーのレベルの差に基づいて、癌予防効果を有する化合物を同定すること
を含む方法。 - 前記成長抑制マーカーのレベルは、変異原性刺激剤に暴露されていないインキュベートされた細胞において測定される、請求項29に記載の方法。

- 個体において癌予防に効果的な化合物を決定する方法であって、
前記個体から細胞を取り出すこと、
前記個体へ少なくとも1つの化合物を投与すること、
前記化合物の投与後に前記個体から細胞を取り出すこと、
投与前及び後に取り出された前記細胞を、変異原性刺激剤にin vitroで暴露すること、
投与前に取り出された暴露された細胞及び暴露されていない細胞において、成長抑制マーカーのレベルを測定すること、並びに
投与前及び後に取り出された細胞において、前記成長抑制マーカーのレベルの暴露後の差に基づいて、前記化合物の癌予防効果を決定すること
を含む方法。 - 前記成長抑制マーカーのレベルは、変異原性刺激剤に暴露されていない投与後に取り出された細胞において測定される、請求項45に記載の方法。
