Description
本発明は、一般にサイトカイン阻害剤に関する。さらに特定的には、本発明は、IL8レセプター結合に対する阻害剤(抗体を包含する)に関するが、この阻害剤は、IL8レセプターのアミノ末端細胞外ドメインと相互作用し、そしてレセプター結合についてIL8および他の関連する天然のリガンドと競合する。
サイトカインは、白血球により生産されるホルモン様仲介物質の群である。この因子は、内因性の生物学的シグナルとして働くが、このシグナルは、マクロファージ、リンパ球、および他の細胞型を含む、局所ならびに全身の両宿主防御機構を増幅するように抗原と協同して作用する。代表的なサイトカインは、種々のインターロイキン、インターフェロン、GROα、GROβおよびGROγ、好中球活性化ペプチド-2(NAP-2)、およびENA-78を包

発明の要旨
本発明は、IL8レセプターのアミノ末端細胞外ドメインとの相互作用を介してそのレセプターへのIL8結合を阻害する物質を提供する。この阻害剤は、IL8レセプター仲介生物
学的活性の有用なモジュレーターである。

(IIi)第3番目のポリペプチドのアミノ酸配列は、アミノ酸配列S-S-T-L-P-P-F-L-L-D-A-A-P-C と実質的に同一のアミノ酸配列を含む;そして
(iv)第4番目のポリペプチドのアミノ酸配列は、アミノ酸配列F-L-L-D-A-A-P-C-E-P-E-S-L-E-I と実質的に同一のアミノ酸配列を含む。
(a)IL8レセプターのアミノ末端細胞外ドメインに結合し得、そしてまたレセプターについてIL8と競合し得る、IL8レセプター結合に対する阻害剤を提供する工程;および

A.定義
本発明を記載するに際して、次の用語が用いられ、そして以下のように定義されることが意図される。

B.一般的方法
本発明の中心は、IL8レセプターに対する種々のリガンド(IL8、GROα、GROβ、GRO
γ、NAP-2および、ENA-78を包含する)の結合を抑制し得、それによりこのような結合によって誘発される生物学的応答を調節し得るIL8阻害剤の発見である。従って、これらの阻害剤は、走化性および好中球の活性化を遮断または調節し得る。この阻害剤は、好中球が疾患(関節リュウマチおよび炎症性腸疾患のような炎症性疾患を包含する)の病変を引き起こすもととなる広範な障害の治療および予防に対して有用であり、そして敗血症性ショックおよび急性呼吸不全症候群(ARDS)のような疾患において起こる組織損傷の治療お
よび/または予防に対して有用である。本発明の阻害剤はまた、癌疾患、ウイルス感染症、細菌感染症、真菌感染症、および原虫感染症の治療における好中球仲介炎症応答の刺激に対する用途が見出され、そしてAIDSならびに他の後天性および遺伝性免疫障害のような
、免疫系が弱められた状態の治療に対する用途が見出される。
端細胞外ドメインの配列は、図1および2に示される。本発明に使用される阻害剤は、この領域に広がる連続した配列に由来する単離されたペプチド配列に対して生じさせた抗体を包含し、そしてその類似体(即ち、アミノ酸置換、付加、または欠失を有する配列)に対して生じさせた抗体をも包含する(これらは、上記に定義されたように、IL8レセプターに結合して、それにIL8が結合することを阻害する能力を保持している)。驚くべきことに、IL8R1およびIL8R2両者のアミノ末端細胞外ドメイン領域由来のペプチドに対し
て生成した抗体および抗体の混合物は、これらレセプターへのIL8の結合を遮断し得る。それゆえに、これらの抗体に対する認識部位(エピトープ)は、IL8認識部位と重複するか、あるいはその部位に近接しているようである。これらの抗体は、標準的な技術を用いて作成される、ポリクローナル、モノクローナル、キメラ、あるいはこれらの機能的フラグメントであり得る。さらに、有用な抗体調製物は、もっと後で詳述されるような、数種類の異なる抗体の混合物を包含し得る。

C. 実験
次の記載は、本発明を実施するための特定の実施態様の例である。本実施例は、説明目的のためにのみ提供され、決して本発明の範囲の限定を意図しない。
ペプチドおよび抗血清の調製
ペプチド合成、類毒素との結合、マウスおよびヒツジの免疫、ならびにピンに結合されたペプチドおよびプレート上に被覆されたペプチドに対するELISA反応性の測定は、ChiRonMimotopes Pty.Ltd.(Clayton,VictoRia 3168,オーストラリア)により、標準的な手順を利用して実施した。特に、ヒトIL8レセプターのIL8R1およびIL8R2のアミノ末端細胞外領域部分とかなりの相同性を有するペプチド(表1および2に示す)を、標準技術を使用して合成した。いくつかのペプチド(表中に示される)を、その抗原性を高めるために、内部システイン残基を介してトキシンと結合させた。N末端のアセチル基またはC末端のβアラニン-ジケトピペラジン基は、タンパク質内のペプチド配列の環境を模擬す
る遮断基である。

、ペプチド#6および#7に対してマウスで、そしてペプチド#10、#11、#12、#13の混合物に対してヒツジで生じさせた(図1、表2)。免疫血清は、ピン上のペプチドおよび/またはプレート上に被覆されたペプチドに対してELISAにより測定されたとき、免疫された
ペプチドに対して高力価を示したが、免疫前血清ならびにペプチド#6および#7に対する抗血清は示さなかった。免疫ペプチド#6および#7に対する抗血清の免疫活性の欠乏についての説明として、技術上の困難性が製造会社により述べられている。この特定の抗血清は続行されず、そしてペプチド#10〜13に対する抗血清を代わりに使用した。
好中球IL8レセプター認識アッセイ
抗体を、好中球を認識する能力について下記のようにテストした。好中球は、本質的には製造者の方法に従い、下記のように改変して、好中球単離培地(NIM)(CaRdinal)を用いて全血より単離した。50mlの試験管にNIM20mlおよび血液30mlを入れて50分間遠心分離した。混入している赤血球を氷冷水中で溶血により除去した。得られた好中球を、ハンクス生理緩衝液(HBS)中に懸濁し、氷冷保存した。
再懸濁し、FACS分析用に染色した。細胞は氷上で2時間インキュベートし、1000RPMで5
分間遠心分離し、さらにPBS+1%BSAで2回洗浄した。洗浄した細胞を、FITC結合F(ab’)2 フラグメントのウサギ抗ヒツジIgG(H+L)抗体(PBS+1%BSA中1:100、JacksonImmu
noReseaRchLaboRatoRies)50μl中に再懸濁した。細胞は氷上で1時間インキュベート
し、1000RPMで5分間遠心分離し、PBS+1%BSAで洗浄し、さらにPBSで洗浄した。直ちに
分析に供する細胞は、PBS500mlに懸濁した。生細胞の染色としてヨウ化プロピジウムを2.5ng/mlの割合で添加した。好中球は1%パラホルムアルデヒド中に固定し、最高72時間後まで分析され得る。標識した細胞をFACSCAN(BectonDickonson)で分析した。
ン由来のペプチドに対して生じた抗血清が、天然レセプターを認識し得ることを示す。IL8R1ペプチド#3に対して生じた抗血清は、免疫ペプチドに対する力価は高いが、好中球
を認識しなかった(データは示していない)。この結果は、リガンド認識におけるIL8R
1ペプチド#3配列の役割については何も示していないが、ペプチド#3は、抗ペプチド抗体とは反対に、抗レセプター抗体を惹起するレセプター模倣性立体構造で十分量存在していなかったということのみを意味している。IL8R2ペプチド#6または#7のいずれに対して
生じた抗血清も免疫ペプチドに対して低い力価しか示さず、検討は続行しなかった。
実施例III
レセプター結合アッセイ
抗体を、IL8のそのレセプターへの結合を遮断する能力について、下記のようにテストした。IL8R1およびIL8R2を、レセプター結合アッセイに用いるために、遺伝子組換えにより生成した。IL8R1およびIL8R2をコードするDNAを、IL8R1(HolmesらScience(1991)253:1278)およびIL8R2(MuRphyおよびTiffany,Science(1991)253:1280)についての公開されている配列に基づくオリゴヌクレオチドプライマーを用いるPCRにより、ヒ
トゲノムDNAから単離した。単離した遺伝子のヌクレオチド配列をジデオキシ配列分析に
より確認した。

層細胞を、Hepes-BSA結合緩衝液(25mMHepes,pH7.5,150mM NaCl,5mM CaCl2,5mM MgCl2,1mg/mLウシ血清アルブミン)中、室温で1時間抗体で前処理した。125IIL8を最終濃度が0.2nMとなるよう添加し、室温で3時間インキュベートした。細胞はHepes-BSA結合緩衝液で1回洗浄し、結合125IIL8をガンマ計数器で測定した。非特異的結合は、非
標識IL81μg/mlの存在下で測定した。データは2回繰り返し測定の平均である。

胞で発現されるIL8R2レセプターへの125IIL8の結合を遮断したが、免疫前抗血清は遮断しなかった(図5)。IL8R2ペプチド#10〜13に対する抗血清は、IL8のIL8R1へ
の結合を阻害しなかった(図3)。従って、この抗血清はIL8R2へのIL8の結合を特異
的に中和し、そしてIL8R2のIL8認識配列の近傍にあるかまたは重なっているエピトー
プを認識した。
プターへのIL8の結合を阻害した(図6)。遊離のペプチド#10はこの抗体による中和を
阻止し、このことから、ペプチド#10は、IL8R2上のIL8認識部位における、またはその近傍にあるエピトープを含むことが確認された(図6)。これはIL8R2のアミノ酸残基
1〜15の領域内の中和エピトープに対応する。抗ペプチド#10抗体が涸渇しても、抗IL8R2ペプチド抗血清がIL8R2へのIL8の結合を阻害する能力はなくならなかった(データ
は示していない)。このことは、IL8R2のアミノ末端細胞外領域の残りの部分、即ちア
ミノ酸残基16〜41内に別のエピトープがあり、これもまた抗IL8R2ペプチド抗血清の中
和活性に寄与していることを示している。これらの結果は、IL8R2のアミノ末端細胞外
ドメインのアミノ酸1〜41がIL8の認識に関係する配列を含むことを示唆している。
Claims (2)
- IL8レセプター1のアミノ末端細胞外ドメインと相互作用し得、かつ該レセプターへの結合についてIL8と競合し得る抗体またはそのフラグメントを含む、IL8レセプター1結合に対する阻害剤であって、
ここで、該抗体は、アミノ酸配列M-S-N-I-T-D-P-Q-M-W-D-F-D-D-Lを含むポリペプチドからなる免疫原で哺乳動物を免疫することによって獲得され得るものである、阻害剤。 - 本明細書に記載の方法によって生産される抗体。
