Abstract
【解決手段】 そのような樹状細胞は、腫瘍、ウイルス、もしくは細菌性抗原をT細胞に提示するのに使われ、予防接種プロトコールに有用となりうる。他のサイトカインを、該活性化され抗原パルスされた樹状細胞と別々に、逐次的にもしくは同時に組み合わせて使用することが可能である。抗原を発現し活性化された樹状細胞を用いて免疫応答を刺激する方法も開示される。

Description
本発明は、樹状細胞(dendritic cell)活性化因子、リンパ球を介した免疫応答をin vivoで昂進する方法、並びに、細胞性および体液性免疫応答の操作に有用な樹状細胞集団に関する。
予防接種は、感染症による死もしくは障害を防ぐ有効な手段である。感染症における本法の成功は、新生物疾患の治療もしくは予防における予防接種の利用に対する興味も刺激してきた。しかしながら、ワクチン利用により達成された数々の成功にもかかわらず、ワクチン開発の分野には未だに多くの挑戦がある。非経口の投与経路、多くの異なる予防接種が要求されること、並びにブースター免疫の必要性、およびその頻度など、すべてが疾患を管理もしくは排除しようとする努力の妨げとなっている。
本発明は、樹状細胞を活性化して抗原提示能を昂進させる方法に関する。活性化され、抗原提示する本発明の樹状細胞は、ワクチンアジュバントとして有用である。
本発明は、抗原パルスされた樹状細胞を活性化するためのCD40Lの利用に向けられている。活性化は、樹状細胞がリンパ球細胞に抗原を提示する能力を昂進し、それによって抗原に対する免疫応答を増大させる。本発明の別の態様は、活性化され、抗原パルスされた樹状細胞の単離およびそのワクチンアジュバントとしての利用である。活性化され、抗原パルスされた樹状細胞は、ex vivoで使用し、抗原特異的なT細胞を作製することもできる。
樹状細胞(dendritic cell)は、顕著な形態と広範な組織分布を持つ不均一な細胞集団を含む。樹状細胞系およびその免疫における役割は、Steinman,R.M., Annu. Rev. Immunol.,9:271−296(1991)(本明細書中に参考文献として援用される)にまとめられている。樹状細胞の細胞表面は変わっており、ベールのような特徴的な突起物があり、細胞表面マーカーCD1a+,CD4+,CD86+もしくはHLA−DR+を有すことから特徴付けられる。樹状細胞は、MHC制限T細胞の感化に関し高い能力を持ち、T細胞発生および寛容期間における自己抗原および免疫中の外来抗原の両方に関し、非常に効果的にin situでT細胞へ抗原提示する。
造血幹細胞および始原細胞のex vivoでの増殖手法が米国特許第5,199,942号(本明細書中に参考文献として援用されている)に記載されている。他の適当な方法については、当該分野では既知である。簡単に述べると、ex vivo培養および増殖は以下のものを含む:(1)患者、末梢血採取物、もしくは骨髄外植体からCD34+造血幹細胞および始原細胞を回収すること;および(2)当該細胞をex vivoで増殖させること。特許第5,199,942号に記載されている細胞性増殖因子に加え、flt3−L,IL−1,IL−3およびc−kitリガンドなどの他の因子を使用することができる。
様々なサイトカインが、樹状細胞のex vivo培養に有用であろう。flt3−Lは、EP 0627487 A2およびWO 94/28391(共に本明細書中に参考文献として援用されている)に記載されている一群のポリペプチドをいう。ヒトflt3−L cDNAは、1993年8月6日にアメリカンタイプカルチャーコレクション(ATCC,アメリカ、メリーランド州、ロックビル)に寄託され、登録番号ATCC 69382を受けている。IL−3は、米国特許第5,108,910号(本明細書中で参考文献に取り入れられている)に記載されているインターロイキン−3ポリペプチドをいう。本発明での使用に適したヒトIL−3蛋白質をコードするDNA配列は、アメリカンタイプカルチャーコレクション(ATCC)より登録番号 ATCC 67747で公に入手可能である。c−kitリガンドは、マスト細胞増殖因子(MGF)、スチール因子もしくは幹細胞因子(SCF)とも呼ばれ、EP 423,980(本明細書中に参考文献として援用されている)に記載されている。
CD40は、腫瘍壊死因子(TNF)/神経成長因子(NGF)受容体ファミリーの一員であり、細胞外領域におけるシステインに富んだモチーフの存在により定義される(Smithら、Science 248:1019,1990;MallettおよびBarclay, Immunology Today 12:220;1991)。このファミリーには、リンパ球抗原CD27,CD30(ホジキンリンパ腫およびReed−Sternberg細胞に見出される抗原)、TNFに対する2種の受容体、4−1BBと呼ばれるマウス蛋白質、ラットOX40抗原、NGF受容体、並びにFas抗原が含まれる。ヒトCD40抗原(CD40)は、分子量30,600を持つ、277アミノ酸から成るペプチドである(Stamenkovicら、EMBO J. 8:1403,1989)。

有用なCD40結合蛋白質は、CD40に結合し、CD40のCD40Lとの結合を阻害する(可溶性CD40のCD40Lとの結合を少なくとも約90%阻害することが観察されることによって決定される)能力を持つものであり、モノクローナル抗体、CD40リガンドおよびその誘導分子が含まれる。CD40表面抗原に対するモノクローナル抗体(CD40 mAb)は、ヒトB細胞に対し様々な生物学的活性を媒介することが示されている(例えば、LEUKOCYTE TYPING IV; A.J. McMichael編、Oxford Unviersity Press、オックスフォード、426頁)。USSN 08/130,541(1993年10月1日出願)(その関連した開示は本明細書中に参考文献として援用されている)は、CD40に特異的に結合する2種のモノクローナル抗体、hCD40m2およびhCD40m3を開示している。他のCD40 mAbとは異なり、hCD40m2(ATCC HB11459;ブダペスト条約下、アメリカンタイプカルチャーコレクション、米国メリーランド州ロックビルに1993年10月6日寄託された)およびhCD40m3は、CD40に結合し、CD40Lを構成的に発現する細胞にCD40が結合するのを阻害することで、hCD40m2およびhCD40m3は、リガンド結合ドメインもしくはその近くでCD40に結合することが示されている。
免疫は、疾患を予防もしくは改善するための病原体に対する防御的免疫応答も含め、1世紀の歴史を持つ非常に効果的な手段である。そのような誘導のために使われてきたワクチンは、生きた弱毒化された微生物、または殺された微生物標品もしくはその分画であることが一般的である。弱毒化生ワクチンは一般的に、殺された微生物もしくは病原体由来の非感染性標品から調製されるもの(即ち、トキソイド、組換蛋白質ワクチン)に比べ、自然の感染時に起こる免疫応答により近く模倣するものと考えられている。しかし、弱毒化ワクチンもまた、病原性の復活という危険性を持っており、特に免疫不調の患者には病気を起こしうる。
本発明は、活性化され抗原パルスされた樹状細胞を含む治療組成物の使用法を提供する。そのような細胞を可溶性サイトカイン受容体もしくはサイトカインまたは他の免疫制御分子と組み合わせて使用することも企図されている。本発明の組成物は、免疫応答を刺激するために投与され、巨丸注入、連続点滴、インプラントからの持続的放出、もしくは他の適当な技術によって与えられうる。典型的には、生理学的に受容可能な担体、賦形剤もしくは希釈剤とともに、抗原パルスされ活性化された樹状細胞を含む組成物の形で、本発明の方法の細胞を投与するであろう。そのような担体は、使われる薬量および濃度において受容者にとって毒性が無いであろう。中性緩衝化生理食塩水もしくは同種の血清アルブミンと混合した生理食塩水が、実際に適した希釈液である。
本発明は好ましくは以下の態様を含む。
1.(a) 樹状細胞を回収し;
(b) (i)抗原の取り込みおよび加工を促進する条件下で培養中に該樹状細胞を抗原に暴露させる、もしくは(ii)該抗原をコードする遺伝子で該樹状細胞をトランスフェクトすることによって、該樹状細胞に該抗原を発現させ;そして
(c) 該抗原を発現する樹状細胞を、可溶性CD40のCD40Lへの結合を最低約90%阻害することが観察されることによって決定されるように、CD40に結合し、CD40のCD40Lへの結合を阻害することができるCD40結合蛋白質に暴露することによって、該抗原を発現する樹状細胞を活性化する
工程により産生される、抗原を発現する活性化された樹状細胞の集団。
(a) SEQ ID NO:2のアミノ酸1から261,35から261,34から225,113から261,113から225,120から261もしくは120から225を含むペプチド;
(b) CD40に結合する(a)によるペプチドの断片;並びに

ここにおいて、アミノ酸194のシステインのかわりに置換されるアミノ酸は、トリプトファン、セリン、アスパラギン酸およびリジンからなるグループから選択される
からなるグループから選択される、態様3に記載の集団。
(b) (i)抗原の取り込みおよび加工を促進する条件下で培養中に該樹状細胞を抗原に暴露させる、もしくは(ii)該抗原をコードする遺伝子で該樹状細胞をトランスフェクトすることによって、該樹状細胞に該抗原を発現させ;
(c) 該抗原を発現する樹状細胞を、可溶性CD40のCD40Lへの結合を最低約90%阻害することが観察されることによって決定されるように、CD40に結合し、CD40のCD40Lへの結合を阻害することができるCD40結合蛋白質に暴露することによって、該抗原を発現する樹状細胞を活性化し;そして
(d) 該活性化され抗原を発現する樹状細胞を該個体に投与する
工程を含む、個体における抗原に対する特異的な免疫反応を刺激する方法。

10.造血幹細胞もしくは始原細胞を該個体から回収し、該造血幹細胞もしくは始原細胞をflt−3リガンド,GM−CSF,IL−4,TNF−α,IL−3,c−kitリガンド, GM−CSFとIL−3の融合体、およびこれらの組み合わせからなるグループから選択される分子と接触させることによって、樹状細胞が得られる、態様9に記載の方法。

(b) (i)抗原の取り込みおよび加工を促進する条件下で培養中に該樹状細胞を抗原に暴露させる、もしくは(ii)該抗原をコードする遺伝子で該樹状細胞をトランスフェクトすることによって、該樹状細胞に該抗原を発現させ;
(c) 該抗原を発現する樹状細胞を、可溶性CD40のCD40Lへの結合を最低約90%阻害することが観察されることによって決定されるように、CD40に結合し、CD40のCD40Lへの結合を阻害することができるCD40結合蛋白質に暴露することによって、該抗原を発現する樹状細胞を活性化し;そして
(d) T細胞に対して該樹状細胞に該抗原を提示させる
工程を含む、個体から抗原特異的T細胞を調製する方法。
17.CD40結合蛋白質が:
(a) SEQ ID NO:2のアミノ酸1から261,35から261,34から225,113から261,113から225,120から261もしくは120から225を含むペプチド;
(b) CD40に結合する(a)によるペプチドの断片;
(c) (a)もしくは(b)のペプチドをコードするDNAに、ストリンジェントな条件下(6×SSC、63℃で一晩ハイブリダイズ;3×SSC、55℃で洗浄)でハイブリダイズするDNAにコードされ、CD40に結合するペプチド;

(b) (i)抗原の取り込みおよび加工を促進する条件下で培養中に該樹状細胞を抗原に暴露させる、もしくは(ii)該抗原をコードする遺伝子で該樹状細胞をトランスフェクトすることによって、該樹状細胞に該抗原を発現させ;
(c) 該抗原を発現する樹状細胞を、可溶性CD40のCD40Lへの結合を最低約90%阻害することが観察されることによって決定されるように、CD40に結合し、CD40のCD40Lへの結合を阻害することができるCD40結合蛋白質に暴露することによって、該抗原を発現する樹状細胞を活性化し;そして
(d) T細胞に対して該樹状細胞に該抗原を提示させる
工程によって産生される抗原特異的T細胞の集団。
(a) SEQ ID NO:2のアミノ酸1から261,35から261,34から225,113から261,113から225,120から261もしくは120から225を含むペプチド;

並びにオリゴマー形成ペプチドから成るグループから選択される、可溶性オリゴマーCD40リガンドである、態様19に記載の集団。
本実施例は、精製された樹状細胞をex vivoで作製する方法を記載する。ヒト骨髄を入手し、CD34+表現型を有す細胞をCD34抗体カラム(CellPro, Bothell, ワシントン州)を用いて分離する。CD34+細胞を、適当な培地、例えば、マッコイのエンハンス培地(McCoy’s enhanced media)(樹状細胞の増殖を促進するサイトカイン(即ち、GM−CSF, IL−4, TNF−αをそれぞれ20ng/ml、または、100ng/mlのflt3−Lもしくはc−kitリガンド、またはそれらの組み合わせ)を含む)で培養する。37℃、10% CO2、湿気でおよそ2週間培養し続ける。それから細胞を、CD1a+, HLA−DR+ およびCD86+に対する抗体を使ってフローサイトメトリーによりソートする。GM−CSF, IL−4およびTNF−αの組み合わせは、2週間の培養後、細胞数を6から7倍に増加させることができ、その内50−80%がCD1a+ HLA−DR+ CD86+である。flt3−Lおよび/もしくはc−kitリガンドの添加は、総細胞の増加、従って、樹状細胞の増加をさらに昂進する。こうした条件下で分離し培養した細胞の表現型分析は、細胞の60−70%がHLA−DR+ CD86+(試験した全ての因子の組み合わせにおいてCD1aを発現する細胞の40−50%)である。
本発明は、樹状細胞を回収し増幅させるための方法を記載する。細胞回収前に、flt3−Lもしくはサルグラモスチン(Leukine(登録商標),Immunex Corporation,ワシントン州シアトル)を個体に投与し血中のPBPCおよびPBSCの数を変動もしくは増加させることができる。CSF−1,GM−CSF,c−kitリガンド,G−CSF,EPO,IL−1,IL−2,IL−3,IL−4,IL−5,IL−6,IL−7,IL−8,IL−9,IL−10,IL−11,IL−12,IL−13,IL−14,IL−15,GM−CSF/IL−3融合蛋白質,LIF,FGFおよびその組み合わせなどの他の増殖因子を、flt3−Lと連続的にもしくは同時に組み合わせて、同様に投与することができる。
本実施例は、CD40L刺激された樹状細胞が同種抗原を提示し、従ってT細胞の増殖をもたらす能力を例示する。CD34+細胞をヒトドナーの骨髄から得て、選択されたサイトカインの存在下で2週間培養し、実質的に実施例1に記載したようにしてフローサイトメトリーにより分離した。混合リンパ球反応液(MLR)中でそれらを使う前に、樹状細胞の増殖を助けるサイトカインを含むマッコイのエンハンス培地中で、CD40Lの可溶性3量体の存在下(1μg/ml)もしくは非存在下で樹状細胞をさらに24時間培養した。
本実施例は、樹状細胞が抗原特異的にT細胞の増殖を刺激する能力を例示する。CD34+細胞を、破傷風トキソイドに対して反応すると考えられるヒトドナーの骨髄から採取し、選択されたサイトカインの存在下で2週間培養し、実質的に実施例1に記載したようにしてフローサイトメトリーによって分離した。これらを破傷風トキソイド(TTX)抗原提示アッセイに使用する前に、樹状細胞の増殖を助けるサイトカインを含むマッコイのエンハンス培地中で、CD40Lの可溶性3量体の存在下(1μg/ml)もしくは非存在下で樹状細胞をさらに24時間培養し、それから精製したTTX(Connaught Laboratory Inc., ペンシルバニア州スイフトウォーター)で、37℃、10%CO2下で24時間パルスした。
本実施例は、CD40Lが、抗原パルスされた樹状細胞の抗原特異的T細胞の刺激を活性化する能力について例示する。CD34+細胞を実施例4に記載したようにして回収し、処理をした。但し、CD40Lとともに培養する前に破傷風トキソイドで24時間パルスした。自己由来の破傷風トキソイド反応性T細胞の誘導、および抗原特異的T細胞増殖アッセイは、実施例4に記載されたようにして行った。結果は、図3に示してあるが、最初に抗原でパルスし、それからCD40Lで培養した樹状細胞は、抗原でパルスしたがCD40L暴露しなかった樹状細胞に比べ、TTX特異的T細胞にTTXを提示した際に同等の増殖刺激を与えるのに要する量が、3倍少なかったことが示された。
Claims (13)
- (a) (i)抗原の取り込みおよび加工を促進する条件下で培養中に該樹状細胞を抗原に暴露させる、もしくは(ii)該抗原をコードする遺伝子で該樹状細胞をトランスフェクトすることによって、該樹状細胞に該抗原を発現させ;そして
(b) 該抗原を発現する樹状細胞を、CD40に結合できるCD40結合タンパク質に暴露することによって、該抗原を発現する樹状細胞を活性化する
工程により産生される、抗原を発現する活性化された樹状細胞の集団であって、
ここにおいて、前記CD40結合タンパク質は、CD40抗体、あるいは、CD40結合ペプチド及びオリゴマ形成ペプチドを含む、可溶性オリゴマーCD40リガンドであって、そして、CD40結合ペプチ緒は、以下の:
(i) SEQ ID NO:2のアミノ酸1から261,35から261,34から225,113から261,113から225,120から261もしくは120から225を含むペプチド;
(ii) CD40に結合する、(i)のペプチドの断片;並びに
(iii) (i)もしくは(ii)のペプチドをコードするDNAに、ストリンジェントな条件下(6×SSC、63℃で一晩ハイブリダイズ;3×SSC、55℃で洗浄)でハイブリダイズするDNAにコードされ、CD40に結合するペプチド
から成るグループから選択される、
前記集団。 - CD40結合蛋白質が、CD40のCD40Lへの結合を少なくとも90%阻害することができる、請求項1に記載の集団。
- 造血幹細胞もしくは始原細胞を、GM−CSF,flt3−L,IL−4,TNF−α,IL−3,c−kitリガンド,GM−CSFとIL−3の融合体、およびこれらの組み合わせからなるグループから選択される分子と接触させることによって、樹状細胞が得られる、請求項1又は2に記載の集団。
- CD40結合タンパク質が:
(a) SEQ ID NO:2に記載されたアミノ酸配列を持ち、アミノ酸194のシステインが別のアミノ酸に置換されているポリペプチド;および
(b) ポリペプチド(a)の断片でCD40に結合するポリペプチド;
ここにおいて、アミノ酸194のシステインのかわりに置換されるアミノ酸は、トリプトファン、セリン、アスパラギン酸およびリジンからなるグループから選択される
からなるグループから選択される、請求項3に記載の集団。 - 前記(a)の樹状細胞が、工程(a)および(b)の前にflt−3リガンドで処理されている、請求項1−4のいずれか1項の集団。
- 免疫応答を刺激するための治療組成物の調製ための、請求項1−5のいずれか1項の活性化された樹状細胞の集団の使用。
- インターロイキン1,2,3,4,5,6,7,10,12および15;顆粒球−マクロファージコロニー刺激因子、顆粒球コロニー刺激因子;インターロイキン−3および顆粒球−マクロファージコロニー刺激因子を含む融合蛋白質;インターフェロン−γ;TNF;TGF−β;flt−3リガンド;可溶性CD40リガンド;可溶性CD83;これらサイトカインの生物学的活性のある誘導体;並びにこれらの組み合わせからなるグループから選択される分子と組み合わせである、請求項6の使用。
- 請求項1−5のいずれか1項の活性化された樹状細胞の集団を含む、免疫応答を刺激するための治療組成物
- インターロイキン1,2,3,4,5,6,7,10,12および15;顆粒球−マクロファージコロニー刺激因子、顆粒球コロニー刺激因子;インターロイキン−3および顆粒球−マクロファージコロニー刺激因子を含む融合蛋白質;インターフェロン−γ;TNF;TGF−β;flt−3リガンド;可溶性CD40リガンド;可溶性CD83;これらサイトカインの生物学的活性のある誘導体;並びにこれらの組み合わせからなるグループから選択される分子と投与される、請求項8の治療組成物。
- 抗原特異的T細胞を調製する方法であって、
(a)請求項1−5のいずれか1項の活性化された樹状細胞の集団を提供し;そして
(b)活性化された樹状細胞がT細胞へex vivoで抗原を提示することを可能にする
工程を含む、前記方法。 - 請求項10の方法により得られるT細胞の集団。
- 請求項10のT細胞の集団の、免疫応答を刺激するための治療組成物の調製のための使用。
- 請求項11のT細胞の集団を含む、免疫応答を刺激するための治療組成物。
