Abstract
【解決手段】本発明は、PTHrPの1以上のアミノ酸残基をそれに相当するPTHの残基へ置換することにより、効力あるPTH-2受容体アゴニストまたはアンタゴニストに変換された新規PTHrP類似体に関する。また、これらPTHrP類似体を用いる、PTH-2受容体の変更された作用を伴う種々の病状を治療する方法も記載する。

Description
本発明は、副甲状腺ホルモンに関連する新規ペプチド(PTHrP)類似体に関する。さらに詳しくは、本発明は、類似体にPTH-2受容体アンタゴニストまたはアゴニスト特性を与える1以上のアミノ酸置換基を有するPTHrP類似体に関する。
副甲状腺ホルモン(PTH)はカルシウムホメオスタシスの主要なレギュレーターであり、その主な標的細胞は骨および腎臓で生じる。カルシウム濃度の調節は胃腸系、骨格系、神経系および心臓血管系の正常な機能に必要である。PTHの合成 および放出は主に血清のカルシウムレベルにより制御され;低レベルはホルモンの合成および放出を刺激し、高レベルは抑制する。PTHは3箇所のカルシウム交換部位:消化管、骨および腎臓で血液中へのカルシウム流入を直接、または非直接的に促進することによりかわるがわる血清カルシウムレベルを維持する。PTHは活性型のビタミンDの腎臓での合成を有利にすることによりカルシウムの正味の胃腸吸収に寄与する。PTHは骨芽細胞を阻害することにより、また間接的には骨再吸収細胞である骨芽細胞の分化を刺激することにより骨からのカルシウムの再吸収を促進する。それはまた、腎臓における少なくとも3つの主要な作用:尿細管カルシウムの再吸収刺激、リン酸塩浄化の増強、および活性型ビタミンDの合成を完了する酵素の増強促進を仲介する。PTHは、受容体が介在するPTHによるホスホリパーゼCの活性化も報告されているが(Hruska et al., J. Clin. Invest. 79:230(1987)、主に受容体が介在するアデニル酸シクラーゼの活性化を通じてこれらの作用を発揮する。

ヒトPTH/PTHrP受容体またはヒトPTH-2受容体をコードするプラスミドでトランスフェクトしたCOS-7細胞を、放射活性リガンドとして[Nle8,21,Y34]rPTH-(1-34)NH2を、また競合物質として表示された非標識リガンドを用いる競合結合研究で評価した。アッセイは室温で2時間行った。データは表示された実験数(n)から得られたIC50値の平均(±標準誤差)である。

ペプチドおよび試薬
PTH/PTHrPハイブリッドリガンドの製造法および同定は従前に記載した(Gardella, T.J. et al., J. Biol. Chem. 270:6584-6588(1995))。[Nle8,21,Tyr34]rat(r)PTH(1-34)NH2はBachem Fine Chemicals(Torrance, CA)から入手した。アンタゴニスト[Leu11,D-Tyr12]hPTHrP(7-34)NH2はPeninsula Laboratories(Belmont, CA)から入手した。他の総てのペプチドは、固相化学およびF-moc保護基を用いて、Massachusetts General Hospital (Boston, MA)の生体高分子合成施設により製造された。125I-[NleTyr8,21,Tyr34]rPTH-(1-34)NH2はクロラミン-Tヨード化によって製造し、HPLCで精製した(Shigeno, C. et al., J. Biol. Chem. 263:3872-3878(1988))。[125I]-Na(2,000 Ci/mmol)はDupont/New EnglandNuclear(Boston, MA)から入手した。ダルベッコ改変イーグル培地(Dulbecco'smodified Eagle's medium)(DMEM)、EGTA/トリプシンおよび100x抗生物質混合物(10,000単位/mlペニシリンおよび10mg/mlストレプトマイシン)はGIBCO製であり;ウシ胎児血清(FBS)はHyclone Laboratories(Logan, UT)製であった。
ヒトPTH/PTHrP受容体(Schipani, E. et. al., Endocrinology 132:2157-2165 (1993)およびヒトPTH-2受容体(Usdin, T. et al., J. Biol. Chem. 270:15455-15458(1995))をコードするcDNAを各々、発現ベクターpcDNA-1およびpcDNAI/Amp(InVitrogen, San Diego, CA)に乗せた。COS-7細胞を培養し、従前に記載したようにトランスフェクトした(Lee, C. et al., Mol. Endocrinol. 9:1269-1278(1995))。24ウェルプレート中で、塩化セシウム/臭化エチジウム勾配遠心分離により精製したプラスミドDNA(200ng/ウェル)を用い、細胞をトランスフェクトした。リガンド結合およびcAMP集積アッセイはトランスフェクション後3日めに行い、この時までに細胞密度は500,000±100,000細胞/ウェルに達していた。
従前に記載したように結合反応を行った(同上)。各ウェル(最終容量:300ml)は26fmolの125I-[Nle8,21,Tyr34]rPTH(1-34)NH2は(100,000 CPM)および種々の量(0.4-300pmol)の非標識競合リガンドを含み、双方とも結合緩衝液(50mMトリス塩酸, pH7.7, 100mM NaCl, 5mM KCl, 2mM CaCl2, 5%熱不活性化ウマ血清,0.5%熱不活性化ウシ胎児血清)で希釈した。インキュベーションは、4℃にて6時間行ったスキャッチャード解析を含む実験以外は、室温にて2時間であった。結合反応の終了時に、細胞を0.5mlの結合緩衝液で3回すすぎ、0.5mlの5M NaOHで分解し、次いで前ライゼートを計測した。1μMの[NleTyr8,21,Tyr34]rPTH-(1-34)NH2を含有するウェル中で測定した非特異的結合のトレーサー(NSB)を添加総計の1%以下であった。最大特異的結合(B0)を、非標識PTHリガンドの不在下で細胞に結合した総放射性からNSBを差し引いて算出した。
トランスフェクトしたCOS-7細胞を500mlの結合緩衝液ですすいだ。次いで、200μlのIBMX緩衝液(2mM IBMX, 1mg/mlウシ血清アルブミン, 35mM Hepes-NaOH, pH7.4を含有するDMEM)と100mlの結合緩衝液もしくは種々の量のPTHまたはPTHrP類自体を含有する結合緩衝液を加えた。このプレートを室温にて60分間インキュベートした。次いで、緩衝液を回収し、粉末状のドライアイス上にプレートを置き、0.5mlの50mM塩酸を添加することにより細胞を分解した。この酸ライゼートをdH2Oで1/30に希釈し、アリコート85-50μl)をラジオイムノアッセイによりcAMP含量について分析した。EC50値(関連する受容体を伴うリガンドに関して得られた最大応答(Emax)の50%が得られるリガンド用量)log[リガンド]に対するlog(E/Emax-E)(ここで、Eは相当するリガンド用量で測定されたcAMP応答である)のプロットから決定した。アンタゴニスト研究については、最大刺激用量付近(1.5nM)の[Tyr34]hPTH-(1-34)NH2アゴニストペプチドを含むIBMX緩衝液の添加の2分前に、100μlの結合緩衝液中のCOS-7細胞に阻害剤ペプチド[Leu11,D-Trp12]hPTHrP(7-34)NH2または[Trp23,Tyr36]hPTHrP-(1-36)NH2を適用した。次いで細胞を室温で30分間インキュベートし、生じた細胞内cAMPをRIAにより測定した。データはAssayZapソフトウエア(Elsevier Science Publishers BV, Cambridge, United Kingdom)を用いて分析した。
ヒトPTH/PTHrPおよびヒトPTH-2受容体を発現するCOS-7細胞において、PTH-(1-34)([Tyr34]hPTH-(1-34)NH2)およびPTHrP-(1-36)([Tyr36]hPTHrP-(1-36)NH2)について得られた結合およびcAMPシグナリング応答を図1に示す。PTH/PTHrP受容体で、PTH-(1-34)およびPTHrP-(1-36)の双方は、放射性リガンド125I-[NleTyr8,21,Tyr34]rPTH-(1-34)NH2の結合を完全に阻害したが、PTH-(1-34)はPTHrP-(1-36)の4.8倍高い効力であった(各々IC50s=39および285nM, p<0.001; 図1Aおよび表1)。PTH-2受容体を発現する細胞を用いた場合、PTH-(1-34)結合は高い見かけの親和性を有し、一方、PTHrP-(1-36)結合は弱い効力しか有しておらず、それはPTH-(1-34)のそれより100倍弱いものであった(図1Bおよび表1)。
2つのリガンドのPTH-2受容体との見かけの結合親和性における差異にあずかるPTHおよびPTHrP領域の位置決定のため、発明者らは図3に示された一連のPTH/PTHrPハイブリッド類似体を評価した。いくつかのハイブリッドリガンド(すなわちハイブリッド1および5)からは、いずれかの親リガンドとは異なり、PTH/PTHrP受容体との結合に関してでさえほとんど競合しなかったので、あまり情報は得られず、このことはこれら類似体がいずれかの親リガンドとは異なっていることを示すものである。同様に、これらのハイブリッドリガンドの弱い結合力は、従前にラット骨肉腫細胞(ROS17/2.8)を用いた研究でも観察された。これらの結果は、PTHrPのアミノ末端1-14位、およびPTHの15-34領域の残基の間の不適合性によって引き起こされたリガンド構造における変化と解釈される(Gardella, T.J. et al., J. Biol. Chem. 270:6584-6588(1995))。PTH-2受容体に対する現行の研究に関して、最も多くの情報が得られるハイブリッド類似体はハイブリッド-3およびハイブリッド-4であった。ハイブリッド-4であるPTHrP-(1-21)/PTH-(22-34)は強力に(IC50=11nM)PTH-2受容体と結合し、同様にPTH-(1-34)とも結合した(表1)。2つの残基によるPTHrP配列の延長により、ハイブリッド-3であるPTHrP-(1-23)/PTH-(24-34)が得られ、これはPTH/PTHrP受容体との結合に関してほとんど競合せず(IC50=1,000nM)、PTH-2受容体とは極めて弱い力でしか結合しなかった(IC50=〜6,000nM)。このリガンドにおいては、ハイブリッド-3の結合プロフィールはPTHrP-(1-36)のそれに類似していた。
ハイブリッド3および4の構造ならびに結合特性を比較することにより、発明者らは22位および/または23位における発散残基がPTH-2受容体結合選択性の一因となっているという仮説を出すに至った。まず、受容体結合における残基22および23の役割を試験するために、Phe-22および/またはPhe-23をそれに相当するPTHのGluおよびTrp残基で置換した短いPTHrP-(15-36)断片を作製した。非置換親ペプチドPTHrP-(15-36)は、PTH-2受容体との結合に関して弱くではあるが競合した(図4B)。Glu-22およびTrp-23のGluおよびTrpへの置換を組み合わせることにより、結合力はかなり改良された。この増強の大部分はPhe-23からTryへの単一の置換によるものと説明できた(表1、図4B)。非置換または置換PTHrP(15-36)断片のいずれかのPTH/PTHrP受容体に対する結合は、これら競合アッセイの中では検出できなかった(図4A)。
PTHまたはPTHrPの23位における前記置換は、PTH-2受容体によるcAMPシグナリングに対する効果はほとんど持たないか、または全く持たない;すなわち、[Trp23]PTHrP-(1-36)は依然として不活性であり、[Phe23]PTH-(1-34)はほとんど完全なアゴニストであった(図6Bおよび6D;表2)。PTH-2受容体を用いるcAMP産生の刺激におけるハイブリッド-2であるPTH-(1-14)/PTHrP-(15-34)の高い能力は、ハイブリッド-1であるPTHrP-(1-14)/PTH-(15-34)の弱い活性に匹敵し(表2)、このことはPTHおよびPTHrPの1-14配列内の残基がこの受容体を用いる2つのリガンドのシグナリング特性の変調に関与していることを示した。
前記で論じた研究は、PTHrPがPTH-2受容体に効果的に結合してこれを活性化できないことが2つの残基、すなわち23位のフェニルアラニンおよび5位のヒスチジンによるものであることを証明する。PTH-2受容体を用いた場合に十分効力のあるPTHにおいては、これらの残基は各々トリプトファンおよびイソロイシンに置換されている。PTHrPの残基5および23をこれに相当するPTHのイソロイシンおよびトリプトファンに変換することにより、PTHrPを完全かつ効力あるPTH-2受容体アゴニストへと変換させることができた。
A. et al., Eur. J. Biochem. 91:201-208(1978))お よびPTHrP類似体(Barden, J.A. & Kemp, B.E., Eur. J. Biochem. 184:379-394 (1989); Ray, F.R. et al., Eur. J. Biochem, 211:205-211(1993))に関するNMR研究では、いずれかのリガンドのおけるこの位置の芳香族残基が分子内疎水性相互作用に関与していることが示唆されている。従って、残基23はリガンド構造を維持すると同時に、受容体-結合相互作用を変調するために重要であるかも知れない。
本発明のなおさらなる態様によれば、患者のPTH-2受容体の活性化を阻害するのに十分な治療上有効量のPTHrP類似体を患者に投与することを含んでなる、変更されたまたは過剰に作用するPTH-2受容体に起因する内科疾患を治療する方法が提供される。好ましい具体例では、この方法に用いるPTHrP類似体はPTHrPの23位にトリプトファンからフェニルアラニンへの単一のアミノ酸置換を有する。特定の具体例では、PTHrP類似体は[Trp23]PTHrP-(1-36)である。

Claims (5)
- PTH−2受容体アゴニストであり、かつPTH/PTHrP受容体アゴニストであるPTHrP類似体であって、アミノ酸残基5および23において変更がなされている配列番号2のアミノ酸配列を有するPTHrP類似体(但し、PTHrP残基5における変更がインロイシンからヒスチジンヘのアミノ酸置換であり、かつ、PTHrP残基23における変更がトリプトファンからフェニルアラニンヘのアミノ酸置換である場合を除く)。
- PTH‐2受容体選択性アンタゴニストであるPTHrP類似体であって、アミノ酸残基23において変更がなされている配列番号2のアミノ酸配列を有するPTHrP類似体(但し、PTHrPアミノ酸残基23における変更がトリプトファンからフェニルアラニンへの置換である場合を除く)。
- 請求項1のPTHrP類似体を含んでなる、骨粗鬆症の治療用医薬組成物。
- 請求頃2のPTHrP類似体を含んでなる、PTH−2受容体の変更されたまたは過剰な作用に起因する内科疾患の治療用医薬組成物。
- PTH−2受容体アゴニストであり、かつPTH/PTHrP受容体アゴニストであるPTHrP類似体であって、アミノ酸残基5および23において変更がなされている配列番号2のアミノ酸配列を有し、PTHrP残基5における変更がインロイシンからヒスチジンヘのアミノ酸置換であり、かつ、PTHrP残基23における変更がトリプトファンからフェニルアラニンヘのアミノ酸置換であるPTHrP類似体を含んでなる、PTH−2受容体の変更されたまたは過剰な作用に起因する内科疾患の治療用医薬組成物。
