Abstract
Description
本発明は、ある種のGタンパク質共役型受容体、特に、OGR1、GPR4およびTDAG8(TDAG8はGPR65とも呼ばれる)がプロトン感受性受容体(プロトン感受性GPCR)として働くという本発明者らの驚くべき発見に基づくものである。よって、本発明は、プロトン感受性機能を有するある種のGPCRの新規な使用、そのようなポリペプチドをコードするポリヌクレオチド、組換え材料およびそれらの製造方法に関する。このようなポリペプチドおよびポリヌクレオチドは、限定されるものではないが、プロトンホメオスタシスが変更されている疾病および医学的状態、例えば、高レベルのプロトン、すなわち水素イオンが関与する疾病および医学的状態、例えば骨粗鬆症、特になどの過剰な骨損失の疾病、特に老年性骨粗鬆症および腎不全による骨粗鬆症をはじめとするある種の疾病の処置方法に関して注目されている。プロトン感受性GPCRは、骨代謝の他、呼吸や心血管機能の調節、ならびに炎症および虚血のような、血液供給の悪化(detoriation)と関連した病態に関与する可能性がある。

CCL39ハムスター繊維芽細胞、HEK 293ヒト胎児腎細胞、およびMG63ヒト骨肉腫細胞(全てATCC = American type culture collection, Manassas, USAからの細胞株)を、重炭酸緩衝DMEMとハムのF12培地(10%ウシ胎児血清および抗生物質を添加)の1:1混合物中、CO2雰囲気下、pH7.4で増殖させる。ヒト小柱骨由来前骨芽細胞の一次培養物を確立し、先に詳細に記載したように培養する(Sottile V, et al., 2002, Bone, 30:699-704)。ヒトOGR1の発現ベクターを、ヒトゲノムDNA(U48405,NM_003485)由来のこの受容体のcDNAをpcDNA3.1(+)/myc−His(Invitrogen, Basel, Switzerland)にクローニングすることによって調製する。部位特異的突然変異誘発を、Stratagene (Basel, Switzerland)製のQuick Changeキットを用いて行う。安定トランスフェクションのためには、ベクターをPvuIで線状化する。安定トランスフェクションおよび一時的トランスフェクションは、Effectene試薬(Qiagen, Basel, Switzerland)を用いて行う。受容体を発現する安定細胞調製物を、抗生物質G418(400μg/ml)で選択した後に単離する。導入遺伝子の発現および膜局在を、FITC標識抗myc抗体(Zymed/Stehelin & Cie, Basel, Switzerland)を用いて免疫細胞化学を行うことにより確認する。
リン酸イノシトール(IP)形成アッセイ:バッファーおよびpH:IP形成実験用の塩 溶液を、広いpH範囲をカバーするように、HEPES単独(20mM)またはHEPES/EPPS/MES(各8mM)のいずれかで緩衝させる。HEPESは4−(2−ヒドロキシエチル)ピペラジン−1−エタンスルホン酸であり、EPPSはN’−(2−ヒドロキシエチル)ピペラジン−N’−3−プロパンスルホン酸であり、MESは2−(N−モルホリノ)エタンスルホン酸である。全ての溶液のpHを、注意深く較正したpHメーター(Metrohm, Herisau, Switzerland)を用い、室温で示された値に調整する。この報告の全てのデータは室温でのpHに対して示されている。本発明者らの較正実験によれば、37℃でのpHを得るためには、pH6.8〜7.8の範囲のHEPESバッファーに関しては0.15pH単位を差し引かなければならない。IP形成アッセイ。24ウェルプレートで増殖させた密集細胞培養を、血清フリーDMEM培地中で24時間、ミオ[3H]イノシトール(100MBq/ml;ART/Anawa Trading, Wangen-Duebendorf, Switzerland)で標識する。次に、細胞を、130mMのNaCl、0.9mMのNaH2PO4、5.4mMのKCl、0.8mMのMgSO4、1.8mMのCaCl2、25mMのグルコースを含有する緩衝塩溶液中、37℃でインキュベートする。イノシトールモノホスファターゼ活性を遮断するためにリチウム(20mM)を加え、IP1を蓄積させる(Berridge MJ, et al., 1982, Biochem J, 206:587-595; Berridge MJ and Irvine RF, 1989, Nature, 341:197-205)。示されている場合には、ウシトロンビンまたはSPC(ともにSigma, Buchs, Switzerland製)を、リチウム添加の1分前に加える。特に断りのない限り、インキュベーションを20分間続ける。次に、細胞を氷冷した蟻酸で抽出し、従前に記載されているように(Seuwen K, et al., 1988, EMBO J, 7:161-168)厳密にバッチカラムクロマトグラフィーを用い、遊離イノシトールから全IPを分離する。
受容体モデル:OGR1と他のロドプシン様GPCRの間には高い類似性が示されていることから、公開されているロドプシン構造(Palczewski K, et al., 2000, Science 289:739-745)を鋳型としてホモロジーモデルを構築する。SCWRLプログラム(Bower MJ, et al., 1997, J Mol Biol, 267:1268-1282)を用い、OGR1一次配列をウシロドプシン(bRHO)に対してアラインし、示されているアミノ酸位に関して、bRHOに存在する側鎖をOGR1の対応する側鎖に置換する。距離依存性誘電関数ε=1*rを有するAMBER電場のparm96.datパラメーター(Cornell WD, et al., 1995, J Am Chem Soc, 117:5179-5197)、エネルギーの精密化のための本発明者ら所有のWit!Pソフトウエアのミニマックスモジュール、およびAMBER6パッケージのsander_classicモジュールを用い、得られた構造を分子力学および動力学を適用して精密化する。全てのコンピューター処理の間、鋳型のCα原子を調和電位で固定することにより、またはそれらの位置を0.5A内に拘束する(位置エネルギーの最小化)によりその動きを拘束する。細胞外ループと全てのアミノ酸側鎖は、その電場の電位内で自由に動けるままとする。残基CYS94とCYS172の間にジスルフィド結合を導入する。細胞内ループは含まれていない。
RT−PCR発現プロファイリング:酸フェノール法を用い、細胞培養物から全RNAを調製する。RNAをDNアーゼで処理し、スーパースクリプトII (Life Technologies/Invitrogen, Basel, Switzerland)を用いて逆転写する。OGR1とグリセルアルデヒド3リン酸デヒドロゲナーゼ(GDPH)に関する反応を並行して、Expand High Fidelity Taq (Roche, Basel, Switzerland)により、次の温度周期プロトコール:94℃にて30秒の変性、65℃(OGR1)または55℃(GPDH)にて45秒のアニーリング、72℃にて50秒の伸長;OGR1は36サイクル、GPDHは30サイクルを用いて設定する。GPDHはmRNA量の内部標準として測定する。OGR1では、PCR反応産物をクローンニングし、シーケンシングにより確認する。次のプライマーを用いた:OGR1フォワード:5’−CTGAGCCCATGAGGAGTGTG−3’、リバース:5’−GGタグACGCCAGCAGCAGG−3’;GPDHフォワード:5’−TTAGCACCCCTGGCCAAGG−3’、リバース:5’−CTTACTCCTTGGAGGCCATG−3’。
組換え受容体をFITC標識抗myc抗体(No. 132511, Zymed/Stehelin & Cie, Basel, Switzerland)を用いて検出する。原形質膜マーカーであるアネキシンVを検出するため、アレキサ標識抗体(No. A13202, Juro Supply, Luzern, Switzerland)を用いる。核を色素H33258(Sigma, Buchs, Switzerland)で染色する。
OGR−1ノックアウトマウスの作成:
相同組換えのためのターゲッティングベクターの作成:マウスOGR−1転写物mCT51440は、マウスゲノムCeleraデータベースで、mCG51257と呼ばれるマウス第12染色体上の遺伝子座と一致することが確認された。相同組換えおよびOGR−1遺伝子のノックアウトのためのターゲッティングベクターの作成のために用いるゲノムDNAを増幅するために、Celera配列情報に従ってプライマーを設計する。2つのフランキングゲノム領域の増幅のためのプライマーの配列および条件は以下の通りである。
方法:細胞質カルシウムの過渡電流を、カルシウムインジケーターFluo−3および蛍光画像プレートリーダー(Molecular Devices/BucherBiotech, Basel, Switzerland)を用いて記録する。OGR1でトランスフェクトされたCCL39細胞に、5mMプロベニシドを含有する完全培地中、37℃で1時間、その色素のアセトキシメチルエステル(2μg/ml)を加えて多剤耐性輸送体を阻害する。その後、細胞を洗浄し、室温で45分間、130mMのNaCl、0.9mMのNaH2PO4、5.4mMのKCl、0.8mMのMgSO4、1.8mMのCaCl2、25mMのグルコース、5mMのHEPES、pH7.6を含有する緩衝塩溶液中で維持する。受容体アンタゴニストを同定するため、試験化合物を、このインキュベーションの最後の15分間に加える。リーダーに移行した後、ベースラインを記録し(Fb)、次に、酸性化をもたらす、適量のより強力なバッファー(20mMのHEPES,pH6)を添加することで細胞を刺激する。あるいは、アゴニスト分子を同定するために、この段階で試験化合物を加える。蛍光の読み取り値Fを、F’=(F−Fb)/Fbで計算してノーマライズする。
ヒトGPR4はゲノムDNAから、プライマーGPR4FとGPR4R(それぞれ、5’ CACC ATG GGC AAC CAC ACG TGG GAG GGC TGC 3’と5’ TCA TTG TGC TGG CGG CAG CAT CTT CAG CTG CA 3’)を用いて増幅する。このPCR反応混合物は、0.2mMのdNTP、1×PCRバッファー(1.5mMのMgCl2、0.5単位のTaq DNAポリメラーゼ、40pmolの各プライマーおよび滅菌水を含有する)を全量50μlで含む。この反応のための鋳型はヒトゲノムDNA(Promega, Southampton, UK)である。PCRは、バイオメトラT3サーモサイクラーを用い、次のサイクル条件:95℃にて2分の変性の後、95℃15秒の変性、60℃15分のアニーリング、および72℃1分の伸長を、35サイクルを用いて行う。72℃で最終伸長を7分行う。この1.1kbのPCR産物を精製し、pcDNA3.1 D/V5−His−TOPO(Invitrogen, Paisley, UK)へクローニングする。
cAMP形成アッセイ:24ウェルプレートで増殖させた密集細胞培養物を、血清フリーDMEM培地中で4時間、[3H]アデニン(100MBq/ml; Amersham, Dubendorf, Switzerland)で標識する。次に、IPアッセイに関して上記したように(実施例2)、緩衝塩溶液中、37℃で細胞をインキュベートする。示されている場合には、ホスホジエステラーゼ阻害剤であるイソブチルメチルキサンチン(IBMX,1mM)を加えてcAMPを蓄積させる。インキュベーション時間は15分である。次に、細胞を氷冷トリクロロ酢酸で抽出し、バッチカラムクロマトグラフィーを用い、遊離アデニンおよびATPからcAMPを分離する(Salmon Y, 1979, Adv. Cycl. Nucleot Res, 10: 35-55)。
実施例8に記載のように、GPR4とcAMPルシフェラーゼリポーターを同時発現する細胞を、アッセイを行う前日に、白色の96ウェルプレートに、ウェル当たり10〜20,000細胞でプレーティングし、5%CO2の存在下、37℃で一晩インキュベートする。細胞を、適宜試験化合物(いずれの自家製または市販の化合物ライブラリーを用いてもよい)を含有し、適当なpHの200μlのHBSバッファー(130mMのNaCl、0.9mMのNaH2PO4、0.8mMのMgSO4、1.8mMのCaCl2、5.4mMのKCl、25mMのグルコース、20mMのHEPES)で一度洗浄する。プレートを、CO2を含まないインキュベーター中、37℃で4〜5時間インキュベートする。4〜5時間後、細胞からバッファーを吸引し、細胞を200μlのHBS pH7.4で一度洗浄する。100μlのHBS pH7.4および100μlのSteady Gloルシフェラーゼ試薬(Promega, Southampton, UK)を加えることで細胞を溶解し、回転プラットフォーム上に激しく回転させながら25〜30分間置く。発光は照度計を用いて測定する。酸性pHバッファー(pH6.8またはpH7.0)では、高い発光luminescenceシグナルが生じる。アンタゴニストはこのアッセイにおいて、例えば酸性pHバッファー(pH6.8またはpH7.0)中で、化合物を含まない対照に比べて発光シグナルが低下することを特徴とする。
RNAの抽出および第一鎖cDNAの合成:RNeasy抽出キット(Qiagen, Crawley, UK)を製造業者のプロトコールに従って用い、細胞からRNAを調製する。用いた細胞は一次ヒト気管支上皮細胞(HBEC)、分化HBEC、一次ヒト肺繊維芽細胞、一次ヒト気管支平滑筋細胞(BSMC)、ヒト末梢血T細胞およびヒト末梢血好中球である。一次ヒト細胞はBiowhittakerから購入するか、またはその旨示されている場合には、標準的な手順に従ってヒト末梢血から単離されるかのいずれかである。第一鎖cDNAは、第一鎖cDNA合成キット(Roche, Lewes, UK)で提供されている試薬およびプロトコールを用い、細胞から単離した全RNAから調製する。
組換え受容体をFITC標識抗myc抗体(No. 132511, Zymed/Stehelin & Cie, Basel, Switzerland)を用いて検出する。原形質膜マーカーアネキシンVを検出するため、アレキサ標識抗体(No. A13202, Juro Supply, Luzern, Switzerland) を用いる。核を色素H33258(Sigma, Buchs, Switzerland)で染色する。
アッセイは、従前に公開されている方法(Frevert CW, et al., J Immunology Methods, 1998, 213:41)に従い、96ウェルプレート形式で行う。細胞バッファーは全て、Invitrogen (Paisley, UK)から入手する。カルセイン−AM色素はMolecular Probes (Invitrogen, Paisley, UK)から入手する。従前に記載されているように(Haslett C, et al., 1985, Am J Path, 119:101)、好中球を単離する。要するに、クエン酸塩添加全血を4%デキストラン−T500と混合し、氷上で30分間放置して赤血球を除去する。顆粒球(PMN)をフィコール−パーク密度勾配遠心分離により末梢血単核細胞から分離する。PMNペレットの赤血球混入は低張ショック溶解により除去する。単離された顆粒球(好中球)を蛍光色素カルセイン−AM(5μg)で標識する。標識した好中球を、次に、DMSO中に希釈した試験化合物(0.001〜1000nM)と混合し、室温で10分間インキュベートする。アゴニスト(pH6.8またはpH7.0に緩衝)を、96ウェル走化性チャンバーの底に入れる。このプレートにポリカーボネートフィルター(5μm)をかけ、細胞およびアンタゴニストを用いる場合には、このフィルターの上に乗せる。5%CO2の加湿インキュベーターにて、37℃で90分間細胞を移動させる。このインキュベーション器官の終了時に、移動した細胞を、マルチウェル蛍光プレートリーダー(Fluoroskan II, Labsystems)を用い、485nm励起および538nm発光で定量した。陽性対照細胞(アンタゴニストを添加しない細胞)は、細胞の最大走化応答を示す。一方、アゴニストを含まない陰性対照(無刺激)、すなわち、pH>=7.4は下のチャンバーに加える。陽性対照と陰性対照の違いは、その細胞の走化活性を表す。このアッセイでは、アンタゴニストは、化合物を含まない対照に比べて、酸性のpHに対する走化応答の低下を特徴とする。
アゴニスト(pH用量反応)曲線を次のように求める:1.2mlクラスターチューブで、顆粒球アリコート(4〜5×105細胞80μl)と10μlのアッセイバッファー(1×PBS、pH7.4、+0.1%BSA)を混合する。サンプルを穏やかに振盪し、37℃で5分間インキュベートする(水浴中)。その後、10μlアッセイバッファーを加えるゼロ対照以外の全ての試験管にアゴニストを加える。サンプルを穏やかに振盪し、水浴中、37℃でさらにインキュベートする。次に、250μlの氷冷、至適化した0.25%CellFix(商標)溶液(Becton Dickinson, Oxford, UK)を各試験管に加えて反応を終わらせ、分析まで細胞の形状変化を維持する。試験管を穏やかに振盪し、FACSCaliburフローサイトメーターでデータを採取する前の10分間、氷上でインキュベートする。まず、FSC/SSCプロットを取った後、その最初のプロットから、ゲートを設けた顆粒球を用い、FSC/FL−2プロットを取った。その後はより高い自己蛍光が得られるので、FL−2プロット上で好酸球から好中球を識別する。
血管新生の可能性のある治療標的を、内皮細胞の認知されているマーカー遺伝子と相関した発現を示す遺伝子を選択することにより同定する。このマーカー遺伝子セットとしては、限定されるものではないが、CDH5、VWF、KDR、TIE、TEK、PTPRB、ANGPT2(the Human GVenomeOrganisation/Gene Nomenclature Committee (HUGO/HGNC)によって承認された遺伝子記号)を含む。

実施例8に記載のように、TDAG8とcAMPルシフェラーゼリポーターを同時発現する細胞を、白色の96ウェルプレートに1ウェル当たり10〜20,000細胞でプレーティングする。アッセイを行う前日、培地を血清フリーDMEMに交換し、細胞を、5%CO2の存在下、37℃で一晩インキュベートする。細胞を、適当なpHの100μlのHBSバッファー(130mMのNaCl、0.9mMのNaH2PO4、0.8mMのMgSO4、1.8mMのCaCl2、5.4mMのKCl、25mMのグルコース、20mMのHEPES)で一度洗浄する。次に、試験化合物(例えば、いずれの自家製または市販の化合物ライブラリーからのものでもよい)を含有するこの適当なHBS100μlを細胞に加える。プレートを、CO2を含まないインキュベーター中、37℃で4〜5時間インキュベートする。4〜5時間後、細胞からバッファーを吸引し、細胞を100μlのPBSで一度洗浄する。15μlの溶解バッファー(5mMのトリスリン酸、4mMのDTT、0.4mMのEDTA、2%のグリセロール、0.2%のトリトンX−100)を加え、1分間振盪し、室温で20分間インキュベートすることで細胞を溶解する。50μlの基質バッファー(E1483, Promega, Wallisellen, Switzerland)の自動添加時の光シグナルの測定を、同時基質分注・シグナル測定を備えたLabsystems Luminoskan RS (BioConcept, Allschwill)で行う。このアッセイにおけるアゴニストは発光の増強をもたらし、一方、アンタゴニストは発光の低下をもたらす。例えば、アンタゴニストはこのアッセイにおいて、例えば酸性pHバッファー(pH6.8またはpH7.0)中、またはヒトTDAG8の最大の半分の活性化において、化合物を含まない対照に比べて発光シグナルが低下することを特徴とする。この受容体を安定発現するCCL39 CRE−luc細胞のヒトTDAG8によるルシフェラーゼ誘導の最大の半分の活性化はpH7.28+/−0.02(平均値+/−標準誤差;N=7)で見られる。
ヒト一次末梢血単核細胞(PBMNC;単球、リンパ球およびその他の血液細胞種からなる)を、10%ウシ胎児血清を添加したMEMalpha培地で培養する。破骨細胞の分化を誘導するため、いくつかの培養物にM−CSF(25ng/ml,R&D Systems, Abingdon, UK)、RANKリガンド(50ng/ml,Insight Biotechnology, Wembley, UK)、TGFβ1(5ng/ml,R&D Systems, Abingdon, UK)、デキサメタゾン(1μM,Sigma, Buchs, Switzerland)を含有するサイトカインカクテルを添加する。処理後17日で成熟破骨細胞が見られる。
ヒト一次末梢血単核細胞(PBMNC)を、10%ウシ胎児血清を添加したMEMalpha培地で培養する。破骨細胞の分化を誘導するため、培養物にM−CSF(25ng/ml,R&D Systems, Abingdon, UK)、RANKリガンド(50ng/ml,Insight Biotechnology, Wembley, UK)、TGFβ1(5ng/ml,R&D Systems, Abingdon, UK)、デキサメタゾン(1μM,Sigma, Buchs, Switzerland)を含有するサイトカインカクテルを添加する。破骨細胞の分化に対するpHの影響を検討するため、分化中、細胞を種々のpHの培地で維持し、酒石酸耐性酸性ホスファターゼに対して染色を行った後、顕微鏡下で成熟破骨細胞を計数する。
配列はGCGプログラム「Gap」(Devereux J, et al., 1984, Nucl Acids Res, 12:387-395.)を用いてアラインする。用いたタンパク質配列は、Refseqデータベース受託番号として

である。

Claims (15)
- プロトンホメオスタシスのバランスがとれていない疾病および医学的状態のための薬剤の開発における、単離されたプロトン感受性GPCRポリペプチドの使用。
- プロトンホメオスタシスのバランスがとれていない疾病および医学的状態のための薬剤の開発における、単離されたプロトン感受性GPCRポリペプチドの使用であって、該ポリペプチドが
(a)ヒトOGR1(受託番号:NM_003485.1)、ラットOGR1(受託番号:XM_234483)、マウスOGR1(受託番号:NM_175493)、ウシOGR1(受託番号:NM_174329)、好ましくは、ヒトOGR1(受託番号:NM_003485.1)、ヒトGPR4(受託番号:NM_005282)、マウスGPR4(受託番号:NM_175668)、ヒトTDAG8(受託番号:NM_003608)およびマウスTDAG8(受託番号:NM_008152)のポリヌクレオチド配列を含むポリヌクレオチドによりコードされる単離されたポリペプチド;
(b)配列番号1のポリペプチド配列の少なくとも20%の同一性を有するポリペプチド配列を含む単離されたプロトン感受性GPCRポリペプチド;
(c)配列番号3のポリペプチド配列の少なくとも20%の同一性を有するポリペプチド配列を含む単離されたプロトン感受性GPCRポリペプチド;
(d)配列番号4のポリペプチド配列と少なくとも20%の同一性を有するポリペプチド配列を含む単離されたプロトン感受性GPCRポリペプチド;
(e)配列番号1、配列番号3または配列番号4のポリペプチド配列を含む単離されたポリペプチド;
(f)配列番号1のポリペプチド配列と少なくとも20%の同一性を有する、単離されたプロトン感受性GPCRポリペプチド;
(g)配列番号3のポリペプチド配列と少なくとも20%の同一性を有する、単離されたプロトン感受性GPCRポリペプチド;
(h)配列番号4のポリペプチド配列と少なくとも20%の同一性を有する、単離されたプロトン感受性GPCRポリペプチド;
(i)配列番号1、配列番号3、または配列番号4のポリペプチド配列;および
(l)CCL39ハムスター繊維芽細胞においてpH依存性リン酸イノシトール形成を示すか、またはCHOK1 CRE−luc細胞もしくはCCL39 CRE−luc細胞においてcAMPルシフェラーゼリポーターアッセイでpH依存性シグナルを示す(a)〜(i)のポリペプチド
からなる群から選択される、使用。 - 該ポリペプチドが配列番号1、配列番号3または配列番号4のポリペプチド配列を含む、請求項1または2に記載の単離されたポリペプチドの使用。

(a)ヒトOGR1(受託番号:NM_003485.1)、ラットOGR1(受託番号:XM_234483)、マウスOGR1(受託番号:NM_175493)、ウシOGR1(受託番号:NM_174329)、好ましくは、ヒトOGR1(受託番号:NM_003485.1)、ヒトGPR4(受託番号:NM_005282)、マウスGPR4(受託番号:NM_175668)、ヒトTDAG8(受託番号:NM_003608)およびマウスTDAG8(受託番号:NM_008152)のポリヌクレオチド配列を含む単離されたポリヌクレオチド;
(b)配列番号1のポリペプチド配列と少なくとも20%の同一性を有するプロトン感受性GPCRポリペプチド配列をコードする単離されたポリヌクレオチド;
(c)配列番号3のポリペプチド配列と少なくとも20%の同一性を有するプロトン感受性GPRCポリペプチド配列をコードする単離されたポリヌクレオチド;
(d)配列番号4のポリペプチド配列と少なくとも20%の同一性を有するプロトン感受性GPRCポリペプチド配列をコードする単離されたポリヌクレオチド;
(f)ヒトOGR1(受託番号:NM_003485.1)、ラットOGR1(受託番号:XM_234483)、マウスOGR1(受託番号:NM_175493)、ウシOGR1(受託番号:NM_174329)、好ましくはヒトOGR1(受託番号:NM_003485.1)、ヒトGPR4(受託番号:NM_005282)、マウスGPR4(受託番号:NM_175668)、ヒトTDAG8(受託番号:NM_003608)およびマウスTDAG8(受託番号:NM_008152)のポリヌクレオチド配列を含む単離されたポリヌクレオチド;

(h)CCL39ハムスター繊維芽細胞においてpH依存性リン酸イノシトール形成を示すか、またはCHOK1 CRE−luc細胞もしくはCCL39 CRE−luc細胞においてcAMPルシフェラーゼリポーターアッセイでpH依存性シグナルを示す(a)〜(g)のポリヌクレオチド
からなる群から選択される、使用。 - プロトンホメオスタシスのバランスがとれていない疾病および医学的状態の予防および/または処置のための薬剤の製造を目的とする、請求項1〜4のいずれか一項に記載のポリペプチドと特異的に結合する抗体の使用;
プロトンホメオスタシスのバランスがとれていない疾病および医学的状態の予防および/または処置のための、請求項1〜4のいずれか一項に記載のポリペプチドと特異的に結合する抗体を含む医薬組成物;または
プロトンホメオスタシスのバランスがとれていない疾病および医学的状態の予防および/または処置方法であって、請求項1〜4のいずれか一項に記載のポリペプチドと特異的に結合する、有効量の抗体をそのような予防および/または処置を必要とする被験体に投与することを含む方法。 - 請求項1〜4のいずれか一項に記載のポリペプチドのプロトン感受活性と拮抗する化合物を同定するためのスクリーニング方法。
- 請求項1〜4のいずれか一項に記載のポリペプチドのプロトン感受活性を促進する化合物を同定するためのスクリーニング方法。
- 請求項1〜4のポリペプチドの機能または発現レベルを刺激または阻害する化合物を同定するためのスクリーニング方法であって、
(a)候補化合物と直接的または間接的に結合している標識の手段により、候補化合物と、該ポリペプチド(または該ポリペプチドを発現する細胞または膜)またはその融合タンパク質との結合を定量的または定性的に測定または検出すること;
(b)標識競合物の存在下で、候補化合物と、該ポリペプチド(または該ポリペプチドを発現する細胞または膜)またはその融合タンパク質との結合の競合を測定すること;
(c)該候補化合物が該ポリペプチドの活性化または阻害によって生じるシグナルをもたらすかどうかを、該ポリペプチドを発現する細胞または細胞膜に適当な検出系を用いて試験すること;または
(d)細胞における該ポリペプチドをコードするmRNA、または該ポリペプチドの産生に対する候補化合物の作用を、例えばELISAアッセイを用いて検出すること
からなる群から選択される方法を含む、方法。 - プロトンホメオスタシスのバランスがとれていない疾病および医学的状態を予防および/または処置する方法であって、そのような予防および/または処置を必要とする被験体に、請求項7の方法から得ることができる有効量のアンタゴニストを投与することを含む、方法。
- プロトンホメオスタシスのバランスがとれていない疾病および医学的状態を予防および/または処置する方法であって、そのような予防および/または処置を必要とする被験体に、請求項8の方法から得ることができる有効量のアゴニストを投与することを含む、方法。
- プロトンホメオスタシスのバランスがとれていない疾病および医学的状態を予防および/または処置するための、請求項7の方法から得ることができるアンタゴニストを含む医薬組成物。
- プロトンホメオスタシスのバランスがとれていない疾病および医学的状態を予防および/または処置するための、請求項8の方法から得ることができるアゴニストを含む医薬組成物。
- 請求項1〜4のいずれか一項に記載のポリペプチドに対する抗体を含む診断キット。
- プロトンホメオスタシスのバランスがとれていない疾病および医学的状態の予防および/または処置のための医薬製剤を含む診断キットであって、該医薬製剤が請求項1〜4のいずれか一項に記載のポリペプチドに対する抗体を含む診断キット。
