Abstract
Description
本発明は、カノーラタンパク質単離物の調製および養殖におけるその使用に関する。
カノーラタンパク質単離物は、前述の米国特許出願に一般的に記載されているように、バッチプロセスまたは連続プロセスまたは半連続プロセスの何れかによって、カノーラ油糧種子粗粉から単離することができる。
実施例1:
本実施例は、カノーラタンパク質単離物を提供するための、本発明の手順を例示する。
本実施例では、カノーラタンパク質単離物の実験室スケールのサンプルの調製について述べる。
本実施例では、実施例1および2で生成されたカノーラタンパク質単離物のタンパク質分析を提供する。
本開示を要約すると、養殖の用途を含めた多くの用途を有するカノーラ油糧種子タンパク質単離物の、新規の調製方法が提供される。本発明の範囲内で変更が可能である。
Claims (39)
- カノーラタンパク質単離物を調製する方法であって、
(a)カノーラ油糧種子粗粉を抽出して、前記カノーラ油糧種子粗粉中のタンパク質を可溶化し、タンパク質含有量が約5から約40g/L、pHが約5から約6.8であるタンパク質水溶液を形成すること、
(b)前記カノーラ油糧種子粗粉の抽出残渣から前記タンパク質水溶液を分離すること、
(c)選択膜技術を用いることによりイオン強度を実質上一定に保ちながら、前記タンパク質水溶液のタンパク質濃度を少なくとも約50g/Lまで増加させて濃縮したタンパク質溶液を得ること、および
(d)前記濃縮したタンパク質溶液を乾燥させて、タンパク質含有量が乾燥重量に基づき少なくとも約90wt%(N × 6.25)のカノーラタンパク質単離物を得ることを含む方法。 - 前記カノーラタンパク質単離物が少なくとも約100wt%(N × 6.25)d.h.のタンパク質含有量を有する、請求項1に記載の方法。
- 前記カノーラタンパク質単離物が、2Sカノーラタンパク質約25から約55wt%、7Sカノーラタンパク質約45から約75wt%、12Sカノーラタンパク質約0から約15wt%であるカノーラタンパク質プロフィールを有する、請求項1に記載の方法。
- 前記カノーラタンパク質単離物が、2Sカノーラタンパク質約40から約50wt%、7Sカノーラタンパク質約50から約60wt%、12Sカノーラタンパク質約1から約5wt%であるカノーラタンパク質プロフィールを有する、請求項2に記載の方法。
- バッチ式で実施され、前記カノーラ油糧種子粗粉の前記抽出を、イオン強度が少なくとも約0.05、pHが約5から約6.8の水性塩溶液を用いて、少なくとも約5℃の温度で実施する、請求項1に記載の方法。
- 前記塩溶液が約0.1から約0.6のイオン強度を有する、請求項5に記載の方法。

- 連続式で実施され、前記抽出ステップが、
(i)前記カノーラ油糧種子粗粉を、イオン強度が少なくとも約0.5でpHが約5から約6.8の水性塩溶液と、約5°から約65℃の温度で連続的に混合し、
(ii)約10分までの時間、前記カノーラ油糧種子粗粉からタンパク質を抽出しながら、前記混合物をパイプで連続的に搬送し、タンパク質濃度が約5から約40g/Lのタンパク質水溶液を形成することによって実施される、請求項1に記載の方法。 - 前記塩溶液が約0.1から約0.8のイオン強度を有する、請求項12に記載の方法。
- 前記塩溶液が約5.3から約6.2のpHを有する、請求項12に記載の方法。
- 前記混合ステップの前記水性塩溶液中の油糧種子粗粉の濃度が約5から約15wt%である、請求項12に記載の方法。
- 前記温度が少なくとも約35℃である、請求項12に記載の方法。
- 前記タンパク質水溶液が約10から約30g/Lのタンパク質含有量を有する、請求項12に記載の方法。
- 前記水性塩溶液が酸化防止剤を含む、請求項12に記載の方法。
- カノーラ種子粗粉の残渣からのタンパク質水溶液の前記分離に続いて、タンパク質水溶液を色素除去ステップにかける、請求項1に記載の方法。
- 前記色素除去ステップをタンパク質水溶液のダイアフィルトレーションによって実施する、請求項19に記載の方法。
- 前記色素除去ステップを、色素吸着剤をタンパク質水溶液と混合し、続いて色素吸着剤をタンパク質水溶液から除去することによって実施する、請求項19に記載の方法。
- 前記色素吸着剤が粉末状活性炭である、請求項21に記載の方法。
- 前記油糧種子粗粉を水で抽出し、それに続いて、得られるタンパク質水溶液に塩を添加して、少なくとも約0.05のイオン強度を有するタンパク質水溶液を得る、請求項1に記載の方法。
- 前記濃縮ステップを限外ろ過によって実施して、タンパク質濃度が少なくとも約200g/Lの濃縮タンパク質溶液を得る、請求項1に記載の方法。
- 前記濃縮したタンパク質溶液を、抽出ステップに用いたと同じイオン強度を有する水性塩溶液を使用したダイアフィルトレーションにかける、請求項1に記載の方法。
- 前記ダイアフィルトレーションを、約2から約20容のダイアフィルトレーション溶液を用いて実施する、請求項25に記載の方法。
- 前記ダイアフィルトレーションを、約5から約10容のダイアフィルトレーション溶液を用いて実施する、請求項26に記載の方法。
- 前記ダイアフィルトレーションステップの少なくとも一部を酸化防止剤の存在下で実施する、請求項25に記載の方法。
- 前記濃縮したタンパク質溶液を、色素除去ステップにかける、請求項1に記載の方法。
- 前記色素除去ステップを粒状活性炭またはポリビニルピロリドンを用いて実施する、請求項29に記載の方法。
- 前記濃縮したタンパク質溶液を低温殺菌ステップにかける、請求項1に記載の方法。
- 前記低温殺菌ステップを、濃縮したタンパク質溶液を約55°から約70℃の温度に約10から約15分間加熱することで実施する、請求項31に記載の方法。
- 乾燥させたカノーラタンパク質単離物を、養殖に使用するための飼料組成物として処方することを含む、請求項1に記載の方法。
- 前記飼料組成物をサケ科の魚に給餌するために処方する、請求項33に記載の方法。
- 請求項1の方法によって作製されたカノーラタンパク質単離物を含む、養殖用の飼料組成物。
- 前記飼料組成物をサケ科の魚に給餌するために処方する、請求項35に記載の飼料組成物。
- タンパク質含有量が少なくとも約90wt%で、かつ、2Sカノーラタンパク質約25から約55wt%、7Sカノーラタンパク質約45から約75wt%、12Sカノーラタンパク質約0から約15wt%であるカノーラタンパク質プロフィールを有するカノーラタンパク質単離物を含む、養殖用の飼料組成物。
- タンパク質含有量が少なくとも約100wt%で、かつ、2Sカノーラタンパク質約40から約50wt%、7Sカノーラタンパク質約50から約60wt%、12Sカノーラタンパク質約1から約5wt%であるカノーラタンパク質プロフィールを有する前記カノーラタンパク質単離物を含む、請求項37に記載の飼料組成物。
- サケ科の魚に給餌するために処方される請求項37に記載の飼料組成物。
