Description
本発明は、カノーラタンパク質単離物の調製および養殖におけるその使用に関する。
(a)カノーラ油糧種子粗粉を抽出して、該カノーラ油糧種子粗粉中のタンパク質を可溶化し、タンパク質含有量が5〜40g/Lであり、pHが5〜6.8であるタンパク質水溶液を形成すること、
(b)該カノーラ油糧種子粗粉の抽出残渣から該タンパク質水溶液を分離すること、
(c)選択膜技術を用いてイオン強度を実質上一定に保ちながら、該タンパク質水溶液のタンパク質濃度を少なくとも50g/Lまで増加させて濃縮したタンパク質溶液を得ること、および
(d)該濃縮したタンパク質溶液を乾燥させて、タンパク質含有量が乾燥重量に基づき少なくとも90wt%(N×6.25)のカノーラタンパク質単離物を得ること
からなることを特徴とする方法である。
カノーラタンパク質単離物は、前述の米国特許出願に一般的に記載されているように、バッチプロセスまたは連続プロセスまたは半連続プロセスの何れかによって、カノーラ油糧種子粗粉から単離することができる。
実施例1:
本実施例は、カノーラタンパク質単離物を提供するための、本発明の手順を例示する。
本実施例では、カノーラタンパク質単離物の実験室スケールのサンプルの調製について述べる。
本実施例では、実施例1および2で生成されたカノーラタンパク質単離物のタンパク質分析を提供する。
本開示を要約すると、養殖の用途を含めた多くの用途を有するカノーラ油糧種子タンパク質単離物の、新規の調製方法が提供される。本発明の範囲内で変更が可能である。
Claims (21)
- カノーラタンパク質単離物を調製する方法であって、
(a)カノーラ油糧種子粗粉を抽出して、該カノーラ油糧種子粗粉中のタンパク質を可溶化し、タンパク質含有量が5〜40g/Lであり、pHが5〜6.8であるタンパク質水溶液を形成すること、
(b)該カノーラ油糧種子粗粉の抽出残渣から該タンパク質水溶液を分離すること、
(c)選択膜技術を用いてイオン強度を実質上一定に保ちながら、該タンパク質水溶液のタンパク質濃度を少なくとも50g/Lまで増加させて濃縮したタンパク質溶液を得ること、および
(d)該濃縮したタンパク質溶液を乾燥させて、タンパク質含有量が乾燥重量に基づき少なくとも90wt%(N×6.25)のカノーラタンパク質単離物を得ること
からなることを特徴とする方法。 - 前記カノーラタンパク質単離物が少なくとも100wt%(N×6.25)のタンパク質含有量を有する、請求項1に記載の方法。
- 前記カノーラタンパク質単離物が、
2Sカノーラタンパク質 25〜55wt%、
7Sカノーラタンパク質 45〜75wt%、及び
12Sカノーラタンパク質 0〜15wt%
であるカノーラタンパク質プロフィールを有する、請求項1または2に記載の方法。 - 前記カノーラタンパク質単離物が、
2Sカノーラタンパク質 40〜50wt%、
7Sカノーラタンパク質 50〜60wt%、及び

- 前記水性塩溶液が0.1〜0.6のイオン強度を有する、請求項5に記載の方法。
- 前記水性塩溶液が5.3〜6.2のpHを有する、請求項5または6に記載の方法。
- 前記工程(a)におけるカノーラ油糧種子粗粉の抽出を、前記水性塩溶液を10〜30分間撹拌しながら実施する、請求項5〜7のいずれか1項に記載の方法。
- 前記工程(a)におけるカノーラ油糧種子粗粉の抽出中の前記水性塩溶液中のカノーラ油糧種子粗粉の濃度が5〜15wt%である、請求項5〜8のいずれか1項に記載の方法。
- 前記工程(a)におけるカノーラ油糧種子粗粉の抽出から得られる前記タンパク質溶液が、10〜30g/Lのタンパク質濃度を有する、請求項5〜9のいずれか1項に記載の方法。
- 前記水性塩溶液が酸化防止剤を含む、請求項5〜10のいずれか1項に記載の方法。
- 連続式で実施され、前記抽出ステップが、
(i)前記カノーラ油糧種子粗粉を、イオン強度が少なくとも0.05でpHが5〜6.8の水性塩溶液と、5〜65℃の温度で連続的に混合し、
(ii)10分までの時間、該カノーラ油糧種子粗粉からタンパク質を抽出しながら、該混合物をパイプで連続的に搬送し、タンパク質含有量が5〜40g/Lのタンパク質水溶液を形成することによって実施される、請求項1〜4のいずれか1項に記載の方法。 - 前記水性塩溶液が0.1〜0.6のイオン強度を有する、請求項12に記載の方法。
- 前記水性塩溶液が5、3〜6.2のpHを有する、請求項12または13に記載の方法。
- 前記工程(i)の混合における前記水性塩溶液中のカノーラ油糧種子粗粉の濃度が5〜15%w/vである、請求項12〜14のいずれか1項に記載の方法。
- 前記工程(i)の混合における温度が少なくとも35℃である、請求項12〜15のいずれか1項に記載の方法。
- 前記工程(ii)において、前記タンパク質水溶液が10〜30g/Lのタンパク質含有量を有する、請求項12〜16のいずれか1項に記載の方法。
- 前記水性塩溶液が酸化防止剤を含む、請求項12〜17のいずれか1項に記載の方法。
- 前記カノーラ油糧種子粗粉を水で抽出し、それに続いて、得られたタンパク質水溶液に塩を添加して、少なくとも0.05のイオン強度を有するタンパク質水溶液を得る、請求項1〜4のいずれか1項に記載の方法。
- 前記塩を添加した後のタンパク質水溶液が、0.1〜0.6のイオン強度を有する、請求項19に記載の方法。
- 前記濃縮ステップを限外ろ過によって実施して、タンパク質濃度が少なくとも200g/Lの濃縮タンパク質溶液を得る、請求項1〜20のいずれか1項に記載の方法。
