Claims (22)
- 以下の(a)〜(i)を特徴とするタンパク質単離物の製造方法;
(a)油料種子を粉砕して油と油料種子粕を生成させ、
(b)その油料種子粕から溶媒抽出して残留油を回収し、
(c)抽出した油料種子粕から脱溶媒された油料種子粕を与えるために、真空下で15℃〜50℃の温度で溶媒を除去し、
(d)前記脱溶媒された油料種子粕中のタンパク質の可溶化を生じさせてpHが5ないし6.8の水性タンパク質溶液を形成するために抽出し、
(e)残留油料種子から水性タンパク質溶液を分離し、
(f)選択性膜技術を使用することによって、イオン強度を実質的に一定に保ちながら、前記水性タンパク質溶液の濃度を増加させて、濃縮されたタンパク質溶液を得、
(g)前記濃縮されたタンパク質溶液を、温度が15℃未満の冷水中に稀釈して、少なくとも部分的にミセルの形で分離したタンパク質粒子を形成させ、
(h)タンパク質ミセルを沈降させて、無定形で、粘着性のある、ゼラチン状の、グルテン様タンパク質ミセル集塊物を形成させ、そして
(i)乾燥重量ベースで少なくとも90重量%(N×6.25)のタンパク質含量を有する、タンパク質ミセル集塊物を上澄液から回収する。 - 前記方法がバッチモードで達成され、前記油料種子粕の抽出が、イオン強度が少なくとも0.10で、pHが5〜6.8であり、そして前記水性タンパク質溶液が5〜40g/Lのタンパク質含量を有することを特徴とする、請求項1に記載の方法。
- 前記油料種子粕の抽出が、イオン強度が0.15〜0.6、pHが5.3〜6.2であり、前記水性タンパク質溶液が10〜30g/Lのタンパク質含量であることを特徴とする、請求項2に記載の方法。
- 前記油料種子粕の抽出が、前記塩水溶液の攪拌を伴って10〜30分間、前記抽出ステップの間の前記塩水溶液中の油糧種子粕の濃度が5〜15%w/vで行われることを特徴とする、請求項2または3に記載の方法。
- 前記プロセスが連続式で実施され、前記抽出ステップが
(i)油料種子粕を、イオン強度が少なくとも0.10、pHが5〜6.8の塩水溶液と、温度が5〜65℃で連続的に混合し、
(ii)その油料種子粕からタンパク質を抽出してタンパク質の含量が5〜40g/Lのタンパク質の水溶液を形成するため10分までの時間、前記混合物をパイプを通して連続的に搬送すること、
によって実施されることを特徴とする請求項1に記載の方法。 - 前記塩水溶液が、イオン強度が0.15〜0.8、pHが5.3〜6.2であり、温度が少なくとも35℃であり、および前記タンパク質水溶液がタンパク質の含量が5〜40g/Lであり、塩水溶液中の油糧種子粕の濃度が5〜15w/vであることを特徴とする、請求項5に記載の方法。
- 前記油料種子粕の前記抽出が、イオン強度が少なくとも0.10、pHが3〜5または6.8〜9.9を有する塩の水溶液を用いて実施され、次いで前記残渣油料種子粕からの水性タンパク質溶液の分離の後に、タンパク質水溶液のpHが5〜6.8に調節されることを特徴とする請求項1に記載の方法。
- 前記油料種子粕がカノーラ油料種子粕であり、残渣カノーラ種子粕からの水性タンパク質溶液の前記分離の後に、水性タンパク質溶液が色素除去ステップを行うことを特徴とする、請求項1〜7のいずれかに記載の方法。
- 前記色素除去ステップが、
(a)水性タンパク質溶液の限外濾過によって実施されるか、または
(b)色素吸着剤と水性タンパク質溶液と混合し、次いで水性タンパク質溶液から色素吸収剤を分離する
ことによって実施されることを特徴とする請求項8に記載の方法。 - 前記油料種子粕が水で抽出され、次いでその水性タンパク質溶液に塩を加えてイオン強度が少なくとも0.10を有する水性タンパク質溶液を得ることを特徴とする請求項1に記載の方法。
- 前記濃縮ステップが、タンパク質含量が少なくとも200g/Lを有する濃縮タンパク質溶液を得るために限外濾過によって実施されることを特徴とする請求項1〜10のいずれかに記載の方法。
- 前記濃縮タンパク質溶液が、濃縮タンパク質溶液の粘度を下げるため、少なくとも20℃の温度まで、しかしそれより高いと濃縮タンパク質溶液の温度がミセルの形成を不可能にする温度をこえることのないように加温されることを特徴とする請求項11に記載の方法。
- 前記方法がバッチベースで行われ、前記濃縮されたタンパク質溶液が、溶液を所望の希釈度に達するために必要な体積を有し、温度が好ましくは10℃未満の水の中に加えることにより、15倍以下まで希釈されることを特徴とする請求項1〜4および6〜12のいずれかに記載の方法。
- 前記方法が連続ベースで行われ、前記濃縮されたタンパク質溶液が、10℃未満の温度の冷水と連続的に混合されて15倍以下の濃縮タンパク質溶液を与えることを特徴とする請求項1および5〜12のいずれかに記載の方法。
- 回収されたタンパク質ミセル塊がタンパク質粉末に乾燥される請求項1〜14のいずれかに記載の方法。
- 前記油料種子粕がカノーラ種子粕であり、そして、上澄液からタンパク質ミセル塊が回収された後、上澄がバッチ、セミ連続または連続ベースで操作されて、さらなるタンパク質単離物を回収することを特徴とする請求項1〜15のいずれかに記載の方法。
- 前記上澄液が
(a)上澄を100〜400g/Lのタンパク質濃度まで濃縮し、そして濃縮された上澄を乾燥する、
(b)上澄を100〜400g/Lのタンパク質濃度まで濃縮し、濃縮された上澄を回収したタンパク質ミセル塊と混合し、その混合物を乾燥する、または
(c)上澄を100〜400g/Lまで濃縮し、その濃縮された上澄と回収されたタンパク質ミセル塊の少なくとも一部とを混合し、そして得られた混合物を乾燥する、
ことにより操作されることを特徴とする、請求項16に記載の方法。 - 前記タンパク質濃度が、200〜300g/Lである、請求項17に記載の方法。
- 前記稀釈、沈降および回収ステップの代替法として、濃縮タンパク質溶液を透析してその塩含量を低下させ、タンパク質ミセルを形成させ、そして透析された濃縮タンパク質溶液から乾燥重量基準で少なくとも100重量%(N×6.25)のタンパク質含量を有するタンパク質単離物を、透析された濃縮タンパク質を乾燥することによって回収することを特徴とする、請求項1に記載の方法。
- 前記油料種子粕が、カノーラ油料種子粕、菜種粕、またはマスタード種子粕であることを特徴とする、請求項1〜19のいずれかに記載の方法。
- 前記溶媒除去ステップが、温度15〜20℃における真空下での空気−脱溶媒法によって実施されることを特徴とする、請求項1〜20のいずれかに記載の方法。
- 前記タンパク質ミセル塊が、乾燥重量基準で少なくとも100重量%(N×6.25)のタンパク質含量を有することを特徴とする、請求項1〜21のいずれかに記載の方法。
