本発明は、患者での転移を検出するための方法を提供する。いくつかの実施形態では、この方法は、造影剤に連結させられる抗体、プローブ、および低分子を含んでいる組成物に投与して、上記患者での前記造影剤の局在を検出することを含む。いくつかの実施形態では、造影剤に結合した抗EphA2抗体は1種類以上の転移を検出するために、患者に投与される。標識抗体は細胞上で高密度のレセプターに結合し、それによって腫瘍細胞上で蓄積する。標準の撮像技術を使用して、腫瘍の部位を検出することができる。

いくつかの実施形態では、本発明は、転移またはSREを調整する方法を提供するもので、EphA2治療に感受性のある患者を最初に同定することを含み、この方法は、造影剤に連結させられる抗体、プローブ、プライマーまたはオリゴヌクレオチドを含んでいる組成物を、それを必要としている患者に投与する工程と、患者で遺伝子または遺伝子産物の証拠の有無を検出することが挙げられる。いくつかの実施形態では、上記遺伝子がEphA2である。患者でEphA2発現の証拠が存在することは、EphA2治療の候補である患者であることを示す。また、患者のEphA2発現の証拠が示されなければ、EphA2治療の候補ではない患者であることを示す。この方法は、患者に組成物を投与する工程と、造影剤に連結したEphA2オリゴヌクレオチドまたは抗体を比較し、患者のSREまたは転移を示す臨床指標の有無を検出する工程とを有する。臨床指標が存在することは、SREまたは転移がEphA2治療の候補である患者であることを示し、患者の臨床指標の証拠が示されなければ、EphA2治療の候補ではない患者を示す。
いくつかの実施形態では、本発明は、SREおよび/または転移に関連する遺伝子または遺伝子産物を撮像および/または検出するためのキットを提供する。いくつかの実施形態では、遺伝子はEphA2である。本発明のキットは、本発明の方法を実行するために、指示書と同様に、検出可能な抗体、低分子、オリゴヌクレオチド、デコイレセプター、ミメティック、またはプローブを含む。任意に、キットは以下の一つ以上を含むものであってもよい。すなわち、コントロール(正および/または負)、コントロールのための容器、写真、または正および/負の結果の典型的な例の記述。
正常ヒト間充織幹細胞(MSC, BioWhittaker(登録商標)からのカタログ#PT−2501)の1バイアル(750,000細胞/バイアル)を、10−7Mのデキサメタゾン、0.05mMのリン酸アスコルビン酸塩、10mMのβグリセロリン酸塩を有する完全なMSCGMを含む骨形成培地に播種し、一晩、細胞培養皿に付着させた。幹細胞は、10,000細胞/cm2の密度で播種した。翌日、サブコンフルエントな癌細胞をトリプシン処理し、計数するために、骨形成培地中に再懸濁した。癌細胞を、骨形成培地で40,000細胞/mlに希釈した。間充織幹細胞からの培地を除去し、幹細胞が前日有したのと同じ密度(10,000細胞/cm2)で癌細胞を加えた。
(アルカリホスファターゼ染色)
培地は、培養細胞から除去した。この細胞を、10%の中性緩衝ホルマリンで3〜5分間固定した。3〜5分後に、ホルマリンを除去し、リン酸緩衝食塩水(PBS)で置換した。次いで、このPBSを、アルカリホスファターゼ染色液で置換した。アルカリホスファターゼ染色液は、Fast Blue RR(Sigma(登録商標)、カタログ#FBS−25)の1カプセル(12mg)を48mlの水に加えることによって調製した。この溶液を混合した後に、溶液を濾過した。AS−MXリン酸ナフトール(Sigma(登録商標)、カタログ#85−5)の2mlを、この濾過済み溶液に加えた。この染色液を、37℃で約30分間インキュベート可能にし、次いで、PBSですすぎ洗いした。次いで、アルカリホスファターゼ染色液を、顕微鏡の下で視覚化した。
この細胞層を、PBS(Mg及びCa無しの)で洗った。100μlの細胞消化緩衝液(アッセイ緩衝液[PH9.0で1.5Mのトリス、1mMのZnC12、1mM のMgC12]の1:10希釈液+1%のトリトンX−100(登録商標))を、96穴のプレートの各々の穴に加えた。次いで、この消化緩衝液を、37℃で約30分間インキュベートした。次いで、溶解物を新しい96穴のプレートまで移送した。50μlの基質(10mlのH2O中に1カプセルのSigma(登録商標) 104)、50μlのアルカリホスファターゼ緩衝液(PH9.0で1.5Mのトリス、1mMのZnC12、1mMのMgC12)、及び10μlの細胞溶解物を、新しい96穴のプレートに加えた。混合物を37℃で15分間インキュベートした。反応を100μlのNaOHの追加により停止させた。次いで、O.D.を、410ナノメートルで読み込んだ。
細胞を10%の中性緩衝ホルマリンで5分間固定した。5分後に、固定液を除去し、細胞を脱イオン水ですすぎ洗いした。細胞に、5%の水性の硝酸銀溶液を加えた。次いで、この細胞を、細胞培養皿の蓋を除去して、1時間紫外線光に曝露した。反応は、室温で2〜3分間5%の水性のチオ硫酸ナトリウムを追加して停止させた。次いで、細胞を洗って、室温で保存した。
間充織幹細胞を、骨形成培地中、10,000細胞/cm2の密度で96穴の皿中に播種し、37℃で一晩付着させた。同時に、癌細胞(PC3、LNCaP、またはMDA231)をサブコンフルエントに播種した。翌日、癌細胞をトリプシン処理し、40,000細胞/mlの濃度で、骨形成培地中に再懸濁した。次いで、培地を、宿主細胞を含む96穴のプレートから除去した。次いで、癌細胞を10,000細胞/cm2の密度で、宿主細胞培養に加えた。アルカリホスファターゼ活性を、4日目に測定した。カルシウム沈着は、2〜3日ごと培地を変えて11〜14日間細胞をインキュベートすることにより分析した。カルシウム沈着は、11〜14日目に、フォン・コッサ染色液を用いて視覚化した。
間充織幹細胞(MSC)を、単独、或いはPC3(前立腺癌細胞)またはMDA231(胸部癌細胞)の存在下、骨形成培地中で4日間、培養した。4日目に、細胞を固定し、アルカリホスファターゼ活性のために染色した(図1、及び図2)。図1で分かるように、PC3の存在下でのみ、宿主細胞が刺激されて骨芽細胞になった。それは暗い汚れの存在によって示されている。アルカリホスファターゼ活性も、細胞溶解物を生成して測定した。図2で分かるように、アルカリホスファターゼ活性は、PC3細胞の存在下で、細胞が存在しない時よりかなり大きい。
骨転移の際、その発現が調整される遺伝子を同定するために、以下の処置を実行した。間充織幹細胞を、単独、またはPC3細胞の存在下、骨形成培地中で、4乃至11日間培養した。妥当な時間に、全RNAが、細胞から単離した。RNAを逆転写にかけ、ロシュ・ライト・サイクラー(登録商標)に特異的な遺伝子の発現について解析した。図4で分かるように、アルカリホスファターゼ遺伝子、オステオカルシン、及びCBFA2 RNAの発現は、骨転移の際、増加した。負のコントロール(NC1及びNC2)の発現は、骨転移の際、増加しなかった。
共培養でのバクテリオファージ選択のために、TG1大腸菌宿主株の新しい培養を1%のブドウ糖を有するL培地中、600ナノメートル(OD600)で0.8の光学濃度まで増殖させた。
骨移植および/またはSREの調節因子を同定するために、候補化合物を宿主細胞と癌細胞との共培養と接触させる。候補化合物を細胞の共培養に、約1ピコモルないし約5Mの濃度で添加する。同時に、コントロール共培養を、候補化合物が無い状態でインキュベートする。日毎に、骨移植またはSREの生物学的マーカーを測定して、候補化合物が骨移植および/またはSREの調節因子であるかどうかを判断する。細胞のRNA、DNA、およびタンパク質を単離して細胞の遺伝的プロフィールを同定する。遺伝的プロフィールをコントロールプロフィールと比較する。骨移植および/またはSREを示す変化が、骨移植および/またはSREの調節因子を示す。
1日目、PC3細胞を、10%胎仔ウシ血清(FCS)および1xPen/Strepを含むRPMIに16,000細胞/cm2の密度で播種した。2日目、細胞を未処理またはEphA2に対するアンチセンス・オリゴヌクレオチドによる形質移入もしくは逆コントロールオリゴヌクレオチドによる形質移入をおこなった。3日目、培地を骨形成培地に変えて、10,000細胞/cm2の濃度で間充織幹細胞を培養物に添加した。培養物をさらに3日間、37℃、5%CO2条件下でインキュベートした。3日目に、細胞をアルカリホスファターゼに対して固定および染色あるいは定量的アルカリホスファターゼ分析のために溶解した。図9に示すように、結果は、アンチセンス・オリゴヌクレオチドの存在下ではアルカリホスファターゼ接触および定量的分析の減少が見られた。
PC3細胞を、EphA2に対するアンチセンスまたは逆コントロールオリゴヌクレオチドで処理し、次に3%Matrigel(登録商標)オーバーレイを有するMatrigel(登録商標)に置いた。図13に示すように、アンチセンス・オリゴヌクレオチドは、PC3細胞の増殖を阻害した。その一方で、コントロールオリゴヌクレオチドは阻害しなかった。
PC3細胞は、EphA2に対するアンチセンスもしくは逆コントロールオリゴヌクレオチドで処理した。この細胞を0.35%軟寒天に播種し、培養7日後にアラマーブルーを用いて定量した。

PC3細胞コロニー形成阻害の結果生ずるこれらのアンチセンス・オリゴヌクレオチドは、EphA2が、転移性表現型の生成または維持に役割を果たし、または、SREに関与することを示す。
複数個体の正常組織の全RNAを貯蔵し、逆転写し、EphA2に対するプライマーを用いて定量的PCRにかけた。6つの癌および腫瘍周辺正常組織から得たLCM組織由来の増幅RNAを逆転写し、EphA2に対するプライマーを用いた定量的PCRにかけた。図12に示すように、癌のmRNAレベルが、腫瘍周辺レベルよりも約2倍高いことがわかった。大腸癌試料のEphA2レベルは、正常試料の約2倍高く現れた。
癌性細胞のポリヌクレオチドによって示される差動的発現遺伝子の発現は、転移性表現型の促進等腫瘍形成における遺伝子生成の役割と機能とを確かめるために、アンチセンス・ノックアウト技術を用いて分析が可能である。
細胞増殖の阻害に対する遺伝子発現の効果は、転移性乳癌細胞系統(MDA−MB−231またはMDA231(「231」))、SW620大腸結腸直腸癌細胞、SKOV3細胞(ヒト卵巣悪性腫瘍細胞系統)、またはまたはLNCaP、PC3、22Rvl、MDA−PCA−2bまたはDU145前立腺癌細胞で、評価することができる。
抗体認識および調製のためのEphA2の線形エピトープは、当技術分野で知られている数多くの方法のいずれかによって同定し得る。いくつかの例の方法は、抗原のアミノ酸配列に由来するペプチドの抗体結合能を調べることを含む。結合の評価は、BIACOREまたは酵素免疫測定法方法を用いておこなうことができる。他の技術として、平面固体キャリア(「チップ」)上で、抗体に対してペプチド・ライブラリーを露出させること、ならびに固相スクリーニングで用いられる多数の方法のいずれかを介して結合を検出することが挙げれられる。さらに、ファージ提示を用いて、いくつかのバイオ・パニングのラウンド後、エピトープの選択によるペプチドのライブラリーをスクリーニングすることに用いることができる。
