Abstract
Description
Claims (16)
- 全血サンプルの個体の赤血球に含まれるヘモグロビン含量を測定する方法であって、
(a)赤血球を含む全血サンプルを球形化溶液の中に希釈し、それにより当該サンプル中の赤血球を実質的に球形にするステップと;
(b)球形にされた前記赤血球を、一度に1個、光学式検査ゾーンを通過させるステップと;
(c)各赤血球がその光学式検査ゾーンを通過するとき、当該赤血球に所定の波長の光ビームを照射するステップと;
(d)照射されたそれぞれの赤血球から所定の角度範囲内に反射された照射光のレベルを検出するステップと;
(e)検出された反射光のレベルに基づき、照射された各赤血球のヘモグロビン含量を決定するステップを含む、前記方法。 - 前記溶液が、その中にある赤血球の形状を実質的に球形にする洗剤を含む、請求項1に記載の方法。
- 前記洗剤がn-ドデシル-n,n-ジメチル-3-アンモニオ-1-プロパン・スルホネートを含む、請求項2に記載の方法。
- 前記溶液が、前記赤血球の内部にあるヘモグロビン分子と結合する酸素含有ガスをさらに含み、反射率が実質的に一様なヘモグロビン複合体を形成する、請求項2に記載の方法。
- 前記ガスが一酸化炭素を含む、請求項4に記載の方法。
- 前記溶液が、前記赤血球の内部にあるヘモグロビン分子と結合する酸素含有ガスを含み、反射率が実質的に一様なヘモグロビン複合体を形成する、請求項1に記載の方法。
- 前記ガスが一酸化炭素を含む、請求項6に記載の方法。
- ヘモグロビン含量を決定する前記ステップを、コンピュータのメモリに、異なるさまざまな全血サンプルの平均赤血球ヘモグロビンと、前記所定の波長での光照射に対する前記サンプルの平均細胞反射率の間の関係を記憶させ、各赤血球の測定された反射率と記憶させた前記関係に基づいて赤血球ヘモグロビンを決定することによって実行する、請求項1に記載の方法。
- 前記所定の波長が610nm〜710nmである、請求項1に記載の方法。
- 前記波長が約635nmである、請求項1に記載の方法。
- 個体の赤血球のヘモグロビン含量を測定する装置であって、
(a)赤血球が一度に1個通過できる光学式検査ゾーンを規定する光学式フロー・セルと;
(b)所定の波長の照射ビームを発生させるレーザーを備えていて、それぞれの赤血球が前記光学式フロー・セルの光学式検査ゾーンを通過するときにそのビームを各赤血球に照射する光学系と;
(c)照射されたそれぞれの赤血球から所定の角度範囲内に反射された照射光のレベルを検出する反射率検出器と;
(d)検出された反射光のレベルに基づき、照射された各赤血球のヘモグロビン含量を決定する計算手段とを備える、前記装置。 - 前記反射率検出器が、照射された赤血球から反射された照射光を受け取る位置にある光回収端と、高ゲイン光検出器に隣接する位置にある光放出端とを有する複数の光ファイバーを備える、請求項11に記載の装置。
- 前記光ファイバーの光回収端が、照射ビームに対して同心状に配置された複数の環状アレイとして配置されている、請求項12に記載の装置。
- 前記高ゲイン光検出器が光電子増倍管を備える、請求項12に記載の装置。
- 前記所定の角度範囲が、前記照射ビームが伝播する軸線に対して測定される約4°〜10°である、請求項11に記載の装置。
- 前記計算手段が、(i)異なるさまざまな全血サンプルの平均赤血球ヘモグロビンと、前記所定の波長での光照射に対する前記サンプルの平均細胞反射率の間の関係を記憶させてあるメモリと、(ii)各赤血球で測定した反射率と記憶させた前記関係に基づき、照射された各赤血球のヘモグロビン含量を決定する手段とを備える、請求項11に記載の装置。
