Abstract
Description
本発明は、一般的には、ワクチンと、感染性疾患の処置または予防に関する。より具体的には、本発明は、プロテオソームアジュバント、または麻疹ウイルス抗原とともに処方されるプロテオソーム:リポ糖アジュバントを含む組成物、ならびにそれらの治療的使用に関する。
麻疹は、伝染性の強い疾患であり、毎年およそ4000万人がこれに感染し、これによって90万人を超える人が死亡している(WHO/UNICEF,Joint WHO/UNICEF statement on Vitamin A for Measles,Weekly Epidemiology Record 19:133,1987)。2001年には、世界保健機構(WHO)とUNICEFによって、開発途上国向けのワクチン接種キャンペーンを通じて、麻疹による死亡数を2005年までに少なくとも50%減少させるためのプログラムが発表された。これは、世界中の80%以上において80%以上が確保されることによって達成される(すなわち;非特許文献1)。現在使用されている生の弱毒化ワクチンは有効ではあるが、深刻な限界がある。具体的には、中和母体抗体の存在が原因で、しばしば、9ヶ月未満の月齢の子供の防御ができないことがある(非特許文献2;非特許文献3)。麻疹ウイルス(MV)に伴う死亡の30%から50%が、この受攻期に生じる。母体抗麻疹抗体力価は非常に幅広く変化し、受動的に獲得した抗体は、通常、3から4週間の半減期を有する。結果として、乳児は、誕生から1歳までの間のほぼいずれかの時点で、麻疹にかかりやすくなる(非特許文献4)。

本明細書中に記載される発明は、プロテオソームをベースとするMVワクチン(プロテオソーム:リポ糖(LPS−MVワクチン)の鼻腔内投与が、気道と全身的な抗体応答の両方において粘膜応答を刺激することができるという驚くべき発見に関係する。さらに、本明細書中に記載されるワクチン組成物の動物(マウスおよび幼若アカゲザル(rhesus macaque)を含む)への投与により、組成物は宿主または被験体に安全に投与することができ、観察できる毒性作用または有害な作用を何ら伴わないことが示される。プロテオソームと1つ以上の麻疹ウイルス抗原を含む免疫原性組成物が、本明細書中でさらに詳細に議論される。これらは、麻疹感染を処置または予防するというような治療的な使用、ならびにそれらを調製するための方法に適している。

1つ以上のMV免疫原を含む本明細書中に記載される免疫原性組成物は、免疫応答、例えば、防御免疫応答を誘発するために使用することができる。本発明により、本明細書中に記載されるように、1つ以上のMV免疫原またはその断片、融合タンパク質、多価免疫原、あるいは、そのような免疫原の混合物を、MVに特異的な免疫応答(細胞性および/または体液性)(これは、防御免疫応答である場合もある)を誘発するために十分な用量で、被験体に投与することによって、MVによる感染を処置または予防するための方法が提供される。MVポリペプチド免疫原およびその変異体、またはそのような免疫原の混合物は、本発明の方法において使用される場合には、アジュバント、例えば、プロジュバントまたはOMP−LPSを含む組成物の一部であることが好ましい。1つの実施形態においては、本発明の免疫原性組成物にはさらに、1つ以上の別の微生物抗原、例えば、ウイルス抗原、細菌抗原、寄生虫抗原、またはそれらの組み合わせが含まれ得る。例えば、MV免疫原性組成物には、風疹抗原およびおたふく風邪抗原もまた含まれる場合がある。
(プロテオソームの調製)
免疫原(例えば、麻疹ウイルス抗原)は、ヒトまたは動物被験体の防御免疫応答を誘発することができる本発明のワクチン組成物を形成するように、プロテオソームとともに処方され得る。プロテオソームはアジュバントとして有用であり、グラム陰性細菌から精製される外膜タンパク質から構成される。プロテオソームを調製するための方法は、例えば、Mallett et al.,Infect.Immun.63:2382,1995;米国特許第6,476,201 B1号;米国特許出願公開番号2001/0053368;ならびに米国特許出願公開番号2003/0044425に記載されている。簡単に説明すると、フェノールで死滅させたグループB 2型髄膜炎菌(Neisseria meningitidis)のペーストを、1Mの塩化カルシウム中の6%のEmpigen(登録商標)(EBB)(Albright and Wilson,Whithaven,Cumbria,UK)の溶液で抽出した。抽出物をエタノールで沈殿させ、1%EBB−Tris/EDTA−生理食塩水中に溶解させ、その後、硫酸アンモニウムで沈殿させた。沈殿したプロテオソームを1%のEBB緩衝液中に再度溶解させ、透析濾過し、0.1%のEBB緩衝液中で−70℃で保存した。
(リポ糖の調製)
フローチャート2(図2)の例は、フレクスナー赤痢菌(S.flexneri)またはプレジオモナス・シゲロイデス(P.shigelloides)からのLPSの単離および精製のためのプロセスを示す。このプロセスは、他のグラム陰性細菌(赤痢菌属(Shigella)、プレシオモナス属(Plesiomonas)、エシェリキア属(Escherichia)、またはサルモネラ属(Salmonella)の種を含む)からLPSを調製するために同様に使用することができる。細菌を醗酵によって300L中で増殖させた後、細菌を堆積させ、細胞ペーストを、細菌ペースト1グラムあたり3mLの0.9MのNaCl、0.005MのEDTA、および10mgのリゾチームで再度水和した。リゾチームでの消化を、室温で1時間進行させた。その後、0.025MのMgCl2中の50U/mlのBenzonase(DNase)を添加し、DNase消化を室温で30分間進行させた。その後、懸濁液を14,000から19,000psiでマイクロフルイダイザーを通過させることによって砕いた。新しいDNase(50U/mL)を添加し、懸濁物の消化を、さらに30分間室温で進行させた。消化した細胞懸濁物を水浴中で68℃に加熱し、等量の90%のフェノール(これもまた68℃に加熱した)を添加し、その後、混合液を震盪させながら68℃で30分間インキュベートした。混合液を4℃で遠心分離して、水相と有機相を分離させた。水相を回収し、有機相をWFI(注射用蒸留水)で68℃で30分間再度抽出した。混合液を4℃で遠心分離し、第2の水相を回収し、2つの回収した水相を合わせた。核酸を沈殿させるために、10mMのCaCl2を含む20%のエタノールをプールした水相に添加した。混合液を4℃で一晩攪拌し、次いで、沈殿した核酸を、10,000×gで30分間の遠心分離によって堆積させた。上清を回収し、濃縮し、30,000MW中空線維カートリッジを使用して、0.15MのNaCl、0.05MのTris、0.01MのEDTA、および0.1%のEmpigen(登録商標)BB、pH8.0の中に透析濾過した。最後に、LPSを、0.22μMのMillpak(登録商標)60フィルターユニットを使用して滅菌濾過し、滅菌の保存容器に等分し、−80℃で保存した。
(プロテオソーム:リポ糖アジュバントの調製および特徴づけ)
本発明のプロテオソームアジュバント処方物を、おそらくは非共有的である会合を可能にするためにプロテオソームとLPSを混合することによって製造した。LPSは、多数のグラム陰性細菌、例えば、赤痢菌属(Shigella)、プレシオモナス属(Plesiomonas)、エシェリキア属(Escherichia)、またはサルモネラ属(Salmonella)の種のいずれかから導くことができる(実施例2を参照のこと)。これを、フローチャート3(図3)に記載したように、実施例1のプロテオソームと混合する。簡単に説明すると、プロテオソームとLPSを4℃で一晩融解させ、TEEN緩衝液中で1%のEmpigen(登録商標)BBになるように調節した。2つの成分を、約10:1から約1:3の間のプロテオソーム:LPSの最終wt/wt比を生じる量で、室温で15分間混合した。プロテオソーム:LPS混合物を適切な大きさ(例えば、Size9)の10,000 MWCO中空線維カートリッジ上でTNS緩衝液(0.05MのTris、150mMのNaCl、pH8.0)中に透析濾過した。浸透液中のEmpigen(登録商標)含有量が<50ppm(Empigen(登録商標)Turbidity Assayによるか、またはBradford Reagent Assayによる)になった時点で、透析濾過を停止した。バルクのアジュバント(本明細書中ではOMP−LPSと呼ぶ)を濃縮し、5mg/mLのタンパク質となるように(Lowryアッセイによる)調製した。最後に、アジュバントを、0.22μmのMillipak 20フィルターユニットを使用して滅菌濾過した。バルクのアジュバントを、滅菌の保存容器に等分し、凍結させた。
(麻疹ウイルス抗原の調製)
米国でワクチンの目的に使用されている麻疹ウイルス(MV)の弱毒化株であるMoraten株の増殖を、0.01〜0.001の感染多重度(MOI)でVeroミドリザル腎臓細胞を感染させることによって行った。ここでは、感染させた細胞を、10レベルの工業用チャンバー(Nalge Nunc International,Rochester,NY)で培養した。MOIは、不完全な妨害を生じる粒子の発生を最少にするために低くした。感染細胞の培養を、有意な細胞病理学が検出されるまで(例えば、約3〜5日)モニターし、その時点で、感染細胞の培養物を、1回の凍結−融解サイクルに供して細胞を破壊させた。細胞の破片を2100×gで20分間、4℃での遠心分離によって除去した。細胞を含まないMVを含む上清を回収し、0.45μmのフィルターと、その後に0.22μmのフィルターを連続して通過させて濾過した。その後、濾過した上清を4℃で2時間、14,000rpmで限外濾過した。堆積した麻疹ウイルス粒子をリン酸緩衝化生理食塩水(PBS)中に再度懸濁させ、次いで超音波処理を行った。タンパク質濃度を、2シストロン性酸性タンパク質アッセイを使用して決定し(Pierce Biotechnology,Rockford,IL)、標準曲線を、ウシ血清アルブミン画分V(BSA)とウシγグロブリン画分II(BGG)の混合物を使用して作製した。
(麻疹ウイルス抗原調製物の分析)
ドデシル硫酸ナトリウム(SDS)−ポリアクリルアミドゲル電気泳動(SDS−PAGE)を、ウイルス調製物中のMVヘマグルチニン(H)タンパク質と融合(F)タンパク質の存在を評価するために行った(図4を参照のこと)。ウイルス試料の段階希釈物を、10%のポリアクリルアミドゲル上での電気泳動によって分離し、タンパク質のバンドを、クマシーブリリアントブルーG−250(Kodak,Rochester,NY)での染色によって視覚化した。クマシーブリリアントブルーでの染色によって検出された個々のMV抗原の相対的な量と、試料の全タンパク質含有量の割合として決定したHタンパク質とFタンパク質の量を、Scion imageソフトウェアを使用する定量的デンシトメトリーによって決定した。タンパク質調製物中のHタンパク質とFタンパク質の量は44.4%であった。
(プロテオソーム:LPSと麻疹ウイルス抗原を含む処方物の調製)
本発明の処方物を、実施例3によるプロテオソーム:LPSアジュバント(本明細書中ではOMP−LPSとも呼ぶ)を実施例4によるMV抗原と、最適な安定性と免疫学的結果を促進する割合で混合することによって調製した。いくつかの場合には、プロテオソーム:LPSアジュバントとの処方の前に、ウイルス抗原調製物を、1%の界面活性剤(例えば、Empigen BBまたはMega−10)を含むように調製し、続いて、透析し、その後、プロテオソーム:LPSアジュバントと混合した。
(プロテオソームと麻疹ウイルス抗原を含む処方物の調製)
本発明の処方物を、実施例1によるプロテオソームを、実施例4によるMV抗原と、安定性と免疫学的結果について最適な処方物となる割合で混合することによって調製した。プロテオソームとの処方の前に、ウイルス抗原調製物を、実施例6に開示したように1%の界面活性剤(例えば、Empigen BBまたはMega−10)を含むように調製した。
(麻疹ウイルス抗原ワクチン処方物の分析)
ワクチン処方物を、SDS−PAGE(クマシーブリリアントブルー染色と免疫ブロット分析)によって、そして免疫金顕微鏡法によって分析した。SDS−PAGE分析の前に、ワクチン処方物を10,000で15秒間遠心分離した。ワクチン処方物の可溶性の画分(上清)と不溶性の画分(ペレット)を回収した。不溶性の画分をPBS中に再度懸濁させ、その後、β−メルカプトエタノールを含む試料緩衝液を添加した。プロテオソーム処方MVワクチン組成物の場合は、ワクチン処方物の可溶性画分中のプロテオソームOMPの存在をモニターし、これらの実験において、良好な処方プロセスの指標として確保した。クマシーブリリアントブルー染色を使用して、可溶性画分と不溶性画分中にタンパク質が存在することを検出し、免疫ブロット分析を使用して、透析した調製物の中にMV Hタンパク質とFタンパク質が存在することを確認した。MV抗原を含むプロテオソームのワクチン処方物、およびMV抗原を含むOMP:LPSが、検出することができる量のMV Hタンパク質とFタンパク質を含むことを見出した(図5A)。
(麻疹ウイルス抗原ワクチン処方物でのマウスの免疫化)
免疫化を、グループあたり5匹のマウスを含む21のグループの10週齢のBALB/c雌マウスに対して行った。それぞれの実験について、全てのマウスを2日おきに、体重と、毒性の兆候(例えば、毛の状態、猫背の姿勢、脂性肌、眼の分泌物、および脱水状態)について、実験手順が終わるまで評価した。BALB/cマウスを粘膜内(IM)および/または鼻腔内(IN)より、プロテオソーム:MV抗原処方物、またはプロテオソーム:LPS:MV抗原処方物で1日目および14日目に免疫化した。全てのワクチン処方物には、実施例4で調製した0.4μgのMV抗原を含めた。IN免疫化については、マウスを最初にイソフランの吸入によって軽く麻酔し、その後、鼻腔内に25μl(1つの鼻腔について12.5μl)を送達する自動導入チャンバーを使用して、ワクチンまたは対照処方物を投与した。IM免疫化については、25μlのワクチン処方物を、後肢に注射することによって投与した。全ての場合において、対照マウスを、緩衝液(PBS)のみ、MV抗原のみ、プロテオソームのみ、またはプロテオソーム:LPS(例えば、OMP:LPS)のみでINまたはIMによって免疫化した。
(ELISAによる麻疹ウイルスに特異的な抗体の分析)
血清と粘膜MV特異的抗体応答を、定量的ELISAによって測定した。血清試料について、全IgG、IgGイソ型(IgG1、IgG2a、IgG2b)およびIgAを測定した。鼻洗浄液および肺洗浄液については、IgAのみを評価した。U底の96ウェルマイクロタイタープレート(Greiner)を、炭酸緩衝液(pH9.6)中に希釈した1μg/mlのMV抗原で、4℃で一晩コーティングした。プレートを、PBS 0.1%のTween−20(PBS−T)中の2%の脱脂粉乳でブロックし、その後、試料の希釈物を2連で添加し、37℃で2時間インキュベートした。二次抗体には、ヤギ抗マウスIgG西洋ワサビペルオキシダーゼ(HRP;Pharmingen BD)、ヤギ抗マウスIgG1HRP,ヤギ抗マウスIgG2a−HRP、またはヤギ抗−マウスIgG2b−HRP(Southern Biotechnologies Associates)を含めた。ラット抗マウスIgA−ビオチン(Pharmingen,BD)を、IgAの検出のための二次抗体として使用し、続いて、ストレプトアビジン−HRP(Jackson Immunoresearch Laboratories)を使用した。アッセイを、TM Blue基質(Serological Corporation)の添加によって完了させた。反応を、0.2Mの硫酸(Sigma)を使用して停止させた。450nmで記録した光学密度の値の平均と標準偏差(SD)を、自動マイクロプレートリーダー(Bio−Rad Laboratories,Richmond,CA)から計算した。試験試料中の抗体濃度を、精製したマウスIgG抗体(Sigma,St.Louis,MO)または精製したマウスIgA(Bethyl Laboratories,Montgomery,TX)を使用してそれぞれのプレート上でインキュベートした標準曲線から計算した。値は、血清または洗浄液1ミリリットルあたりの特異的抗体のナノグラム数を示す(図6を参照のこと)。
(プラーク減少中和アッセイによる麻疹ウイルスに特異的な抗体の分析)
試料(血清および/または洗浄液)中に含まれるMV特異的抗体の、MVの増殖を中和する能力を、以前に記載された(Ward et al.,Diagn.Microbiol.Infect.Dis.33:147,1999)プラーク減少中和(PRN)アッセイによって評価した。簡単に説明すると、Vero細胞を、90〜95%の細胞集密度が得られるように24ウェルプレート(Falcon,BD Biosciences,Mississauga,Ontario,Canada)にプレートした。試料をPRN中で使用する前に、56℃で40分間熱不活化させた。試料を希釈し、37℃で90分間、MVとともにインキュベートし、その後、70%の細胞集密度のVero細胞の2連のウェルを、100μlの10倍の段階希釈物に感染させた。LebovitzのL15培地(Gibco Life Technologies,Grand Island,NY)中の16%のメチルセルロースの重層を、感染させた細胞に適用し、その後、プレートを5%のCO2中で37℃で4日間インキュベートした。4%のニュートラルレッドの溶液を添加して単層を染色し、その後、さらに24時間置いた。最後に、細胞の単層を、3.7%のホルマリンで10分間固定させ、目で見ることができるプラークを数えて、プラーク形成単位の数を決定した(図7を参照のこと)。それぞれの試料を2連で評価した。PRN指数を、Kaber法を使用して決定して、中和の50%終点を計算した。以下の式を使用してPRN値を計算した:最高希釈の逆数のlog10−[(平均プラーク数/ウイルス対照による平均プラーク数の和−0.5)×希釈係数のlog10]。
(麻疹ウイルス抗原ワクチン処方物の安全性)
MV抗原ワクチン処方物を、マウスにおいて安全性(すなわち、毒性)について評価した(実施例9もまた参照のこと)。マウスを、プロテオソームまたはOMP−LPSとともに処方したMV抗原の2または3用量のいずれかで、鼻腔内に(IN)免疫化した。さらに、プロテオソームとともに処方したMV抗原を使用して、粘膜内経路(IM)を通じてマウスを免疫化した。毒性がないことを、本明細書中に開示したワクチン処方物のいずれかを使用して検出した。マウスを毎日観察し、体重を測定した。INまたはIMのいずれかで投与したワクチン処方物については、全てのマウスにおいて行動の変化は記録されず、体重においても統計学的に有意な変動(例えば、+/−1.0グラムより大きい)は検出されなかった。これらのデータは、本発明のワクチン処方物が、おそらく、ヒトである被験体に用いる場合にも安全であることを示唆している。
(麻疹ウイルス抗原ワクチン処方物での免疫化後の血清IgG抗体応答)
MV抗原ワクチン処方物の全身性免疫を誘発する能力を、MV特異的抗体についての定量的ELISAによる血清試料の分析によって評価した。これらの実験では、マウスを図6に示した(矢印)ように、1日目、14日目、および/または28日目に免疫化した。これらの実験では、2用量または3用量のINで投与したプロテオソームMV抗原ワクチン処方物によって、測定可能な量のIgGの統計学的に有意な増加が誘導された(図6)。IMで投与した場合には、プロテオソーム−MVワクチン処方物によって、2用量のワクチンを投与した後、全てのマウスにおいて血清IgGの測定可能な増加が誘発され、これは、3用量の投与後には有意に増加した(図6)。血清IgGの検出可能なレベルはまた、IMで投与した3用量のMVのみを投与したマウスにおいても観察された。OMP−LPS−MV抗原ワクチン処方物でINで免疫化したマウスは、3用量のワクチンの後に有意なレベルのIgGを生じた。2用量のOMP−LPS−MV抗原処方ワクチンの投与によっては、検出可能な血清IgG応答は誘発されなかった。血清IgGは、全ての動物において免疫化の前には検出されず、PBS対照を投与したグループ、MV抗原のみをINで投与したグループ(2または3用量)、ならびにOMP−LPSのみを投与したグループ(2または3用量)を含む対照グループにおいては全ての時点で検出できないままであった(図6)。
(麻疹ウイルス抗原ワクチン処方物での免疫化後の粘膜抗体応答)
鼻洗浄液および肺洗浄液中のIgAの検出のためのELISAを使用して、鼻および気道の粘膜免疫を誘発するMVワクチン処方物の能力を決定した。鼻洗浄および肺洗浄を、最後の免疫化の8日後に行った。有意なレベルのMV特異的IgAが、3用量のプロテオソーム:MVワクチン処方物をINで投与したマウスにおいて検出されたが、そのようなレベルは2用量のワクチンを投与したマウスにおいては検出されなかった(図6)。対照的に、IMで投与した2用量または3用量のプロテオソーム:MV処方ワクチンによっては、検出可能なIgA応答は誘発されなかった(図6)。INで投与した3用量のOMP−LPS−MV抗原処方ワクチンで免疫化したマウスは、有意なレベルのMV特異的IgAを誘発し、力価は肺洗浄液試料において6000ng/mlに達した。IgAレベルは、鼻洗浄液よりも肺洗浄液中でより高かった(図6)。このことは、INで投与されたOMP−LPS−MV抗原ワクチン処方物によって、気道において粘膜免疫応答が誘発されたことを示唆している。IMで投与されたOMP−LPS−MV抗原処方ワクチン(2用量)は、検出可能なIgA応答を誘発しなかった。IgAのレベルは、MV抗原のみをINで投与したグループ、MV抗原のみをIMで投与したグループ(2用量または3用量)、PBS対象、およびOMP−LPSのみをINで投与したグループ(2用量または3用量)を含む対照グループから得られた鼻洗浄液および肺洗浄液の両方において低いままであったか、または検出できなかった。
(麻疹ウイルス抗原ワクチン処方物で免疫化したマウスに由来する血清および洗浄液試料の免疫中和活性)
プラーク減少中和アッセイを使用して、血清試料、さらには、鼻洗浄液および肺洗浄液試料を、最後の免疫化の8日後に採取した際の免疫中和活性について分析した。これらの実験では、INで投与したプロテオソーム−MV抗原処方ワクチンは、低いが有意なMV中和を示した(図7)。IMで投与したプロテオソーム−MVワクチン処方物は、粘膜IgA応答を誘発せず(図6)、結果として、ウイルスの中和は、MVを鼻または肺洗浄液試料に接触させた場合にも観察されなかった。IMで投与したプロテオソーム−MVワクチン処方物(3用量)によって得られた血清試料は、MVの増殖を中和することが示された(図7)。IMで投与したプロテオソーム−MV抗原ワクチン(3用量)と比較して、有意に低い中和レベルが、プロテオソーム−MV抗原処方物(2用量)をIMで投与したマウスと、MV抗原のみをIMで投与(3用量)したマウスに由来する血清試料を用いて観察された(図4)。これは、ELISAによって測定したIgGレベル(図6)と一致した。さらに、INで投与した3用量のOMP−LPS−MV抗原ワクチン処方物により、中和血清抗体の生産が誘発された。同様の結果が、INで投与されたプロテオソーム−MV処方物についても観察された。免疫中和活性の検出可能なレベルはまた、肺洗浄液試料においても観察された(図7)。MV抗原のみをINで投与したグループ(2または3用量)、PBSのみを投与したグループ、およびOMP−LPSのみを投与したグループ(2または3用量)のような対照グループから得られた試料は、MV中和活性を有していなかった。これらのデータは、MV抗原ワクチン処方物が、MV感染とその合併症からワクチンを接種した被験体を防御できる免疫応答を誘発することを示唆している。
(麻疹ウイルス抗原ワクチン処方物で免疫化したマウスにおける免疫応答)
種々のIgG抗体イソ型の優位は、特異的なタイプの免疫応答に関係している。血清IgG1は体液性免疫に関与しているTH2型応答に関係しており、一方、血清IgG2aは細胞性免疫に関与しているTH1型応答に関係している。イソ型特異的IgG1またはIgG2a抗体の濃度を、最後の免疫化の8日後に回収した血清においてELISAによって測定した。図8は、各実験群について、血清試料中のIgG1とIgG2aレベルと、それぞれのIgG1/IgG2a比を示す。血清IgG応答が対照よりも有意に高いグループについては、プロテオソームまたはOMP:LPSを含むMVの処方物により、MVのみをINまたはIMで投与したグループと比較して低いIgG1/IgG2a比が得られ、これは、プロテオソームおよびOMP:LPSがMV特異的免疫応答を1型の表現形へと再度向けることに有効であることを示している。
(麻疹ウイルス抗原に対する血清抗体の特異性の分析)
血清抗体の抗原特異性を、免疫ブロット分析によって決定した。MV H、F、およびMタンパク質を検出するために、比較用免疫ブロットを設計した(図9)。MV中和抗体を有している血清(INで投与したOMP−LPS−MV抗原ワクチン処方物(3用量)で免疫したマウスから得た)を、非中和抗体を有している血清(IMで投与したプロテオソーム−MV抗原ワクチン処方物(2用量)で免疫化したマウスから得た)と比較した。高い中和活性を有している抗体を含む血清は、MVタンパク質に特異的に結合することができた。対照的に、低い中和活性を有している血清試料は、MV Hタンパク質を検出することができず、MV Mタンパク質を弱く認識した。MV Fタンパク質の認識は、同様の分子の大きさの交差反応性であるVero細胞タンパク質の存在が原因で、非特異的なバックグラウンドの結合を上回って検出することは困難であった。それにもかかわらず、F0/F1バンドの強さは、免疫ブロットを中和抗体と接触させた場合には、非中和抗体と比較して高かった。まとめると、これらの結果は、中和活性のレベルが、MV抗原の認識、例えば、MV Hタンパク質の認識のレベルと相関関係にあることを示している。
(種々の用量の麻疹ウイルス抗原−プロテオソーム:LPS処方物を投与した動物の免疫応答)
この実施例では、種々の鼻腔内用量のプロテオソーム:LPS(IVX908)とともに処方された麻疹ウイルスワクチンを投与したマウスにおける、粘膜および全身性中和抗体免疫応答について記載する。
麻疹ウイルス分割抗原を以下のように調製した。Moratenワクチン株MV(R.Wittes,Connaught Laboratory,Mississauga,ONから譲り受けた)を、10レベルの細胞工業用チャンバー(Nalge Nunc International,Rochester,NY)を使用して、0.01〜0.001の感染多重度(MOI)でVeroミドリザル腎臓細胞中で増殖させた。細胞変性効果がピークの時に、フラスコを1回凍結−融解させた。細胞の破片を遠心分離(2100×g、4℃で20分間)によって除去し;プールした上清を最初に0.45μmのフィルター、その後、0.22μmのフィルターを通して濾過した。濾液を超遠心分離し(10,000×g、4℃で2時間)、ペレットを再度懸濁し、リン酸緩衝化生理食塩水(PBS)溶液中で超音波処理した。タンパク質濃度を、ウシ血清アルブミン画分Vとウシγグロブリン画分II(Pierce Bicinchoninic Acid Protein Assay,Pierce Biotechnology,Rockford,IL)の混合物を用いた標準曲線に基づいて測定した。IVX908との処方の前に、1%の界面活性剤(Mega−10、Bachem AG)をMV抗原調製物に添加し、その後、これを、PBSに対して、Slide−A−Lyzer透析カセット(Pierce,Rockford,IL)中で7日間かけて透析した。
MV分割抗原の段階希釈物を、10%のポリアクリルアミドゲル上での電気泳動によって分離し、タンパク質のバンドを、クマシーブルーG−250(Kodak,Rochester,NY)で視覚化した(図10A)。MV Hタンパク質に相当する80kDaのバンドが観察された。予想されたように、種々のFタンパク質のバンドが得られた。F0は60kDaのタンパク質として同定され、タンパク質分解によって処理されたF1サブユニットは41kDaのタンパク質バンドとして見られた。クマシー染色によって同定された他のMV抗原には、Nタンパク質(50〜60kDa)とMタンパク質(38kDa)が含まれていた。MV抗原調製物中の残りのVeroタンパク質の存在(例えば、Vero細胞溶解物中でのみ検出された約70kDaの濃いバンド)もまた、クマシー染色によって観察された。
IVX908(Protollin(商標)としてもまた公知である)を、cGMPガイドラインにしたがって製造した。これは、以前に記載されたように(Rima et al.,Curr.Topics Microbiol.Immunol.191:65−83(1995))透析濾過によって調製された期待される赤痢菌(S.flexneri)ワクチンのプロテオソーム−赤痢菌(S.flexneri)2a LPSロットと同じであった。LPSに対するプロテオソームポーリンの比は、およそ1:1wt/wtであった。髄膜炎菌(N.Meningtidis)ポーリン(Porin)A、ポーリンB、およびクラスIVタンパク質は、それぞれ、全プロテオソームタンパク質含有量の約20%、75%、および5%を占めた。IVX908を、動物への投与の直前に、1:1の比でMV抗原調製物と混合した。
ワクチン処方物を、ニッケルグリッドの上のairfuge上で5分間遠心分離した。次いで、グリッドを1%のBSAのブロッキング溶液中に5分間浸した。モノクローナル抗H(Chemicon International,Temecula,CA)を、一次抗体として使用した。室温で1時間のインキュベーションの後、グリッドをさらに、1%のBSAで5分間ブロックし、その後、抗マウスIgG−Gold−10nm(Aurion,Wageningen,Netherlands)に対して室温で1時間接触させた。グリッドをPBSと再蒸留水で洗浄し、その後、風乾させた。最後に、グリッドをPTA 3%(pH6.0)(ホスホタングジック酸(phosphotungsic acid)で着色し、日立7100電子顕微鏡を使用して観察した。代表的な電子顕微鏡写真を図10Cに示す。IVX908は、種々の大きさ(100nmから300nm)の丸型の膜構造を示した(図1C)。IVX908構造の表面との金粒子の接近した会合は、ワクチン処方物中でMV抗原がIVX908と会合していることを示した。IVX908と会合していないH抗原もまた観察された。
全ての動物の処理は、マギル大学(McGill University)のAnimal Care and Use Committee(プロトコール#4481)によって承認された。10週齢の雌のBALB/cマウスの21個のグループを使用した(1つのグループあたり5匹の動物)。体重と毒性の兆候(すなわち、毛の状態、猫背の姿勢、脂性肌、眼の分泌物、脱水状態)を、2日ごとに、実験手順が終わるまでモニターした。表1は、研究した種々の実験グループを記載する。ワクチン処方物には、1用量あたり1μg、3μg、および6μgの濃度でMV抗原を含めたが、IVX908の濃度は、全ての処方物について3μg/用量の定量に維持した。PBSを、全てのワクチン処方物について希釈剤として使用した。対照のグループには、IVX908のみ(3μg/用量)、MV抗原のみ(1、3、または6μg/用量)、Vero細胞タンパク質のみ(6μg/用量)、IVX908−Vero細胞タンパク質(6μg/用量)、およびビヒクルPBSを投与した。
血清および粘膜のMV特異的抗体応答を、定量的ELISAによって測定した。血清中の全IgGおよび特異的IgGイソ型(IgG1、IgG2a)を測定した。鼻洗浄液および肺洗浄液中のMV特異的IgAレベルを決定した。丸底の96ウェルマイクロタイタープレート(Greiner,MJS Biolynx,Brockville,ON)を、炭酸−重炭酸緩衝液(pH9.6)中に希釈した1μg/mlの超音波処理した全MV抗原で、4℃で一晩コーティングした。プレートを、2%の脱脂粉乳−PBS−Tでブロックし、その後、試料の希釈物を2連で添加し、37℃で2時間インキュベートした。PBS−Tでの洗浄後、二次標識抗体を、37℃で1時間かけて添加した。二次抗体には、ヤギ抗マウスIgG HRP(Pharmingen BD,San Diego,CA)、ヤギ抗マウスIgG1−HRP、ヤギ抗マウスIgG2a−HRP、およびヤギ抗−マウスIgG2b−HRP(Southern Biotechnologies Associates,Birmingham,AL)を含めた。IgAの検出のためには、ラット抗マウスIgA−ビオチン(Pharmingen,BD,San Diego,CA)を二次抗体として使用し、続いて、ストレプトアビジン−HRP(Jackson Immunoresearch Laboratories,West Grove,PA)を使用した。アッセイを、TM Blue基質(Serologicals Corporation,Norcross,GA)の添加によって完了させた。反応を、0.2Mの硫酸(Sigma−Aldrich Canada,Oakville,Ontario)を使用して停止させた。各試料の段階希釈物を測定し、標準曲線の25%〜75%の範囲に入るデータ点の光学密度の値を選択して、最終的な推定濃度を得た。450nmでの光学密度の値の平均と標準偏差(S.D.)の値を、自動マイクロプレートリーダー(Bio−Rad Laboratories,Richmond,CA)から計算した。試験試料中の抗体濃度を、精製したマウスIgG(Sigma−Aldrich Canada,Oakville,Ontario)または精製したマウスIgA(Bethyl Laboratories,Montgomery,TX)を使用してそれぞれのプレートについて作製した標準曲線から計算した。値は、血清または肺/鼻洗浄液1ml中の特異的抗体のngを示す。プロテオソームをベースとするワクチンで免疫化した後のセロコンバーションを、ワクチン前のレベルからの少なくとも4倍の抗体力価の上昇と定義した。
MV中和抗体を、以前に記載された(Ward et al.,Diagn.Microbiol.Infect.Dis.33:147−52(1999))プラーク減少中和(PRN)アッセイによって評価した。簡単に説明すると、Vero細胞を、90〜95%の細胞集密度が得られるように24ウェルプレート(Falcon,BD Biosciences,Mississauga,ON,Canada)に播種した。血清試料を、各実験グループの5匹の動物からプールし、使用前に、56℃で40分間熱不活化させた。血清の段階希釈物を混合し、37℃で90分間、継代数の低いEdmonston MV(25〜35プラーク形成単位)とともにインキュベートした。その後、細胞集密状態のVero細胞の2連のウェルを、100μlの血清の2倍の段階希釈物+MV混合物に感染させた。LiebovitzのL−15培地(Gibco/Life Technologies,Grand Island,NY)中の16%のメチルセルロースの重層を、感染させた細胞に適用し、その後、細胞を5%のCO2中で37℃で4日間インキュベートした。4%のニュートラルレッドの溶液を添加して、単層を染色し、細胞をさらに24時間インキュベートした。その後、細胞の単層を、3.7%のホルマリンで10分間固定した。目で見ることができるプラークを数えて、プラーク形成単位(PFU)の数を決定した。ウイルスのみを陰性対照とし、そして麻疹ウイルスワクチンをワクチン接種した個体に由来するヒトの血清を陽性対照とした。PRN値をKaber法を使用して得、中和の50%終点を決定した。PRN値は、プラークの数が≧50%減少した血清希釈の逆数のlog2として表される。比較のために、ヒトの血清防御は、PRN値>120として定義されている(Chen et al.,J.Infect.Dis.162:1036−42(1990))。PRN値を抗体濃度に対して標準化した。2用量および3用量のIVX908:MV抗原ワクチンを投与した後に動物から得られた血清試料の中和活性のグラフによる表示を、それぞれ、図13Aおよび図13Bに示す。全てのMV分割抗原濃度で、2用量のIVX908−MVが、有意な血清中和活性を誘発するために十分であった。さらなる用量のIVX908−MVによって、血清中和応答が増強された。鼻洗浄液および肺洗浄液中に存在する抗体による有意な中和はまた、最も高いMV分割抗原濃度(6μg)を投与したグループにおいて、および3μg/用量の肺洗浄液中でも観察された。対照グループに由来する血清および粘膜試料は、いずれの時点においても中和活性を有していなかった。
血清を、3用量の高い中和活性を有している、1用量あたり6μgのIVX908−MVを投与した動物から回収し、MV抗原に特異的な抗体を検出するための免疫ブロットによって分析した。HおよびF MV抗原に特異的に結合するモノクローナル抗体を使用したMV分割抗原の免疫ブロット分析(上記の調製方法を参照のこと)もまた行った。MV分割抗原、Veroタンパク質、およびOMPプロテオソームの調製物をSDS−PAGEによって分離し、分離した抗原の免疫ブロットを上記のように行った。図14に示すように、IVX908−MVで免疫化したマウスから回収した血清は、麻疹ウイルスHタンパク質(80kDa);麻疹ウイルスF0タンパク質(50〜60kDa);麻疹ウイルスF1タンパク質(41kDa);髄膜炎菌(N.meningitidis)OMP Por A(45kDa);および髄膜炎菌(N.meningitidis)OMP Por B(33kDa)を認識した。
マウスにおいては、血清IgG1はTH2型応答に関係しており、一方、血清IgG2aはTH1型応答に関係している(Maassen et al.,Vaccine 21:2751−57(2003))。最後のワクチン用量の10日後に、脾臓を全てのマウスから得た。単核細胞を、70μmのNylon細胞ストレーナー(BD Falcon)を通して処理することによって脾臓から単離し、単細胞懸濁液を得た。1つの実験グループあたり5匹の動物(表1を参照のこと)に由来する脾細胞をプールした。脾細胞によるIFNγ分泌のMV特異的刺激を、ELIPSOTによって定量した(Enzyme Linked ImmunoSPOT)(MABTECH,Nacka、Sweden)。脾細胞を、炭酸/重炭酸緩衝液(pH9.6)中に希釈した抗IFNγモノクローナル抗体クローンAN−18の5μg/mlでコーティングしたMultiScreen(商標)Immunobilon−Pベース96ウェルプレート(Millipore,Billerica,MA)中に100,000細胞/ウェルの密度で播種した。脾細胞を、種々の濃度のMV分割抗原(0.1から10μg)で72時間刺激した。PHA(5μg/mL)を陽性対照として使用した。Veroタンパク質調製物(10μg/mL)と培養培地を陰性対照として使用した。結果を、陰性対照の値を引き算した後、スポット形成細胞(SFC)/100万個の脾細胞として表す。陰性対照は、ほとんどの実験において1つのウェルあたり5未満のスポットを生じた(平均=1.3±1.2)。実験用のウェルは、5より多いスポット/ウェルが存在する(平均よりも>3 SD)場合に、ポジティブとみなした。Veroタンパク質調製物のみによって誘導されたスポットの平均数(陰性対照値)を、種々の濃度のMV分割抗原によって誘導されたスポットの平均数から引き算し、これを、100,000個の細胞あたりのサイトカインスポット形成T細胞サブセットの数に対して較正した(図15)。MV分割抗原の、対照グループ(IVXのみ、MVのみ、PBS)に由来する脾細胞とのインキュベーションによっては、スポット形成は生じなかった。これらのデータは、鼻腔内に投与されたIVX908−MVがMV特異的IFNγ応答を誘導する能力を有していることを示している。値は、3連の実験の平均を表す(*p<0.05 不等分散の片側t検定)。

この実施例では、麻疹DNAワクチンで免疫し、その後、IVX908を鼻腔内に追加免疫した幼若アカゲザルにおける免疫応答を記載する。この研究の全ての動物を、実験用動物の適切な世話についての手順およびプロトコールにしたがって世話し、処置した。
Claims (47)
- アジュバントと、1つ以上の麻疹ウイルス抗原とを含む免疫原性組成物であって、該アジュバントは、プロテオソームとリポ糖とを含み、少なくとも1つの麻疹抗原がHタンパク質である、免疫原性組成物。
- 前記アジュバントと、Fタンパク質およびHタンパク質を含む2つ以上の麻疹ウイルス抗原とを含む、請求項1に記載の免疫原性組成物。
- 1つ以上の麻疹ウイルス抗原が組換え麻疹抗原である、請求項1に記載の免疫原性組成物。
- 1つ以上の麻疹抗原が麻疹分割抗原である、請求項1に記載の免疫原性組成物。
- 前記麻疹分割抗原が、Moraten株、Schwarz株、Zagreb株、またはEdmonston株に由来する、請求項4に記載の免疫原性組成物。
- プロテオソームタンパク質の重量パーセントとしてのリポ糖の最終含有量が約10%から500%までの範囲である、請求項1に記載の免疫原性組成物。
- 前記プロテオソームと前記リポ糖とが同じ細菌から得られる、請求項1に記載の免疫原性組成物。
