シラカンバ樹液の比較対照として、ヒト表皮細胞に分化促進を高める作用が知られているカルシウム溶液(塩化カルシウム;以下、単に「カルシウム溶液」と記す。)を用い、また、培地のみで生育したものを対照としてコレステロール産生に与える作用について調べた。正常ヒト新生児表皮細胞を前培養後、培地の浸透圧低下を防ぐために培地量に対してシラカンバ樹液30v/v%、20v/v%、10v/v%、1v/v%、各それぞれの等量分を凍結乾燥により1/10倍に濃縮して添加した。また、比較対照においてはカルシウム溶液を元々の培地中に含有されるカルシウムイオン濃度の0.03mM/mLに加えて総量で1.5mM/mLとなるように添加し、それぞれの培地条件で一週間培養した後、培地を取り除きPBS(−)で1回洗浄してから表皮細胞を回収した。このヒト表皮細胞に、水、クロロホルム及びメタノール各2.5mLを添加して混合した後、クロロホルム層を単離、乾固した。抽出した脂質をHPTLCプレート〔シリカゲルG60(20×10cm)Art.5641;メルク社製〕にて、混合溶媒(クロロホルム:メタノール:酢酸=190:9:1)で2回展開した。40%硫酸を噴霧後180度で15分加熱することにより発色させた後、TLC上のコレステロールをデンシトメーターで比色定量した。培地のみで生育させたコントロールを1.0として、コレステロール産生促進率を算出し、その結果を表1に示す。
