Claims (15)
- 平形支持体(2)と、この平形支持体(2)上に配置されている捕捉分子を含むスポット(4)のアレイ(3)とを備え、各スポット(4)には捕捉分子と検体溶液によりもたらされる標的分子との間の結合事象を検出するためのマイクロ電極装置(5)が付設されているDNAチップにおいて、電極装置(5)が、標的分子にとって透過性である親水性反応層(14)内に少なくとも部分的に埋め込まれ、反応層(14)内には固定された捕捉分子が3次元に分布させられていることを特徴とするDNAチップ。
- 反応層(14)の厚みが2〜100μmであることを特徴とする請求項1記載のDNAチップ。
- 電極幅と電極間隔との合計をLとするとき、反応層はほぼ1〜5Lの範囲にある厚みを有することを特徴とする請求項2記載のDNAチップ。
- 電極幅および電極間隔が1μm(=1000nm)の範囲にあり、反応層が2〜10μmの厚みを有することを特徴とする請求項2又は3記載のDNAチップ。
- マイクロ電極装置が2極システムであり、反応層が約3μmの厚みを有することを特徴とする請求項3記載のDNAチップ。
- マイクロ電極装置が4極システムであり、反応層が約7μmの厚みを有することを特徴とする請求項1乃至5の1つに記載のDNAチップ。
- 反応層(14)が約95℃まで熱的に安定していることを特徴とする請求項1乃至6の1つに記載のDNAチップ。
- 反応層(14)が捕捉分子の共有結合のための結合グループを含んでいることを特徴とする請求項1乃至7の1つに記載のDNAチップ。
- 反応層(14)はヒドロゲルであることを特徴とする請求項1乃至8の1つに記載のDNAチップ。
- 結合グループとして、無水マレイン酸および/またはアクリル酸(メタクリル酸)グリシジルを有するアクリルアミドをベースとするラジカルに架橋結合可能なヒドロゲルが用いられることを特徴とする請求項6記載のDNAチップ。
- インターディジタル構造の電極装置(5)が設けられていることを特徴とする請求項1乃至10の1つに記載のDNAチップ。
- インターディジタル構造の電極装置(5)は2極のマイクロ電極システムであることを特徴とする請求項11記載のDNAチップ。
- インターディジタル構造の電極装置(5)は4極のマイクロ電極システムであることを特徴とする請求項11記載のDNAチップ。
- 平形支持体(2)は半導体層およびこの半導体層に結合された絶縁層(13)を含み、絶縁層(13)は半導体層とは反対側の面に電極装置(5)および反応層(14)を担持することを特徴とする請求項1乃至13の1つに記載のDNAチップ。
- 半導体層はシリコン層(12)であることを特徴とする請求項14記載のDNAチップ。
