Abstract
Description
本出願は2002年7月11日に出願された米国仮出願No.60/395,179の利益を請求し、ここにそのすべてを参考文献として組み込む。
本発明は政府認可番号NIH T32 HL 07284-26およびR01HL 64381の援助により行われた。政府は本発明に関して、幾分かの権利を有する。
さらに背景技術として、超音波研究の領域において、気体充填マイクロバブルがグレースケール及びドップラー超音波のコントラストを顕著に向上する。コントラストの増強は、マイクロバブルに取り込まれている気体とそれを取り巻く血液との音波のインピーダンスのずれに起因する。超音波変換器が放射する音波パルスは異なるインピーダンスを有する物質の界面で後方に反射(後方散乱)されるが、インピーダンスのずれが大きいと超音波の後方散乱の割合が大きくなり、超音波の後方散乱の割合が大きくなるとより大きな超音波コントラストが達成される。超音波の音波パルスがマイクロバブルに当たるとバブルの気相の容積振動(volumetric oscillation)が誘導される。この振動は高度に非線形であり、大きな後方散乱を起こすので、変換器アレイにより検出することができる(Forsberg及びShi,2001)。マイクロバブルの振動により形成された後方散乱は、研究中の組織構造により形成されるものよりも数桁も大きくなりうる(Klibanov,1999)。
さらなる態様及び本発明の利点は、ここに記載する事項から明らかであろう。
i)抗体及び抗体フラグメント、高い特異性と高いアフィニティを有する抗体を含む。従来の及び/又は遺伝子学的に設計された抗体を利用することができる。マイクロバブル組成物のヒトへの使用にはヒト抗体が用いられ、標的化分子に対して起こりうる免疫反応を回避しまたは減じることができる。
ii)細胞接着分子、そのレセプター、サイトカイン、成長因子、ペプチドホルモン、ペプチド擬似物及びこれらの断片。
iii)非ペプチドアゴニスト/アンタゴニストまたは細胞接着分子、サイトカイン、成長因子及びペプチドホルモンのためのレセプターの非生物活性バインダー。
iv)オリゴヌクレオチド及び修飾オリゴヌクレオチド、ワトソン−クリックまたは他の型の塩基ペアにより、DNAまたはRNAに結合するアプタマーを含むがこれに限定されない。
v)プロテアーゼ基質/阻害剤。プロテアーゼは多くの病理学的状態に関与する。
vi)種々の小さい分子、種々の生物学的レセプターに結合しうることが知られている生物活性化合物を含む。
vii)基質の結合パートナーとしての不活性化されたプロテアーゼ。
M−S−V
ここでMはマイクロバブル膜、Sはスペーサー腕、及びVは標的化分子である。
スペーサー腕の接着した標的化分子と変形可能な非球形(例えば、鈍鋸歯状、しわ状、折り畳み状など)膜の両方を有するマイクロバブルは、標的化したものが表面に結合するときに特に優位性を示す。標的化分子が標的に結合する能力は、結合部位のレセプターの構造を適応させる膜の変形性とスペーサー腕の柔軟性の両方により増強される。
セクション1:材料及び方法
1.1 試薬
P−セレクチンFc融合タンパク質(P−セレクチン.Fc)を凍結乾燥形にてR&D Systems(Minneapolis,MN)より得た。凍結乾燥されタンパク質750μgをDulbeccoのリン酸緩衝液(DPBS)(Invitogen, UK)15μLに溶解してP−セレクチン.Fcの部分試薬を作成し、これを液体窒素中で凍結し、4ヶ月未満−20℃で保存した。ビオチン化の前にアンチ−P−セレクチン抗体Rb40.34ストックを10,000ダルトンMWCO透析カートリッジ(Pierce, Rockford, IL)を用いてDPBS中で一昼夜透析した。抗体−マイクロバブル架橋系のためのN-ヒドロスクシンイミド−ビオチン(NHS-ビオチン)とストレプトアビジンをSigma(St. Louis, MO)から得た。ユーロピウムラベルしたストレプトアビジンとDELFIA溶液をWallac Oy (Turku, Finland)から得た。抗体濃度を測定するためのアドバンストタンパク質アッセイ試薬をCytoskelton, Inc (Denver, CO)から得た。流フローチャンバー(flow chamber)実験における非特定的接着を防止するためのTBS中のブロッカーカゼインをPierce (Rockford, IL)から得た。Tween-20をJ.T.Baker (Phillipsburg, NJ)から得た。Coulter容器用の希釈剤としてのIsoton-IIをBeckman Coulter(Miami, FL)
から得た。Kimura(0.05%wt/volトルイジンブルー、22%エタノール中0.9%NaCl、及び0.07Mリン酸緩衝液、pH6.4:全てSigma (St.Louis, MO)より)を血液カウントにおける白血球の変色に用いた(Olsemら、2001)。
in vitroでの接着能の実験は、平行板型フローチャンバー(GlycoTech, Rockville, MD)にて行った。上板と下板の間の距離はガスケットの厚みから測定し、0.254mmで、フローパス幅は2.5cmであった(ガスケット”B”)。P−セレクチン.Fc基質をおいた35mm直径のポリスチレン皿(Corning, Corning, NY)がフローチャンバーの上板の役割をした。マイクロバブルの浮力のため、特別に誂えたホルダーを用いてフローチャンバーを逆さに用いた。減圧と周囲に巻くゴムバンドにより皿をフローチャンバーの上にしっかりと置き、フローチャンバー全体を逆さにしたホルダーに挿入した。吸い込み状態にあるシリンジポンプ(Harvard Apparatus, Cambridge, MA)を用いてマイクロバブル分散液を吸引して規定の剪断速度でフローチャンバーに通した。実験はLeitz Laborlux II顕微鏡(Rockleigh,NJ)で一部エピ蛍光剤(epifluorescence)を使用し、40倍の対物レンズ(Olympus, Tokyo, Japan)で視覚化した。フローチャンバー実験から得られたデータをその後のオフライン分析のために標準的なVHSカセット(Sony, Tokyo, Japan)またはデジタルビデオカセット(Sony, Tokyo, Japan)に記録した。逆平行板型フローチャンバーの線図を図1に示す。
5匹の雄のC57B1/6と、P−セレクチンノックアウトマウス(P-/)をHilltop Labs (Scottsdale, PA)から入手し、U.Vaの小さな動物施設にて飼った。全てのマウスは生後8〜12週で見たところ健康だった。全ての動物実験はバージニア大学動物保護及び利用委員会制度(institutional animal care and use committee)により承認されたものである(プロトコル#2474)。
ハイブリドーマから得たRb40.34上清をDPBS中4℃で一昼夜透析した。各々2mgのモノクローナル抗体(mAb)をビオチン化するために45μgのNHSビオチンを10μLDMSOに溶解した。混合物を2時間4℃でインキュベートし、次いで一昼夜透析し結合しなかったビオチンを除去した。
35mmのポリスチレン皿をメタノールで洗浄し、P−セレクチン.Fcタンパク質の装着の前に空気中で乾燥させた。各洗浄済みの皿の中心に、全ての皿について同じ円のテンプレートを用いて1cm直径の円をフェルトペンで描いた。P−セレクチン.Fc融合タンパク質の必要重量(25,150または250ng)をDPBSで200μLに希釈し、皿の円を描いた部分に吸着させた。次いで皿を用意した蓋とぬらしたペーパータオルで覆い過剰な蒸発を防いだ。結合は一昼夜4℃で行った。その後皿をDPBS中0.05%のTween-20で5回洗浄し、結合しなかったタンパク質を除去した。皿表面への非特定的な接着を防ぐため、皿をTBSに2mLのカゼインを溶解したもので少なくとも2時間室温でインキュベートしてブロックした。全ての皿をブロックし、そして同じ日に使用し、フローチャンバー中で適切に封できない欠陥のある皿は廃棄した。負の対照として幾つかのブロックした皿を1.0mLのDPBS中の1.0mgRb40.34で少なくとも30分間インキュベートし、0.05%Tween-20で5回洗浄した。あるいは幾つかの皿はP−セレクチン.Fcの代わりにカゼインでインキュベートし、次いで洗浄し、ブロックした。
本実験で使用したマイクロバブルはジステアロイルホスファチジルコリン(DSPC)の脂質殻でデカフルオロブタン(C4F10)気体核をカプセル化したものである。ポリエチレングリコール(PEG)のブラシで脂質殻の周りを取り囲んだ。ビオチン化C4F10充填マイクロバブルを前記されたとおり(Kimら、2000;Lindnerら、2001)調製した。簡単に述べると、C4F10気体をホスファチジルコリン、PEGステアレート、及びビオチン−PEG−DSPCの水性分散液に高出力ソニケーションにより分散させた。これにより脂質−カプセル化されたC4F10マイクロバブルが形成した;マイクロバブルに取り込まれなかった脂質を遠心浮遊法を繰り返して除去した。2つのマイクロバブルの群を調製した:蛍光プローブ(Di-OまたはDi-I)を脂質殻に取り込んだ、蛍光ラベルしたものと、ラベル化していないものである。それぞれのマイクロバブル群を別々に4℃で2ヶ月間以内で保存し、顕微鏡観察とCoulterサイズ分析で検出された気体の損失は最小限であった。
既知の濃度(25、150または250ng/200μg)のP−セレクチン.Fcを上述の通り35mmの皿に吸着させた。カゼイン中で2時間のブロッキングの後1.0mLDPBS中0.96μgのビオチン化Rb40.34を各皿に加えた。溶液の量は皿の全表面を覆うのに充分であった。室温で30分間のインキュベートの後、0.05%Tween-20で7回洗浄して結合しなかった抗体を除去し、1.0mL中ユーロピウムラベルしたストレプトアビジンを各皿に加えた。室温で30分間のインキュベートの後、皿を7回洗浄して結合しなかったストレプトアビジンを除去した。0.9mLのDELFIA増強溶液(enhancement solution)を各皿に加え、室温で5分間インキュベートした。反応物を各皿から集め、96穴のマイクロタイタープレート(穴当たり300μL)に入れ、SPECTRAmax Gemini XSデュアルスキャニングマイクロプレート分光蛍光計(Molecular Devices, Sunnyvale, CA)を用いた時間分解分光蛍光計に用いた。プレートを360nmで励起し、250〜1250μ秒のタイマー間隔の間610nmで読んだ。カゼインのみでインキュベートしたスキャニングプレートにより非特定的な抗体の接着のレベルを測定した。Rb40.34のビオチン化は、アビジン−HABA置換法(Green, 1965)を用いて1モルの抗体当たりおよそ0.3モルのビオチンであると測定された。吸着されたP−セレクチン.Fcのサイト密度(site densities)は、Rb40.34がP−セレクチン.Fc二量体の各ヘッドと1対1で結合し、ストレプトアビジンとRb40.34が1対1で結合すると仮定して計算した。
抗体ラベル化したしわ状のマイクロバブルと球形のマイクロバブルとを同数ずつ混合し、希釈してC4F10飽和DPBS緩衝液1mL当たりおよそ3×106バブルとした。マイクロバブルの緩衝液の飽和によりマイクロバブルへのあるいはマイクロバブルからの気体の移動を防いだ。混合したマイクロバブル分散液を磁気撹拌棒で連続的に撹拌し、全ての実験において確実に均一にした。混合したマイクロバブル分散液を望ましい剪断速度で10cmの0.6mm直径のチューブを通じてフローチャンバーに吸い込んだ。各実験の間中、P−セレクチン.Fcが吸着された1cm直径の円の中心近傍の一つの視野(110μm×150μm)を観察した。エピ蛍光発光を低強度透過光に維持し、これにより蛍光ラベル化したしわ状のマイクロバブルとラベル化していない球形マイクロバブルとの区別が可能になった。各皿を単一の剪断速度で実験の間中観察した。各実験をおよそ10分間続き、あるいは視野が結合マイクロバブルで飽和するまで行った。各皿の単一の視野につき、各群のマイクロバブルの総流量75〜300が観察された。
各群の接着マイクロバブルの数を数えることによりオフラインで接着能を測定した。マイクロバブルを一時的に接着している、すなわち結合したマイクロバブルが10秒以内で剥がれるものか、あるいは強固に接着している、すなわち結合したマイクロバブルが10秒以上持ちこたえたものかのいずれかにクラス分けした。接着マイクロバブルの数をマイクロバブルの流量で表面の結合距離(1マイクロバブル直径)内に標準化し、接着能を算出した。
マイクロバブル接着行動の継続時間における分布を上記のフローチャンバー実験用に集めた。ビデオを1/3の速度で再生して短命の接着行動の検出を可能にした。バブルが静止したままでいるフレーム数を数え、これをビデオのフレームレートで割り、行動の継続時間を算出した。少なくとも10秒間接着したままのマイクロバブルを「強固に接着」と勘定し、10秒間を超えては分析しなかった(これらのバブルの大部分が残りの実験では結合したままであることが観察された)。一瞬結合したバブルの休止時間(pause time)を各P−セレクチン.Fcサイト密度でプールして(pool)、そしてこれをプロットした。
マウスに0.5μgのマウス腫瘍壊死因子(TNF-α)(Sigma, St. Louis, MO)を陰嚢内に手術前2時間に投与し、挙睾筋でのP-セレクチンの発現が最大になるようにした。マウスは、125mg/kg体重のケタミン(Parke-Davis, Morris Plains, NJ)、12.5mg/kg体重のキシラジン(Phoenix Scientific, St. Joseph, MO)及び0.025mg/kg体重の硫酸アトロピン(Elkins-Sinn, Cherry Hill, NJ)を腹膜内注射により麻酔にかけた。電気温パッドを用意して体温を38℃に維持した。気管にPE90チューブ(Becton Dickinson, Sparks, MD)を挿管し、自発呼吸を促進させ、そして右頚静脈にPE20チューブのカニューレを挿管した。マイクロバブル分散液を頚部カニューレを通じて投与し、各実験の最後に頚動脈から血液試料を採取した。
Di-Iラベル化した殻を両群に用いて上記の実験に記載したようにビオチン化マイクロバブルを調製した。100倍の水浸漬型対物レンズでの光の透過を可能とした修正フローチャンバーを構築した。標準的なCorningの35mmポリスチレン皿から円形の1cm半径の穴を切り出し、200μgのアビジンを吸着させてあるプラスチックカバーガラスを穴に適合させた。このような変更を加えた皿を数本のゴムバンドと通常のフローチャンバー減圧によりフローチャンバー台に固定した。修正フローチャンバーの線図を図2に示す。各マイクロバブル群をそれぞれ別々にフローチャンバーに引き込んだ。45s−1での5分間(分析に充分なバブルが吸着される時間)の引き込みの後、バブル分散液をC4F10飽和DPBSで置換した。およそ5〜20の接着マイクロバブルを取り囲む5つの視野が観測され、写真を撮った。そして剪断速度を5分間で122.5g−1に増加し、その間にさらに5つの視野が観察された。剪断速度を5分間で306.3s−1に増加し、またさらに5つの視野が観察された。カメラを流れの方向に沿わせるように注意した。各接着マイクロバブルの変形指数を、マイクロバブルにおける流れの方向に沿った最も長い軸の長さを、流れの方向に垂直な方向に沿った最も長い軸の長さで割り、算出した。
上記の通り鈍鋸歯状のマイクロバブルを静水圧により調製した。24.0×106または120×106球形マイクロバブル(未処理)またはしわ状マイクロバブルの分散液を注射グレードの0.9%NaClの0.5Lバッグに注入した。高調波造影を用い線形の超音波プローブによって、生理食塩水バッグ中でのマイクロバブルの音波レスポンス(echogenicity)を測定した。
球形及びしわ状マイクロバブルの接着能を、各フローチャンバー実験において賦与した3つの壁剪断速度において比較した。Excel ver.9.0表計算ソフト(Microsoft)を用いて2標本t−検定を行った。有意性はp=0.05となった。10の小静脈における接着したマイクロバブルの平均数をin vivo実験で比較した。同様の2標本t−検定をこれらのデータについて行った。
実施例1−5に記載したように調製した1mLの洗浄ビオチン化鈍鋸歯状マイクロバブルを、ストレプトアビジン(1μm直径、Molecular Probes, Eugene, OR)で被覆された〜1.108の蛍光ポリスチレンラテックスビーズと混合して30分間混合のためにインキュベートすることができる。次いで材料を逆さに置いたシリンジまたはフラスコに入れそして通常の重力でまたは〜500rpm(r=15cm)のバケツローターで遠心してマイクロバブルと接着した微粒子の浮遊物を得ることができる。あるいはマイクロバブルを抗体または他のリガンドまたは標的化分子で被覆し、特定の抗原を有する細胞混合物と混合することができる。抗原を有する細胞のマイクロバブルへの選択的な接着は、バブル−細胞複合体の浮遊物を作りだし、そして意図した細胞を含み、水性媒体中のバブル及び堆積物に結合しない、抗原を有していない細胞を含まないマイクロバブル層浮遊物の選択的な濃縮を可能とするだろう。マイクロバブル表面上の鈍鋸歯形状の存在によりマイクロバブルが標的細胞に接着するのが容易になり、接着能と結合力を改善する。
2.1 マイクロバブルの圧壊
脂質殻がバブル表面を通っての気体の拡散のバリアを提供する。マイクロバブルへのゆっくりとした均一な正の圧力の負荷により、気体内容物が脂質殻を通過して拡散する。脂質単分子層殻の表面部分は気体核の表面よりももはや過剰となり、殻が外側に曲がりしわや折り畳みを形成する。このしわが外側に突き出た二重層を形成する。しわ状マイクロバブルが飽和C4F10溶液内で保存されれば、濃度勾配は再膨張(re-inflation)に対抗するので、しわは安定である。予想されるしわの構造の線図は図3に表される。マイクロバブルへの圧力の負荷は脂質単分子層(黄色)の曲がりを生起し、二重層のしわを形成する。PEG分子は赤で、固定化Rb40.34抗体は黒で示される。
異なる3つの濃度の組み換え型マウスP-セレクチン(P-セレクチン.Fc)基質についてフローチャンバー実験を行った。サイト密度対吸着されたP-セレクチン質量のグラフを、皿表面のP-セレクチン.Fcタンパク質について作成し、これを図6に示す。サイト密度25ng、150ng、及び250ngは、それぞれ31,97及び133サイト/μm2である。グラフは非線形で、0.785cm2(1cm円の面積)当たりおよそ300ngP-セレクチンにおいて飽和していることがわかる。非特定的なバックグラウンドの接着が14サイト/μm2と等しいシグナルを生じた。
2.3.1 強固な接着能及び一時的な接着能
この実験では、接着行動に2つのタイプが観察された:一時的な接着と強固な接着である。造影には強固な接着が望ましい。なぜなら意図した標的部分にマイクロバブルが集合するからである。P−セレクチン標的化マイクロバブルの接着能を3つのP−セレクチン.Fcサイト濃度につき3つの剪断速度でフローチャンバー法にて測定した。結果を図7に示す。強固な接着能は剪断速度にほぼ線形に減少することが見て取れる:剪断速度33.6s−1、最高のP−セレクチン濃度(250ng/平行板)において最大7.6%から、122s−1、最低濃度(25ng/平行板)において2%にまで減少する(図7、A、B、C)。測定された各サイト密度でしわ状マイクロバブルは球形マイクロバブルと比べて実質的に(p<0.05)より多くの強固な接着能を示した(33.6s−1と76.6s−1)。アイソタイプ対照抗体(Iso Ab)を有する標的化しわ状及び球形マイクロバブル、または純粋カゼイン基質に注入されるP−セレクチン標的化マイクロバブルは、殆ど無視できる程の強固な接着しか生じなかった(<2%)(図7D)。
強固な接着行動の割合を、強固な接着能を捕捉能(capture efficiency)で割って算出した。この割合は、マイクロバブルが強固な(一時的の逆)接着行動を形成する傾向、あるいは同様にどのように良好な捕捉行動が安定化され、そして強固な接着に変化するかを表すものである(図8)。強固な接着の割合は各剪断速度におけるP−セレクチン.Fcサイト密度の関数としてプロットする。しわ状マイクロバブルの強固な接着の割合は、各剪断速度において250〜150ngのP−セレクチンでだいたい一定であり、25ngP−セレクチンでは若干減少する。球形マイクロバブルは、全ての条件において強固な接着の割合がしわ状マイクロバブルのそれよりも低いが、同様のサイト依存性傾向を示した。
2つのタイプの接着行動が観察された:バブルが少なくとも10秒間接着する強固な接着と、バブルが離れるまでに10秒間未満しか接着しない一時的な接着である。少なくとも10秒間接着したバブルの大半はもっと長い時間結合したままであることが観察されたが、10秒間を強固な接着のカットオフとして選択し、これはフローチャンバー実験にて通常の持続時間である。フローチャンバー実験から得た休止時間分布の分析により一時的な接着行動及び強固な接着行動は2つの異なる群を形成し、しわ状及び球形バブルに異なる群を生じる(図9A〜9C)。これにより、強固及び一時的接着について10秒間をカットオフ時間として使用する正当性が明らかである。
種々の剪断条件下の球形及びしわ状マイクロバブルの変形度を測定して、しわ状マイクロバブルの強固な接着能が増強されるのは、結合することができるバブル表面領域における変形が誘導する増加によるものであるという仮説を検証した。接着能を測定した全ての剪断速度にわたってしわ状マイクロバブルは球形マイクロバブルよりも実質的により多く変形しており、これは図10に示されている。1を超える変形指数は流れの方向への伸長(バブルに対する剪断力により変形が引き起こされていることを示す)と、P−セレクチン.Fc基質と接触しているマイクロバブル領域での対応する増加を表している。よりたくさんのP−セレクチン.Fc:Rb40.34結合が形成されると、強固な接着が安定化される。しかし変形指数は、セクション2.4.2に記載された、剪断速度に対する強固な接着能の依存性とは相関関係がないように見える:変形指数は剪断と共に増加するが強固な接着能は剪断と共に減少する。この矛盾点は、結合したバブルに対して剪断力が反対の振る舞いをすることに起因するのであろう。剪断速度を増すと剪断力がバブルを圧す力の大きさが増加し、表面の変形の増加を引き起こす。しかしこの剪断力は結合したバブルに対して平行板表面から離そうとする、引き離す力としてはたらく。2つの相対する効果の相対的な大きさは、接着バブルの剪断引っ張りに対する感受性と脂質殻が変形される相対的な容易性から見積もられる。
しわ状及び球形マイクロバブルのin vivoでの接着能は、炎症中のモデルマウスにより検討した。結果を図11に示す。剪断速度500〜3000s−1下のサイトカインで刺激した小静脈についてマイクロバブルの接着を検討した。4匹のc57B1/6マウスにおいてしわ状マイクロバブルが球形マイクロバブルよりも実質的に大きな接着を示した。P−セレクチン欠損マウス(P-/-)における標的化マイクロバブルの使用あるいはアイソタイプ対照抗体(Iso Ab)でのマイクロバブルの標的化は、両方のマイクロバブル群についておよそ2倍の接着にまで減少させた。球形マイクロバブルはしわ状マイクロバブルよりも直径で〜0.5μmサイズが大きかったが、このサイズの違いが内皮表面への送達の違いに寄与したとは考えられない。さらに接着したしわ状及び球形マイクロバブルにサイズまたは伸長の違いは特に見られなかった。P-/-マウスまたはアイソタイプ対照実験で接着したマイクロバブルは、Lindnerらが報告(2001)したように、マクロファージに接着するようであった。
等濃度で0.5Lバッグの注射グレードの0.9%NaClに注入された場合、視覚的に測定されるように、鈍鋸歯状マイクロバブルにより提供されるコントラストは、円形マイクロバブルのそれとおよそ等しく、測定範囲内ではバブル濃度と共に音波応答性が増加する。

- マイクロバブルが約1〜約10マイクロメートルの平均直径を有する、請求項1に記載のマイクロバブル組成物。
- 標的がレセプターであり、標的結合分子がレセプターに結合する、請求項1に記載のマイクロバブル組成物。
- レセプターが細胞外マトリクスタンパク質、接着分子、レセプターに結合するG−プロテイン、細胞表面タンパク質、サイトカイン、糖タンパク、ペプチド、脂質、糖脂質、炭水化物またはこれらの組合せからなる群から選択される、請求項6に記載のマイクロバブル組成物。
- 標的化分子がペプチド、擬ペプチド、アプタマー、タンパク質、抗体と抗体フラグメント、オリゴ糖、及び小さい有機分子からなる群から選択される、請求項1に記載のマイクロバブル組成物。
- 液体キャリヤー中の、気体充填マイクロバブルの懸濁液を含む、標的に結合するのに有用なマイクロバブル組成物であって、該マイクロバブルが表面突起を有するマイクロバブル膜を実質的に有し、該マイクロバブル膜が標的に結合する標的結合分子をさらに含む、前記マイクロバブル組成物。
- 表面突起が膜の折り畳みを含む、請求項9に記載のマイクロバブル組成物。
- マイクロバブル膜が脂質、タンパク質または界面活性剤を含み、マイクロバブルが約1〜約10マイクロメートルの平均直径を有する、請求項9に記載のマイクロバブル組成物。

- 標的化結合分子がペプチド、擬ペプチド、アプタマー、タンパク質、抗体と抗体フラグメント、オリゴ糖及び小さい有機分子からなる群から選択される、請求項9に記載のマイクロバブル組成物。
- レセプターが細胞外マトリクスタンパク質、接着分子、レセプターに結合したG−プロテイン、細胞表面タンパク質、サイトカイン、糖タンパク、ペプチド、脂質、糖脂質またはこれらの組合せからなる群から選択される、請求項13に記載のマイクロバブル組成物。
- 液体キャリヤー中の、気体充填マイクロバブルの懸濁液を含む、標的に結合するのに有用なマイクロバブル組成物であって、該マイクロバブルが主に非球形マイクロバブル膜を有し、該非球形マイクロバブル膜が剪断応力下で、対応する球形マイクロバブル膜よりも高い変形度を示し、該マイクロバブル膜が標的に結合する標的結合分子を含む、前記マイクロバブル組成物。
- マイクロバブル膜が脂質、タンパク質、ポリマーまたは他の界面活性剤またはこれらの組合せを含む、請求項16に記載のマイクロバブル組成物。
- 気体が実質的に血液に不溶である、請求項16に記載のマイクロバブル組成物。
- マイクロバブルが約1〜約10マイクロメートルの平均直径を有する、請求項16に記載のマイクロバブル組成物。
- 標的が細胞膜結合レセプターであり、標的化分子がレセプターに結合する、請求項16に記載のマイクロバブル組成物。
- マイクロバブルを標的に結合させる方法であって、請求項1、9及び16のいずれか1項に記載のマイクロバブル組成物に標的を接触させることを含む、前記方法。
- マイクロバブル組成物のマイクロバブル膜がスペーサー腕により接着された標的化分子を含む、請求項22に記載の方法。
- 標的に向けられたマイクロバブル組成物の製造方法であって、
液体キャリヤーに懸濁され、球形マイクロバブル膜を有する、気体充填マイクロバブルを形成し;
球形マイクロバブルを非球形マイクロバブルに変化させ;そして
該マイクロバブル膜に、標的に結合する標的化分子を接着あるいは導入する
ことを含む、前記方法。 - 標的化分子が、該変化より前に膜に接着あるいは導入される、請求項24に記載の方法。
- 標的化分子が、該変化より後に膜に接着あるいは導入される、請求項24に記載の方法。
- 該変化が、球形マイクロバブル膜内からの気体の部分的放出を含む、請求項24に記載の方法。
- 該変化が、球形マイクロバブル膜を圧力にかけることを含む、請求項27に記載の方法。
- 該圧力が、静水圧、超音波、またはマイクロバブル膜の浸透圧差により適用される、請求項38に記載の方法。

- 患者において超音波造影を行う方法であって:
患者に請求項34に記載の超音波造影剤を導入し;そして
該組成物を基準に超音波像を現像する
ことを含む、前記方法。 - 請求項32に記載の治療用組成物を患者に投与することを含む、患者の治療的処置を行う方法。
