Description

−毛細血管を通ることを可能にするために、8ミクロンよりも小さな粒子サイズ、
−無毒、又は許容できる毒性、
−十分な反射強調、
−製造及び臨床使用の容易さ、及び
−高度に特有のターゲット付けを可能にすること、
である。
a)それら粒子の中心が中空ではない
b)以下に説明されるコーティング層を除き、粒子が1つ以上の金属から本質的に構成される、すなわち中実の金属コアから形成されると共に、たんぱく質、多糖類又は他の構造化合物のような非金属化合物(外面層を除く)とは関連しない、すなわちそれらは中実の金属粒子である
ことを意味している本質的に中実の金属粒子である。
ある層の反射強調は、数理モデルを用いて計算されることができ、ここで
'r(k)'は非圧縮性材料の振幅反射係数ある、
't'は、例えば水のような媒体1、例えば超音波コントラスト層/造影剤のような媒体2、及び例えば基板のような媒体3の間における複素伝達係数である、
'r'は、例えば水のような媒体1、例えば超音波コントラスト層/造影剤のような媒体2、及び例えば基板(図1参照)のような媒体3の間における複素反射係数である、
'k'は、前記コントラスト層における超音波の波数である、
'd'は、前記コントラスト層の厚さである。
図3は、周波数を関数とした50nmのプラチナ層、50nmのタングステン層、50nmの金層及び50nmのタンタル層の論理的な予測反射強調を示す。
50nmの金が2μmのポリカーボネート(PC)フォイル上に蒸着されている。22MHzのトランスデューサを装備するTaberna Pro Medicum社のデジタル超音波イメージングシステムは、蒸着した金層を備える及び備えない前記PCフォイルの反射を測定するのに用いられる。2umのPCフォイル及び50nmの蒸着した金を備える2umのPCフォイルのまとめられた反射強度(ピークエリア)が図4に示される。
2μmのポリカーボネート(PC)フォイルは、50nmの銀粒子の層で被覆され、これら粒子は30nmのサイズを持つ。22MHzのトランスデューサを装備するTaberna Pro Medicum社のデジタル超音波イメージングシステムは、金層を備える及び備えないPCフォイルの反射を測定するのに用いられる。その結果が図5に示される。
Claims (15)
- 35×105g/cm2sよりも上の音響インピーダンスを持つ中実の金属ナノ粒子を超音波反射体として有する超音波造影剤であって、前記金属ナノ粒子は、中実の金属コアから形成され、該粒子の凝集を減少させるためのコーティングを有し、前記金属は、金又は銀であり、前記金属ナノ粒子は、前記コーティングを除き非金属化合物に結合されない、超音波造影剤。
- 50×155g/cm2sよりも上の音響インピーダンスを持つ請求項1に記載の超音波造影剤。
- 前記中実の金属ナノ粒子は1nmから100nmの間に直径を有する請求項1に記載の超音波造影剤。
- 前記中実の金属ナノ粒子は1nmから50nmの間に直径を有する請求項1に記載の超音波造影剤。
- 前記金属は非磁性である請求項1に記載の超音波造影剤。
- 前記金属は貴金属である請求項1に記載の超音波造影剤。
- 前記金属ナノ粒子は、1つ以上のコーティングを有する請求項1に記載の超音波造影剤。
- 前記コーティングは、天然若しくは合成炭水化物、合成ポリアミノ酸、又は生理学的に許容される合成ポリマー若しくはそれらの派生物を有する請求項1乃至7の何れか一項に記載の超音波造影剤。
- 前記1つ以上のコーティングは治療薬剤を有する請求項7に記載の超音波造影剤。
- 1つ以上のバイオターゲット特異性分子が、前記金属粒子の表面に結合していることを特徴とする請求項1に記載の超音波造影剤。
- 前記バイオターゲット特異性分子は、細胞マーカー、病原体、異種薬剤及び/又は毒性薬剤から成るグループから選択されるターゲットを認識する請求項10に記載の超音波造影剤。
- 前記バイオターゲット特異性分子は、抗体又はその抗体の断片である請求項10に記載の超音波造影剤。
- 超音波造影剤の製造における請求項1乃至12の何れか一項に記載の金属ナノ粒子の使用。
- 請求項1乃至12の何れか一項に記載の超音波造影剤を投与された動物又は人間の超音波イメージング方法。
- 分離した組織サンプル又は器官をイメージングする方法において、前記1乃至12の何れか一項に記載の超音波造影剤を前記組織サンプル又は器官に投与すると共に、当該サンプル又は器官の超音波イメージング検査を行うことを有する方法。
