本明細書中に記載された発明は、新規な遺伝子およびそのコードするタンパク質(84P2A9と称される)に関し、そして84P2A9を発現する種々の癌、特に前立腺癌の管理において有用な診断および治療の方法および組成物に関する。
ガンは、冠状動脈疾患(coronary disease)に次いで、第2の主なヒトの死亡原因である。全世界的に、数百万の人々が、毎年癌により死亡している。米国単独では、癌は毎年50万人をはるかに超える人々の死亡原因であり、毎年140万あまりの新たな症例が診断されている。心臓疾患での死亡は、有意に減少している一方で、癌に起因する死亡は、一般に増加の傾向にある。来世紀の初期には、癌は死亡の主原因になると予測されている。

本発明の別の局面は、84P2A9関連タンパク質およびそのポリペプチドフラグメントを提供する。84P2A9タンパク質の特定の実施形態は、図2に示されるようなヒト84P2A9のアミノ酸配列の全てまたは部分を有するポリペプチドを含む。あるいは、84P2A9タンパク質の実施形態は、図2に示されるヒト84P2A9のアミノ酸配列における変異を有する改変体ポリペプチドを含む。
本発明の別の局面は、84P2A9関連タンパク質およびポリペプチドに結合する抗体を提供する。好ましい抗体は、84P2A9関連タンパク質に特異的に結合し、そして非84P2A9タンパク質と結合しない(または、弱く結合する)。別の実施形態において、抗体は、84P2A9抗体関連タンパク質およびそのホモログと結合する。

個体における84P2A9遺伝子および84P2A9遺伝子産物の状態を評価するアッセイは、この個体に由来する生物学的サンプルの増殖または腫瘍形成能力に関する情報を提供し得る。例えば、84P2A9 mRNAは、正常前立腺組織に比べて前立腺癌(ならびに、例えば図4〜8に示される他の癌組織)においてかなり多く発現されるので、生物学的サンプル中の84P2A9 mRNA転写物または84P2A9タンパク質の相対レベルを評価するアッセイは、84P2A9調節不全と関連する疾患(例えば、癌)を診断するために使用され得、そして適切な治療選択肢を規定するに有用である予後情報を提供し得る。

BJら、Biochem.Biophys.Res.Commun.1999,261:646〜51)を使用して試験され得る。代表的には、84P2A9タンパク質は、抗84P2A9抗体を使用して、84P2A9を発現している前立腺癌細胞株から免疫沈降され得る。あるいは、Hisタグに対する抗体が、(上記のベクターを使用して)84P2A9を発現するように操作された細胞株において使用され得る。免疫沈降した複合体は、ウエスタンブロッティング、タンパク質の35S−メチオニン標識、タンパク質微量配列決定、銀染色および2次元ゲル電気泳動のような手順によってタンパク質の会合に対して試験され得る。
通常前立腺および精巣関連のタンパク質であり、そして、また、前立腺の癌(および他の癌)において発現される84P2A9の同定は、このような癌の処置に対する多くの治療アプローチを展開する。本明細書中で議論されるように、転写因子としての84P2A9の機能は、腫瘍促進遺伝子を活性化すること、または腫瘍形成をブロックする遺伝子を抑制することに関与する可能性がある。

第1の種類の治療アプローチ内で、本発明は、その結合パートナーへの84P2A9の結合または他のタンパク質との84P2A9の結合を阻害するための、種々の方法および組成物、ならびに84P2A9機能を阻害するための方法を包含する。

遺伝子移入および遺伝子治療の技術は、84P2A9を合成している腫瘍細胞に、治療用ポリヌクレオチド分子(すなわち、アンチセンス、リボザイム、内部抗体をコードするポリヌクレオチドおよび他の84P2A9阻害分子)を送達するために使用され得る。多数の遺伝子治療アプローチが、当該分野で公知である。84P2A9アンチセンスポリヌクレオチド、リボザイム、84P2A9転写と干渉し得る因子などをコードする組換えベクターは、このような遺伝子治療アプローチを使用して標的腫瘍細胞に送達され得る。
上記のように、84P2A9の発現プロフィールにより、84P2A9が、進行した前立腺癌および転移した前立腺癌で高度に発現されることが示される。この発現パターンは、癌−精巣(Cancer−Testis)(CT)抗原またはMAGE抗原(黒色腫および他の癌において上方制御される、精巣特異的遺伝子である)を連想させる(Van den EyndeおよびBoon,Int J Clin Lab Res.27:81〜86,1997)。84P2A9の組織特異的発現および癌での高い発現レベルに起因して、MAGE抗原は、現在癌ワクチンの標的として検討されている(Durrant,Anticancer Drugs 8:727〜733,1997;Reynoldsら、Int J Cancer 72:972〜976,1997)。
上記で記載された診断適用および治療適用において使用するために、本発明によって、キットもまた提供される。このようなキットは、1つ以上のコンテナ(容器)(例えば、バイアル、チューブなど)を厳重に閉じ込めて受容するように区画化されるキャリアを備え得る。コンテナ手段のそれぞれは、本方法において、使用されるべき別々の要素の1つを含む。例えば、コンテナは、検出可能に標識された、または検出可能に標識され得るプローブを含み得る。このようなプローブは、それぞれ84P2A9関連タンパク質または84P2A9遺伝子またはメッセージに特異的な抗体またはポリヌクレオチドであり得る。キットが、標的核酸配列を検出するために核酸ハイブリダイゼーションを利用する場合、キットはまた、標的核酸配列の増幅用のヌクレオチドを含むコンテナ、および/またはレポーター分子(例えば、酵素標識、蛍光標識、または放射性同位体標識)に結合されるレポーター手段(例えば、ビオチン結合タンパク質(例えば、アビジンまたはストレプトアビジン)を含むコンテナを有し得る。
本発明の種々の局面を、以下に続くいくつかの実施例によってさらに記載および例示する。これらはいずれも、本発明の範囲の制限を意図するものではない。
(材料および方法)
(LAPC異種移植片およびヒト組織)
LAPC異種移植片を、Cheales Sawyers博士(UCLA)から入手し、記載のように生成した(Kleinら、1997、Nature Med.3:402−408)。アンドロゲン依存性LAPC−4異種移植片およびアンドロゲン非依存性LAPC−4異種移植片(それぞれLAPC−4 ADおよびAI)ならびにLAPC−9 ADおよびAI異種移植片を雄性SCIDマウスにおいて増殖させ、そしてレシピエント雄に小組織塊として継代させた。LAPC−4 AIおよびLAPC−9 AI異種移植片は、LAPC−4 AD腫瘍またはLACD−9腫瘍からそれぞれ誘導した。AD腫瘍を有する雄性マウスを去勢し、そして2〜3ヶ月間飼育した。腫瘍が再増殖した後、この腫瘍を採集し、そして去勢雄SCIDマウスにおいてまたは雌性SCIDマウスにおいて継代させた。RNAおよびタンパク質の分析のためのヒト組織をUCLA(Los Angeles,CA)のヒト組織リソースセンター(Human Tissue Resource Center)(HTRC)およびQualTek,Inc.(Santa Barbara,CA)から得た。良性の前立腺過形成組織サンプルは患者由来であった。
ヒト細胞株(例えば、HeLa)を、ATCCから入手し、そして5%ウシ胎児血清を有するDMEM中で維持した。
腫瘍組織および細胞株を、10ml/g組織または10ml/108細胞を用いてTrizol試薬(Life Technologies、Gibco BRL)においてホモジナイズし、総RNAを単離した。ポリA RNAを、QiagenのOligotex mRNA MiniキットおよびMidiキットを用いて総RNAから精製した。総RNAおよびmRNAを、分光光度計分析(O.D. 260/280nm)によって定量し、そしてゲル電気泳動によって分析した。

抑制サブトラクティブハイブリダイゼーション(SSH)を使用して、前立腺癌で示差的に発現され得る遺伝子に対応するcDNAを同定した。SSH反応は2つのLAPC−4 AD異種移植片からのcDNAを利用した。詳細には、84P2A9 SSH配列を差引き(subtraction)から同定した。ここで、LAPC−4 AD腫瘍(去勢後3日)由来のcDNAを、インタクトな雄で増殖したLAPC−4 AD腫瘍由来のcDNAから差引き(サブトラクト)した。インタクトな雄において増殖したLAPC−4 AD異種移植片腫瘍をを「テスター」cDNAの供給源として使用し、一方、LAPC−4 AD腫瘍(去勢後3日)由来のcDNAを、「ドライバー」cDNAの供給源として使用した。
SSH反応から生じる遺伝子フラグメントを増幅するために、2つのPCR増幅を行った。第一のPCR反応において、1μlの希釈最終ハイブリダイゼーション混合物を、最終容量25μlの1μlのPCRプライマー1(10μM)、0.5μl dNTP混合物(10μM)、2.5μlの10×反応緩衝液(CLONTECH)、および0.5μlの50×Advantage cDNA polymerase Mix(CLONTECH)に添加した。PCR1を、以下の条件を用いて実施した:75℃で5分、94℃で25秒、次いで27サイクルの以下:94℃で10秒、66℃で30秒、72℃で1.5分。5つの別の第一のPCR反応を各実験について行った。産物をプールし、そして水で1:10に希釈した。第二のPCR反応については、プールし、そして希釈した第一のPCR反応物からの1μlを、プライマーNP1およびNP2(10μM)をPCRプライマー1の代わりに使用したことを除き、PCR1について用いたものと同じ反応混合物に添加した。PCR2を、10〜12サイクルの以下:94℃で10秒、68℃で30秒、72℃で1.5分を用いて実施した。PCR産物を、2%アガロースゲル電気泳動を用いて分析した。
第一鎖cDNAを、Gibco−BRL Superscript Preamplificationシステムを用いるオリゴ(dT)12〜18プライミングによって1μgのmRNAから生成した。製造者プロトコルを使用し、そしてこれは、42℃で50分間の逆転写酵素とのインキュベーション、続いて37℃で20分間のRNAse H処理を包含した。反応を完了した後、この容積を、標準化の前に水で200μlに増大させた。16の異なる正常ヒト組織由来の第一鎖cDNAを、Clontechから入手した。
上述の材料および方法に記載の2つのSSH実験は、多数の候補遺伝子フラグメントクローン(SSHクローン)の単離に至った。全ての候補クローンを配列決定し、そして、対応する遺伝子の正体に関する情報を提供し、そして示差的な発現について特定の遺伝子を分析するための決定を導くのを補助するために、主な公的な遺伝子およびESTデータベース中の全配列に対する相同性分析に供した。一般に、検索したデータベースのいずれかにおいていかなる公知の配列とも相同性を有さず、従って新規な遺伝子を表すと考えられる遺伝子フラグメント、ならびに以前に配列決定された発現配列タグ(EST)に対して相同性を示す遺伝子フラグメントを、RT−PCRおよび/またはノーザン分析によってディファレンシャル(示差的)発現分析に供した。
2347塩基対(bp)の全長84P2A9 cDNAクローン(クローン1)を、LAPC−4 AD cDNAライブラリー(Lambda ZAP Express,Stratagane)からクローニングした(図2)。このcDNAは、504アミノ酸のオープンリーディングフレーム(ORF)をコードする。配列分析によって、6つの可能性のある核局在化シグナルの存在が示され、そしてPSORTプログラム(http://psort.nibb.ac.jp:8800/form.html)を用いて核であることが予測される。タンパク質配列は、ヒト脳タンパク質KIAA1152(337アミノ酸領域にわたって39.5%同一性)と相同であり、そしてLUCA15腫瘍サプレッサータンパク質と相同であるドメインを示す(42アミノ酸領域にわたって64.3%同一性)(GenBank登録番号#P52756)(図3)。84P2A9
cDNAは、プラスミドp84P2A9−1として、アメリカンタイプカルチャーコレクション(ATCC;Manassas,VA)に2000年1月5日に寄託され、そして登録番号PTA−1151を割り当てられた。
正常ヒト組織における84P2A9 mRNA発現を、プローブとして標識された84P2A9 SSHフラグメント(実施例1)を用いる、全部で16の異なる正常ヒト組織を含む、2つの多重組織ブロット(Clontech;Palo Alto、California)のノーザンブロッティングによって分析した。RNAサンプルを、βアクチンプローブを用いて定量的に標準化した。この結果は、正常精巣および前立腺における、2.4kbおよび4.5kbの転写物の発現を実証した(図4)。
ポリクローナル抗体は、例えば、免疫剤および所望の場合アジュバントの1回以上の注射によって、哺乳動物中で惹起できる。代表的には、免疫剤および/またはアジュバントは、複数の物質または腹膜注射によって哺乳動物に注射される。代表的に、ペプチドは、84P2A9のコード領域から設計され得る。代替的に(あるいは)、免疫剤は、84P2A9タンパク質の全てもしくは一部、またはそれらの融合タンパク質を含み得る。例えば、84P2A9アミノ酸配列は、当該分野で周知の種々の公知の融合タンパク質パートナーのいずれか1つ(例えば、マルトース結合タンパク質、LacZ、チオレドキシンまたは免疫グロブリン定常領域)に融合され得る(例えば、Current Protocols In Molecular Biology,第2巻、16ユニット、Frederick M.Ausubulら、編.,1995;Linsley,P.S.,Brady,W.,Urnes,M.,Grosmaire,L,Damle,N.,およびLedbetter,L.(1991)J.Exp.Med.174.561−566)。他のこのような組換え細菌タンパク質としては、グルタチオン−S−トランスフェラーゼ(GST)、および84P2A9のHISタグ化融合タンパク質(適切なアフィニティーマトリックスを用いて誘導細菌から精製され得る)が挙げられる。

(pGEX構築物を用いる組換え84P2A9の産生)
細菌細胞において、84P2A9を発現するために、84P2A9の一部を、pGEX−6P−1中にクローニングすることによって、グルタチオンS−トランスフェラーゼ(GST)遺伝子に、融合した(Amersham Pharmacia Biotech,NJ)。全ての構築物を作製してN末端で融合したGSTおよびC末端の6つのヒスチジンエピトープとともに組換え84P2A9タンパク質配列を精製した。ヒスチジンコドンをORFの3’末端でクローニングプライマーに付加することによって、6つのヒスチジンエピトープタグを生成した。PreScissionTM認識部位によって、84P2A9からGSTタグの切断を可能にする。アンピシリン耐性遺伝子およびpBR322起点によってE.coli中でのプラスミドの選択および維持が可能になる。この構築物において、84P2A9のアミノ酸1〜151を含有するフラグメントを、pGEX−6P−1にクローニングした。さらなる構築物を84P2A9タンパク質のpGEX−6P−1にまたがる領域(例えば、アミノ酸1〜504およびアミノ酸151〜504)中に作製し得る。
哺乳動物系において組換え84P2A9を発現するために、完全長の84P2A9 cDNAを、例えば、C末端に6Hisタグを提供する発現ベクター(pCDNA3.1 myc−his、InVitrogen)中にクローン化し得る。これらの構築物は、293T細胞中にトランスフェクトされ得る。トランスフェクトされた293T細胞溶解液は、ウェスタンブロットにおいて上記実施例4に記載された抗−84P2A9ポリクローナル血清でプローブされ得る。
哺乳動物細胞において84P2A9を発現するため、1512bp(504アミノ酸)の84P2A9 ORFを、完全な翻訳開始Kozakコンセンサス配列をとともに、pcDNA3.1/V5−His−TOPO(Invitrogen,Carlsbad,CA)中にクローニングした。タンパク質発現は、サイトメガロウイルス(CMV)プロモーターから駆動される。組換えタンパク質は、V5エピトープおよび6つのヒスチジン(C末端に融合された)を有する。pcDNA3.1/V5−His−TOPOベクターはまた、ウシ成長ホルモン(BGH)ポリアデニル化シグナルおよび転写終止配列(mRNAの安定性を強化する)を含み、そしてエピソーム複製のためのSV40起点およラージT抗原を発現する細胞株中の単一ベクターレスキューを伴う。ネオマイシン耐性遺伝子が、このタンパク質を発現する哺乳動物細胞の選択を可能にし、そしてアンピシリン耐性遺伝子およびColE1起点が、E.coli中でのプラスミドの選択および維持を可能にする。
構成的に84P2A9を発現する哺乳動物細胞株を生成するために、1551bp(517アミノ酸)ORFをpSRa構築物中にクローニングする。pSRa構築物の293T−10A1パッケージング株へのトランスフェクション、または293細胞中のpSRaおよびヘルパープラスミド(φ )の同時トランスフェクションによって、それぞれ、アンホトロピックおよびエクトトロピックなレトロウイルスを生成する。このレトロウイルスを用いて、種々の哺乳動物細胞株を感染し得、これによってクローニングした遺伝子84P2A9の宿主細胞株への組み込みを生じる。タンパク質発現は、長末端反復配列(LTR)から駆動される。ネオマイシン耐性遺伝子が、このタンパク質を発現する哺乳動物細胞の選択を可能にし、そしてアンピシリン耐性遺伝子およびColE1起点が、E.coliにおけるプラスミドの選択および維持を可能にする。さらなるpSRa構築物を作製して、N末端およびC末端の両方のGFPおよび全長84P2A9タンパク質のmyc/6HIS融合タンパク質を生成する。
バキュロウイルス発現系における組換え84P2A9タンパク質を生成するために、84P2A9 cDNAを、N末端にHis−タグを提供する、バキュロウイルス転移ベクターpBlueBac4.5(Invitrogen)中にクローニングする。詳細には、pBlueBac−84P2A9を、ヘルパープラスミドpBac−N−Blue(Invitrogen)とともに、SF9(Spodoptera frugiperda)昆虫細胞中に同時トランスフェクトし、組換えバキュロウイルスを生成する(詳細についてはInvitrogenの指示マニュアルを参照のこと)。次いで、バキュロウイルスを細胞上清から収集し、そしてプラークアッセイにより精製する。
GeneBridge4照射ハイブリッドパネル(Walterら、1994,Nat.Genetics 7:22)(Research Genetics,Huntsville AI)を用いて、84P2A9の染色体局在化を決定した。以下のPCRプライマーを用いて、94P2A9を局在化した:

93照射ハイブリッドパネルDNAについて得られたマッピングベクターは以下であった:
000010001100101100100000110001001001000010001010011000100000001001001001010000000100000010000。このベクターおよびマッピングプログラムは、http://www.genome.wi.mit.edu/cgi−bin/contig/rhmapper.plで、染色体1q32.3上の84P2A9に位置する(D1S1602−D1S217)。
84P2A9が細胞中の既知のシグナル伝達経路を直接的または間接的に活性化するか否かを決定するために、ルシフェラーゼ(luc)を基礎にした転写レポーターアッセイを、84P2A9を発現する細胞中で実施する。これらの転写レポーターは、良く特徴付けられたシグナル伝達経路の下流にある、既知の転写因子のためのコンセンサス結合部位を含む。これらのレポーターおよびそれらの関連転写因子、シグナル伝達経路、および活性化刺激物の例を、以下に列挙する。

84P2A9媒介効果は、mRNA発現を示す細胞中でアッセイされ得る。ルシフェラーゼレポータープラスミドは、脂質媒介トランスフェクション(TFX−50、Promega)により導入され得る。相対的転写活性であるルシフェラーゼ活性の指標は、細胞抽出物のルシフェリン基質とのインキュベーションにより測定され、そして反応の化学発光がルミノメーター中でモニターされる。
84P2A9にMAbを生成するために、代表的には、Balb Cマウスを、完全フロイントアジュバント中に混合された、約10〜50μgのタンパク質免疫原を用いて腹腔内に免疫する。タンパク質免疫原としては、細菌およびバキュロウイルスが産生した組換え84P2A9タンパク質および哺乳動物が発現したヒトIgG FC融合タンパク質が挙げられる。次いでマウスに、フロイントの不完全アジュバント中に混合された10〜50μgの抗原で2〜4週間毎に、引き続いて免疫を行う。あるいは、Ribiアジュバントを最初の免疫に用いる。さらに、DNAベースの免疫プロトコールを用いる。このプロトコールでは、84P2A9 cDNA単独もしくはIgG FC融合をコードするpCDNA3.1のような哺乳動物発現ベクターを用いて、プラスミドDNAの直接注射によってマウスを免疫する。このプロトコールを単独で、そしてタンパク質免疫原と組み合わせて用いる。免疫後7〜10回採血をして、免疫応答の力価および特異性をモニタリングする。一旦、適切な反応性および特異性(ELISA、ウエスタンブロットおよび免疫沈降分析によって決定)を得ると、次いで、当該分野で周知の確立された手順で融合およびハイブリドーマ生成を行う(HarlowおよびLane,1988)。
前立腺癌における84P2A9の発現は、この遺伝子が腫瘍進行において機能的役割を有するという証拠を提供する。84P2A9は、腫瘍形成または腫瘍形成をブロックする遺伝子を抑制するのに関与する活性化合物遺伝子に関与する転写因子として機能する可能性がある。84P2A9機能は、インビトロアプローチを用いて哺乳動物細胞中で評価され得る。哺乳動物発現については、84P2A9は、pcDNA 3.1 myc−His−タグ(実施例5)およびレトロウイルスベクターpSRαtkneo(Mullerら、1991、MCB 11:1785)を含む多くの適切なベクター中にクローン化され得る。このような発現ベクターを用い、84P2A9は、NIH3T3、rat−1、TsuPr1および293Tを含むいくつかの細胞株で発現され得る。84P2A9の発現は、抗−84P2A9抗体を用いてモニターされ得る(実施例4および9を参照のこと)。
腫瘍細胞増殖に対する84P2A9タンパク質の影響は、腫瘍を保有するマウス中の遺伝子過剰発現によりインビボで評価され得る。例えば、SCIDマウスに、各脇腹上に、tkNeo空ベクターまたは84P2A9を含む、PC3、TSUPR1、またはDU145細胞のいずれかの1×106を皮下注射し得る。少なくとも以下の2つのストラテジーが用いられ得る:(1)ポリオーマウイルス、鶏痘ウイルス(1989年7月5日に公開されたUK2,211,504)、(アデノウイルス2のような)アデノウイルス、ウシパピローマウイルス、トリザルコーマウイルス、サイトメガロウイルス、レトロウイルス、B型肝炎ウイルスおよびシミアンウイルス40(SV40)のようなウイルスのゲノムから、または例えばアクチンプロモーターまたは免疫グロブリンプロモーター(このようなプロモーターが宿主細胞系と適合するという条件で)の異種哺乳動物プロモーターから得られた構成的プロモーターのようなプロモーターの調節下の、構成的84P2A9発現、ならびに(2)(このようなプロモーターが宿主細胞系と適合するという条件で)ecdysone、tetなどのような、誘導性ベクター系の制御下の調節発現。次いで、腫瘍容積を、触知可能な腫瘍の出現時にモニターし、そして、経時的に、84P2A9発現細胞が、より速い速度で成長するか否か、および84P2A9発現細胞により生成される腫瘍が、改変された病原性の特徴(例えば、増大した転移、血管形成、化学的治療薬物に対して減少した応答性)を示すか否かを決定する。さらに、マウスに、1×105の同じ細胞を同所的に移植し得、84P2A9が、前立腺における局所成長に対する影響、または特に肺、リンパ節、および骨髄に特異的に転移する細胞の能力に対する影響を有するか否かを決定する。
84P2A9の配列分析は、核局在化シグナルの存在を示した。84P2A9の細胞位置は、細胞生物学で広く用いられる細胞下分画技法を用いて評価され得る(Storrie Bら、Methods Enzymol.1990;182:203−25)。前立腺細胞株または精巣細胞株は、核、細胞質ゾルおよび膜画分に分離され得る。異なる画分中の84P2A9発現は、ウェスタンブロッティング技法を用いて試験され得る。
