Description
本明細書中に記載された発明は、新規な遺伝子およびそのコードされたタンパク質(24P4C12と称される)に関し、そして24P4C12を発現する種々のガン、特に前立腺癌の管理において有用な診断および治療方法および組成物に関する。
ガンは、心臓疾患(coronary disease)に次いで、第2の主なヒトの死亡原因である。全世界的に、数百万の人々が、毎年癌により死亡している。米国単独では、癌は毎年50万人をはるかに超える人々の死亡原因であり、毎年140万人あまりが新たな症例として診断されている。心臓疾患での死亡は、有意に減少している一方で、癌に起因する死亡は、一般に増加の傾向にある。来世紀の初期には、癌は死亡の主原因になると予測されている。
本発明は、複数の膜貫通領域および前立腺癌における発現により特徴付けられる、遺伝子およびタンパク質の新規なファミリーに関する。より詳細には、本発明は、新規な遺伝子およびタンパク質(24P4C12と称される)を提供する。24P4C12遺伝子は、13の膜貫通ドメインを含み、かつ12の膜貫通ドメインを含むマウスおよびC elegans遺伝子に相同性を有する710アミノ酸のタンパク質をコードする。ヒト24P4C12遺伝子のコード領域全体および部分的非コード領域のヌクレオチド配列、ならびにコードされたアミノ酸配列は、図1A〜1Dに示される(配列番号1、2)。RT−PCRおよびノザンブロッティング分析により、正常結腸、前立腺、腎臓および肺において、ならびに前立腺癌異種移植片において24P4C12の発現が示される。24P4C12タンパク質の膜貫通特性は、前立腺癌におけるその発現と合わせると、24P4C12が、例えば、インビボで24P4C12タンパク質に結合し得、かつこれを調節し得る抗体および他の低分子を用いた、前立腺癌治療の標的であることを示唆する。さらに、前立腺癌細胞の細胞表面にこれが局在するので、24P4C12タンパク質を検出し得る抗体および他の薬剤は、前立腺癌画像化法において有用であり得る。例えば、24P4C12転写産物を検出し得るポリヌクレオチドプローブおよびプライマーを使用した種々の他の分子検出アッセイはまた、前立腺癌および潜在的な他の癌を診断し、モニターし、予知し、および病期分類する際における使用を見出し得る。

(項目39) 前記癌が前立腺癌である、項目37または38に記載の使用。
(項目40) 薬学的組成物であって、該薬学的組成物は、項目10もしくは11に記載の24P4C12ポリペプチドまたはその免疫原性部分、項目26に記載のベクター、項目1に記載のポリヌクレオチドに相補的なアンチセンスポリヌクレオチド、項目1に記載のポリヌクレオチドを切断し得るリボザイム、項目13〜22のいずれか1項に記載の抗体もしくはそのフラグメントまたは組換えタンパク質、あるいは項目36に記載の24P4C12特異的結合薬剤、および必要に応じて生理学的に受容可能なキャリアを含有する、薬学的組成物。
(項目41) 24P4C12を発現する癌を有する患者を処置する方法であって、該方法は、項目26に記載のベクターを該患者に投与する工程を包含し、その結果、該ベクターが、単鎖モノクローナル抗体コード配列を該癌細胞に送達し、そして該コードされた単鎖抗体が、該癌細胞において細胞内で発現される、方法。

H38087は、NCBIにおけるtblastn手段を使用して、24P4C12アミノ酸配列を用いてdBESTデータベースを検索することによって、24P4C12のファミリーメンバーとして同定された。相同タンパク質のタンパク質フラグメントをコードするESTが同定された。これらのうちの1つであるH38087は、精巣ライブラリーからクローニングされた。cDNA(クローンGTB6)は、2738bpの大きさ(配列番号6)であり、そして11の推定膜貫通ドメインを有する(図7A〜7Dにおいて、下線を引かれ、そしてその図において、152〜220、311〜379、743〜811、830〜895、995〜1060、1133〜1201、1394〜1459、1556〜1624、1655〜1723、1859〜1924および19880〜2056として番号を付される)704のアミノ酸タンパク質(配列番号7)をコードする。5’非翻訳領域の58の塩基対は、非常にGCに富み(87%)、これは、この遺伝子は、翻訳調節エレメントを含み得ることを示す。24P4C12およびH38087のアミノ酸配列は、全体の配列に対して、44%同一であり、そして56%相同である(図8)。

mRNAおよびPSA mRNAの発現が、試験される。好ましい実施形態において、サンプルにおける24P4C12 mRNA過剰発現とPSA mRNA過剰発現との一致は、前立腺癌、前立腺癌感受性、または前立腺腫瘍の発生もしくは存在の指標を提供する。
本明細書中に開示される24P4C12タンパク質配列により、当業者は、種々の当該分野で受け入れられるプロトコルのいずれか1つを通じて、この配列と相互作用する分子を同定し得る。例えば、種々のいわゆる相互作用捕捉系(「ツーハイブリッドシステム」ともいわれる)のうちの1つを利用し得る。このような系において、相互作用する分子は、転写因子およびレポーター遺伝子の直接的な発現を再構成し、次いで、その発現をアッセイする。代表的な系は、真核生物転写アクチベーターの再構成を通じて、インビボでタンパク質間相互作用を同定し、そして例えば、米国特許第5,955,280号、同第5,925,523号、同第5,846,722号および同第6,004,746号に開示されている。
24P4C12の前立腺腫瘍関連タンパク質としての同定は、このような癌の処置に対する多くの治療アプローチを開放する。上記で議論されるように、24P4C12は、増殖シグナルの活性化または調節に関与するレセプターとして機能し、そして治療のために標的とされ得る細胞表面上にエピトープを提示することが可能である。

上記で記載または示唆された診断適用および治療適用において使用するために、本発明によって、キットもまた提供される。このようなキットは、1つ以上のコンテナ手段(例えば、バイアル、チューブなど)を厳重に閉じ込めて受容するように区画化されるキャリア手段を備え得る。コンテナ手段のそれぞれは、本方法において、使用されるべき別々の要素の1つを含む。例えば、コンテナ手段の1つは、検出可能に標識された、または検出可能に標識され得るプローブを含み得る。このようなプローブは、それぞれ24P4C12(および/またはH38087)タンパク質または24P4C12(および/またはH38087)遺伝子またはメッセージに特異的な抗体またはポリヌクレオチドであり得る。キットが、標的核酸配列を検出するために核酸ハイブリダイゼーションを利用する場合、キットはまた、標的核酸配列の増幅用のヌクレオチドを含むコンテナ、および/またはレポーター分子(例えば、酵素標識、蛍光標識、または放射性同位体標識)に結合されるレポーター手段(例えば、ビオチン結合タンパク質(例えば、アビジンまたはストレプトアビジン)を含むコンテナを有し得る。
(材料および方法)
(LAPC異種移植片)
LAPC異種移植片を、Cheales Sawyers博士(UCLA)から入手し、記載のように生成した(Kleinら、1997、Nature Med.3:402−408;Craftら、1999、Cancer Res.59:5030−5036)。アンドロゲン依存性LAPC−4異種移植片およびアンドロゲン非依存性LAPC−4異種移植片(それぞれLAPC−4 ADおよびAI)をそれぞれインタクトな雄SCIDマウスまたは雄去勢マウスにおいて増殖させ、そしてレシピエント雄に小組織塊として継代させた。LAPC−4 AI異種移植片は、LAPC−4 AD腫瘍から誘導して得た。AI異種移植片を生成するために、LAPC AD腫瘍を有する雄マウスを去勢し、そして2〜3ヶ月間飼育した。LAPC腫瘍が再増殖した後、この腫瘍を採集し、そして去勢雄SCIDマウスにおいてまたは雌SCIDマウスにおいて継代させた。
ヒト細胞株(例えば、HeLa)を、ATCCから入手し、そして5%ウシ胎児血清を有するDMEM中で維持した。
腫瘍組織および細胞株を、10ml/g組織または10ml/108細胞を用いてTrizol試薬(Life Technologies、Gibco BRL)においてホモジナイズし、総RNAを単離した。ポリA RNAを、Qiagen’s Oligotex mRNA MiniキットおよびMidiキットを用いて総RNAから精製した。総RNAおよびmRNAを、分光光度計分析(O.D. 260/280nm)によって定量し、そしてゲル電気泳動によって分析した。

SSH反応から生じる遺伝子フラグメントを増幅するために、2つのPCR増幅を行った。第一のPCR反応において、1μlの希釈最終ハイブリダイゼーション混合物を、最終容量25μlの1μlのPCRプライマー1(10μM)、0.5μl dNTP混合物(10μM)、2.5μlの10×反応緩衝液(CLONTECH)、および0.5μlの50×Advantage cDNA polymerase Mix(CLONTECH)に添加した。PCR1を、以下の条件を用いて実施した:75℃で5分、94℃で25秒、次いで27サイクルの94℃で10秒、66℃で30秒、72℃で1.5分。5つの別の第一のPCR反応を各実験について行った。産物をプールし、そして水で1:10に希釈した。第二のPCR反応については、プールし、そして希釈した第一のPCR反応物からの1μlを、プライマーNP1およびNP2(10μM)をPCRプライマー1の代わりに使用したことを除き、PCR1について用いたものと同じ反応混合物に添加した。PCR2を、10〜12サイクルの94℃で10秒、68℃で30秒、72℃で1.5分を用いて実施した。PCR産物を、2%アガロースゲル電気泳動を用いて分析した。
第一鎖cDNAを、Gibco−BRL Superscript Preamplificationシステムを用いるオリゴ(dT)12〜18プライミングによって1μgのmRNAから生成した。製造者プロトコルを使用し、そしてこれは、42℃で50分間の逆転写酵素とのインキュベーション、続いて37℃で20分間のRNAse H処理を包含した。反応を完了した後、この容積を、標準化の前に水で200μlに増大させた。16の異なる正常ヒト組織からの第一鎖cDNAを、Clontechから入手した。

半定量発現分析を、軽度のバンド強度を与えるサイクル数でPCR産物を比較することにより達成した。
上述の材料および方法に記載のSSH実験は、多数の候補遺伝子フラグメントクローン(SSHクローン)の単離に至った。全ての候補クローンを配列決定し、そして、対応する遺伝子の正体に関する情報を提供し、そして示差的な発現について特定の遺伝子を分析するための決定を導くのを補助するために、主な公的な遺伝子およびESTデータベース中の全配列に対する相同性分析に供した。一般に、検索したデータベースのいずれかにおいていかなる公知の配列とも相同性を有さず、従って新規な遺伝子を表すと考えられる遺伝子フラグメント、ならびに以前に配列決定された発現配列タグ(EST)に対して相同性を示す遺伝子フラグメントを、RT−PCRおよび/またはノーザン分析によってディファレンシャル発現分析に供した。
24P4C12遺伝子をコードする全長cDNAを、正常な前立腺のライブラリーから単離し、そして配列決定した。単離されたクローンの2つ(24P4C12−GTE9(24P4C12遺伝子のコード領域のほとんどを含む)および24P4C12−GTE5(24P4C12遺伝子のコード領域全体を含む)と称する)をそれぞれ、ATCC(Manassas、Virginia)に、プラスミドp24P4C12−GTE9およびp24P4C12−GTE5として、1999年2月2日および26日に寄託し、そしてそれぞれATCC受託番号207084および207129で承認されている。これらの2つのクローン、ならびに24P4C12コード領域のほとんどをコードする別のクローン24P4C12−GTE4は、図1A〜1D(配列番号1)に示されるような24P4C12の完全なコード配列および部分的な非コード配列を生成するように合わせられた重複するヌクレオチド配列を有した。
正常ヒト組織における24P4C12 mRNA発現を、プローブとして標識された24P4C12 SSHフラグメント(実施例1)を用いる、全部で16の異なる正常ヒト組織を含む、多重組織ブロットのノーザンブロッティングによって分析した(Clontech;Palo Alto、California)。RNAサンプルを、βアクチンプローブを用いて定量的に標準化した。ノーザンブロット分析は、主に前立腺および結腸において発現を示し、腎臓においてやや低い発現が検出され、そして膵臓、肺および胎盤において顕著により低い発現が検出された。
24P4C12に対するポリクローナル血清を生成するために、24P4C12タンパク質配列のアミノ酸1−14(MGGKQRDEDDEAYG;配列番号71)に対応するペプチドを合成した。このペプチドをキーホールリンペットヘモシアニン(KLH)にカップルさせ、そして以下のようにウサギを免疫するために用いた。最初にウサギを完全フロイントのアジュバント中に混合した200μgのペプチド−KLHで免疫した。次いで、不完全フロイントアジュバント中の200μgのペプチド−KLHを2週間毎に注射した。各免疫の後約7−10日に採血した。ELISAおよびウェスタンブロッティング分析を用いて免疫ペプチドおよび24P4C12タンパク質に対するウサギ血清の力価および特異性それぞれを測定した。アフィニティー精製した抗−24P4C12ポリクローナル抗体は、免疫ウサギからの粗血清をAffigel 15(BioRad)に共有結合した24P4C12ペプチドを含むアファニティーマトリックス上を通過させることにより調製した。PBSを用いてマトリックスを十分に洗浄した後、24P4C12ペプチドに特異的な抗体を、低pHグリシン緩衝液(0.1M、pH2.5)で溶出し、そしてPBSに対して透析した。
組換え24P4C12を発現するために、完全長の24P4C12 cDNAを、C末端に6Hisタグを提供する発現ベクター(pCDNA3.1 myc−his、InVitrogen)中にクローン化し得る。これらの構築物は、293T細胞中にトランスフェクトされ得る。トランスフェクト293T細胞溶解液は、ウェスタンブロットにおいて実施例4に記載された抗−24P4C12ポリクローナル血清でプローブされ得る。
バキュロウイルス発現系における組換え24P4C12タンパク質を生成するために、24P4C12 cDNAを、N末端にHis−タグを提供する、バキュロウイルス転移ベクターpBlueBac4.5(Invitrogen)中にクローン化する。詳細には、pBlueBac−24P4C12を、ヘルパープラスミドpBac−N−Blue(Invitrogen)とともに、SF9(Spodoptera frugiperda)昆虫細胞中に同時トランスフェクトし、組換えバキュロウイルスを生成する(詳細についてはInvitrogenの指示マニュアルを参照のこと)。次いで、バキュロウイルスを細胞上清から収集し、そしてプラークアッセイにより生成する。
H38087を、NCBI中のtblastnツールを用いて24P4C12アミノ酸配列を用いdBESTデータベースをサーチすることにより24P4C12のファミリーメンバーとして同定した。相同性タンパク質のタンパク質フラグメントをコードするESTを同定した。これらのうちの1つ、H38087は、精巣ライブラリーからクローン化した。このcDNA(クローンGTB6)は、サイズが2738bpであり、そして11の推定の膜貫通ドメインをもつ704アミノ酸タンパク質をコードする(図7A−7D;配列番号6、7)。5’非翻訳領域の58塩基対は、非常にGCリッチ(87%)であり、この遺伝子が翻訳調節エレメントを含み得ることを示す(図7A−7D)。24P4C12(配列番号2)およびH38087(配列番号7)のアミノ酸配列は、完全配列に亘って44%同一および56%相同である(図8)。発現分析は、H38087が遍在的に発現されることを示す(図9)。最高の発現レベルが精巣で検出される。発現はまた、すべてのLAPC異種移植片中で観察される。H38087は、遍在的に発現されるので、24P4C12−特異的治療剤を試験するためのコントロールとして供し得る。H38087機能に影響する24P4C12特異的治療剤は、正常細胞に対して毒性であり得る。しかし、H38087ではなく、24P4C12に選択的に影響する治療剤は、正常細胞に対してより少ない毒性であり得る。従って、H38087タンパク質は、24P4C12に対する治療様式の前臨床試験ツールとして有用である。
24P4C12が細胞中の既知のシグナル伝達経路を直接的または間接的に活性化するか否かを決定するために、ルシフェラーゼ(luc)を基礎にした転写レポーターアッセイを、24P4C12を発現する細胞中で実施する。これらの転写レポーターは、良く特徴付けられたシグナル伝達経路の下流にある、既知の転写因子のためのコンセンサス結合部位を含む。これらのレポーターおよびそれらの関連転写因子、シグナル伝達経路、および活性化刺激物の例を、以下に列挙する。
1.NFκB−luc、NFκB/Rel;Ik−キナーゼ/SAPK;成長/アポトーシス/ストレス
2.SRE−luc、SRF/TCF/ELK1;MAPK/SAPK;成長/分化

24P4C12媒介効果は、mRNA発現を示す細胞中でアッセイされ得る。ルシフェラーゼレポータープラスミドは、脂質媒介トランスフェクション(TFX−50、Promega)により導入され得るルシフェラーゼ活性、相対的転写活性の指標は、細胞抽出物のルシフェリン基質とのインキュベーションにより測定され、そして反応の化学発光がルミノメーター中でモニターされる。
24P4C12モノクローナル抗体を生成するために、24P4C12タンパク質を含むグルタチオン−S−トランスフェラーゼ(GST)融合タンパク質を合成し、そして免疫原として用いる。あるいは、24P4C12は、C末端6XHisおよびMYCタグをもつ24P4C12をコードするCMV駆動発現ベクター(pcDNA3.1/mycHIS、Invitrogen)でトランスフェクトされた293T細胞中で首尾良く発現され得る。細胞中で発現されたHISタグ24P4C12は、標準的な技法を用いるニッケルカラムを用いて精製され得る。
前立腺およびその他の癌における24P4C12の発現は、この遺伝子が腫瘍進行および/または腫瘍開始において機能的役割を有するという証拠を提供する。24P4C12が、増殖シグナルを活性化することに関与するレセプターとして機能する可能性がある。24P4C12機能は、インビトロアプローチを用いて哺乳動物細胞中で評価され得る。哺乳動物発現には、24P4C12は、pcDNA 3.1 myc−His−タグ(実施例5)およびレトロウイルスベクターpSRαtkneo(Mullerら、1991、MCB 11:1785)を含む多くの適切なベクター中にクローン化され得る。このような発現ベクターを用い、24P4C12は、PC−3、NIH3T3、LNCaPおよび293Tを含むいくつかの細胞株で発現され得る。24P4C12の発現は、抗−24P4C12抗体およびノザンブロット分析を用いてモニターされ得る(実施例4および9を参照のこと)。
腫瘍細胞増殖に対する24P4C12タンパク質の影響は、腫瘍をもつマウス中の遺伝子過剰発現によりインビボで評価され得る。例えば、SCIDマウスに、各脇腹上に、tkNeo空ベクターまたは24P4C12を含む、PC3、TSUPR1、またはDU145細胞のいずれの1×106を皮下し得る。少なくとも以下の2つの戦略が用いられ得る:(1)ポリオーマウイルス、鶏痘ウイルス(1989年7月5日に公開されたUK2,211,504)、(アデノウイルス2のような)アデノウイルス、ウシパピローマウイルス、トリザルコーマウイルス、サイトメガロウイルス、レトロウイルス、B型肝炎ウイルスおよびシミアンウイルス40(SV40)のようなウイルスのゲノムから、または例えばアクチンプロモーターまたは免疫グロブリンプロモーター(このようなプロモーターが宿主細胞系と適合するという条件で)から得られた構成的プロモーターのようなプロモーターの調節下の、構成的24P4C12発現、および(2)(このようなプロモーターが宿主細胞系と適合するという条件で)ecdysone、tetなどのような、誘導性ベクター系の制御下の調節発現。次いで、腫瘍容積を、触知可能な腫瘍の出現時にモニターし、そして、経時的に、24P4C12発現細胞が、より速い速度で成長するか否か、および24P4C12発現細胞により生成される腫瘍が、改変された病原性の特徴(例えば、増大した転移、血管形成、化学的治療薬物に対して減少した応答性)を示すか否かを測定する。さらに、マウスに、1×105の同じ細胞を同所的に移植し得、24P4C12が、前立腺における局所成長に対する影響、または特に肺、リンパ節、および骨髄に特異的に転移する細胞の能力に対する影響を有するか否かを決定する。
24P4C12の配列分析は、膜貫通ドメインの存在を示した。24P4C12の細胞位置は、細胞生物学で広く用いられる細胞下分画技法を用いて評価され得る(Storrie Bら、Methods Enzymol.1990;182:203−25)。前立腺細胞株は、核、細胞質ゾルおよび膜画分に分離され得る。異なる画分中の24P4C12発現は、ウェスタンブロッティング技法を用いて試験され得る。
