Description

本明細書中に記載される発明は、PHOR−1と呼ばれる新規の遺伝子およびそのコードされるタンパク質、ならびに診断および治療方法、ならびにPHOR−1を発現する種々の癌(特に、前立腺癌)の管理において有用である組成物に関する。

本明細書中に記載されているPHOR−1 cDNA配列は、PHOR−1遺伝子産物(単数または複数)をコードする他のポリヌクレオチドの単離、ならびにPHOR−1遺伝子産物のホモログをコードするポリヌクレオチドの単離、PHOR−1遺伝子産物の選択的スプライシングされたイソ型、対立遺伝子改変体、および変異形態の単離を可能にする。PHOR−1遺伝子をコードする全長のcDNAを単離するために使用され得る種々の分子クローニング方法が周知である(例えば、Sambrook,J.ら、Molecular Cloning:A Laboratory Manual、第2版、Cold Spring Harbor Press、New York、1989;Current Protocols in Molecular Biology、Ausubelら編、Wiley and Sons,995)。例えば、λファージクローニング方法論が、商業的に利用可能なクローニング系(例えば、λZAP Express、Stratagene)を使用して便利に使用され得る。PHOR−1遺伝子のcDNAを含有しているファージクローンが、標識されたPHOR−1 cDNAまたはそのフラグメントでプローブすることによって同定され得る。例えば、1つの実施形態においては、PHOR−1 cDNA(図1A〜D;配列番号1)またはその一部が合成され得、そしてPHOR−1遺伝子と重複しかつそれに対応する全長のcDNAを回収するためのプローブとして使用され得る。PHOR−1遺伝子自体が、ゲノムDNAライブラリー、細菌の人工染色体ライブラリー(BAC)、酵母の人工染色体ライブラリー(YAC)などを、PHOR−1 DNAプローブまたはプライマーを用いてスクリーニングすることによって単離され得る。

本発明の別の局面は、PHOR−1タンパク質およびそのポリペプチドフラグメントを提供する。本発明のPHOR−1タンパク質は、本明細書中で特に同定されるもの、ならびに、以下に概説される方法に従って過度の実験を行うことなく単離/生成されそして特徴付けられ得る程度の、対立遺伝子改変体、保存的置換改変体、およびホモログを含む。種々のPHOR−1タンパク質またはそのフラグメントの一部を組合せた融合タンパク質、ならびにPHOR−1タンパク質と異種ポリペプチドとの融合タンパク質もまた、含まれる。このようなPHOR−1タンパク質は、まとめて、PHOR−1タンパク質、本発明のタンパク質、またはPHOR−1と呼ばれる。本明細書中で使用される場合は、用語「PHOR−1ポリペプチド」は、少なくとも10アミノ酸の、好ましくは少なくとも15アミノ酸の、ポリペプチドフラグメントまたはPHOR−1タンパク質をいう。

PHOR−1またはその改変された形態をコードする核酸もまた、トランスジェニック動物または「ノックアウト」のいずれか動物を作製するために使用され得る。次いでこれは、治療的に有用な試薬の開発およびスクリーニングに有用である。トランスジェニック動物(例えば、マウスまたはラット)は、トランスジーンを含む細胞を有している動物である。トランスジーンは、動物または動物の祖先に、出産前(例えば、胚の段階)に導入された。トランスジーンは、それからトランスジェニック動物が生じる細胞のゲノム中に組み込まれるDNAである。1つの実施形態においては、PHOR−1をコードするcDNAは、PHOR−1をコードするDNAを発現する細胞を含有しているトランスジェニック動物を作製するために使用される確立された技術およびゲノム配列に従って、PHOR−1をコードするゲノムDNAをクローン化するために使用され得る。
本発明の別の局面は、PHOR−1ポリヌクレオチドおよびPHOR−1タンパク質およびそれらの変異体を検出するための方法、ならびにPHOR−1を発現する細胞を同定するための方法に関する。高度に組織に限定されたPHOR−1の発現パターンは、この分子が、転移性の疾患についての診断マーカーとして作用し得ることを示唆する。この状況においては、PHOR−1遺伝子産物の状態は、進行した段階の疾患についての罹患性、進行速度、および/または腫瘍の積極性を含む種々の因子を推定するために有用な情報を提供し得る。以下に詳細に議論されるように、患者のサンプル中のPHOR−1遺伝子産物の状態は、免疫組織化学的分析、種々のノーザンブロッティング技術(インサイチュハイブリダイゼーションを含む)、RT−PCR分析(例えば、レーザー捕捉微小解剖サンプル)、ウェスタンブロット分析、および組織アレイ分析を含む、当該分野で周知の種々のプロトコールによって分析され得る。
個体中のPHOR−1遺伝子およびPHOR−1遺伝子産物の状態を評価するアッセイは、この個体に由来する生物学的サンプルの増殖または腫瘍形成の可能性についての情報を提供し得る。例えば、PHOR−1 mRNAが、前立腺、腎臓、子宮、頚部、胃、および直腸のガンにおいてこのように高度に発現されるが、ほとんどの正常な細胞中ではそうではないので、生物学的サンプル中のPHOR−1 mRNA転写物またはタンパク質の相対的なレベルを評価するアッセイが、PHOR−1の脱調節に関連する疾患(例えば、ガン)を診断するために使用され得、そして適切な治療の選択を定義することにおいて有用な予後の情報を提供し得る。同様に、生物学的サンプル中でのPHOR−1ヌクレオチドおよびアミノ酸配列の完全性を評価するアッセイもまた、この状況において使用され得る。
本明細書中に開示されるPHOR−1タンパク質配列は、当業者が、PHOR−1と相互作用するタンパク質、低分子、および他の試薬、ならびに種々の当該分野で受容されているプロトコールの任意の1つを通じてPHOR−1によって活性化される経路を同定することを可能にする。例えば、当業者は、種々のいわゆる相互作用捕捉システム(「ツーハイブリッドアッセイ」とも呼ばれる)の1つを利用し得る。このようなシステムにおいては、相互作用する分子が、転写因子を再構成し、レポーター遺伝子の発現を指向し、次いでその発現がアッセイされる。代表的なシステムは、真核生物の転写活性化因子の再構成を通じてインビボでのタンパク質−タンパク質の相互作用を同定し、そしてこれは、例えば、米国特許第5,955,280号、同第5,925,523号、同第5,846,722号、および同第6,004,746号に開示されている。
前立腺ガンタンパク質としてのPHOR−1の同定は、前立腺ガンの処置のための多数の治療アプローチへの道を開く。上記に議論されているように、PHOR−1はG−タンパク質結合レセプター(GPCR)であり、そしてその発現は、コロニーの増殖を誘導し、そしてcAMPおよびチロシンのリン酸化を調節する。さらに、PHOR−1は、治療のために標的化され得る細胞表面のエピトープを示す。
PHOR−1の細胞表面での発現は、この分子が抗体に基づく治療ストラテジーについての魅力的な標的であることを示す。PHOR−1がガン細胞上で発現され、そしてほとんどの正常な細胞上では発現されないので、PHOR−1免疫反応性組成物の全身的な投与は、標的ではない器官および組織への免疫治療分子の結合によって引き起こされる毒性、非特異性、および/または非標的効果を伴わずに、良好な感受性を示すことが予想される。PHOR−1の細胞外ドメインと特異的に反応する抗体は、毒素もしくは治療薬との結合体として、または細胞の増殖もしくは機能を阻害し得る裸の抗体としてのいずれかで、PHOR−1を発現するガンを全身的に処置するために有用であり得る。
本発明は、PHOR−1のその結合パートナーまたはリガンドに対する結合を阻害する種々の方法および組成物、またはPHOR−1機能を阻害するための他のタンパク質(単数または複数)および方法とのその組合せを含む。PHOR−1のN末端を標的化する分子(例えば、以下の実施例に記載される抗体)は、それらがおそらく、PHOR−1上のリガンド結合部位を標的化するので、タンパク質機能を阻害するために特に魅力的である。
1つのアプローチにおいては、PHOR−1に結合し得、それによってPHOR−1がその結合パートナー(単数または複数)に対して会合/結合すること、または他のタンパク質(単数または複数)と会合することを妨げ得る、組換え分子が、PHOR−1機能を阻害するために使用される。このような組換え分子は、例えば、PHOR−1特異的抗体分子の反応性の部分(単数または複数)を含み得る。特定の実施態様においては、PHOR−1結合パートナーのPHOR−1結合ドメインが、ヒトのIgG(例えば、ヒトのIgG1)のFc部分に対して連結された2つのPHOR−1リガンド結合ドメインを含有している二量体の融合タンパク質中に操作され得る。このようなIgG部分は、例えば、CH2およびCH3ドメイン、ならびにヒンジ領域を含み得るが、CH1ドメインは含まない。このような二量体の融合タンパク質は、可溶性の形態で、PHOR−1の発現に関連しているガン(前立腺、乳房、膀胱、肺、骨、結腸、膵臓、精巣、頚、および卵巣のガンを含むがこれらに限定されない)を罹患している患者に対して投与され得る。ここでは、二量体の融合タンパク質はPHOR−1に特異的に結合し、それによって結合パートナーとのPHOR−1の相互作用をブロックする。このような二量体の融合タンパク質はさらに、公知の抗体連結技術を使用して多量体のタンパク質へと結合させられ得る。
別のアプローチにおいては、PHOR−1に特異的に結合する単鎖抗体をコードする組換えベクターが、遺伝子導入技術を通じてPHOR−1を発現する細胞中に導入され得る。ここでは、コードされる単鎖の抗PHOR−1抗体は、細胞内で発現され、PHOR−1タンパク質に結合し、そしてそれによってその機能を阻害する。このような細胞内単鎖抗体を操作するための方法は、周知である。このような細胞内抗体はまた、「イントラボディー(intrabodies)」としても公知であり、細胞内の特定の部分に対して特異的に標的化され得、それによって処置の阻害活性が集められる場所にわたって制御を提供する。この技術は、当該分野で良好に適用されている(概要については、RichardsonおよびMarasco、1995、TIBTECH、第13巻)。イントラボディーは、他の豊富な細胞表面レセプターの発現を実質的に排除することが示されている。例えば、Richardsonら、1995、Proc.Natl.Acad.Sci.USA 92:3137−3141;Beerliら、1994、J.Biol.Chem.289:23931−23936;Deshaneら、1994、Gene Ther.1:332−337を参照のこと。
別のクラスの治療アプローチにおいては、本発明は、PHOR−1遺伝子の転写を阻害するための種々の方法および組成物を提供する。同様に、本発明はまた、タンパク質へのPHOR−1 mRNAの翻訳を阻害するための方法および組成物を提供する。
遺伝子導入および遺伝子治療技術が、PHOR−1を合成する腫瘍細胞に対して治療用のポリヌクレオチド分子(すなわち、アンチセンス、リボザイム、イントラボディーをコードするポリヌクレオチド、および他のPHOR−1阻害分子)を送達するために使用され得る。多数の遺伝子治療アプローチが当該分野で公知である。PHOR−1アンチセンスポリヌクレオチド、リボザイム、PHOR−1の転写を妨害し得る因子などをコードする組換えベクターが、このような遺伝子治療アプローチを使用して腫瘍細胞を標的化するために送達させられ得る。
本発明はさらに、PHOR−1タンパク質またはそのフラグメントからなるガンのワクチン、ならびにDNAに基づくワクチンを提供する。前立腺および腫瘍に制限されたPHOR−1の発現に関して、PHOR−1のガンワクチンは、標的以外の組織に対して非特異的な影響を生じることなく、PHOR−1を発現するガンを特異的に予防しそして/または処置することにおいて有効であると予想される。抗ガン治療における使用のための体液によっておよび細胞によって媒介される免疫性を作成するためのワクチンにおける腫瘍抗原の使用は、当該分野で周知であり、そしてヒトのPSMAおよびげっ歯類のPAP免疫原を使用して、前立腺ガンにおいて使用されている(Hodgeら、1995、Int.J.Cancer 63:231−237;Fongら、1997、J.Immunol.159:3113−3137)。このような方法は、PHOR−1タンパク質もしくはそのフラグメント、またはPHOR−1をコードする核酸分子、およびPHOR−1免疫原を発現し得そしてPHOR−1免疫原を適切に提示し得る組換えベクターを使用することによって、容易に行われ得る。
上記に記載されているかまたは示唆されている診断用および治療用の適用における使用については、キットがまた本発明によって提供される。このようなキットは、閉じられた制限の中に、1つ以上の容器手段(例えば、バイアル、チューブなど)を受容させられる仕切られたキャリア手段を含み得る。それぞれの容器手段は、この方法において使用される別々のエレメントの1つを含有している。例えば、1つの容器手段は、検出可能な標識であるかまたは検出可能に標識され得るプローブを含み得る。このようなプローブは、それぞれPHOR−1タンパク質またはPHOR−1遺伝子またはメッセージについて特異的な抗体またはポリヌクレオチドであり得る。キットが標的の核酸を検出するために核酸のハイブリダイゼーションを利用する場合には、キットはまた、標的の核酸配列の増幅のためのヌクレオチド(単数または複数)を含有している容器、および/あるいはレポーター手段(例えば、レポーター分子(例えば、酵素標識、蛍光標識、または放射性同位元素標識)に対して結合させられたビオチン結合タンパク質(例えば、アビジンまたはストレプトアビジン))を含有している容器を有し得る。

(材料および方法)
(LACP異種移植片:)
LACP異種移植片を、Dr.Charles Sawyers(UCLA)から入手し、そして記載されているように作成した(Kleinら、1997、Nature Med.3:402−408;Craftら、1999、Cancer Res.59:5030−5036)。アンドロゲン依存性および非依存性のLACP−4異種移植片(それぞれ、LAPC−4 ADおよびAI)ならびにLAPC−9異種移植片(それぞれ、LAPC−9 ADおよびAI)を、インタクトな雄性のSCIDマウスまたは去勢した雄性中でそれぞれ増殖させ、そしてレシピエントの雄性においてかなりの量の小さい組織として継代した。LAPC−4 AI異種移植片は、LAPC−4 AD腫瘍から誘導し、そしてLAPC−9 AI異種移植片は、LAPC−9 AD腫瘍から誘導した。AI異種移植片を作成するために、LAPC AD腫瘍を保有している雄性のマウスを去勢し、そして2〜3ヶ月間維持した。LAPC腫瘍の再増殖後、腫瘍を回収し、そして去勢した雄性または雌性のSCIDマウス中に移した。
ヒトの細胞株(例えば、HeLa)を、ATCCから入手し、そして10%のウシの胎児の血清を有しているDMEM中で維持した。
腫瘍組織および細胞株を、Trizol試薬(Life Technologies,Gibco BRL)中で、10ml/g組織または10ml/108個の細胞を使用して、全RNAを単離するためにホモジナイズした。ポリA RNAを、QuiagenのOligotex mRNA Mini and Midiキットを使用して全RNAから精製した。全mRNAを、分光光度分析(O.D.260/280nm)によって定量し、そしてゲル電気泳動によって分析した。
以下のHPLC精製したオリゴヌクレオチドを使用した。
抑制サブトラクティブハイブリダイゼーション(SSH)を、アンドロゲン非依存性のガンと比較して、アンドロゲン依存性の前立腺ガンにおいてアップレギュレートされ得る遺伝子に対応しているcDNAを同定するために使用した。
SSH反応によって得られた遺伝子フラグメントを増幅するために、2回のPCR増幅を行った。最初のPCR反応においては、1μlの稀釈した最終のハイブリダイゼーション混合物を、25μlの最終容量中の1μlのPCRプライマー1(10μM)、0.5μlのdNTP混合物(10μM)、2.5μlの10×反応緩衝液(CLONTECH)、および0.5μlの50×Advantage cDNAポリメラーゼMix(CLONTECH)に対して、添加した。PCR1を、以下の条件を使用して行った:75℃で5分間、94℃で25秒間、次いで27サイクルの、94℃で10秒間、66℃で30秒間、72℃で1.5分間。5個の別々の最初のPCR反応を、それぞれの実験について行った。生成物をプールし、そして水で1:10に稀釈した。第2回目のPCR反応については、プールしそして稀釈した最初のPCR反応物を、プライマーNP1およびNP2(10μM)をPCRプライマー1の代わりに使用したことをのぞいて、PCR1について使用したものと同じ反応混合物に対して添加した。PCR2を、10〜12サイクルの、94℃で10秒、68℃で30秒、72℃で1.5分を使用して行った。PCR産物を、2%のアガロースゲル電気始動を使用して分析した。
第1鎖のcDNAを、1μgのmRNAからオリゴ(dT)12〜18プライミングを用いて、Gibco−BRL Superscript Preamplification systemを使用して作製した。製造業者のプロトコールを使用し得、そしてこのプロトコールは、逆転写酵素を用いた42℃で50分間のインキュベーション、続く20分間の37℃でのRNase H処理を含んだ。反応の完了後、容量を、較正の前に水を用いて200μlに増大させた。16個の異なる正常なヒトの組織に由来する第1鎖のcDNAを、Clontechから入手した。
いくつかのSSH実験を、材料および方法(上記)に記載するように行い、そして多数の候補の遺伝子フラグメントクローンの単離を導いた。全ての候補のクローンを配列決定し、そして、対応する遺伝子の正体についての情報を提供するため、および異なる発現について特定の遺伝子を分析するための指針を補助するために、主要な一般的な遺伝子およびESTデータベース中の全ての配列に対する相同性分析に供した。一般的には、任意の検索されるデータベース中のいずれの公知の配列遺伝子フラグメントに対しても相同性を有さず、従って、新規の遺伝子を提示すると考えられる遺伝子フラグメント、ならびに以前に配列決定された発現される配列タグ(EST)に対して相同性を示す遺伝子フラグメントを、RT−PCRおよび/またはノーザン分析による異なる発現の分析に供した。
3136bpの全長のcDNA(GTH10)を、前立腺ライブラリーから単離し、これによって、317アミノ酸のORFを明らかにした(図1A〜D)。タンパク質配列は、7個の膜貫通ドメインを明らかにし、そしてこれは嗅覚に関係しているGタンパク質結合レセプター(GPCR)に対して相同性を有する(Ramingら、1993、Nature 361:353;Malnicら、1999、Cell 96:713)。101P3A11に対して最も相同である配列は、RA1cとして公知の脳中で発現されるラットの嗅覚レセプターである(Ramingら、1998、Receptor Channels 6:141)。これは、299の残基の重複でRA1cと59.9%同一である。RA1cのヒトホモログはおそらく、HPRAJ70であり、これは、Human Genome Siences(第US5756309号、第WO96/39435号)によって同定された前立腺特異的GPCRである。101P3A11と共に両方の遺伝子のアラインメントを図2に示す。全長の101P3A11 cDNAは、298の残基の重複でHPRAJ70に対して59.4%同一であるアミノ酸配列をコードする。脳の嗅覚レセプターとの101P3A11の相同性は、本発明者らがこの遺伝子を嗅覚レセプター−1の前立腺ホモログ(Prostate Homologue of Olfactory Receptor−1(PHOR−1))と命名することを導いた。このレセプターのファミリーのメンバーであるタンパク質は、細胞外アミノ末端、3個のさらなる細胞外ループ、3個の細胞内ループ、および細胞内カルボキシル末端を示す。PHOR−1の第2の細胞外領域は、残基90(NST)で1つの可能性のあるNグリコシル化部位を示し、このことは、このタンパク質がグリコシル化され得ることを示唆している。
正常ヒト組織中でのPHOR−1 mRNAの発現を、最初に、全部で16個の異なる正常ヒト組織を含有している、2つの複数の組織ブロット(Clontech;Palo Alto,California)のノーザンブロッティングによって、プローブとして標識した101P3A11 SSHフラグメント(実施例1)を使用して、分析した。RNAサンプルを、β−アクチンプローブを用いて定量的に較正した。この分析の結果を図6A〜Cに示す。3.5kbの転写物の発現を、正常な前立腺中でのみ検出した。
(前立腺ガンの異種移植片および臨床的な標本のノーザン分析)
前立腺ガン組織中でのPHOR−1の発現を分析するために、ノーザンブロッティングを、LAPC異種移植片に由来するRNAおよび前立腺ガンのサンプルのパネルに由来するRNAについて、それらの適合する正常な隣接している前立腺組織とともに行った。結果は、LAPC−4 AD、LAPC−9 AD、およびLAPC−9 AI中での高いレベルのPHOR−1の発現を示す。より低い発現が、正常な前立腺中で検出され、そしてLAPC−4 AI中では発現は見られない(図8)。前立腺ガンの患者に由来する臨床的な標本の4個の正常/腫瘍の適合する対の分析は、全ての患者のサンプル中でPHOR−1の発現を示した(図8)。興味深いことに、4個の適合する対のうちの3個において、PHOR−1の発現は、適合する正常な隣接している組織と比較して腫瘍サンプル中で5〜20倍高かった。これらの結果は、前立腺特異的PHOR−1がガンにおいてアップレギュレートされ、そして前立腺ガンの病因において機能的な役割を有し得ることを示唆する。
ノーザン分析およびドットブロット分析は、前立腺ガンの腫瘍の隣接している正常な組織と比較した場合に、10個の前立腺ガンの腫瘍のうちの8個において(Gleasonスコア6から9)、PHOR−1のアップレギュレーションを示した(図9A〜B)。患者に由来する増幅したcDNA(Clontech,CA)を使用するドットブロットは、3/3の前立腺ガンの患者、6/14の腎臓ガンの患者,2/8の子宮ガンの患者、1/1の頚ガンの患者、3/8の胃ガンの患者、および7/7の直腸ガンの患者において、PHOR−1のアップレギュレーションを示す(図10)。
アンチセンスPHOR−1リボプローブを使用するRNAインサイチュ分析は、正常な前立腺(4/4)、PIN(1/1)、および前立腺ガン(6/6)の患者において、有意な腺状の上皮および基底細胞の発現を示した。PHOR−1センスリボプローブは、わずかな染色から全く染色なしまでを有した。PINおよび前立腺ガンにおけるRNAのインサイチュ染色を、図11A〜Bにッ示す。ガン細胞中での染色強度は、一般的には、正常な腺において観察される染色強度よりも高かった(図12A〜B)。RNAのインサイチュの結果はまた、前立腺組織中で観察される発現が、腺上皮、基底細胞、およびガン細胞中であることを実証する。
(pGEX構築物)
細菌細胞中でPHOR−1を発現させるために、PHOR−1の一部を、pGEX−2TまたはpGEX−6P−1(Amersham Pharmacia Biotech,NJ)中にクローニングすることによって、グルタチオンS−トランスフェラーゼ(GST)遺伝子に対して融合させた。全ての構築物を、C末端の6個のヒスチジンエピトープを有するかまたは有さないで、N末端に融合したGSTとともに組換えのPHOR−1タンパク質配列を生じるように作製した。6個のヒスチジンエピトープタグを、ORFの3’末端でクローニングプライマーにヒスチジンコドンを付加することによって、作製した。トロンビンまたはPreScissionTM認識部位によって、PHOR−1またはpGEX−2TおよびpGEX−6P−1構築物のぞれぞれからのGSTタグの切断を可能にする。アンピシリン耐性遺伝子およびpBR322起点によって、E.coliにおけるプラスミドの選択および維持を可能にする。以下のPHOR−1のフラグメントを、pGEX−6P−1中にクローン化した: アミノ酸128から238 アミノ酸188から317 アミノ酸100から295。
細菌細胞中でPHOR−1を発現させるために、PHOR−1の一部を、マルトース結合タンパク質(MBP)遺伝子に対して、pMAL−c2XおよびpMAL−p2X(New England Biolabs,MA)中にクローニングすることによって融合させた。全ての構築物を、N末端に融合させたMBPおよびC末端に6個のヒスチジンエピトープを有する組換えPHOR−1タンパク質配列を生じるように作製した。6個のヒスチジンエピトープタグを、3’クローニングプライマーに対してヒスチジンコドンを付加することによって作製した。第Xa因子認識部位によって、PHOR−1からのGSTタグの切断を可能にする。pMAL−c2XおよびpMAL−p2Xベクターを、それぞれ、細胞質または細胞質周辺中の組換えタンパク質を発現するように最適化する。細胞質周辺での発現は、ジスルフィド結合を伴うタンパク質の折り畳みを増強する。PHOR−1のアミノ酸86から310を、pMAL−c2XおよびpMAL−p2X中にクローン化した。
(pcDNA4/HisMax−TOPO構築物)
哺乳動物の細胞中でPHOR−1を発現させるために、951bpのPHOR−1 ORFを、pcDNA4/HisMax−TOPO Version A(cat#k864−20、Invitrogen、Carlsbad,CA)中にクローン化した。タンパク質の発現は、サイトメガロウイルス(CMV)プロモーターおよびSP163翻訳エンハンサーから駆動される。組換えタンパク質は、N末端に融合されたXpressTMおよび6個のヒスチジンエピトープを有する。組換えタンパク質のC末端は、終止コドンの前にベクター配列によって生じる28個のアミノ酸の融合を有する。pcDNA4/HisMax−TOPOベクターはまた、エピソームの複製のためのSV40起点およびラージT抗原を発現する細胞株中での単純なベクターレスキューとともに、mRNAの安定性を増強するために、ウシ成長ホルモン(BGH)ポリアデニル化シグナルおよび転写終結配列を含む。ゼオシン耐性遺伝子は、タンパク質およびアンピシリン耐性遺伝子を発現する哺乳動物細胞の選択を可能にし、そしてColE1起点は、E.coli中でのプラスミドの選択および維持を可能にする。

構成的にPHOR−1を発現する哺乳動物細胞株を作製するために、951bpのPHOR−1 ORFをpSRα構築物中にクローン化し、そして安定な細胞株を作製した。両栄養性およびエコトロピックなレトロウイルスを、pSRα構築物の293T−10A1パッケージング株中へのトランスフェクション、またはpSRαとヘルパープラスミド(j−)の293細胞中への同時トランスフェクションによって、それぞれ作製する。レトロウイルスは、種々の哺乳動物細胞株を感染させるために使用し得、それによってクローン化した遺伝子であるPHOR−1の宿主細胞株中への組込みを生じる。タンパク質の発現は、長末端反復(LTR)から駆動される。ネオマイシン耐性遺伝子は、タンパク質およびアンピシリン耐性遺伝子を発現する哺乳動物細胞の選択を可能にし、そしてColE1起点は、E.coli中でのプラスミドの選択および維持を可能にする。抗FLAG抗体を使用する検出を可能にするためにC末端にFLAGタグを融合させたさらなるpSRα構築物を作製した。FLAG配列(5’gat tac aag gat gac gac gat aag 3’)(配列番号33)をORFの3’末端のクローニングプライマーに付加した。
バキュロウイルス発現系で組換えPHOR−1タンパク質を作製するために、PHOR−1 cDNAを、バキュロウイルス転移ベクターpBlueBac 4.5 (Invitrogen)中にクローン化する。これは、N末端にHisタグを提供する。詳細には、pBlueBac−−PHOR−1を、組換え体のバキュロウイルスを作製するために、SF9(Spodoptera frugiperda)昆虫細胞中に、ヘルパープラスミドpBac−N−Blue(Invitrogen)とともに同時トランスフェクトする(詳細については、Invitrogenの説明マニュアルを参照のこと)。次いで、バキュロウイルスを、細胞上清から回収し、そしてプラークアッセイによって精製する。
3個の免疫原を、PHOR−1に特異的に結合する抗体試薬を作製するために誘導した。2個の抗原は、PHOR−1タンパク質配列のアミノ酸1〜14(MVDPNGNESSATYF;配列番号8)およびアミノ酸262〜274(VHRFSKRRDSPLP;配列番号9)をコードするペプチドであった。これらのペプチドは、それぞれ、PHOR−1タンパク質の細胞外N末端およびPHOR−1タンパク質の推定の6番目と7番目の膜貫通ドメインの間の細胞外ループをコードすると推定される。これはリガンド結合部位と予想される。第3の免疫原が、PHOR−1タンパク質配列のアミノ酸86〜310を含有しているグルタチオン−S−トランスフェラーゼ(GST)融合タンパク質であった。この融合タンパク質を、以下のプライマー(それぞれ、配列番号34および配列番号35)を用いて、PHOR−1のcDNAクローンのヌクレオチド388〜1062のPCRによって媒介される増幅によって作製した:
得られた生成物を、pGEX−2T GST融合ベクター(Pharmacia)のEcoRIおよびXhoI制限部位中にクローン化した。組換えのGST−PHOR−1融合タンパク質を、グルタチオンセファロース親和性クロマトグラフィーによって、誘導した細菌から精製した。
PHOR−1に対するポリクローナル血清を作製するために、キーホールリンペットヘモシアニン(KLH)に対して結合させた精製したGST−融合タンパク質およびペプチドを、以下のように個体のウサギを免疫化するために使用した。ウサギを、200μgの融合タンパク質または完全フロイントアジュバント中で混合したKLH−ペプチド抗原を用いて免疫化した。次いで、ウサギに、不完全フロイントアジュバント中の200μgの免疫原を用いて2週間ごとに注射した。試験採血を、それぞれの免疫化の約7〜10日後に行った。それぞれのペプチド抗血清の力価は、それぞれの免疫原に対してELISAによって決定した場合には、少なくとも1×105であった。GST−融合血清の力価は、少なくとも1×106であった。
PHOR−1に対するmAbを作製するために、Balb Cマウスを、200μgの完全フロイントアジュバント中に混合したGST−PHOR−1融合タンパク質を用いて腹腔内で免疫化した。次いで、引き続いて、マウスを、フロイント不完全アジュバント中に混合した200μgの抗原を用いて2〜4週間ごとに免疫化した。3回の免疫化後には、これらのマウスに由来する試験採血の力価は、ELISAによって決定した場合には、少なくとも1.2×106であり、そしてGST融合体中に存在する同じアミノ酸をコードするMBP融合タンパク質を使用するウェスタンブロッティングによって示されるように、PHOR−1アミノ酸配列を特異的に認識した(図15)。LNCaP細胞およびLAPC9異種移植片細胞のフローサイトメトリー分析は、それらの同種の予備免疫血清と比較して免疫化したマウスの血液の組合せを用いて染色した場合に、蛍光のシフトを示し、このことは、PHOR−1タンパク質の細胞表面での検出を実証する(図16A〜B)。
293T細胞を、発現プラスミドpCDNA4 HIS/MAX(Invitrogen)で一過性にトランスフェクトした。pCDNA4 HIS/MAX中では、PHOR−1cDNAが、アミノ末端で、2つのエピトープタグ、Express、およびHisGに融合されている。空のベクターコントロールおよびPHOR−1でトランスフェクトした細胞を、N末端のExpressエピトープを特異的に認識するmAbでか、またはPHOR−1配列のN末端の14個のアミノ酸に対して指向されたウサギのpAbで染色した。図13Aおよび図14A〜Bに示すように、PHOR−1でトランスフェクトした細胞は、抗Express mAbおよびpAbの両方を用いたコントロール細胞と比較して、特異的な蛍光のシフトを示す。このシフトは、PHOR−1が原形質膜の外側に暴露されたN末端を伴って細胞の表面で発現されることを示す。これは、公知のGタンパク質結合レセプターのトポロジーと一致する。これらの細胞によるPHOR−1の発現をさらに、免疫組織化学によって確認した(次の実施例を参照のこと)。
(方法)
ホルマリンで固定し、パラフィンに包埋した組織のブロックを、4ミクロンに切片化し、そして正に荷電させたCapillary Gap顕微鏡スライド(Ventana Medical Systems,Inc.,Tucson,AZ)上に配置した。キシレン中での脱蝋、続くアルコール系列による水和の後、組織の切片を、クエン酸ナトリウム(10mM、pH6.0)の存在下で20分間、蒸し器の中で前処理し、続いて抗体反応を最適化するために、10分間のプロテイナーゼK(1:40)インキュベーションした。5分間の冷却後、スライドを、ビオチン−ストラプトアビジン−ペルオキシダーゼ技術を使用して免疫染色した。簡潔には、スライドを、5分間、ブロッキング血清(正常なヤギ)、続いて、2μl/mlの抗PHOR−1ウサギモノクローナル一次抗体(25分)、ビオチニル化した二次抗体ヤギ抗ウサギIgG(25分)、内因性ペルオキシダーゼブロッキング(3×1.5分)、およびペルオキシダーゼ酵素に結合させたストレプトアビジン複合体(Vector Labs,Burlingame,CA)(10分)中でインキュベートした。各インキュベーションとインキュベーションとの間に切片を緩衝液中でリンスした。DAB−ジアミノベンジジンクロモゲン(QualTek Molecular Labs)を、反応を進行させるために使用した−これによって、茶色の沈殿を生じた。引き続きスライドを、ヘマトキシリンで対比染色し、そしてカバースライドをかけた。
PHOR−1タンパク質の内因性発現が、抗PHOR−1(PEPTIDE1:アミノ酸1〜14)、ウサギのポリクローナル抗体(図18A〜F)の免疫組織化学的分析において実証される。前立腺ガンでの染色は、正常な前立腺において観察される染色よりも多かった。染色は、正常な前立腺の内腔の上皮内の先端に局在化される(図18Eおよび18F)。前立腺ガンで観察される染色はまた、中程度の段階のガンについては低く先端に局在化され(図18Bおよび18C)、そしてより進行した前立腺ガンの細胞の全てにわたって局在化される(図18A)。前立腺ガン細胞株LNCaPもまた、ほとんど全ての細胞において同様の染色を示す(図18Dおよび19F)。
pSRαレトロウイルスベクター中のneoまたはPHOR−1のいずれかを安定に発現するPC3細胞を、1%のFBS中で一晩増殖させた。次いで、細胞を、未処理のままにしたか、または10%のFBSで3分間処理した。細胞を溶解させ、そして抗ホスホチロシン(UBI、Lake Placid,NY)(図20A)、または抗ホスホロ−ErK(Cell Signal、Beverly,MA)mAb(図20B)を用いたウェスタンブロッティングによって分析した。抗Grb2 mAb(Transduction Laboratories,San Diego,CA)のオーバーレイは、等しいタンパク質の充填を示す(図20C)。図20Dにおいて、PHOR−1の発現を、ノーザンブロッティングによって評価した(PC3−PHOR−1細胞によるPHOR−1の発現の免疫組織化学的実証については、図19Dもまた参照のこと)。RNAを、コントロールのPC3−neo細胞およびPHOR−1で安定に形質導入したPC3細胞から抽出し、そしてRNAのブロットをPHOR−1プローブ(クローンGTH10のXba−Ecor1フラグメント)を使用してハイブリダイズした。LAPC4異種移植片に由来するRNAを、ポジティブコントロールとして使用した(図20E)。結果は、PHOR−1 mRNAが、レトロウイルスで形質導入されたPC3−PHOR−1細胞中で発現されるが、コントロール細胞中では発現されないこと、そして一旦発現されると、PHOR−1はPC3細胞のリン酸化のパターンを変化させることを示す。
親の細胞およびPHOR−1を発現する細胞を、cAMPの細胞質での蓄積を誘導するそれらの能力について比較した。293T細胞を、空のpcDNA4 HIS MAXベクターまたはpcDNA4 HIS MAX PHOR−1でトランスフェクトした。細胞を、1%のウシ胎仔血清(FBS)中で一晩飢餓させ、そして培地のみ、または10%のFBSの存在下でインキュベートした。細胞を溶解させ、そしてcAMP含有量について、製造業者の推奨(Linco Research,St Charles,MI)に従って酵素結合イムノアッセイ(EIA)によって分析した。結果を表1に示し、そしてこれはPHOR−1の発現がFBSに応答してcAMP濃度を変化させることを示す。
嗅覚のレセプターを含む、全ての特徴付けたGPCRは、cAMP経路を活性化することによって機能する。リガンドの非存在下では、GPCRは通常は、不活性な状態である。リガンドの結合または過剰発現の際に、GPCRは活性な立体構造を獲得し、そしてGタンパク質と複合体を形成する。この相互作用は、Gタンパク質サブユニットの解離およびアデニル酸シクラーゼの活性化を生じ、これによってcAMPの蓄積を生じる(Birnbaumer L,Cell 1992、71:1069)。cAMPの増強された産生は、GPCRの効果を媒介するいくつかの下流のシグナル伝達経路の活性化を生じる。293T細胞中でのPHOR−1の発現がFBSに応答してcAMPの蓄積を可能にするこの実施例における実証は、これらの条件下でPHOR−1がGPCRとして機能することを示す。
PHOR−1を安定に発現するNIH−3T3細胞を、軟寒天中でコロニーを形成するそれらの能力について分析した。neoまたは活性化Rasを安定に発現するNIH−3T3細胞を、ネガティブコントロールおよびポジティブコントロールとして、それぞれ使用した。実験を、2連で行った。アッセイを、細胞のプレーティングの4週間後に評価した。結果を、図21および表2に示す。
コロニーの計数は、PHOR−1がneoコントロールと比較してコロニーの形成において3倍の増大を誘導することを示す。この有意な増大は、2つの別々の実験において観察された。これらの結果は、NIH3T3細胞中でのPHOR−1の発現が、強力なオンコジーンであるRasによる5倍の増大と比較して、コロニーの形成において3〜4倍の増大を誘導することを示し、このことは、PHOR−1が有意な形質転換能力を有することを示唆している。
PHOR−1の染色体配置を、GeneBridge4放射線照射ハイブリッドパネル(Walterら、1994、Nat.Genetics 7:22)(Research Genetics,Huntsville A1)を使用して決定した。以下のPCRプライマーを、PHOR−1を配置するために使用した(それぞれ、配列番号31および配列番号32):
このベクター、およびhttp://www−genome.wi.mit.edu/cgi−bin/contig/rhmapper.plでのマッピングプログラムによって、11p15.5で、染色体11のテロメアにPHOR−1を配置した。
PHOR−1が細胞中の公知のシグナル伝達経路を直接または間接的に活性化するかどうかを決定するために、ルシフェラーゼ(luc)に基づく転写レポーターアッセイを、PHOR−1を発現する細胞中で行う。これらの転写レポーターは、十分に特徴付けられたシグナル伝達経路の下流に存在する公知の転写因子についてのコンセンサスな結合部位を含む。レポーターおよびそれらの会合する転写因子の例、シグナル伝達経路、および活性化の刺激を以下に列挙する。
前立腺ガン中でのPHOR−1の発現は、この遺伝子が腫瘍の進行および/または腫瘍の開始において機能的な役割を有するという証拠を提供する。レセプターとしてのPHOR−1の機能が、増殖シグナルを活性化することに関係することは可能である。PHOR−1機能を、インビトロでのアプローチを使用して哺乳動物細胞中で評価し得る。哺乳動物での発現のために、PHOR−1を多数の適切なベクター(pcDNA 3.1 myc−His−tagおよびレトロウイルスベクターpSRαtkneo(Mullerら、1991、MCB 11:1785)を含む)中にクローン化し得る。このような発現ベクターを使用して、PHOR−1を、いくつかの細胞株(PC−3、NIH 3T3、LNCaP、および293Tを含む)中で発現させ得る。PHOR−1の発現を、抗PHOR−1抗体およびノーザンブロット分析を使用してモニターし得る。
PHOR−1タンパク質の腫瘍細胞の増殖に対する効果を、腫瘍を保有しているマウス中での遺伝子の過剰発現によってインビボで評価し得る。例えば、SCIDマウスを、tkNeo空ベクターまたはPHOR−1を含有している、PC細胞3、TSUPR1細胞、またはDU145細胞のいずれかの1×106を用いて、いずれかの側腹部に皮下注射し得る。少なくとも2つのストラテジーを使用し得る:(1)ポリオーマウイルス、鶏痘ウイルス(1989年7月5日に公開された、第UK 2,211,504号)、アデノウイルス(例えば、アデノウイルス2)、ウシパピローマウイルス、鳥類の肉腫ウイルス、サイトメガロウイルス、レトロウイルス、B型肝炎ウイルス、およびSV40のようなウイルスのゲノムから、または異種の哺乳動物プロモーター(例えば、アクチンプロモーター、またはイムノグロブリンプロモーター)(そのようなプロモーターが宿主細胞システムと適合性である限りは)から得られる構成的なプロモーターのようなプロモーターの調節下での、構成的なPHOR−1の発現、ならびに(2)誘導可能なベクターシステム(例えば、エジクソン、tetなど)(そのようなプロモーターが宿主細胞システムと適合性である限りは)の制御下での調節された発現。次いで、腫瘍の容量を、PHOR−1を発現する細胞がより速い速度で増殖するかどうか、およびPHOR−1を発現する細胞によって産生された腫瘍が、変更された積極性(例えば、増強された転移性、血管形成、化学療法剤に対する減少した応答性)の特徴を示すかどうかを決定するために、明確な腫瘍の出現時点およびその後に経時的にモニターし得る。さらに、マウスに、PHOR−1が、前立腺中での局所的な増殖、または細胞が転移する(特に、肺、リンパ節、および骨髄に対して)能力に対して影響を有するかどうかを決定するために、正常位で同じ細胞の1×105を用いて移植し得る。
PHOR−1は、嗅上皮およびニューロン中で発現される嗅覚のレセプターの大きなファにリーに対して相同である。PHOR−1と相同であるさらなる遺伝子を同定する試みにおいては、PHOR−1のタンパク質配列を、公のEST(発現配列タグ)データベース(dBest)中のファミリーのメンバーを同定するための電子的なプローブとして使用した。NCBI(National Center for Biotechnology Information)の「tblastn」機能を使用して、dBestデータベースを、PHOR−1タンパク質配列を用いて問い合わせした。この分析によって、1つの新規のファミリーのメンバーを明らかにした(図22)。EST、AI138213を、ヒトの胎盤のライブラリーから単離し、そしてこれは、PHOR−1のカルボキシル末端領域に相同である。これは、95個のアミノ酸の重複にわたってPHOR−1と49.5%の同一性を示す。この新規のファミリーのメンバーを、前立腺および前立腺癌のサンプル中での発現について分析し、そしてヒトのcDNAライブラリーからクローン化する。
ここで、(A)はアデニンを示し、(C)はシトシン、(G)はグアニン、(T)はチミン、(R)はアデニンまたはグアニン、(Y)はシトシンまたはチミン、(S)はシトシンまたはグアニン、(D)はアデニンまたはグアニンまたはチミン、(H)はアデニンまたはシトシンまたはチミン、(N)はアデニンまたはグアニンまたはシトシンまたはチミンを示す。
Claims (5)
- 配列番号:2のPHOR−1タンパク質に免疫反応性の抗体又はその抗原結合性フラグメントであって、ガン細胞に対して毒性の治療薬に結合された、前記抗体又はその抗原結合性フラグメント。
- 細胞に対して毒性の治療薬が放射性同位元素、化学療法剤、及び毒素からなる群より選択される、請求項1記載の抗体又はそのフラグメント。
- 放射性同位元素が131I、90Y、186Re、及び212Biからなる群より選択され、
化学療法剤がタキソール、アクチノマイシン、マイトマイシン、エトポシド、テノポシド、ビンクリスチン、ビンブラスチン、コルヒチン、ゲロニン、及びカリヒマイシンからなる群より選択され、
毒素がジフテリア毒素、エノマイシン、フェノマイシン、Pseudomonas外毒素(PE)A、PE40、アブリン、アブリンA鎖、ミトゲリン、モデシンA鎖、及びα−サルシンからなる群より選択される、請求項2記載の抗体又はそのフラグメント。 - 以下の工程を包含する、試験組織におけるガンの存在の有無を決定する方法:
(a)該試験組織のサンプルにおける配列番号:2のPHOR−1タンパク質又はそれをコードするmRNAのレベルを決定する工程;
(b)ガンの存在しない対応する組織における該タンパク質又は該mRNAのレベル値を提供する工程;及び
(c)工程(b)で提供されたレベルに対して工程(a)で決定されたタンパク質又はmRNAのレベルを比較する工程であって、
それにより、(b)と比較した(a)のレベルが大きい場合は該試験組織にガンが存在することを示し、(b)と比較した(a)のレベルが大きくない場合は該試験組織にガンが存在しないことを示す、前記方法。 - PHOR−1タンパク質のレベルが、サンプルをPHOR−1タンパク質と特異的に結合する媒介物(agent)に接触させる工程、及び該サンプルに対する該媒介物の結合のレベルを検出する工程、を含む方法によって決定される、請求項4に記載の方法であって、該媒介物が抗体又はその抗原結合性フラグメントである、前記方法。
