【解決手段】 マイコバクテリウム・ツベルクローシス菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、かつ特定の配列を有する塩基配列に含まれる連続する少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・ツベルクローシス菌の同定方法。

好ましくは、核酸増幅反応時の副反応生成物はピロリン酸である。
好ましくは、乾式分析素子を用いてピロリン酸の検出を行う。
本発明の抗酸菌属の検出方法は、各菌種に特異的な配列を3'末端に有するプライマーを用いることを特徴とする。本発明の方法を利用することにより、抗酸菌属を同定することが可能である。本発明の方法の好ましい形態では、各菌に特異的なプライマーを用いて核酸増幅反応を行わせ、その伸長反応の有無を検出する。検出する方法の具体例として、電気泳動、質量分析、液体クロマトグラフィー等の増幅産物を直接測定する方法や、ポリメラーゼ伸長反応の際、生成するピロリン酸を検出する方法が挙げられる。
菌特異的なプライマーを配列表に示す配列の少なくとも3塩基を3'末端に含むよう設定する。プライマーを2本以上設定する場合、1本がその部位であればよい。そして、上記プライマーを用いてポリメラーゼ伸長反応を行わせる。
本発明において使用する各抗酸菌に特異的と特異的なプライマーは、16SrRNA遺伝子配列の中で、各種間の遺伝子配列が異なる可変領域(およそ130番目から200番目)である。本発明で使用するプライマーの好ましい塩基数は5〜60塩基である。特に好ましくは15〜40塩基である。
本発明の方法で行う核酸の増幅には、これまで開発されてきた各種の方法を使用することができる。本発明で用いることができる核酸増幅法としては、PCR(特公平4−67960号、特公平4−67957号)、LCR(特開平5−2934号)、SDA(Strand Displacement Amplification:特開平5−130870号)、RCA(Rolling Circle Amplification:Proc.Natl.Acad.Sci, Vol.92, 4641-4645 (1995))、ICAN(Isothermal and Chimeric primer-initiated Amplification of Nucleic Acids)、LAMP(Loop-Mediated Isothermal Amplification of DNA;Bio Industry, 第18巻、2号(2001))、NASBA(Nucleic acid Sequence-based Amplification method; Nature, 350, 91 (1991))、及びTMA(Transcription mediated amplification method; J.Clin Microbiol.第31巻、3270(1993))等が挙げられる。
本発明の方法は、菌種同定を行うことが可能な核酸増幅用プライマーを用いているので、検出手段は、増幅産物量が定量できるものであれば特に制限されない。

本発明において使用することのできる乾式分析素子は、一層または複数層の機能層からなる分析素子であって、その少なくとも一層(または複数の層に渡って)に検出試薬を含有させ、層内での反応により生じた生成色素を、分析素子の外から反射光あるいは透過光により比色定量するものである。
本発明で使用することのできるピロ燐酸定量用乾式分析素子は、公知の多種の乾式分析素子と同様の層構成とすることができる。乾式分析素子は、前記(C)項(検出)における、式2または式3の反応を行うための試薬の他、支持体、展開層、検出層、光遮蔽層、接着層、吸水層、下塗り層その他の層を含む多重層としてもよい。このような乾式分析素子として、例えば特開昭49−53888号公報(対応米国特許3,992,158)、特開昭51−40191号公報(対応米国特許4,042,335)、及び特開昭55−164356号公報(対応米国特許4,292,272)、特開昭61−4959号公報(対応EPC公開特許0166365A)の各明細書に開示されたものがある。
Piを定量検出するための一連の反応における最後の「呈色反応」に用いる酵素に応じて、ペルオキシダーゼ(POD)を用いる方法とグルコース−6−燐酸デヒドロゲナーゼ(G6PDH)を用いる方法がある。以下、これらの方法の具体例を説明する。

無機燐(Pi)を、プリンヌクレオシドホスホリラーゼ(PNP)により、イノシンと反応させ、生じたヒポキサンチンをキサンチンオキシダーゼ(XOD)により酸化して尿酸を生成する。この酸化過程で生じる過酸化水素(H2O2)を用いて、ペルオキシダーゼ(POD)により、4−アミノアンチピリン(4−AA)とフェノールとを酸化縮合させてキノンイミン色素を形成し、これを比色する。
無機燐(Pi)、コカルボキシラーゼ(TPP)、フラビンアデニンジヌクレオチド(FAD)、Mg2+の存在下で、ピルビン酸をピルビン酸オキシダーゼ(POP)により酸化してアセチル酢酸を生成する。この酸化過程で生じる過酸化水素(H2O2)を用いて、上記(1−1)の場合と同様に、ペルオキシダーゼ(POD)により、4−アミノアンチピリン(4−AA)とフェノールとを酸化縮合させてキノンイミン色素を形成し、これを比色する。
(2−1)
無機燐(Pi)とグリコーゲンとをホスホリラーゼを用いて反応させ、グルコース−1−燐酸(G−1−P)を生成させる。生じたグルコース−1−燐酸をホスホグルコムターゼ(PGM)により、グルコース−6−燐酸(G−6−P)にする。グルコース−6−燐酸とニコチアミドアデニンジヌクレオチド(NAD)との存在下、グルコース−6−燐酸デヒドロゲナーゼ(G6PDH)により、NADを還元してNADHにし、これを比色する。
無機燐(Pi)とマルトースとをマルトースホスホリラーゼ(MP)を用いて反応させ、グルコース−1−燐酸(G−1−P)を反応させる。以下、上記(2−1)と同様に、生じたグルコース−1−燐酸をホスホグルコムターゼ(PGM)により、グルコース−6−燐酸(G−6−P)にする。グルコース−6−燐酸とニコチアミドアデニンジヌクレオチド(NAD)との存在下、グルコース−6−燐酸デヒドロゲナーゼ(G6PDH)により、NADを還元してNADHにし、これを比色する。
酸性下で無機燐(燐酸塩)と水溶性モリブテン酸イオンとを錯化させた「燐モリブテン酸塩(H3[PO4Mo12O36])を直接定量する「直接法」と、上記直接法の反応に続いて、還元剤により、Mo(IV)からMo(III)として、モリブテン青(Mo(III))を定量する「還元法」とがある。水溶性モリブテン酸イオンの例としては、モリブテン酸アルミニウム、モリブテン酸カドミウム、モリブテン酸カルシウム、モリブテン酸バリウム、モリブテン酸リチウム、モリブテン酸カリウム、モリブテン酸ナトリウム、モリブテン酸アンモニウムなどが挙げられる。還元法で使用される代表的な還元剤の例としては、1,2,4アミノナフトールスルホン酸、硫酸第一鉄アンモニウム、塩化第一鉄、塩化第一スズ−ヒドラジン、硫酸−p−メチルアミノフェノール、N,N−ジメチル−フェニレンジアミン、アスコルピン酸、マラカイトグリーンなどが挙げられる。
(1) 支持体上に試薬層を有するもの。
(2) 支持体上に検出層、試薬層をこの順に有するもの。
(3) 支持体上に検出層、光反射層、試薬層をこの順に有するもの。
(4) 支持体上に第2試薬層、光反射層、第1試薬層をこの順に有するもの。
(5) 支持体上に検出層、第2試薬層、光反射層、第1試薬層をこの順に有するもの。
以下の実施例により本発明をさらに詳細に説明する。しかしながら、本発明は実施例によって限定されるものではない。
(1)試料の調製
既に菌種が、Mycobacterium tuberculosis(Mtb)、Mycobacterium avium(Ma)、Mycobacterium intracellulare(Mi)と同定されている培養菌サンプルを準備し、集菌洗浄後、R.Boomらの方法(Journal of Clinical Microbiology 28巻3号495頁(1990))に従って、ゲノムDNAを抽出した。
上記(1)で調製したDNA溶液を用いて、以下の条件でPCR増幅反応を行った。

t2(upper:Mtb検出用):5'-ataccggataggaccacg-3' (配列番号10)
a2 (upper: Ma検出用) :5'-ataccggataggacctca-3' (配列番号11)
i2(upper:Mi検出用) : 5'-aataccggataggaccttt-3'(配列番号12)
M2 (lower:3菌種共通) :5'-tgcttcttctccacctacc-3'(配列番号13)
各菌検出用プライマーと検体を以下の表1に記載の組み合わせでPCR反応を行った。
ゼラチン下塗層が設けられている厚さ180μmの無色透明ポリエチレンテレフタレ−ト(PET)平滑フイルムシ−ト(支持体)上に下記の表3に記載の組成(a)の水溶液を、以下の被覆率となるように塗布し、乾燥して試薬層を設けた。
前記(2)におけるPCR増幅反応後の溶液をそのまま、上記(3)で製作したピロ燐酸定量用乾式分析素子上に各々20μL点着し、ピロ燐酸定量用乾式分析素子を37℃にて5分間インキュベ−ション後、波長650nmにて支持体側から測定して得られた5分後の反射光学濃度(ODR)、及び、あらかじめ作成してあった反射光学濃度をピロリン酸濃度に読み替える検量線を用いてピロリン酸濃度(mM)に読み替えた数値を表6に示した。
(1)試料の調製
既に菌種が、M.tubeclosis(Mtb)、M.avium(Ma)、M.intaracellulare(Mi)、M.kansasii(Mk)と同定されている培養菌サンプルを準備し、集菌洗浄後、R.Boomらの方法(Journal of Clinical Microbiology 28巻3号495頁(1990))に従って、ゲノムDNAを抽出した。
上記(1)で調製したDNA溶液を用いて、以下の条件でPCR増幅反応を行った。

M5 (lower) :5'- cgtcctgtgcatgtcaaa -3'(配列番号28)

- マイコバクテリウム・ツベルクローシス菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、かつ配列番号1に記載の塩基配列に含まれる連続する少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・ツベルクローシス菌の同定方法。
- マイコバクテリウム・ツベルクローシス菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、配列番号2に記載の塩基配列に含まれる連続する少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・ツベルクローシス菌の同定方法。
- 配列番号3に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号3における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・ツベルクローシス菌の同定方法。
- マイコバクテリウム・アビウム菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、配列番号4に記載の塩基配列に含まれる少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・アビウム菌の同定方法。
- マイコバクテリウム・アビウム菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、配列番号5に記載の塩基配列に含まれる少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・アビウム菌の同定方法。
- 配列番号6に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号6における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・アビウム菌の同定方法。
- マイコバクテリウム・イントラセルラー菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、配列番号7に記載の塩基配列に含まれる少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・イントラセルラー菌の同定方法。
- マイコバクテリウム・イントラセルラー菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、配列番号8に記載の塩基配列に含まれる少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・イントラセルラー菌の同定方法。
- 配列番号9に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号9における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・イントラセルラー菌の同定方法。
- マイコバクテリウム・カンサシイ菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、かつ配列番号23に記載の塩基配列に含まれる連続する少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・カンサシイ菌の同定方法。
- マイコバクテリウム・カンサシイ菌の16SrRNA遺伝子配列中の可変領域に対応する塩基配列から成り、配列番号24に記載の塩基配列に含まれる連続する少なくとも3塩基を3’末端に有する核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・カンサシイ菌の同定方法。
- 配列番号25に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号25における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる核酸増幅用プライマーを用いて核酸増幅反応を行うことを含む、マイコバクテリウム・カンサシイ菌の同定方法。
- 核酸増幅反応時の副反応生成物を検出することを特徴とする、請求項1から12の何れかに記載の方法。
- 核酸増幅反応時の副反応生成物がピロリン酸であることを特徴とする請求項13に記載の方法。
- 乾式分析素子を用いてピロリン酸の検出を行う、請求項14に記載の方法。
- 配列番号3に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号3における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる、マイコバクテリウム・ツベルクローシス菌を同定するために使用する核酸増幅用プライマー。
- 配列番号6に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号6における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる、マイコバクテリウム・アビウム菌を同定するために使用する核酸増幅用プライマー。
- 配列番号9に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号9における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる、マイコバクテリウム・イントラセルラー菌を同定するために使用する核酸増幅用プライマー。
- 配列番号25に記載の塩基配列に含まれる連続する少なくとも15塩基以上の塩基配列であって、配列番号25における26番目の塩基であるG以降の3’側の塩基を少なくとも3塩基以上含む塩基配列からなる、マイコバクテリウム・カンサシイ菌を同定するために使用する核酸増幅用プライマー。
- 請求項16から19の何れかに記載の少なくとも一種の核酸増幅用プライマー、少なくとも一種のデオキシヌクレオシド3リン酸、少なくとも一種のポリメラーゼ、及び乾式分析素子を含む、抗酸菌属検出用キット。
