[Test 1] Interleukin-1α (IL-1α) production inhibition test (sample solution)

(Cells and culture conditions)
Normal human neonatal epidermal keratinocytes (Kurabo NHEK (F) KK-4009) were used as the cells. EpiLife-KG2 (Kurabo M-EPI-2150) was used as a culture solution, and the cells were cultured under conditions of 5% CO ₂ and 37 ° C.
(Method)
Epidermal keratinocytes were seeded at a concentration of 3 × 10 ⁴ cells / mL in a 24-well plate at 1 mL / well, and cultured until confluent. Thereafter, the medium was replaced with a basal medium in which Hydrocortisone and BPE were removed from EpiLife-KG2, and the sample solution was added at a concentration of 50 μL / mL. After 24 hours, the cells were washed with phenol red-free MEM, and then irradiated with 50 mJ of UVB. Thereafter, 0.5 mL / well of basal medium was added and further cultured for 24 hours, and then the medium was collected, and the amount of IL-1α in the culture supernatant was quantified using Human IL-1α Immunoassay KIT (GT 8035). In order to observe the effect on cell proliferation, the number of cells was counted using a Cell Counting Kit (Dojindo 343-07623). As a control, a system without UV irradiation and no sample was prepared and compared. IL-1α production was calculated separately by preparing a calibration curve using the IL-1α standard. (Test results)
As shown in FIG. 1, the button extract and Kawaramugi extract, which are the active ingredients of the present invention, exhibited a strong IL-1α production inhibitory effect that exceeded the anti-inflammatory agent Dexamethasone. That is, the product of the present invention is expected to prevent inflammation and activation of melanocytes associated with IL-1α and to have an anti-inflammatory action and a whitening action. In addition, although the influence which it has on the cell proliferation of each sample was observed, the sample which affects cell proliferation was not found.

[Test 2] Safety test (1) Primary skin irritation test Each plant extract obtained in Extraction Examples 1 to 7 is prepared with an aqueous solution so that the solid content concentration is about 1.0%, and this is shaved on the back. 0.03 mL per 2 × 2 cm ² skin of Hartley guinea pigs (female, 3 animals per group, body weight around 340 g) was applied to each animal. (2) Cumulative skin irritation test Prepare an aqueous solution of each plant extract obtained in Extraction Examples 1 to 7 so that the solid concentration is about 2.0%, and pluck the flank (2 × 2 cm ² ). 0.03 mL was applied to each animal once a day, 5 times a week, per 2 × 2 cm ² of skin of a Hartley guinea pig (female, 3 mice per group, weight around 320 g). Judgment was made on the day of each application and the day after the last application by the scoring method of primary irritation using erythema and edema as indices. As a result, in all animals, no erythema and edema were observed for 2 weeks after application, and the animals were judged negative.

(3) Single-dose toxicity test Prepare an aqueous solution of each plant extract obtained in Extraction Examples 1 to 7 so that the solid content concentration is about 1.0%. A ddy mouse (male and female, 1 group, 5 animals, 5 weeks old) fasted for 16 hours was orally administered with a sonde, and the onset, degree, etc. of toxic symptoms were observed over time. As a result, no abnormalities were observed in all mice for 14 days, and no abnormalities were found in the anatomical results. Therefore, LD50 was 2,000 mg / kg or more, and it was determined to be a safe substance.
